Diagnostic Device Model for Insecticide susceptibilities of Beet Armyworm, Spodoptera exigua(Hubner) (파밤나방(Spodoptera exigua (Hiibner)) 살충제 감수성 진단장치모형)

  • Simple diagnostic kits for monitoring insecticide susceptibility of beet armyworm, Spodoptera exigua (Hiibner) were developed and applied to the field populations. The operation of the kits was based on the correlations between enzyme activities of esterase (EST) and acetylcholinesterase (AChE) and the insecticide susceptibilities. Four different kinds of diagnostic kits (ED, EM, AD, and AM) were designed and classified by diagnostic enzymes (E for esterases and A for acetylcholinesterase) and inhibitors (D for dichlorvos and M for monocrotophos). Diagnostic inhibitor concentrations were 1 mM for ED, 10 mM for EM, 100 mM for AD, and 100 mM for AM. Resistant larvae which were not inhibited by the diagnostic amounts of insecticides developed positive staining (red color), but susceptible~ s howed negative (no color). An insect was used for both EST and AChE diagnostic kits, but different in their samples: hemolymph for EST and the head for AChE. These four diagnostic kits were applied to 1 1 different populations which showed variations of insecticide susceptibilities. Four kits were different in the capability discriminating the insecticide susceptibilites according to insecticides: ED to bifenthrin, AD to methomyl, and ED and AM to chlorpyrifos-methyl. These diagnostic devices can be used for insecticide-resistance management program for this insect pest. It also provide a technical guide to insect pest management for farmers, directors, and researchers.

Zanthoxylum Piperitum Attenuates the Allergic Inflammation in vivo and in vitro

  • Natural products have received revived interest via traditional remedies or alternative medicine used for the treatment of various diseases. Zanthoxylum piperitum (ZP) has been utilized in traditional medicine for various medicinal purposes. The present study was conducted to evaluate whether ZP modulates allergic inflammation both in vivo and in vitro. We examined the pharmacological effects of ZP on 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) symptoms in mice. Additionally, in order to clarify the anti-inflammatory mechanisms of ZP, we elucidate the effect of ZP on the expression levels of inflammatory cytokines and nuclear factor-κB (NF-κB) in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). The results demonstrated that ZP attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, ZP suppressed the expression of inflammatory cytokines and activation of NF-κB in AD-like skin lesions and stimulated HMC-1. These results provide experimental evidence that ZP may be useful candidate for treating allergic inflammation including AD.

Is the Peak-Affect Important in Fast Processing of Visual Images in Printed Ads?: A Comparative Study on the Affect Integration Theories

  • This study investigates how affects elicited by visual images in print ads are integrated to form a liking for the ads. Assuming a sequential rather than simultaneous processing of still-cut images, we adopt the 'think-aloud' method to capture consumers' spontaneous responses to visual images. We hypothesize that not only would consumers show mixed affects toward a still-cut visual image but that they would also integrate their serial affects heuristically rather than simply averaging the affects as suggested by the compensatory hypothesis. By comparing the effects of two contradictory affect integration hypotheses (i.e., peak-affect and mood-maintenance) with compensatory integration, using a single regression model, we found that peak-negative along with mood maintenance integration of serial affects for a print ad works best in the formation of ad liking. The results also support our initial premise that people can have mixed valence even toward a still-cut ad.

  • In order to introduce to engineers the suitable calculation techniques of TBM advance rate (ad.) and ultimately promote to understand the designing process, this study was carried out. We analyzed the 17 bored data of TBM which applied to the roadway and water supply tunnels in Korea. From this analysis, it was able to how that the average utilization is 30.83% md the correlation equation of Ad and TBM´s diameter (D) is Ad(m/month) = 506.05ㆍ $e^{-0.1162}$$\times$D than the correlation coefficient ($R^2$) is 0.76. In the object of the W tunnel of Seoul-Busan highspeed railway, the Ad of TBM 5.0mø was analyzed by the variety of empirical models and upper correlation equation. Average Ad of the empirical models was calculated to be larger than one of the upper equations. But considering only the results of 3.0~5.0mø TBM in the 17 bored data, the average Ad by the models belongs to the similar range of bored data. Therefore, when the reliability and representative of parameters are decreased, a reliability test should be carried out through the comparison a variety of empirical models with the upper correlation equation.

  • For the safety evaluation of adenovirus-mediated gene transfer, we investigated differential gene expressions after transfecting adenoviral vector containing p16 tumor suppressor gene (Ad5CMV-p16) into human non-small cell lung cancer cells. In the previous study, we showed adenovirus-mediated $p16^{INK4a}$ gene transfer resulted in significant inhibition of cancer cell growth. We investigated gene expression changes after transfecting Ad5CMV-p16, Ad5CMV (null type, a mock vector) into A549 cells by using cDNA chip and oligonucleotide microarray chip (1200 genes) which carries genes related with signal transduction pathways, cell cycle regulations, oncogenes and tumor suppressor genes. We found that $p16^{INK4a}$ gene transfer down regulated 5 genes (cdc2, cyclin D3, cyclin B, cyclin E, cdk2) among 26 genes involved in cell cycle regulations. Compared with serum-free medium treated cells, Ad5CMV-p16 changed 27 gene expressions, two fold or more on oligonucleotide chip. In addition, Ad5CMV-p16 did not seem to increase the tumorigenicity-related gene expression in A549 cells. Further studies will be needed to investigate the effect of Ad5CMV-p16 on normal human cells and tissues for safety evaluation.

  • Various anatomical MRI imaging biomarkers for Alzheimer's Disease (AD) identification have been recognized so far. Cortical and subcortical volume, hippocampal, amygdala volume, and genetics patterns have been utilized successfully to diagnose AD patients from healthy. These fundamental sMRI bio-measures have been utilized frequently and independently. The entire possibility of anatomical MRI imaging measures for AD diagnosis might thus still to analyze fully. Thus, in this paper, we merge different structural MRI imaging biomarkers to intensify diagnostic classification and analysis of Alzheimer's. For 54 clinically pronounce Alzheimer's patients, 58 cognitively healthy controls, and 99 Mild Cognitive Impairment (MCI); we calculated 1. Cortical and subcortical features, 2. The hippocampal subfield, amygdala nuclei volume using Freesurfer (6.0.0) and 3. Genetics (APoE ε4) biomarkers were obtained from the ADNI database. These three measures were first applied separately and then combined to predict the AD. After feature combination, we utilize the sequential feature selection [SFS (wrapper)] method to select the top-ranked features vectors and feed them into the Multi-Kernel SVM for classification. This diagnostic classification algorithm yields 94.33% of accuracy, 95.40% of sensitivity, 96.50% of specificity with 94.30% of AUC for AD/HC; for AD/MCI propose method obtained 85.58% of accuracy, 95.73% of sensitivity, and 87.30% of specificity along with 91.48% of AUC. Similarly, for HC/MCI, we obtained 89.77% of accuracy, 96.15% of sensitivity, and 87.35% of specificity with 92.55% of AUC. We also presented the performance comparison of the proposed method with KNN classifiers.

Anti-atopic Effects of Castanea crenata Inner Shell Extracts Fermented by Lactobacillus bifermentans (Lactobacillus bifermentans로 발효한 율피의 항아토피 효과)

  • Atopic dermatitis (AD) is a common skin disease characterized by chronic and relapsing inflammatory dermatitis with immunological disturbances. In spite of the continuous increase in the incidence of AD, it is regrettable that till date there is no effective treatment to treat the same. Therefore, the present study was designed to examine the possible anti-atopic effects of Castanea crenata inner shell extracts fermented by Lactobacillus bifermentans (FCS) in 2,4-dinitrochlorobenzene (DNCB) induced AD in NC/Nga mice. Based on the results of HPLC analysis, we found that FCS contains anti-inflammatory factors such as gallic acid (10.18 mg/g) and ellagic acid (2.14 mg/g). The groups that we have used in this study included 0.1%, 1%, 5% fermented Castanea crenata inner shell extracts (FCS 0.1, FCS 1, FCS 5), 1,3-butylene glycol treated control (AD), and normal mice. After topical FCS treatment, we observed that the clinical severity score for AD was lower in both the FCS 1 and FCS 5 groups than the AD group. We also proved beyond doubt that there was improvement of melanin, erythema and skin moisture indices in the FCS 5 group. Spleen index and gene expression levels of pro-inflammatory cytokines such as IL-$1{\beta}$ and TNF-${\alpha}$ were significantly decreased in the FCS 5 group compared to the AD group (P<0.05). Further, we also found that the level of serum immunoglobulin E (IgE) in the FCS-treated group was decreased in a concentration-dependent manner. The results of our study suggest that FCS can be effectively used as a cosmeceutical ingredient for both the prevention and improvement of AD.

Influence of Age at Weaning on Caecal Content Characteristics and Post-weaning Performance and Health of Rabbits

  • This study chiefly aimed to ascertain the effect of age at weaning on caecal content characteristics, post-weaning performance and health of rabbits. A secondary aim was to study the effect of dietary restriction on performance and health of rabbits. After delivery, 30 litters of 8 pups each, were housed in separate cages. Fifteen litters were weaned 24 days from birth (group W 24) while the other 15 were weaned at 28 days (group W28). After weaning, 53 rabbits of group W24 and 56 of group W28 were fed ad libitum (group LIB), while 53 rabbits of group W24 and 57 of group W28 were restricted (group RES). All the rabbits were kept in individual cages. Fourteen other rabbits of groups W24 (7 fed ad libitum and 7 restricted) and 7 of group W28 were sacrificed to obtain data regarding caecum weight and caecal content. Age at weaning affected neither the state of health nor the final live weight. In comparison with those of group W28, the rabbits weaned at 24 days had a lower daily gain until 35 days of age but gain was higher subsequently. From the 28th day, however, the feed intake and the feed conversion ratio of the group W24 rabbits were always better than those of W28. The restricted diet until 42 days resulted in a lower weight both at 42 days (1,078 vs. 1,164 g) and at 70 days (2,192 vs. 2,294 g). Indeed, from the 42nd day, despite receiving feed ad libitum the rabbits of the restricted group continued to ingest less feed (120.8 vs. 127.2 g) than those of the ad libitum group, and had a slightly lower daily gain (39.8 vs. 40.3 g). The measurements carried out on the caecum highlighted significant differences among the groups in caecal content (33.1 vs. 31.7 vs. 25.3 g respectively for groups W24LIB, W28 and W24RES), for total volatile fatty acids (53.2 vs. 50.6 vs. 44.5 mmol/l respectively for groups W24LIB, W28and W24RES) for ammonia (7.0 vs. 7.2vs 8.8 mmol/l respectively for groups W24LIB, W28 and W24RES), for propionate (5.7 vs. 6.3 vs. 8.1% respectively for groups W24LIB, W28 and W24RES) and for propionate/butyrate (0.33 vs. 0.36 vs. 0.49 respectively for groups W24LIB, W28 and W24RES).

  • Objectives : The Present study was carried out to investigate the preventive effect of Anti-diabetic prescription 1(AD-1) on streptozotocin(STZ)-induced diabetic nephropathy. Methods : AD-1 consists of eleven herbs that have an effect on diabetes mellitus. AD-1 was given to rats with the combination of oral administration and herbal-acupuncture stimulation. The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with AD-1 treatment. Experimental diabetes was induced by the injection of STZ(60mg/kg) to e rat via the peritoneum. The effect of AD-1 on STZ-induced diabetic nephropathy was observed by measuring the serum level of creatinine and BUN. Urine secretion of albumin for 24 hours and urine level of glucose measures too. Anti-oxidative stress of AD-1 administration in living body was estimated by measuring lipid peroxide and GSH content in cortex of kidneys. Results : STZ induced increase of serum creatinine, BUN and albumin secretion were lowered by AD-1 treatment. Conclusion : The AD-1 treatment showed protective effect on rat diabetic nephropathy model, and action mechanism of the effect was thought to be concerned with anti-oxidative stress.

Prediction of hub genes of Alzheimer's disease using a protein interaction network and functional enrichment analysis

  • Alzheimer's disease (AD) is a chronic, progressive brain disorder that slowly destroys affected individuals' memory and reasoning faculties, and consequently, their ability to perform the simplest tasks. This study investigated the hub genes of AD. Proteins interact with other proteins and non-protein molecules, and these interactions play an important role in understanding protein function. Computational methods are useful for understanding biological problems, in particular, network analyses of protein-protein interactions. Through a protein network analysis, we identified the following top 10 hub genes associated with AD: PTGER3, C3AR1, NPY, ADCY2, CXCL12, CCR5, MTNR1A, CNR2, GRM2, and CXCL8. Through gene enrichment, it was identified that most gene functions could be classified as integral to the plasma membrane, G-protein coupled receptor activity, and cell communication under gene ontology, as well as involvement in signal transduction pathways. Based on the convergent functional genomics ranking, the prioritized genes were NPY, CXCL12, CCR5, and CNR2.
