Sprague Dawley 흰쥐에서 테스토스테론에 의하여 유발된 전립선 비대증에 미치는 산수유 추출물의 영향 (Effects of Corni Fructus on Testosterone-induced Benign Prostatic Hyperplasia in Sprague Dawley Rats)

  • 노년기 비뇨기 계통에 가장 흔한 증상의 하나인 전립선 비대증은 요도를 둘러싼 전립선의 주위의 평활근과 상피세포의 과다 증식에 의한 것이다. 산수유는 강력한 항산화 효과로 인하여 다양한 질병의 예방 및 치료에 효과적이라고 보고되었지만 전립선 비대증에 대한 효능은 아직 알려지지 않았다. 본 연구에서는 산수유 열수 추출물이 testosterone에 의하여 유도되는 전립선 비대증에 미치는 영향을 조사하였다. 실험동물 내재성 testosterone의 영향을 배제하기 위해 거세를 하였으며, 전립선 비대증을 유도하기 위해, testosterone propionate (TP)를 피하 주사하였다. 산수유 추출물은 TP 주입과 함께 매일 경구 투여하였고, $5{\alpha}$-reductase type 2의 선택적 억제제인 finasteride를 양성 대조군으로 사용하였다. 본 연구의 결과에 의하면, 산수유 추출물 투여군에서는 finasteride 처리군에서와 마찬가지로 혈청 내 dihydrotestosterone 농도가 억제되었으며 전립선 무게 증가와 조직병리학적 변화가 유의하게 감소되었다. 산수유 추출물은 또한 전립선 조직 및 혈청에서 각각 TP에 의해 증가된 $5{\alpha}$-reductase type 2의 발현 및 농도를 유의적으로 억제하였다. 아울러 산수유 추출물은 TP에 의하여 유도된 AR, AR의 co-activator 및 세포증식 마커 단백질들의 발현 증가뿐 만 아니라 prostate-specific antigen의 수치와 발현도 감소시켰다. 결론적으로 산수유 추출물은 전립선 비대억제를 위한 식의약 소재로서의 개발 가능성이 매우 높음을 의미한다.

배양 간세포내에서의 콜레스테롤 합성에 대한 담즙산의 저해효과 (Inhibitory Effects of Bile Acids on the Cholesterol Biosynthesis in Cultured Hepatocytes)

  • 간세포내 두가지 microsome효소인 HMG-CoA reductase와 cholesterol-$7{\alpha}$-hydroxylase는 콜레스테롤 합성과 담즙산으로의 분해의 조절효소임은 이미 알려진 사실이다. 본 실험에서는 콜레스테롤의 대사산물인 4종류 담즙산들이 간세포내의 콜레스테롤 합성 및 HMG-CoA reductase활성에 미치는 효과를 조사하였다. 간세포의 배양에서 간세포로의 담즙산 흡수는 배지내 농도(1mM~10mM)와 배양시간(1/2~3hr)의 차이에 따라 비례적으로 증가하였다. 콜레스테롤 합성저해에 대한 담즙산의 효과는 배지내 담즙산의 농도 및 배양시간에 따라 크게 감소하였다. 분리시킨 microsome내 HMG-CoA reductase의 활성에 대한 담즙산의 저해효과는 insulin 투여에 의하여 효소활성을 촉진시킨 경우에서도 뚜렷하였으며 콜레스테롤 합성 역시 저하되었다. 분리시킨 간세포막 내 $Na^+$,$K^+$-ATPase의 활성은 1.8~2.5mM정도의 배지내 담즙산 농도까지 증가함을 보였으나 cholic acid 흡수는 상기 효소의 활성에 거의 영향을 주지 않는 것으로 나타났다. 이러한 이유는 아직 불분명하나 1차 대사산물인 cholic acid의 흡수는 단순확산에 의한 것으로 사료되며 이에 대한 더 많은 연구가 요구된다.

Albizzia julibrissin Suppresses Testosterone-induced Benign Prostatic Hyperplasia by Regulating 5α-Reductase Type 2 - Androgen Receptor Pathway

  • Albizzia julibrissin (AJ) is an herbal medicine that shows low toxicity, promotes promoting blood circulation and mitigates the inflammation and has mild side effects. Benign prostate hyperplasia (BPH) is one of the most common diseases that occurs in older males and often results in lower urinary tract symptoms. This study was conducted to evaluate the protective effect of AJ against BPH using LNCaP cells and Sprague Dawley rats treated with testosterone. Treatment with AJ extract reduced the expression of androgen receptor (AR) and prostate-specific antigen (PSA) in vitro. In vivo, rats were divided into 6 groups: 1 (Normal Control); 2 (Testosterone propionate (TP) alone); 3 (TP + finasteride); 4 (TP + AJ 10 mg/kg); 5 (TP + AJ 50 mg/kg); 6 (TP + AJ 300 mg/kg). The groups treated with AJ showed reduced the relative prostate weights and BPH-related proteins were altered, with decreased AR, PSA and proliferating cell nuclear antigen (PCNA) observed by western blot. Histopathological analysis revealed the therapeutic effect of AJ, with a decreased thickness of epithelial cells and reduced level of PCNA and $5{\alpha}$-reductase type 2. These results suggest that AJ extract could ameliorate testosterone-induced benign prostatic hyperplasia.

Microalgae, Tetraselmis tetrathele has Alopecia Prevention and Scalp Improvement

  • The microalga, Tetrathelmis tetrathele, is used in the development of products for the aquaculture, food, and nutraceutical industries. In the present study, we investigated whether the T. tetrathele ethanolic extract (TTE), which has anti-inflammatory properties, can confer protection against alopecia and improve scalp health, influence the proliferation of human keratinocytes, HaCaT cells, and human hair follicle dermal papilla cells (HFDPC), or inhibit 5α-reductase activity. We found that TTE inhibited the production of the inflammatory mediator, nitric oxide (NO), and prostaglandin E2 (PGE2) without cytotoxicity in LPS-stimulated RAW 264.7 cells. In addition, TTE encouraged the proliferation of HaCaT cells and HFDPC. Our results showed that TTE had anti-inflammatory activities, proliferated HaCaT cells and HFDPC, and inhibited 5α-reductase activity. Therefore, we suggest that T. tetrathele could be a potent therapeutic agent for alopecia prevention and scalp improvement.

불포화 지방산과 마그네슘, 칼슘과의 상호작용이 3-Hydroxy-3-Methylglutaryl Co-A reductase 활성에 미치는 영향 (Interaction of dietary Mg(II), Ca(II) and polyunsaturated fatty acids on 3-Hydroxy-3-Methylalutaryl Co-A reductase activity)

  • Interaction of dietary Magnesium, Calcium and Polyunsaturated fatty acid(vegetable oils)on 3-Hydroxy-3-Methylglutaryl Co-A reductase activity was studied for a period of 30 days using isocalories and isonitrogenous as a basal diet . The subject rabbits were divided into 18 feeding groups. The results are summarized as follows: 1. The ratio of ${\alpha}_1-lipoprotein$ par lipalbumin is 0.34 for control group, 0.38 the highest group fed 0.1 Mg(II) 10ml plus perilla oil and basal diet, the lowest 0.25 group fed 0.1M Mg(II) 5ml plus sesame oil and basal diet. 2. The ratio ratio of ${\alpha}_2-lipoprotein$ per lipalbumin is 0.64 for control group. 0.95 as the highest for the group fed 0.1M Ca(II) 15ml plus sesame oil and basal diet, 0.1M Ca(II) 5ml plus perilla oil and basal diet. 3. The ratio of ${\beta}-lipoprotein$ per lipalbumin is 0.71 for control group, the highest 0.81 for the groups fed 0.1M Mg(II) 10ml plus sesame oil and basal diet, the lowest 0.37 for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 4. In serum triglyceride, control group was 129.5mg%, the highest 155.4mg% for the group fed 0.1M Ca(II) 5ml plus sesame oil and basal diet, the lowest 85.7mg% for the group fed 0.1M Mg(II) 10ml plus soybean oil and basal diet. 5. In serum cholesterol, control group was 96.7mg%, the highest 152.5mg% for the group fed 0.1M Ca(II) 10ml plus sesame oil and basal diet, the lowest 80.5mg% for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 6. In case of HMG-CoA reductase activity, control group was 0.95, the highest 0.98 for the group fed 0.1M Ca(II) plus soybean oil and basal diet. 7. Interaction between metal(II) ions and polyunsaturated fatty acid(vegetable oil) are soybean oil>sesame oil>perilla oil, for Mg(II). soybean oil>perilla oil>sesame oil, for Ca(II). Therefore, it is invetigated that the interaction between metal ion and polyunsaturated fatty acid is the higher, the cholesterol level is the lower, and HMG-CoA reductase activity is increased.

토마토 추출액 복합체가 전립선 암 세포와 전립선 비대증에 미치는 영향 (The Effect of the Compound of Tomato Extract to the Prostatic Cancer Cell and the Prostate of the Rat Model of Benign Prostatic Hyperplasia)

  • Benign prostatic hyperplasia (BPH) is one of the common disease in elderly men. Recently old-age population is increased and we are growing more and more interested in clinical importance of BPH. In this study, the effect of PLX, which was the mixture of tomato extract (including 2% of lycopene) and chitooligosaccharide, on prostatic cancer cell and testosterone-induced BPH in adult rats of the Sprague Dawley strain was determined. The cell viability was evaluated by MTT method using L929 and LNCaP cell line, pretreated with various concentrations of PLX. The expression of prostatic specific antigen (PSA) and 5${\alpha$}$-reductase genes were evaluated by realtime PCR using LNCaP cell line and compared various concentrations of PLX with 50 ${\mu}$M of finasteride. An experimental prostatic hyperplasia was induced in male Sprague Dawley rats by giving testosterone for 8 weeks. After 2 weeks from start of giving testosterone, PLX and finasteride were administered orally once a day. The results were analyzed with prostate weight per body weight at 8 weeks. Cell viability of L929 cell line decreased specifically at the concentration of 2000 ${\mu}$g/mf of PLX. The cytotoxicity of PLX to the LNCaP cell line was shown at above 500 ${\mu}$g/ml of PLX. The inhibitory effect of PLX to the expression of PSA and 5${\alpha$}$-reductase genes in LNCaP cell line increased with the concentration of PLX. In vivo study, the results of PLX and finasteride administered group were 3.75${\pm}$0.60 and 3.49${\pm}$0.49 prostate weight ${\times}10^3$/body weight, which were lower than the result of BPH induced group (4.74${\pm}$0.58). These results suggested that PLX may be an effective material in BPH by having the role of the 5a-reductase inhibitor.

자작나무 추출물에서 보이는 모유두(HDP) 세포 성장 촉진 효과와 작용 메커니즘 연구 (Studies on the effect of Betula platyphylla extract on human dermal papilla cell proliferation and its mechanism of action)

  • 자작나무(Betula platyphylla) 수피 추출물에는 생리활성을 보일 수 있는 다수의 terpenoids 물질이 포함되어 있고 염증반응을 완화시키는 치료제로 사용되어 왔다. 본 연구는 탈모예방 효과를 보일 수 있는 천연 물질을 확보하기 위해 자작나무를 여러 용매에서 추출 및 분리 정제하여 몇몇 분획을 획득하였다. 이를 사용하여 사람 모낭 모유두세포(HDP)에 대한 세포 독성 및 성장촉진 효과 그리고 β-catenin, VEGF, IGF1, Cyclin D의 발현변화와 5-α-reductase activity에 미치는 영향을 평가하였다. Ethyl acetate를 이용하여 분리한 4개의 분획 중 H3-2 분획은 알려진 유효물질인 ETB, LPO와 동등수준의 모낭 모유두세포(HDP)에 대한 성장 촉진 효과를 보였고 성장에 관여하는 주요인자들의 발현을 높이는 효과를 보였다. 또한, 5-α-reductase activity에 ETB, LPO가 큰 영향을 보이지 못한 반면 최종분획물인 H3은 5-α-reductase activity 감소활성이 있음을 확인할 수 있었다.

Corni Fructus attenuates testosterone-induced benign prostatic hyperplasia by suppressing 5α-reductase and androgen receptor expression in rats

  • BACKGROUND/OBJECTIVES: Benign prostatic hypertrophy (BPH) is a major cause of abnormal overgrowth of the prostate mainly in the elderly. Corni Fructus has been reported to be effective in the prevention and treatment of various diseases because of its strong antioxidant effect, but its efficacy against BPH is not yet known. This study was designed to evaluate the therapeutic efficacy of Corni Fructus water extract (CF) in testosterone-induced BPH rats. MATERIALS/METHODS: To induce BPH, rats were intraperitoneal injected with testosterone propionate (TP). Rats in the treatment group were orally administered with CF with TP injection, and finasteride, which is a selective inhibitor of $5{\alpha}$-reductase type 2, was used as a positive control. RESULTS: Our results showed that the increased prostate weight and histopathological changes in BPH model rats were suppressed by CF treatment. CF, similar to the finasteride-treated group, decreased the levels of testosterone and dihydrotestosterone by TP treatment in the serum, and it also reduced $5{\alpha}$-reductase expression and concentration in prostate tissue and serum, respectively. In addition, CF significantly blocked the expression of the androgen receptor (AR), AR co-activators, and proliferating cell nuclear antigen in BPH rats, and this blocking was associated with a decrease in prostate-specific antigen levels in serum and prostate tissue. CONCLUSIONS: These results suggest that CF may weaken the BPH status through the inactivation of at least $5{\alpha}$-reductase and AR activity and may be useful for the clinical treatment of BPH.

SRD5A2 Gene Polymorphisms and the Risk of Benign Prostatic Hyperplasia but not Prostate Cancer

  • Background: Testosterone, a primary androgen in males, is converted into its most active form, dihydrotestosterone (DHT), by $5{\alpha}$-reductase type 2 (encoded by the SRD5A2 gene) in the prostate. DHT is necessary for prostatic growth and has five times higher binding affinity than testosterone for androgen receptors. We hypothesized that polymorphic variations in the SRD5A2 gene may affect the risk of benign prostatic hyperplasia and prostate cancer. Materials and Methods: We analyzed SRD5A2 gene polymorphisms in 217 BPH patients, 192 PCa cases, and 171 controls. Genotyping was undertaken using direct DNA sequencing. Genotype data were compared between cases and controls using a Chi square statistical tool. Results: We found that the A49T locus was monomorphic with 'AA' genotype in all subjects. At V89L locus, the presence of 'VV' showed a marginally significant correlation with increased BPH risk (p=0.047). At the $(TA)_n$ locus, longer TA repeats were found to be protective against BPH (p=0.003). However, neither of these polymoprhisms correlated with the risk of PCa. Conclusions: We conclude that A49T is monomorphic in the study population, VV marginally correlates with BPH risk, and longer $(TA)_n$ repeats are protective against BPH. None of these polymorphisms affect the risk of PCa.

Promoting Effect of a Mixture of 8 Herbal Extracts (SPELA 707) on Hair Growth

  • In Korean folk medicine, several herbs, Glycyrrhizae Radix, Persicae Semen, Salviae Radix, Angelicae Gigantis Radix, Zanthoxyli Fructus, Ginseng Radix Alba, Cnidii Rhizoma, and Carthami Flos, are known to enhance blood circulation and have wound healing or anti-inflammatory effects. These pharmacological actions prompted us to investigate whether these herbs might stimulate hair growth. Thus, using a mixture of their extracts called SPELA 707, we investigated their effects and found that SPELA 707 possessed significant hair cycle converting activity from the telogen phase to the anagen phase in C3H mice. Furthermore, we found that SPELA 707 enhanced the hair density in subjects with hair loss and also promoted the conversion of hair into the anagen phase in subjects with androgenetic alopecia. In addition, hair growth promotion effect of SPELA 707 occurred through inhibition of steroid $5{\alpha}$-reductase activity, which is known to block hair growth. Taken together, these results suggest that SPELA 707 has a potential to be used for the treatment of hair loss.
