  • A, B 두 요인의 영향을 받고 있다고 생각되는 rc개 측정치가 있고 이것이 다음과 같이 $r\timesc$ 이차분류표로 정리되었다고 하자. 여기에서 $$y_{ij} = \mu + \alpha_i + \beta_j + \epsilon_{ij}$$ 와 같은 가법모형을 생각한다. 그리고 $\epsilon_{ij}$는 잔여항으로써 평균이 0, 분산이 $\sigma^2$인 정규분포를 한다고 가정하는 것이 보통이다. 또 이것은 모수모형인 경우 $E(y_{ij}) = \mu + \alpha_i + \beta_i, v(y_{ij}) = \sigma^2$임을 의미하는 것으로 생각된다. 그러나 자료에 따라서는 위에서와 같은 가법적 모형을 적용한다는 것이 적당하지 못한 경우가 있다.

Antioxidant and Alpha-Glucosidase Inhibitory Activity of Strychnos nux-vomica Extracts (마전자 추출물의 항산화 및 Alpha-Glucosidase 저해 활성)

  • The physiological activities of Strychnos nux-vomica extracts were investigated through the total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), comet assay, and $\alpha$-glucosidase inhibitory activity. S. nux-vomica extracts were prepared with methanol, ethanol, and acetone. The methanol extract showed the highest phenolic content (71 mg/100 g gallic acid equivalents). Pretreatment with S. nux-vomica extracts resulted in significant reductions in oxidative DNA damage at all of the concentrations tested ($1{\sim}50\;{\mu}g/mL$). The $\alpha$-glucosidase inhibitory activity of a methanol extract was 12.8% at the concentration of 1 mg/mL. Therefore, these results indicate that S. nux-vomica might be a noble potential candidate exhibiting antioxidant and $\alpha$-glucosidase inhibitory activity.

Role of Tumor Necrosis Factor-${\alpha}$ Promoter Polymorphism and Insulin Resistance in the Development of Non-alcoholic Fatty Liver Disease in Obese Children

  • Purpose: Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) polymorphism has been suggested to play an important role in the pathogenesis of non-alcoholic fatty liver disease (NAFLD) in obese adults, and known to be a mediator of insulin resistance. In this study, we evaluated the role of TNF-${\alpha}$ promoter polymorphisms and insulin resistance in the development of NAFLD in obese children. Methods: A total of 111 obese children (M:F=74:37; mean age, $11.1{\pm}2.0$ yrs) were included. The children were divided into 3 groups: controls (group I, n=61), children with simple steatosis (group II, n=17), and children with non-alcoholic steatohepatitis (group III, n=33). Serum TNF-${\alpha}$ levels, homeostasis model assessment of insulin resistance (HOMA-IR), and TNF-${\alpha}$ -308 and -238 polymorphisms were evaluated. Results: There were no differences in TNF-${\alpha}$ polymorphism at the -308 or the -238 loci between group I and group II + III ($p$=0.134 and $p$=0.133). The medians of HOMA-IR were significantly different between group I and group II + III ($p$=0.001), with significant difference between group II and group III ($p$=0.007). No difference was observed in the HOMA-IR among the genotypes at the -308 locus ($p$=0.061) or the -238 locus ($p$=0.207) in obese children. Conclusion: TNF-${\alpha}$ promoter polymorphisms at the -308 and -238 loci were not significantly associated with the development of NAFLD in children; nevertheless, insulin resistance remains a likely essential factor in the pathogenesis of NAFLD in obese children, especially in the progression to NASH.

Anti-inflammatory Effects and Its Mechanisms of NANA (N-Acylneuraminic Acid) Isolated from Glycomacropeptide (유청단백질 Glycomacropeptide에서 분리한 NANA의 안전성 및 염증저하 메카니즘 구명 연구)

  • The focus of this study was to clarify the relation between the nitric oxide (NO) production and cytokine expression including tumor necrosis factor-${\alpha}$ (TNF) and interleukin-6 (IL-6), and also investigated the effect of G-NANA (N-acylneuraminic acid isolates from glycomacropeptide) or S-NANA (Synthetic N-acylneuraminic acid) on LPS stimuli from RAW264.7 cell. The NANA is the predominant sialic acid found in mammalian cells and G-NANA is isolation of GMP (GMP is a valuable bioactive peptide with a varying degree of glycosylation including sialic acid). The lipopolysaccharide (LPS) of Gram-negative bacteria induces the expression of cytokines and potent inducers of inflammatory cytokines such as TNF-${\alpha}$ and IL-6. In this experiment, upon stimulation with increasing concentrations of chitosan, the LPS-stimulated TNF-${\alpha}$ and IL-6 secretion was significantly recovered with in the incubation media of RAW264.7 cells. Consistently, RT-PCR with mRNA and immunoblot analysis with anti-cytokine antiserum including TNF-${\alpha}$ and IL-6 showed that the amount of TNF-${\alpha}$ and IL-6 secretion in the incubation media recovered with the concentration of chitosan. The LPS-stimulated NO secretion was significantly recovered with in the 6 and 12 h incubation media of RAW264.7 cells, too. The recovery effect of G-NANA on IL-6 and NO secretion may be induced via the stimulus of TNF-${\alpha}$ in RAW264.7 cell. These results once again suggest that G-NANA may have the anti-inflammatory effect via the stimulus of TNF-${\alpha}$ in the LPS-stimulated inflammation in RAW264.7 cells.

Two Polymorphs of Structures of $\alpha,\alpha$-Trehalose Octaacetate Monohydrate

  • Structures of two polymorphs of ${\alpha},{\alpha}$-trehalose octaacetate monohydrate, $C_{28}H_{38}O_{19}\;{\cdot}\;H_2O$, have been studied by X-ray diffraction method. ${\alpha},{\alpha}$-trehalose (${\alpha}$-D-glucopyranosyl ${\alpha}$-D-glucopyranoside) is a nonreducing disaccharide. The polymorph I belongs to the monoclinic $P2_1$, and has unit cell parameters of a=10.725(l), b=15.110(4), c=11.199(5) ${\AA}$, ${\beta}=108.16(2)^{\circ}$ and Z=2. The polymorph II is orthorhombic $P2_12_12_1$, with a=13.684(4), b=15.802(4), c=17.990(9) ${\AA}$ and Z=4. The final R and R$_w$ values for monoclinic polymorph I are 0.043 and 0.048 and for orthorhombic polymorph II are 0.116 and 0.118, respectively. Those R values of polymorph II are high because the large thermal motions of acetyl groups and the poor quality of the crystal. The molecular conformations in the two polymorphs are similar. Both D-glucopyranosyl rings have chair $^4C_1$ conformations and atoms of glycosidic chain ${\alpha}(1{\rightarrow}1)$ linkage are coplanar. The primary acetate groups of the pyranose residues assume both gauche-trans conformations. The molecules of two polymorphs have pseudo-C$_2$ symmetry at glycosidic O(1) atom. The bond lengths and angles are normal compared with those in other acetylated sugar compounds. The molecules in the monoclinic crystal are held by the hydrogen bonds with the water molecules and by van der Waals forces.

  • Artificial intelligence (AI), big data, and ubiquitous robotic companions -the three most notable technologies of the 4th Industrial Revolution-are receiving renewed attention each day. Technologies that can be experienced in daily life, such as autonomous navigation, real-time translators, and voice recognition services, are already being commercialized in the field of information technology. In the biosciences field in Korea, such technologies have become known to the local public with the introduction of the AI doctor Watson in large number of hospitals. Additionally, AlphaFold, a technology resembling the AI AlphaGo for the game Go, has surpassed the limit on protein folding predictions-the most challenging problems in the field of protein biology. This report discusses the significance of AI technology and big data on the bioscience field. The introduction of automated robots in this field is not just only for the purpose of convenience but a prerequisite for the real sense of AI and the consequent accumulation of basic scientific knowledge.

Diagnosis of human genetic mutations based on DNA microarray technology

  • In this presentation, we will discuss several recent achievements developed in my laboratory for microarray-based diagnosis of human genetic mutations including HNF-1 and BRCA1 mutations. To determine the presence of the genetic mutations in a human sample, we prepared allele-specific oligonucleotide chips from selected mutation sites and generated target probes using a tow-step method for Cy-3 DNA $samples^{1)}$ or in vitro transcription of promoter-tagged PCR products for Cy-3 RNA $samples^{2)}$. Hybridization of the target probes to the chips successfully identified all of the genotypes for the tested sites. For more reliable diagnosis, we also employed single base extension (SBE) reaction and zip-code microarray technique for our strategy. Particularly we developed an efficient PNA zip-code microarray for the detection of $HNF-1{\alpha}$ $mutations^{3)}$. Using multiplex SBE reactions and zip-code strategy, we were able to correctly diagnose several mutation sites in exon 2 of $HNF-1{\alpha}$ with a wild-type and mutant including a MODY3 patient. These works represent successful applications of DNA microarray technology for the diagnosis of human genetic mutations.

Inhomogeneity of Hot Rolling Texture in Cu/Nb Added Ultra Low Carbon Steels

  • The texture and microstructure in Cu/Nb added ultra low carbon steels through the different thickness layer were studied after hot rolling. It was found that the two ultra low carbon steels all show the inhomogeneity of hot rolling texture and the Cu-added ultra low carbon steel was far more inhomogeneous than Nb-added one. In the center layer, the strong ${\alpha}\;fibre,\;{\gamma}\;fibre$ textures and the shear textures including 001<110>, 111<112> were founded. Near the surface, the ${\alpha}\;fibre$ texture and the orientation texture caused by a typical plane-strain deformation condition of bcc metals were observed.

  • Application of the spectrophotometer to the analysis of 17 ${\alpha}-ethinyl$ estradiol and $17{\alpha}-ethinyl-19-nortestosterone$ acetate mixture in oral contraceptive has been accomplished. It is used Beckman Du Spectrophotometer as a apparatus. The petroleum ether extract of ethinyl estradiol is determined at $535\;m{\mu}$ and the chloroform extract of norethindrone acetate is determined at $380\;m{\mu}$ respectively. This analytical method is formed Lambert Beer's law. This method can be used to the analysis of ethinyl estradiol aid norethindrone acetate mixture in commercial dosage form of routine assay.

  • Two new diterpenoids, orientalin A (1), and B (2), have been isolated together with six known compounds, kirenol (3), $ent-16{\beta},17-dihydroxykauran-19-oic$ acid (4), $ent-16{\beta},17-dihydroxykauran-19-oic$ $acid-16{\beta},l7-acetonide$ (5), $3,7-dimethylquercetin$ (6), ${\beta}-sitosterol$ (7), and daucosterol (8) from the ethanol extract of Siegesbeckia orientalis (Compositae). Their chemical structures have been elucidated as $ent-15-acetoxy-2{\alpha},16,19-trihydroxypimar-8(14)-ene$ (1), $ent-16-acetoxy-2{\alpha},15,19-trihydroxypimar-8(14)-ene$ (2), respectively, on the basis of chemical and spectral evidence.
