넙치에서 분리된 phosphoinositide 3-kinase γ 유전자의 클로닝 및 특성 연구 (Cloning and Characterization of Phosphoinositide 3-Kinase γ cDNA from Flounder (Paralichthys olivaceus))

  • Phosphoinositide 3-kinase (PI3K)는 항산화 제어반응, 심근세포 성장, 및 세포 내 특수반응 뿐만 아니라 세포분화, 생장, 운동, 식균 및 내항작용, 세포 골격유지에 관여하는 등 세포 신호체계에서 핵심 역할을 하는 효소이다. PI3K는 세 그룹으로 나누어지며 type I PI3K는 leukocyte에서 우선적으로 발현되고 G-proteins의 ${\beta}{\gamma}$ subunits에 의해서 활성화 된다. 본 연구에서는 넙치(Paralichthys olivaceus)의 $PI3K{\gamma}$를 암호화하는 cDNA를 클로닝하였다. 넙치의 $PI3K{\gamma}$는 1,341 bp 염기로 구성되는 한 개의 ORF를 가지며 이 단백질은 447 아미노산으로 구성되어있다. $PI3K{\gamma}$는 zebrafish의 $PI3K{\gamma}$와 89.6%, mouse와는 84.7%, Norway rat와는 84%, human의 $PI3K{\gamma}$와는 74.9%가 아미노산 상동성을 나타내었다. $PI3K{\gamma}$유전자의 대장균에서 발현을 위하여 pET-44a(+)-PI3K 재조합 DNA를 구축하여 대장균에서 발현시킨 결과 49 kDa의 재조합 단백질이 과발현 됨을 확인 할 수 있었다. His-tag affinity chromatography를 이용하여 $PI3K{\gamma}$단백질을 순순 분리하였으며 wortmannin을 이용하여 $PI3K{\gamma}$의 활성을 분석하였다.

Effects of Oral Administration of Phellinus linteus on the Productions of the Th1- and Th2-type Cytokines in Mice

  • Background: The mushroom Phellinus linteus (PL) has been shown to have the anti-tumor and immunostimulatory effects. We hypothesized that the hot water extract of PL (WEPL) exerts its significant immunostimulatory effect by inducing production of the Th1-derived cytokine interferon-${\gamma}$ (IFN-${\gamma}$) by T lymphocytes. Methods: T lymphocytes were isolated from the mice fed with 200 mg/kg of WEPL once a day for 4 weeks, and then stimulated with the mitogen concanavaline A (Con A). IFN-${\gamma}$ gene and intracellular protein expressions were analyzed by RT-PCR and flow cytometry, respectively. The production of IFN-${\gamma}$ was measured by enzyme-linked immunosorbent assay. Results: WEPL significantly enhanced the transcription of IFN-${\gamma}$ mRNA. The effect of WEPL on IFN-${\gamma}$ expression was further supported by a concomitant increase in the number of cells with intracellular IFN-${\gamma}$ protein as well as the secretion of IFN-${\gamma}$. However, WEPL did not modulate either gene expression or protein secretion of interleukin-4, a Th2-associated cytokine, by Con A-stimulated T lymphocytes. Conclusion: Our results demonstrate that one of the potentially beneficial anti-tumor and immunostimulatory effects of WEPL may be mediated through the enhancement of IFN-${\gamma}$ secretion by T lymphocytes.

특발성 폐섬유화증에서 Interferon-${\gamma}$의 효과 (The Efficacy of Interferon(IFN)-${\gamma}$ in Idiopathic Pulmonary Fibrosis)

  • 서 론 : 특발성 폐섬유화증은 원인 모르게 폐 내에 섬유화가 진행하여 진단 후 중간생존기간이 3-5년 정도로 보고되는 치명적인 질환이나 생존기간을 증가시키는 효과가 있는 치료제는 아직 알려져 있지 않다. 최근 스테로이드나 다른 면역억제제 치료에 반응이 없는 IPF에서 INF-${\gamma}$이 효과가 있다는 보고가 있었으나 아직은 많은 논란이 있어 진행된 IPF 환자에서 INF-${\gamma}$ 효과를 관찰하기 위하여 본 연구를 시행하였다. 방 법 : 1999년 1월부터 2001년 3월까지 IPF-UIP로 진단되었던 환자 중 스테로이드제제 및 기존의 면역억제제에 반응을 보이지 않았던 9명 (연령 $55.4{\pm}15.3$세, 남:여=8:1)을 대상으로 INF-${\gamma}$ 200만 unit를 주당 3회 피하주사로 12개월간 저용량 스테로이드와 합께 투여하였다. INF-${\gamma}$ 투여시에도 UIP가 진행되거나 심각한 부작용 시 투여를 중단하였다. 결 과 : 1) 전체 9례 중 5례는 INF-${\gamma}$ 치료를 도중에 중단하였고 총 4례만이 INF-${\gamma}$ 12개월치료를 완료하였으나, 이들에서 UIP의 호전은 관찰되지 않았다 2) 전체 9례 중 5례가 치료 시작 후 12개월 이내에 질병악화로 인해 사망하였고, 1례는 INF-${\gamma}$ 치료종료 후 폐렴으로 사망하였다. 3) INF-${\gamma}$ 치료 시작시점에 생존군에 비하여 사망군의 폐기능이 저하되어 있었다(FVC: 추정 정상치의 $61.3{\pm}5.1$ %, vs. $45.7{\pm}12.3%$, P=0.048, $D_Lco$: $45.0{\pm}5.0$ % vs. $30.8{\pm}11.2$ %, P=0.048). 결 론 : 진행된 IPF에서 INF-${\gamma}$ 치료는 효과가 없었으며, 특히 심한 폐기능의 손상과 저산소증의 동반 시 INF-${\gamma}$의 투여는 주의를 요하는 것으로 사료되었다.

Hyperoside Protects Cells against Gamma Ray Radiation-Induced Apoptosis in Hamster Lung Fibroblast

  • Ionizing radiation, including that evoked by gamma (${\gamma}$)-rays, induces oxidative stress through the generation of reactive oxygen species, resulting in apoptosis, or programmed cell death. This study aimed to elucidate the radioprotective effects of hyperoside (quercetin-3-O-galactoside) against ${\gamma}$-ray radiation-induced apoptosis in Chinese hamster lung fibroblasts, V79-4 and demonstrated that the compound reduced levels of intracellular reactive oxygen species in ${\gamma}$-ray-irradiated cells. Hyperoside also protected irradiated cells against DNA damage (evidenced by pronounced DNA tails and elevated phospho-histone H2AX and 8-oxoguanine content) and membrane lipid peroxidation. Furthermore, hyperoside prevented the ${\gamma}$-ray-provoked reduction in cell viability via the inhibition of apoptosis through the increased levels of Bcl-2, the decreased levels of Bax and cytosolic cytochrome c, and the decrease of the active caspase 9 and caspase 3 expression. Taken together, these results suggest that hyperoside defend cells against ${\gamma}$-ray radiation-induced apoptosis by inhibiting oxidative stress.

The Effect of Gamma Irradiation on PLGA and Release Behavior of BCNU from PLGA Wafer

  • The objectives of this study were to investigate the influence of gamma irradiation for sterilization on poly(D,L-lactide-co-glycolide) (PLGA) with different molecular weight and the effect of gamma irradiation on the release behavior of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU, carmustine) from PLGA wafer with various irradiation doses. The effect of gamma irradiation on PLGA was evaluated by gel permeation chromatography (GPC), differential scanning calorimetry (DSC), and electron paramagnetic resonance (EPR). The weight average molecular weight (M$_{w}$) and glass transition temperature (T$_{g}$) of PLGA decreased after gamma irradiation. The extent of M$_{w}$ reduction was dependent on irradiation dose and PLGA molecular weight. Using EPR spectroscopy, we successfully detected gamma irradiation induced free radicals in PLGA. The gamma irradiation increased the release rate of BCNU from PLGA wafer at applied irradiation doses except 2.5 Mrad of irradiation dose in this study.study.

  • For analytic functions we give an expression for the kernel $K_n$ of the remainder terms for the Gauss-Radau and the Gauss-Lobatto rules with end points of multiplicity r and prove the convergence of the kernel we obtained. 한국인 치주질환 환자에서 Fc ${\gamma}R$ 유전자다형성에 관한 연구 (Fc ${\gamma}R$ genetic polymorphisms of periodontal disease in Korean population)

  • Genomic Project 이후로 다양한 질환에 있어서 유전적인 영향에 관한 연구가 진행되고 있다. 이 연구의 목적은 한국인 치주질환 환자에서 Fc ${\gamma}R$ 유전자의 유전자다형성과 치주질환 특성과의 관련성을 알아보는 것이다. 치주적으로 건강한 한국인 90명(대조군, 남자64명, 여자26명), 중도 만성 치주염환자 40명(severe chronic periodontitis patients; severe CP, 남자 24명, 여자 16명)을 대상으로 임상지수(치주낭 깊이, 입상부착소실, 치은지수, 치태지수, 탐침 후 출혈지수, 치조골소실)를 측정하였다. 또한 이들의 정맥혈에서 추출한 DNA를 PCR(Polymerase Chain Reaction)법, 전기영동법 등을 이용하여 Fc ${\gamma}RIIIa$ , Fc ${\gamma}RIIIb$의 대립유전자의 존재여부를 확인하였다. 이를 바탕으로 각 유전자의 다형성 및 Fc ${\gamma}R$ 복합유전자형 (Fc ${\gamma}R$ composite genotype)을 확인하여, 각 군 간을 비교하였다. 치주질환의 특성과 유전자 다형성과의 관련성을 알아보기 위하여 Fc ${\gamma}R$ 유전자에 대한 유전자다형성을 조사한 결과 다음과 같은 결과를 얻을 수 있었다. 1. Fc ${\gamma}RIIla$에 대한 유전자다형성 연구결과 대조군과 severe CP, AgP군 사이에서, severe CP와 AgP군 사이에서는 대립유전자분포가 서로 유의성 있는 차이를 나타내었지만(p<0.05), Fc ${\gamma}RIIlb$에서는 유의성 있는 차이를 보이지 않았다(p>0.05). 2. Fc ${\gamma}R$ 복합유전자형간의 비교에서 유의성 있는 차이를 발견할 수 없었다(p>0.05). 이와 같은 결과를 종합하여 볼 때 실험대상 한국인 치주염환자에서 Fc ${\gamma}R$ 유전자에 대한 다형성분석에서 Fc ${\gamma}RIIIa$ 대립유전자가 치주염에 대한 감수성과 관련되어 있다고 생각된다. 이 연구의 결과는 유전자의 차이가 치주질환의 감수성 판단의 자료로 활용할 수 있는 가능성을 보여주고 있다.

Gamma Irradiation Up-regulates Expression of B Cell Differentiation Molecule CD23 by NF-κB Activation

  • Gamma irradiation ($\gamma$-IR) is reported to have diverse effects on immune cell apoptosis, survival and differentiation. In the present study, the immunomodulatory effect of a low dose $\gamma$-IR (5~10 Gy) was investigated, focusing on the role of NF-${\kappa}B$ in the induction of the B cell differentiation molecule, CD23/FceRII. In the human B cell line Ramos, $\gamma$-IR not only induced CD23 expression, but also augmented the IL-4-induced surface CD23 levels. While $\gamma$-IR did not cause STAT6 activation in these cells, it did induce both DNA binding and the transcriptional activity of NF-${\kappa}B$ in the $I{\kappa}B$ degradation-dependent manner. It was subsequently found that different NF-${\kappa}B$ regulating signals modulated the $\gamma$-IR-or IL-4-induced CD23 expression. Inhibitors of NF-${\kappa}B$ activation, such as PDTC and MG132, suppressed the $\gamma$-IR-mediated CD23 expression. In contrast, Ras, which potentiates $\gamma$-IR-induced NF-${\kappa}B$ activity in these cells, further augmented the $\gamma$-IR- or IL-4-induced CD23 levels, The induction of NF-${\kappa}B$ activation and the subsequent up-regulation of CD23 expression by $\gamma$-IR were also observed in monocytic cells. These results suggest that $\gamma$-IR, at specific dosages, can modulate immune cell differentiation through the activation of NF-${\kappa}B$, and this potentially affects the immune inflammatory response that is mediated by cytokines.

Poly-gamma-glutamic acid의 방사선 조사에 의한 인대 조직 손상 보호 효과 (Prevention Effect of Poly-gamma-glutamic Acid on Porcine Ligament Tissue Damage Induced by Gamma Irradiation)

  • This study was conducted to determine the prevention effect of poly-gamma-glutamic acid (${\gamma}-PGA$) on tissue damage induced by gamma irradiation for development of xenograft. Porcine tendons were treated at various doses of ${\gamma}-PGA$ (0.1, 0.5, 1 and 5%) and then gamma-irradiated (30 kGy). Prevention effects on tissue damage were measured as the result of tensile strength, hydroxyproline contents and viscosity of ${\gamma}-PGA$. Tensile strength was remarkably decrease in gamma-irradiated porcine ligament, but increased by ${\gamma}-PGA$ treated one. Among the ${\gamma}-PGA$ treatment doses, 1% treated group showed the highest values of tensile strength compared to non-treated group. Hydroxyproline contents was significantly increased by gamma irradiation, but decreased by the ${\gamma}-PGA$ treatment. Particularly, 1 and 5% ${\gamma}-PGA$ treated group were exhibited lower values of hydroxyproline contents than other group. In the result of viscosity, gamma-irradiated ${\gamma}-PGA$ (1%) was remarkably increased. Base on the results, it demonstrated that gamma irradiation induces severe alteration of mechanical property and collagen contents on porcine ligament, but ${\gamma}-PGA$ can effectively prevent these tissue damage.

일부 남성근로자에서 ${\gamma}-GTP$와 관련된 요인에 대한 단면연구 (A Cross-Sectional Study on ${\gamma}-GTP$ and its Related Factors in Male Workers)

  • Objective : To investigate gamma-Glutamyltransferase (${\gamma}-GTP$) and its related factors in male industrial workers. Method : Five hundred and tony male workers without heart disease, diabetes mellitus, renal disease, hepatitis, and other liver diseases were surveyed in October 1998. Blood samples were collected to test for ${\gamma}-GTP$, total-cholesterol and fasting blood glucose. A self-administered questionnaire survey on life style was also done. Results : The total geometric mean value of ${\gamma}-GTP$ was 30.6 U/L. According to a univariate analysis: age, BMI(body mass index, $kg/m^2$), alcohol consumption, current smoking, stress, diastolic blood pressure, and blood total cholesterol were significantly associated with ${\gamma}-GTP$(p<0.05). From a multiple regression analysis: BMI, alcohol consumption, current smoking, diastolic blood pressure and total-cholesterol were significantly related to ${\gamma}-GTP$(p<0.05). Coffee consumption was negatively related to ${\gamma}-GTP$, but not significantly. Conclusion : We recommend that a change in health behavior (i.e. reducing alcohol intake, controlling BMI and not smoking) is necessary to decrease ${\gamma}-GTP$ in male workers.
