The Distribution of Selenium in Proteins of Saccaromyces Cerevisia and Analysis of Selenomethionine in Specific Protein (효모단백질내의 셀레늄 분포 및특정단백질내의 셀레노메티오닌 분석)

  • Selenized yeast (Se yeast) containing $0.1{\%}$(w/w) of selenium was obtained when the yeast was incubated at a selenium concentration of 1$1.14{\times}10_-3 M$ in rich medium. After washing several times, the inorganic selenium on the cell wall was confirmed with MBRT. There was no indication of inorganic selenium on the cell wall when the blue color in MBRT was stayed for 15 minutes. The selenized yeast was sonicated, then the selenium contained protein was obtained after salting out by ammonium sulfate at the concentration $80{\%}$ saturation. The seven protein bands were seperated by SDS-PAGE and the selenium concentration in protein was measured by ICP-AES. Analytical data showed that the large expressed protein band contained a relatively large amount of selenium. The proteins of the 47kDa was contained the concentrations of 69.5 ${\mu}$ Se/g of most many content. The protein (47 kDa) was seperated from PVDF membrane by tank-electroblotting. The isolated protein was hydrolyzed under acid condition and reacted with PITC. The derivatives of amino acids were analyzed by HPLC and compared with the data obtained from regular yeast. The resulting selenium-yeast was analyzed with the selenomethionine concentration of $2{\%}$ comparaed with general amino acids. The goal of this study is to analyze the selenium concentration in protein bands and measure the degree of biotransformation of selenomethionine in a specific protein.

  • In this study, the facilitated transport membranes, poly (ether ether)ketone (SPEEK)-Ag salts layers on top of polycarbonate supports membrane, were prepared and tested for the separation of propylene/propane. SPEEK was synthesised using PEEK and $H_2SO_4$. Experiments were porformed at room temperature and feed pressures up to 30 psig. The SPEEK-Ag salt membranes showed good selectivity for propane over propylene. With increasing the concentration of SPEEK in MeOH, 5~20 wt%, the thickness of SPEEK membrane on the polycarbonate increased. The selectivity and permeance of SPEKK-Ag membrane for propylene/propane were changed by membrane thickness and concentration of Ag salts.

  • Tetracycline (TC) groups, widely used veterinary antibiotics, can enter into environment through animal manure application. TC forms a ligand complex with multivalent metal cations via chelation that can affect sorption and mobility of TC in soil. So far, however, it has been confirmed through the reaction of the soil outside in the aqueous solution and the evaluation of the performance in the soil cultivation process is insufficient. The purpose of this study was to examine effects of copper on TC toxicity to lettuce growth. In this research, $750mg\;kg^{-1}$ of TC and 2.5, 7.5, $17.5mg\;kg^{-1}$ of Cu are treated in soil and lettuce was cultivated in the treated soil. Growth difference of lettuce by treatment was observed. As a result, $750mg\;kg^{-1}$ of TC treated soil showed toxic effect to lettuce and the effect is alleviated by copper treatment.

Influence of the Asian Monsoon on Seasonal Fluctuations of Water Quality in a Mountainous Stream (산간 계류성 하천의 계절적 수질변동에 대한 몬순강우의 영향)

  • The present study was to determine how seasonal rainfall intensity influences nutrient dynamics, ionic contents, oxygen demands, and suspended solids in a lotic ecosystem. Largest seasonal variabilities in most parameters occurred during the two months of July to August and these were closely associated with large spate of rainfall. Dissolved oxygen (DO) had an inverse function of water temperature (r = = = - 0.986, p<0.001). Minimum pH values of<6.5 were observed in the late August when rainfall peaked in the study site, indicating an ionic dilution of stream water by precipitation. Electrical conductivity (EC) was greater during summer than any other seasons, so the overall conductivity values had direct correlation (r = 0.527, p<0.01) with precipitation. Ionic dilution, however, was evident 4 ${\sim}$ 5 days later in short or 1 ${\sim}$ 2 weeks in long after the intense rain, indicating a time-lag phenomenon of conductivity. Daily COD values varied from 0.8 mg $L^{-1}$ to 7.9 mg $L^{-1}$ and their seasonal pattern was similar (r = 0.548, p<0.001) to that of BOD. Total nitrogen (TN) varied little compared to total phosphorus (TP) and was minimum in the base flow of March. In contrast, major input of TP occurred during the period of summer monsoon and this pattern was similar to suspended solids, implying that TP is closely associated (r = 0.890, p<0.01) with suspended inorganic solids. Mass ratios of TN : TP were determined by TP (r= -0.509, p<0.01) rather than TN (r= -0.209, p<0.01). The N : P ratios indicated that phosphorus was a potential primary limiting nutrient for the stream productivity. Overall data suggest that rainfall intensity was considered as a primary key component regulating water chemistry in the stream and maximum variation in water quality was attributed to the largest runoff spate during the summer monsoon.

  • The experiment was conducted in the pots of flooded saline paddy soil to evaluate influence of rice straw and gypsum application on germination and early growth status of directly sowed rice seedlings. 1. Germination percentage of rice seedlings were higher in treatment sowing at 1 day after submergence than that of treatment sowing at 21 days after submergence, and was severely interrupted by rice straw application. 2. Application of rice straw promoted the release of bicarbonate and volatile fatty acid of submerged water, while the amount of sulfate and soluble cations were decreased as compared to control. 3. Germination percentage of rice seedling had significant negative correlations with chemical characters of submerged water, and was highly affected by submerged water at 7 days after sowing. 4. Rice straw application interrupted germination of rice seedling by increasing pH of submerged water over critical level, and gypsum application depressed the early growth of young seedling dues to high salt concentration. 5. The relaease of bicarbonate was remarkedly increased with increasing pH over 7.5 of submerged water.

Arsenic removal from artificial arsenic water using CaAl-monosulfate and CaAl-ettringite (CaAl-monosulfate와 CaAl-ettringite를 이용한 인공비소폐수의 비소 제거 연구)

  • The objective of this study is to remove arsenate from artificially contaminated wastewater using CaAl-ettringite and CaAl-monosulfate which were synthesized in laboratory. The study was carried on the basis of solidification/stabilization of waste using cement. Monosulfate and ettringite are constituents of cement paste. The CaAl-ettringite has a chemical formula of $Ca_6Al_2O_6(SO_4)_3{\cdot}32H_2O$ and has a needle like morphology. Whereas CaAl-monosulfate $Ca_4Al_2O_6(SO_4){\cdot}12H_2O$ has layered double hydroxide structure (LDH) in which the mainlayer consists of Ca and Al and S as interlayer. Ettringite and monosulfate were synthesized by reaction of tricalcium aluminate and gypsum and hydrating this mixture at elevated temperature. The synthesized mineral were characterized by PXRD and FESEM to ensure purity. It was found that concentrations of As(V) in contaminated water were reduced from initial concentration of 1.335 mmol/L to 0.054 mmol/L and 0.300 mmol/L by CaAl-monosulfate and CaAl-ettringite respectively. The post experimental results of PXRD and FESEM analysis indicate that arsenate removal was by ion exchange.

Determination of alkylphenol ethoxylate in water by high performance liquid chromatography/electrospray ionization/mass spectrometry (HPLC/ESI/MS를 이용한 물 중의 알킬페놀에톡실레이트 분석)

  • A method is described for the analysis of short-chain alkylphenol ethoxylates (APEOs), 4-octylphenol-di-ethoxylate (OP2EO) and 4-nonylphenol-di-ethoxylate (NP2EO), in drinking water or wastewater using reversed phase high-performance liquid chromatography with electrospray ionization mass spectrometry. The solvent system was water and methanol containing $10{\mu}M$ trifluoroacetic acid as an ionization solvent. We acidified 1 L of water samples to less than pH 2 with concentrated $H_2SO_4$ and loaded onto Sep-Pak $C_{18}$, and eluted with acetone. The calibration of OP2EO and NP2EO was performed for the concentration range from 20 to 500 ng/L and the correlation coefficients were 0.999 and 0.990, respectively. The limits of detection were 20 ng/L (OP2EO) and 50 ng/L (NP2EO) at a signal-to-noise ratio of 3. Accuracy and precision of this analytical method were 85.8 ~ 122.1% and 8.2 ~ 18.8%, respectively. The proposed method allowed a sensitive and rapid detection of OP2EO and NP2EO and it could be applied for monitoring of APEOs from environmental samples.

Characteristics of Groundwater Quality in Sasang Industrial Area, Busan Metropolitan City (부산시 사상공단지역의 지하수 수질 특성)

  • In urban areas, groundwater pollution is heavily affected by urbanization with land use types. This study aims to characterize groundwater quality and contamination in Sasang industrial area of Busan Metropolitan City where metalworking, machinery and footwear factories are located. Busan Metropolitan City is the highest in the utilization of groundwater resources among the metropolitan cities in Korea. $K^+,\;Na^+,\;Ca^{2+},\;Mg^{2+},\;Cl^-,\;SO_4^{2-}\;and\;HCO_3^-$ concentrations, and electrical conductivity (EC), total dissolved solids (TDS) and salinity are high in the areas near the Nakdong River. The results are attributed to the influence of salt water which intruded into the coastal sediments during sedimentation. In addition, the dominant chemical type of Ca-Cl indicates the influence of salt water in the geological formations as well as anthropogenic pollution. $SiO_2$ ion is interpreted to originate from both water-silicate mineral reactions and the decomposition of cement concretes. Trichloroethylene (TCE) was detected at 12 sites of total 18 sites. However, tetrachloroethylene (PCE) was detected at f sites and 1.1.1-trichloroethane (TCA) at 3 sites. According to the factor analysis, factor 1 was explained by 49.8%, factor 2 19.8%, and factor 3 11.0% with total 80.6% explanation. pH, TDS, salinity, $Ca^{2+},\;K^+,\;Mg^{2+},\;Na^+,\;Al^{3+},\;As^{3+},\;Cl^-\;and\;Fe^{2+}$ were positively highly loaded to factor 1. The chemical components loaded to factor 1 represent the chemical characteristics of both industrial pollution and influence by salt water. Based on the cluster analysis and distribution pattern of chemical components, the concentration of $Na^+,\;Ca^{2+},\;Cl^-,\;SO_4^{2-}\;K^+,\;and\;Mg^{2+}$ is high in the riverside area of the Nakdong River composed of coastal sediments that is influenced by salt water. The downstream area of the Hakjang Stream is judged to be affected by both salt water and artificial pollution. The other part of the study area is interpreted by anthropogenic pollution.

Evaluation of the Giggenbach Bottle Method with Artificial Fumarolic Gases (인공 분기공 가스를 이용한 Giggenbach bottle 법의 평가)

  • We aimed to evaluate the effectiveness of the Giggenbach bottle method and develop the related pretreatment and analytical methods using artificial fumarolic gases. The artificial fumarolic gases were generated by mixing $CO_2$, CO, $H_2S$, $SO_2$, $H_2$, and $CH_4$ gas streams with a $N_2$ stream sparged through an acidic medium containing HCl and HF, with their compositions varied by adjusting the gas flow rates. The resultant fumarolic gases were collected into an evacuated bottle partially filled with a NaOH absorption solution. While non-condensible gases such as CO, $H_2S$, and $CH_4$ accumulated in the headspace of the bottle, acidic components including $CO_2$, $SO_2$, HCl, and HF that were dissolved into the alkaline solution. Like other acidic components, $H_2S$ also dissolved into the solution, but it reacted with dissolved $Cd^{2+}$ to precipitate as CdS when $Cd(CH_3COO)_2$ was added. The non-condensible gases were analyzed on a gas chromatography. Then, CdS precipitates were separated from the alkaline solution by filtration, and they were pretreated with $H_2O_2$ to oxidize CdS-bound sulfide into sulfate. In addition, a portion of the solution was also pretreated with $H_2O_2$ to oxidize sulfite to sulfate. Following the pretreatment, the resultant samples were analyzed for $SO_4^{2-}$, $Cl^-$ and $F^-$ on an ion chromatography. In the meanwhile, dissolved $CO_2$ was analyzed on a total organic carbon-inorganic carbon analyzer without such pretreatment. According to our experimental results, the measured concentrations of the fumarolic gases were shown to be proportional to the gas flow rates, indicating that the Giggenbach bottle method is adequate for monitoring volcanic gas. The pretreatment and analytical methods employed in this study may also enhance the accuracy and reproducibility of the Giggenbach bottle method.

Synthesis of Aniline from Nitrobenzene and Fe(CO)5 with PEG/γ-Al2O3 as Phase Transfer Catalyst (PEG/γ-Al2O3 상이동 촉매상에서 니트로벤젠과 Fe(CO)5로부터의 아닐린 합성)

  • Immobilized polyethylene glycols onto metal oxides such as ${\gamma}-Al_2O_3$, ${\alpha}-Al_2O_3$, $SiO_2$ and $TiO_2$ were used as phase transfer catalysts for the room temperature synthesis of aniline from nitrobenzene and ironpentacarbonyl. The amount of attached PEG molecules increased with specific surface area of metal oxides. Among the immobilized catalysts tested PEG/${\gamma}-Al_2O_3$ showed the highest activity. The reaction rate increased with the chain length of PEG mole-cules and the aqueous NaOH concentration. Mechanistic study carried out using infrared spectrometer revealed that the role of PEG was to increase the formation of $HFe(CO)_4{^-}$ ion, which is known as active species, and its movement from aqueous to organic phase.
