Effect of Gamma Irradiation on the Structural and Physiological Properties of Silk Fibroin

  • This study was conducted to examine the changes in the molecular structure and physiological activities of silk fibroin by gamma irradiation. The results of gel permeation chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that the molecular weight of fibroin was increased depending upon the irradiation dose. Secondary structure of fibroin determined by using circular dichroism revealed that the ratio of $\alpha$-helix was increased up to 10 kGy and then decreased depending upon the irradiation dose. Whereas, the ratio of $\beta$-sheet, $\beta$-turn, and random coil were decreased and then increased with an alteration in the $\alpha$-helix secondary conformation. The 2.2-diphenyl-1-picryl-hydrazil (DPPH) radical scavenging activity of fibroin was increased by gamma irradiation at 5 kGy, but was decreased above 10 kGy depending upon the irradiation dose. Also, the inhibition activities of tyrosinase and melanin synthesis of fibroin were increased by gamma irradiation. These results indicated that gamma irradiation could be used as an efficient method to make fibroin more suitable for the development of functional foods and cosmetics.

Effect of Gamma Rays on the Growth Performance of Bangladesh Clone Tea

  • The experiment was carried out to investigate the effects of gamma radiation on the early growth performance and physiological traits of BT2 clone tea, the most promising cultivar released by Bangladesh Tea Research Institute. The fresh shoot cuttings were irradiated with seven different levels of gamma radiation such as 0, 10, 20, 30, 40, 50 and 60 Gy from Cobalt 60Co source (Dept. of PlantBreeding, Bangladesh Institute of Nuclear Agriculture). Thereafter, the irradiated shoot cuttings were planted in polythene bags and kept under natural conditions. It was observed that callusing was initiated from 8th weeks after placement of tea shoot cuttings in the polythene bags and completed by 12th weeks. The morphological growth of tea shoot cuttings were recorded under varying levels of gamma radiation and growth stages. It was observed that the number of leaves, number of primary branches, base diameter, root length and total leaf area per plant significantly increased with the progress of time and increasing levels of gamma radiation, however, the plant height showed decreasing trend with the increasing levels of gamma radiation, which could be due to the change in chromosomal structure and genetic makeup. After 56 weeks of planting, the plant height, the number of leaves and primary branches per plant, base diameter, root length and total leaf area per plant recorded were 65.70 cm, 30.67, 7.33, 1.48 cm, 23.50 cm, and 1250.67 cm2 per plant respectively under the radiation level 60 Gy, whereas the corresponding figures of the above parameters at the control treatment were 76.21 cm, 18.33, 3.67, 0.92 cm, 17.75 cm and 778.33 cm2 per plant, respectively. A significant relationship was observed among the physiological growth parameters with the increasing levels of gamma radiation. The total dry matter gain, leaf area index, absolute growth rate and relative growth rate were significantly influenced with the rising levels of gamma radiation (up to 60 Gy), whereas the net assimilation rate of individual tea plant non-significantly responded as compared to those of control treatment. Finally after 56 weeks of planting, the maximum total dry weight gain, leaf area index, absolute growth rate, relative growth rate and net assimilation rate recorded under 60 Gay radiation level were 40.25 g/plant/week, 4.25, 1.18 g/week, 0.0621g/g/week and 17.07 g/m2/week respectively.

Ag-V/γ-Al2O3 촉매상에서 탄화수소-Selective Catalytic Reduction에 의한 질소산화물 저감 (DeNOx by Hydrocarbon-Selective Catalytic Reduction on Ag-V/γ-Al2O3 Catalyst)

  • 본 연구에서는 배출가스 중에 포함된 NO를 비선택적 촉매환원법으로 환원시켜 제거하기 위하여 Ag와 V의 함량을 여러 가지로 달리하여 ${\gamma}-Al_2O_3$에 담지한 촉매를 제조하였고, 제조한 촉매에 대하여 온도, 산소농도, 아황산가스농도의 변화에 따른 $NO_x$의 전환율에 대하여 연구하였다. 또한 제조한 촉매의 물성분석을 통하여 촉매의 상태와 $NO_x$의 전환율과의 관계를 알아보았다. $AgV/{\gamma}-Al_2O_3$ 촉매의 경우에는 고온에서는 $Ag/{\gamma}-Al_2O_3$ 촉매보다 낮은 $NO_x$ 전환율을 나타내는 반면에 저온에서는$Ag/{\gamma}-Al_2O_3$ 촉매보다 높은 $NO_x$ 전환율을 나타내었고, 반응가스 중에 $SO_2$가 함유되어 있어도 $NO_x$의 전환율이 낮아지지 않았다. 반응실험 전 후의 촉매에 대하여 X-ray Diffraction, X-ray Photo electron Spectroscopy, Temperature Programmed Reduction, Ultraviolet-Visible Diffuse Reflectance Spectroscopy 등의 분석결과와 반응실험 결과를 비교하여 볼 때 V가 포함됨으로 인하여 Ag의 산화상태가 잘 유지되지 못하여 고온에서는 $NO_x$ 전환율이 낮아지며, $300^{\circ}C$ 이하의 저온에서는 V의 촉매작용으로 인하여 $NO_x$ 전환율이 높아진 것으로 나타났다.

  • 암 억제제인 $p16^{INK4A}$ 단백질의 활성부위 84-104번까지의 21개 아미노산으로 이루어진 펩타이드를 합성하여, 이것의 용액상 구조를 CD, $^1H$ NMR 분광법 그리고, 분자 모델링 방법으로 분석하였다. CDK4 그리고 CDK6와 함께 안정된 complex를 형성하는 p16의 활성 펩타이드(84-104 아미노산)는 in vitro에서 pRb를 인산화하는 CDK4/6의 능력을 차단하고, p16단백질의 기능에서 보여주듯이 G1/S상의 세포 Cycle을 차단한다. NOE를 포함하는 $^3J_{NH{\alpha}}$ 스핀결합 상수, $C_{\alpha}H$ 화학적 이동, 아마이드 화학적 이동의 평균 변화 폭 그리고 온도 계수 등은 p16 펩타이드의 이차구조가 helix-turn-helix의 구조를 구성하는 p16단백질과 유사한 2차 구조를 가지고 있음을 보여주었다. NOE에 근거한 거리 및 이면각을 이용한 3.D 기하구조는 p18이나 p19의 대응하는 부위에 대한 결정구조에서 보여준 바와 같이 아미노산 $Gly^{89}-Leu^{91}$(${\varphi}_{i+1}=-79.8^{\circ}$, ${\varphi}_{i+1}=60.2^{\circ}$)사이에는 ${\gamma}$-회전구조를 형성함을 보여주었다. 이렇게 비교적 단단한 구조를 형성하고 있는 ${\gamma}$-회전구조부위는 p16펩타이드 구조를 안정시키며, CDK를 인식하는 부위로 작용할 수 있다. 이러한 ${\gamma}$-회전구조는 항암제 선도물질을 개발하는데 유용하게 활용될 수 있을 것이다.

  • The composition and the microstructure of the Zn-Cr alloys electroplated in chloride bath with EDTA were studied according to electrolysis conditions. The cathode current efficiency decreased with increasing both Cr/(Cr+Zn) ratio and current density. The Cr content of the alloy deposits increased with Cr/(Cr+Zn) ratio and current density The phase structure of Zn-Cr alloy deposits changed from η-Zn through η-Zn+${\gamma}$'-ZnCr to ${\gamma}$'-ZnCr with increasing Cr content of alloys. The surface morphology of Zn-Cr alloy deposits changed from fine needle shaped crystallites through the mixed structure of needle-shaped and granular one to the colony structure with fine granular crystallites according to the change of phase structure

  • We give pressure stimulation into organic thin films and then manufacture a device under the accumulation condition that the state surface pressure is 10[mN/m]. In processing of a device manufacture, we can see the process is good from the change of a surface pressure for organic thin films and transfer ratio of area per molecule. The structure of manufactured device is Au/Poly-${\gamma}$ Benzyl $_D$-Glutamate/Al and Au/Poly-${\gamma}$ Benzyl $_D$-Glutamate/Au; the number of accumulated layers is 1, 3, 5 and 7. Also, we then examined of the MIM device by means of I-V. The I-V characteristic of the device is measured from 0 to +2[V]. We determined electrochemical measurement by using cyclic voltammetry with a three-electrode system. LB film accumulated by monolayer on an ITO. In the cyclicvoltammetry, An Ag/AgCl reference electrode, a platinum wire counter electrode and LB film-coated ITO working electrode measured in $LiBF_4$ solution, stable up to 0.9V vs. Ag/AgCl.

The Null Distribution of the Likelihood Ratio Test for a Mixture of Two Gammas

  • We investigate the distribution of likelihood ratio test(LRT) of null hypothesis a sample is from single gamma with unknown shape and scale against the alternative hypothesis a sample is from a mixture of two gammas, each with unknown scale and unknown (but equal) scale. To obtain stable maximum likelihood estimates(MLE) of a mixture of two gamma distributions, the EM(Dempster, Laird, and Robin(1977))and Modified Newton(Jensen and Johansen(1991)) algorithms were implemented. Based on EM, we made a simple structure likelihood equation for each parameter and could obtain stable solution by Modified Newton Algorithms. Simulation study was conducted to investigate the distribution of LRT for sample size n = 25, 50, 75, 100, 50, 200, 300, 400, 500 with 2500 replications. To determine the small sample distribution of LRT, I considered the model of a gamma distribution with shape parameter equal to 1 + f(n) and scale parameter equal to 2. The simulation results indicate that the null distribution is essentially invariant to the value of the shape parameter. Modeling of the null distribution indicates that it is well approximated by a gamma distribution with shape parameter equal to the quantity $0.927+1.18/\sqrt{n}$ and scale parameter equal to 2.16.

사람에서 식이의 n6 Gamma-Linolenic Acid와 n3 Alpha-Linolenic Acid가 혈장 지질조성과 혈소판 응집반응에 미치는 영향 (Effect of Dietary n6 Gamma-linolenic Acid and n3 Alpha-linolenic Acid on Plasma Lipid Composition and Platelet Aggregation in Human Subjects)

  • To observe the effect of dietary n6 linoleic acid, n6 gamma-linolenic acid and n3 alphalinolenic acid aon plasma lipid composition and platelet aggregation, twenty college women were divided into 4 groups and treated for 2 weeks with experimental diets supplying fat at 23% cal which were different only in fatty acid composition. Dietary fat was corn oil(CO) as a source of n6 linoleic acid(LA), perilla oil(PO) for n3 alpha-linolenic acid(ALA) and evenign primrose oil(EPO) for n6 gamma-linolenic acid(GLA). Plasma cholesterol level was slightly decreased by PL(13.5g) but significantly increased by equal amount of CO. However, there was similar hypocholeaterolemic effect when double amount of CO(27.0g), was supplemented. Therefore, total fat unsaturation may be more important factor for plasma cholesterol-lowering effect than the structure of fatty acid itself. Plasma cholesterol level was not lowered by supplement of GLA in CO diet. There was similar trend in hypotriglyceridemic effect by PO and CO as in plasma cholesterol. Plasma TG level was rather increased but not significantly by GLA supplement to CO diet. Overall, plasma lipid-lowering effect was greater by ALA than LA and GLA effect was not greater than by LA. GLA supplement did not significantly improve lipid compositions to prevent against CHD. There was no significant change both in fatty acid composition in platelet and ADP-induced platelet aggregation by GLA supplement to corn oil diet and by ALA in PO diet in young women.

  • The effect of interlamellar spacing on microstructural stability at high temperature was studied for unidirectionally solidified ternary $Ni\;/\;Ni_3Al-Ni_3$ Nb and binary $Ni-Ni_3Nb$ eutectic composite. The interlamellar spacing of both alloy systems were varied with the growth rate according to $"{\lambda}^2R=constant"$ relationship. As a result of isothermal heat treatments at high temperature it was considered that coarsening of lamellar structure was due to concentration gradient between the tip with a relatively small radius of curvature and the side of the thick lamellae with a larger radius of the opposite sign. Fault density was increased as the interlamellar spacing decreased. Therefore it is also considered that the higher coarsening rate of the specimen with the smaller interlamellar spacing was due to higher fault density. And the diference of coarsening rate between $Ni\;/\;Ni_3Al-Ni_3Nb$ and $Ni-Ni_3Nb$ eutectic composites was not observed when the interlamellar spacing was similar in size. This means that the presence of ${\gamma}'$ in ${\gamma}\;/\;{\gamma}'\;-{\delta}$ eutectic had no b arrier effect to diffusion through the ${\gamma}$ matrix.

Effect of γ-Irradiation on the Physicochemical Properties of Zein Films

  • To elucidate the effect of gamma-irradiation on the physicochemical properties of zein films, the molecular and mechanical properties of the films were examined after irradiation at various irradiation doses. Gamma-irradiation of zein solutions caused the disruption of the ordered structure of the zein molecules, as well as degradation, cross-linking, and aggregation of the polypeptide chains based on an SDS-PAGE study. Gamma-irradiation increased the solubility of zein and decreased the viscosity due to cleavage of the polypeptide chains. Protein solubility of the zein films in urea/2-mercaptoethanol also increased with increasing irradiation doses. Alterations of the zein molecules by irradiation decreased water vapor per-meability by 12% and increased the elongation of zein films. However, mean tensile strength of the zein films was decreased by gamma-irradiation treatment. Measurement of Hunter color values indicated that irradiation caused a destructive effect on yellow pigments, resulting in a significant decrease in Hunter b values. The microstructure as observed by scanning electron microscopy showed that irradiated zein film had a smoother and glossier surface than the non-irradiated films.
