Quantitation of Flurbiprofen in Isopropyl Myristate by High Performance Liquid Chromatography (고속액체크로마토그래피를 이용한 미리스틴산이소프로필증 플루르비프로펜의 정량)

  • An HPLC procedure with UV detection has been developed for the quantitation of flurbiprofen released into isopropyl myristate used as the receptor phase in an in vitro membraneless drug diffusion cell. The drug and the internal standard (oxaprozin) were extracted from isopropyl myristate with a mixture of dimethylsulfoxide:methanol:water (2:1:1) and quantitated using a reverse phase $C_{18}$ column. The chromatograms were completely free from interfering peaks, and the relative retention times of flurbiprofen and the internal standard were 4.9 and 6.8 min, respectively. Calibration plots were linear over the concentration range of $1-200\;{\mu}g/ml$ of flurbiprofen with correlation coefficients, all higher than 0.99. The mean intra-day precision and accuracy among three replicate sets of the assay in a day were 4.26 and 4.52%, respectively, whereas the mean inter-day precision and accuracy were 3.35 and 3.64%, respectively. The mean recovery of the drug was 92.5% over the calibration range. The method was simple, reliable and accurate for the quantitation of flurbiprofen in unpurified isopropyl myristate.

Unsteady Flow Analysis Around a HAWT System Using Sliding Mesh Technique (미끄럼 격자를 이용한 HAWT 시스템 주위의 비정상 유동장 해석)

  • An unsteady RANS analysis study of the 3-D flow around the NREL Phase VI horizontal axis wind turbine(HAWT) was performed using sliding mesh approach. Two different analysis models such as rotor-only and rotor with tower/nacelle were constructed to investigate the blade/tower interaction. Analysis results for the rotor with tower/nacelle were compared with the corresponding NREL's experimental data which produced fairly good validation of the present CFD model. Comparison of flows around those two models also clearly showed the blade/tower interaction even it was small for upwind configuration. Other visualization results and integrated aerodynamic loads including torque of the blade demonstrated the effective unsteady flow simulation capability of the present CFD model.

  • We present a rapid biogenic method for the production of nanoscale gold particles using pear extract. The formation and stability of pear-derived gold nanoparticles (Pear-AuNPs) were monitored by ultraviolet-visible spectroscopy. Their morphology, elemental composition and crystalline phase were determined by transmission electron microscopy, energy-dispersive X-ray spectroscopy and selected area electron diffraction. The average core size of crystalline Pear-AuNPs was in the range of $10{\pm}5\;nm$ and the observed morphology was spherical. The X-ray photoelectron spectrum showed a strong peak for the pure 'Au' phase. The circular dichroism spectrum indicated the natural capping ability of the pear extract, which generated peptide-gold nanoparticles. These nanoparticles were stable in aqueous solution for two months. A cell viability assay of Pear-AuNPs showed biocompatibility with human embryonic kidney 293 cells. Accordingly, this eco-friendly process for the bio-mimetic production of Pear-AuNPs is nontoxic in nature; consequently, it will find potential application in nano-biotechnology.

Piezoelectric Properties of Y-Substituted Pb(Ni1/3Nb2/3)-PbZrO3-PbTiO3 Ceramics and Their Application to Multilayer Piezoelectric Actuators (Y 치환에 따른 Pb(Ni1/3Nb2/3)-PbZrO3-PbTiO3 세라믹의 압전특성 및 적층형 압전 액츄에이터 응용)

  • The effects of $Y_2$ $O_3$-substitution on the piezoelectric properties of Pb[(N $i_{1}$3/N $b_{2}$3/)$_{0.15}$(Z $r_{1}$2/ $Ti_{1}$2/)$_{0.85}$] $O_3$ceramics were investigated. It was found that $Y^{3+}$ ions incorporate into Pb-sites of the ceramics, resulting in a increased lattice anisotropy and formation of Pb-vacancies. As a result, an orthorhombic-tetragonal phase transition was induced when $\chi$>0.005. At the morphotropic phase boundary of x=0.005, piezoelectric constants( $k_{p}$, $k_{33}$, and $d_{33}$) showed maximum values of 0.53, 0.58, and 350pC/N, respectively. A 30-layer actuator fabricated with the above material showed a maximum strain of 0.12% under 100V DC bias.

  • Purpose: Reliability is the most important factor to detect defects as wind turbines are deployed in large blades. The methods of detecting defects are various, such as non-destructive inspection and thermal imaging inspection. We propose the phased array ultrasonic testing method of non-destructive testing. Methods: We propose the active pressure mechanism for wind power blade. The phase array ultrasonic inspection method is used for fault detection inner blade surface. Controlled pressure of mechanism with respect to z-axis is important for guarantee the result of phase array ultrasonic inspection. The model based control and proposed mechanism are utilized for overall system stability and effectiveness of system. Result: The result of proposed pressure mechanism B is more stable than A. Convergence speed is also faster than A. Conclusion: We confirmed the performance of the proposed constant pressure mechanism through experiments. Non-destructive testing was applied to the specimen to confirm the reliability of detecting defects.

Effects of Hydration Treatments on the Phase Transition of Anodic Aluminum Oxide Layers (알루미늄 양극산화 피막의 상전이에 미치는 수화처리의 영향)

  • Hydration treatments were performed on the pure aluminum substrate at $100^{\circ}C$ followed by anodizing and heat treatments on the layers. The transformation behaviors of the oxide layers according to the hydration treatment were studied using TEM, XRD, RBS etc. Above $90^{\circ}C$ the hydrous oxide film could be formed, which were turned out to be hydrous oxides(AlOOH $nH_2$O). The anodization on the hydrous oxide film was more effective for the transition of amorphous anodic oxides to the crystalline $\Upsilon-Al_2$ $O_3$ comparing with the case for anodizing on the aluminum substrate without hydration treatment And additional heat treatments were also helpful for the acceleration of the transformation of the hydrous oxide to $\Upsilon-Al_2$ $O_3$. During the heat treatment the interface between $\Upsilon-Al_2$ $O_3$and the hydrous oxide layers migrated to the outer side of hydrous layer.

  • An innovative, simple, and rapid assay method based on liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) was developed and validated for the simultaneous determination of eight statin drugs in human urine. A simple sample clean-up procedure using the "dilute and shoot" (DAS) approach enabled a fast and reliable analysis. The influence of the dilution factor was investigated to ensure detectability and reduce the matrix effect. Chromatographic separation was performed on a Phenomenex Kinetex C18 column ($50{\times}3.0mm$ i.d., $2.6{\mu}m$) using an elution gradient of mobile phase A composed of 0.1% acetic acid, and mobile phase B composed of acetonitrile, at a flow rate of 0.35 mL/min. Quantitation was performed on a triple quadrupole mass spectrometer operated in multiple reaction monitoring (MRM) mode using electrospray ionization in positive ion mode. The total chromatographic run time was 15 min. The method was validated for selectivity, sensitivity, recovery, linearity, accuracy, precision, and stability. The present method was successfully applied to the analysis of Rosuvastatin in urine samples after oral administration to healthy human subjects.

Development of New Correlation and Assessment of Correlations for Two-Phase Pressure Drop in Rectangular Microchannels (사각 마이크로채널 내의 2 상 유동 압력강하 상관식의 검증 및 개발)

  • There are two kinds of models in two-phase pressured drop; homogeneous flow model and separated flow model. Many previous researchers have developed correlations for two-phase pressure drop in a microchannel. Most correlations were modified Lockhart and Martinelli's correlation, which was based on the separated flow model. In this study, experiments for adiabatic liquid water and nitrogen gas flow in rectangular microchannels were conducted to investigate two-phase pressure drop in the rectangular microchannels. Two-phase frictional pressure drop in the rectangular microchannels is highly related with flow regime. Homogeneous model with six two-phase viscosity models: $Owen^{(21)}$'s, $MacAdams^{(22)}$'s, Cicchitti et ${al.}^{(23)}$'s, Dukler et ${al.}^{(24)}$'s, Beattie and ${Whalley}^{(25)}$'s, Lin et ${al.}^{(26)}$'s models and six separated flow models: Lockhart and $Martinelli^{(27)}$'s, ${Chisholm}^{(31)}$'s, Zhang et ${al.,}^{(15)}$'s, Lee and ${Lee}^{(5)}$'s, Moriyama and ${Inue}^{(4)}$'s, Qu and $Mudawar^{(8)}$'s models were assessed with our experimental data. The best two-phase viscosity model is Beattie and Whalley's model. The best separated flow model is Qu and Mudawar's correlation. Flow regime dependency in both homogeneous and separated flow models was observed. Therefore, new flow pattern based correlations for both homogeneous and separated flow models were individually proposed.

A Thermodynamical Study on the Phase Formation and Sequence by Ion Beam Mixing in Al/Pd System (이온선 혼합에 의한 Al/Pd계의 상형성 및 전이에 관한 열역학적 연구)

  • Evaporated Al/Pd thin films were irradiated with various doses to produce intermetallic compounds. In order to study the first phase formation and phase sequence, RBS and TEM studies have been used. It was found that the initial phase formed by irradiation of $5{\times}10^{15}Ar^+/cm^2$ was $Al_3Pd_2$, while $1.5{\times}10^{16}Ar^+/cm^2$ gave the subsequent phase of AlPd. This phenomenon was analysed using effective heat of formation (${\Delta}$H') model. The experimental results agree with that predicted by effective heat of formation model. This model has been extended to predict the first phase formation and phase sequence by ion beam mixing in metal/Si systems as well as metal-metal systems.

Effects of chlorine and chlorine compounds on morphology and function of Caulobacter cells (Caulobacter의 세포(細胞)의 형태(形態) 및 기능(機能)의 분화(分化)에 대한 염소 및 염소화합물의 영향(影響))

  • Caulubacter is distinctive in the morphology and replication and ubiquitous in the biosphere, especially in every type of aquatic environment. In water and waste-water treatment processes, chlorine and chlorine compounds have been used as a main disinfectant throughout the world. Therefore, Caulobacter in the waters should be affected by chlorination of the waters. The objective of this study is to determine the effects of the disinfectants on Caulobacter cells and on the developmental processes of the cells. The Caulobacter swarmer cells were disinfected by chlorine at pH 7.0 minutes of the reaction with 2.0 mg/l of infected at pH 10.0. The swarmer cells treated with 2.0 or 4.0 mg/l of chlorine for 15 minutes lost their flagella and were observed by electron microscopy to be damaged on their cell surfaces, discharging some cellular materials. When the chlorinated swarmers and untreated control samples were recultivated in fresh PYE broth medium, the control swarmers multiplicated exponentially after one-hour lag phase, whereas the chlorinated swarmers extended the lag phase to about four hours. During the extended lag phase, the cells were proved by electron microscopy to be grown and be in predivisional step, but no swarmer cell was found. When the stalked cells were chlorinated, almost all the cells were observed to have their stalks broken and some cellular materials discharged. In those samples recultivated, many cells differentiated to possess an abnormally elongated stalk with several crossbands on it. This suggests that the chlorine-shocked Caulobacter cells can develope to abnormal morphology in water environments which they can survive and regrow in.
