Cytotoxic and Anti-oxidant Constituents from the Aerial Parts of Aruncus dioicus var. kamtschaticus

  • Ten compounds (1 - 10), palmitic acid (1), 10-nonacosanol (2), pentacosan-1-ol (3), phytol (4), ${\beta}$-sitosterol (5), ${\beta}$-sitosterol-3-O-${\beta}$-D-glucopyranoside (6), 2,4-dihydroxycinnamic acid (7), hyperoside (8), uridine (9) and adenosine (10), were isolated from the n-hexane and EtOAc-soluble fractions of the aerial parts of A. dioicus var. kamtschaticus (Rosaceae). The structures of these compounds were elucidated on the basis of spectroscopic evidence. All compounds (1 - 10) were isolated for the first time from this plant. Cytotoxicity of 1 - 10 against Jurkat T (T-lymphocytic leukemia cells), HeLa (Human cervical epitheloid carcinoma cells), MCF-7 (Human breast cancer cells), and HL-60 (Human promyelocytic leukemia cells) cell lines was measured. Compound 6 showed good cytotoxicity against HL-60 cell line with $IC_{50}$ value of 8.13 ${\mu}g/mL$. In addition, compounds 7 and 8 exhibited antioxidant activity with $IC_{50}$ values of 16.30 and 12.42 ${\mu}g/mL$, respectively.

  • Levels of sterols including ${\Delta}^7$-dehydrogenase isolated from the tissues of marine animal products (20 species) were determined on 1.5% OV-17 columm of gas-liquid chromatography. The composition showed that the mussels and clams contained various sterols in their tissues : cholesterol, brassicasterol. 24-methylenecholesterol with some minor components such as 22-trans-norcholesta-5,22-dien-3${\beta}$-ol, 22-cis-dehydrocholesterol, 22-trans-dehydrocholesterol, desmosterol, 7-dehydrocholesterol, campesterol, stigmasterol, ${\beta}$-sitosterol, isofucosterol, and 7-dehydrocholesterol which could be converted into vitamin $D_3$ in the skin tissue of animal was present in the muscle of oyster, Crassostrea gigas. On the other hand, the others including gastropoda were predominantly composed of cholesterol. The minor sterols such as 24-methylenecholesterol, stigmasterol and ${\beta}$-sitosterol in the fish intestines are supposed to be derived from dietary plankton. Cholesterol ${\Delta}^7$-dehydrogenase which could convert cholesterol into ${\Delta}^7$-dehydrogenase was present in the pickles of Tricurus haumela intestine.

Phytochemical Study for Botanical Utilization of the Fruits of Malus baccata (자원식물로서 응용을 위한 야광나무 열매의 식물화학적 연구)

  • Very little utilization of the fruits of Malus baccata(Rosaceae) has been employed for food and medicinal plants except for preparing fruit beverages. But, it was estimated as valuable to investigate the chemical components for the botanical resource of this plant. In this study, it was found that the fruits of this plant contained primary long chain alcohol, ${\beta}-sitosterol$, campesterol, ursolic acid and ${\beta}-_D-glucosides$ of ${\beta}-sitosterol$ and campesterol. However, phloretin(dihydrochalcone) and its 5-O-glucoside(phloridzin) known as plant growth regulators in many Rosaceae plants were not found in this plant material by co-TLC analysis with authentic specimens. Although plant sex hormone, estrone, was often contained in relates of M. baccata, e.g., Prunus spp., Crataegus spp. and Malus spp., this compound was not detected in this fruit by comparison with an authentic material. By RIC chromatography, it was suggested that the Soxhlet extraction by the solvent of ether was excellently useful to extract ursolic acid efficiently.

Constituents of Spiraea prunifolia var. simpliciflora (조팝나무 뿌리의 성분 연구)

  • Spiraea prunifolia var. simpliciflora (Rosaceae) is a deciduous. latifoliate shrub growing in most parts of Korea. The roots of this plant have been used for malaria, as antipyretics and emetics. From the roots of this plant, sterol glycoside and two triterpenoids were isolated and the structures were elucidated by chemical and spectroscopic methods. They were identified as $3{\beta}-hydroxyurs-12-ene-28-oic$ acid (ursolic acid.1), $2{\alpha}$, $3{\beta}$, $19{\alpha}-trihy-droxyurs-12-ene-28-oic$ acid (tormentic acid.2) and ${\beta}-sitoste-rol-3-O-{\beta}-D-glucopyranoside$ (3).

Immunomodulatory Effects of β-sitosterol and Daucosterol Isolated from Dioscorea batatas on LPS-stimulated RAW 264.7 and TK-1 Cells (산약에서 분리한 β-sitosterol과 daucosterol의 RAW 264.7 세포와 TK-1 세포에서의 면역 활성 조절 효능)

  • Although many studies on immune modulatory materials have used RAW 264.7 cells, few have used T cell-derived TK-1 cell lines. Moreover, although some studies have investigated the efficacy of plant-derived β-sitosterol, few have examined the immunomodulatory activity of its analogue, daucosterol. In this study, β-sitosterol and daucosterol were isolated from D. batatas and identified by nuclear magnetic resonance spectroscopy. To evaluate the immune-enhancing or inhibitory effects of the isolated phytosterols, the expression levels of the inflammatory response genes COX-2, TNF-α, IL-6, and iNOS were analyzed by RT-PCR. The relative expression levels of TNF-α and iNOS in RAW 264.7 cells were increased more than threefold with β-sitosterol treatment comparing to those of untreated control. In the case of TK-1 cells, the expression level of TNF-α was decreased and the expression level of iNOS was increased in a β-sitosterol concentration-dependent manner. The expression levels of COX-2, TNF-α, and IL-6 increased by approximately 0.7-1.2 times in RAW 264.7 cells treated with daucosterol compared to those of untreated control, but iNOS expression decreased by 0.8-0.18 times. In the case of daucosterol-treated TK-1 cells, the expression levels of TNF-α, IL-6, and iNOS were markedly reduced from those of TK-1 cells treated only with lipopolysaccaride. As a conclusion, β-sitosterol treatment increased TNF-α and iNOS expression levels in RAW 264.7 cells, thus exerting an immune- boosting effect. However, in TK-1 cells, iNOS expression increased while TNF-α expression decreased, indicating an immunosuppressive activity of β-sitosterol. Daucosterol appears to exert an immunosuppressive effect in both macrophages and T cell lines by inhibiting iNOS expression in RAW 264.7 cells and greatly inhibiting the expression of TNF-α, IL-6, and iNOS in TK-1 cells.

  • One new sterol glucoside, $gomphsterol\;{\beta}-D-glucoside$ 1 along with known compounds, ${\beta}-sitosterol$, stigmasterol, campesterol, $stigmasterol-{\beta}-D-glucoside$, friedelin, 3-epi-friedelinol, allantoin, and $chrysoeriol-7-O-{\beta}-D-glucoside$ have been isolated from the aerial parts of Gomphrena globosa (Amaranthaceae). On the basis of spectroscopic (including 2D NMR) and chemical studies, the structure of 1 was elucidated as $(22E,24S)-24-ethylcholesta-7,9(11),22-trien-3{\beta}-ol-3-O-{\beta}-D-glucopyranoside$. Known compounds are reported for the first time from this plant species.

유기 및 관행재배 쌀의 생리활성 성분 분석법 확립 및 함량 비교

  • 관행재배 농산물에 대한 유기재배 농산물의 우수성이 구전되고 있으나 객관적인 과학적 데이터가 부족한 실정이다. 그래서 본 연구에서는 한국인이 주식으로 하고 있는 쌀(동진 1호)을 대상으로 생산이력이 분명한 유기 및 관행재배 시료를 이용하여 독자적으로 확립한 분석법을 이용해 유기 및 관행재배 쌀에 함유된 생리활성성분의 함량 비교를 행하였다. 생리활성성분 분석은 $\gamma$-oryzanol, $\beta$-sitosterol 및 tocopherol류를 대상으로 행하였다. 쌀에 함유된 주요 4종의 $\gamma$-oryzanol류의 동정을 위해 그 4종 화합물들을 ESI-MS 및 NMR 분석을 행한 결과, ODS 컬럼 상에서 cycloartenyl ferulate, 24-methylene cycloartenyl ferulate, campesteryl ferulate 및 $\beta$-sitosteryl ferulate 순으로 용출됨을 확인하였다. 동정된 각 성분들을 HPLC로 분석한 결과, cycloartenyl ferulate와 $\beta$-sitosteryl ferulate의 함량은관행보다 유기재배 쌀에서 유의(p<0.05)하게 높은 값을 보였다. 또한 $\gamma$-oryzanol 중 24-methylene cycloartenyl ferulate와 campesteryl ferulate의 함량은 유의차는 인정되지 않았으나 관행보다 유기재배 쌀에서 더 높은 경향을 보였다. 그리고 $\beta$-sitosterol 함량에 있어서는 유의차가 인정되지 않았으나 관행보다 유기재배 쌀에서 더 높은 경향을 나타냈다. 관행재배와 유기재배 쌀의 tocopherol류의 함량을 분석한 결과, $\alpha$-tocopherol의 함량에 있어서는 관행과 유기재배 쌀 간의 유의차가 관찰되지 않았고, $\beta$- and/or $\gamma$-tocopherols의 함량에 있어서는 유기재배 쌀에서 더 높은 경향이 관찰되었다. 이처럼 쌀에 함유된 생리활성 성분과 그로 인해 발휘되는 기능성 측면에 있어서 유기농산물의 우수성은 유기농산물 재배 농가의 수익 창출에 도움이 되는 홍보자료 제공 및 식품 기능학적 측면에 있어 중요한 기초자료로 활용될 수 있으리라 기대된다. 그러나 본 결과는 본 실험에서 대상으로 한 시료에 한정될 수 있으며, 유기재배 방법과 재배환경 및 조건에 따라 다양한 결과가 얻어질 수 있음에 주의가 요구된다.

Chlorosmaridione; A Novel Chlorinated Diterpene Quinone Methide from Rosemarinus officinalis L.

  • A novel chlorinated diterpene quinone methide; chlorosmaridione (7-chloro-11-hydroxy-abeita-7,9(11),13-triene-6,12-diode) was isolated from petroleum ether extract of the stems of Rosemarinus officinalis L. growing in Egypt. In addition, ${\beta}-sitosterol$, stigmasterol, lupeol acetate, ${\alpha}-amyrin,\;{\beta}-amyrin$, lupeol, acetyloleanolic acid, acetylursolic acid taxodione, horminone, and cryptotanshinone were also identified. Chemical structures of the isolated compounds have been elucidated on the bases of physical, chemical, and spectral data including IR, UV, MS, 1D-and 2D-NMR spectra.

  • A new daucosteryl derivative, verticilloside (1), was isolated from the seeds of Malva verticillata L. (Malvaceae). The structure was determined to be 3-O-[${\beta}$-D-(6'-linoleoyl)glucopyranosyl]-${\beta}$-sitosterol based on spectroscopic analyses ($^1H$ and $^{13}C$-NMR, DEPT, COSY, HMQC, and HMBC) and chemical reactions.

  • From the bulbs of Erythronium japonicum Decaisne(Liliaceae), ${\beta}-sitosterol$ and its $3-O-{\beta}-D-glucoside$ together with fatty acids were isolated. All compounds were identified on the basis of spectral data. Campesterol and stigmasterol were also identified as minor components. Paimitic acid was identified as a major component and stearic, oleic, arachidic, behenic, tricosanoic and lignoceric acids were also characterized as minor ones.
