Purification of Cellulase Produced from Cellulomonas sp. YE-5 (Cellulomonas sp. YE-5가 생산하는 Cellulase의 정제)

  • An extracellular cellulase producing bacterium YE-5 was isolated from soil, and identified as a Cellulomonas sp. by its taxonomical characteristics. The maximal activities of avicelase (0.35 units/ml), CMCase (3.18 units/ml), FPase (0.315 units/ml) and $\beta$-glucosidase (0.882 units/ml) were obtained when this strain was cultured for 48 hrs at $30^{\circ}C$ in a medium containing 0.8% (w/v) Solka floc, 0.06010 (wlv) urea, 0.1% (w/v) $K_2HP0_4$, 0.1% (w/v) $MgS0_4.7H2_0$, 0.2% (w/v) bacto peptone, 0.2% (w/v) yeast extract and pH 6.5. The cellulase was purified by ammonium sulfate fractionation, DEAE-Sepharose column chromatography and Sephadex 6-100 column chromatography from culture filtrate of Cellulomonus sp. YE-5. The molecular weights of purified avieelase, CMCase I, and CMCase II were estimated to be about 95,000 ~ 105,000, 46,000 ~ 47,000 and 120,000 ~ 125,000, respectively.

Formation and Fusion of Protoplasts from the Cellulolytic Fungi, Aspergillus niger MAN-831 and Aspergillus wentii MAW-538 (Cellulase를 생산하는 Aspergillus niger MAN-831과 Aspergillus wentii MAW-538의 원형질체 형성 및 융합)

  • For the effective utilization of cellulosic biomass, conidial protoplast fusion between Aspergillus niger MAN-831(${\beta}-glucosidase$) and A. wentii MAW-538(CMCase and avicelase), which produced potently cellulolytic enzymes was carried out. Optimal conditions for formation and regeneration of protoplast were conidiospore age-5 dyuas. $2-DG-30\mu\textrm{g}/ml$, preincubation time-4 hours, osmotic stabilizer-0.7M KCl, novozyme(7mg/ml)+driselase(2.5mg/ml) and reaction time of enzyme-5 hours. Optimal conditions for protoplast fusion were obtained by treatment of protoplasts with 15mM CaCl2 and 25% polyethylene glycol 4000(pH 6~7) as fusogenic agent at $36^{\circ}C$ for 25~30 minutes. The frequency was then $7.94{\times}10^{-4}$. CMCase, avicelase and ${\beta}-glucosidase$ activity of fusant F-208 strain was 1.5, 1.3, 1.2 times higher than those of parental strains, respectively.

  • To study distribution of bacterial flora and their biochemical characteristics in the Antarctic soilecosystem, these experiments were performed during the austral summer(Feb., 1989) on the King George Island, Antarctica. The numbers of heterotrophic bacterial colonies and extracellular enzyme actibities were estimated from the Antarctic terrestrial soils which were sampled from 17 different locations near Sejong station (Korea) and Teniente Jubany station (Argentina) on the King George Island. The numbers of heterotrophic bacterial colonies were extremely variable with sampling sites and incubation temperatures. Arithmetric average numbers were $2.5\times 10^{4}$, $2.7\times 10^{7}$ , $6.9\times 10^{5}$ CFU/$cm^{3}$ soil at the incubation temperature of $37^{\circ}C$, $25^{\circ}C$ and $4^{\circ}C$, respectively. The activities of extracellular $\alpha$-glucosidase, $\beta$-glucosidase and N-acetyl-$\beta$-glucosaminidase were shown as similar mean percentage in the colonies obtained at different temperatures. Mean value of protease activities, however, was remarkably higher (92%) in the colonies grown at $4^{\circ}C$,.

  • Enzymatic cellulose hydrolysis depends on the several factors such as the structural features (CrI, particle size and surface area, etc.), the nature of cellulase enzyme system, the inhibitory effects of products, and enzyme deactivation. At the presence of products on the initial hydro- lysis rate of cellulose, cellobiose has more severe inhibitory effect than glucose. Othewise, the inhibition effect of products for adsorbed enzyme is related to the glucose and cellobiose conentration hyperbolically. Enzyme deactivation of FPA and ${\beta}-glucosidase$ were expressed by exponential decay profile.

Analysis of gut bacterial diversity and exploration of cellulose-degrading bacteria in xylophagous insects (목질섭식곤충의 장내 세균 다양성 분석 및 섬유소 분해균 탐색)

  • In this study, gut bacterial communities in xylophagous insects were analyzed using the pyrosequencing of 16S rRNA genes for their potential biotechnological applications in lignocelluloses degradation. The result showed that operational taxonomic units (OTUs), species richness and diversity index were higher in the hindgut than in the midgut of all insect samples analyzed. The dominant phyla or classes were Firmicutes (54.0%), Bacteroidetes (14.5%), ${\gamma}-Proteobacteria$ (12.3%) in all xylophagous insects except for Rhinotermitidae. The principal coordinates analysis (PCoA) showed that the bacterial community structure mostly clustered according to phylogeny of hosts rather than their habitats. In our study, the two carboxymethyl cellulose (CMC)-degrading isolates which showed the highest enzyme activity were most closely related to Bacillus toyonensis $BCT-7112^T$ and Lactococcus lactis subsp. hordniae $NCDO\;2181^T$, respectively. Cellulolytic enzyme activity analysis showed that ${\beta}-1,4-glucosidase$, ${\beta}-1,4-endoglucanase$ and ${\beta}-1,4-xylanase$ were higher in the hindgut of Cerambycidae. The results demonstrate that xylophagous insect guts harbor diverse gut bacteria, including valuable cellulolytic bacteria, which could be used for various biotechnological applications.

Antioxidant and Antiobesity Activity of Natural Color Resources (천연색소 소재의 항산화 및 항비만 활성)

  • This study investigated the antioxidant and antiobesity activity of extract powders from the following natural color resources: Polygonum indigo, Black locust, Cochineal, Catechu, Grape, Tesu flower, Henna, Chrysanthemum, Sandalwood Red, Himalayan Rhubarb, and Madder. Total phenol content was the highest in Catechu extract, at 348.25 mg/g. DPPH, ABTS radical scavenging activity and ferric reducing antioxidant power (FRAP) were also higher in Catechu extract. Bleaching inhibition activity in the ${\beta}$-carotene linoleic acid system was the highest in Black locust extract, as was ${\alpha}$-Glucosidase inhibition activity. ${\alpha}$-Amylase inhibition activity was the highest in Catechu extract. Trypsin inhibition activity of Black locust extract was greater than 60%, and ${\alpha}$- chymotrypsin inhibition activity of Catechu extract was greater than 40%. Lipase inhibition activity was the highest Black locust extract, at 52.73%. Viability of 3T3-L1 cells was not affected by treatment with extracts at concentrations of $1.25{\sim}25{\mu}g/ml$. Lipid accumulation in the 3T3-L1 cells was the lowest following treatment with Catechu extract, at 55.8%, and this extract also inhibited adipocyte differentiation. These results suggest that the Catechu and Black locust extracts have high antioxidant and antiobesity activities and can be useful ingredients in functional foods.

Effect of Temperature on Soil Microbial Biomass, Enzyme Activities, and PLFA content during Incubation Period of Soil Treated with Organic Materials (유기물원 항온배양 온도가 토양미생물체량과 효소활성 및 PLFA함량에 미치는 영향)

  • This study was carried out to evaluate the effect of temperature on soil microbial biomass, enzyme activities, and PLFA content in the volcanic(VAS) and the non-volcanic ash soil(NVAS). The soils were treated with organic materials such as organic fertilizer pelleted(OFPL), organic fertilizer powdered(OFPD), pig manure compost(PMC), and food waste compost(FWC). Two grams of organic materials were well mixed with 30g of dried volcanic and non-volcanic ash soil(< 2 mm) with 50% of soil moisture content. And the soils were incubated at 10, 20, $30^{\circ}C$ in incubator. Soils were analysed on the incubation times as followed; soil pH, total nitrogen, organic matter(at 75, 150, 270 days), microbial biomass C and PLFA (at 75, 270 days), microbial biomass N and soil enzyme(at 150, 270 days). pH values of soils treated with PMC and FWC had no changes on soil type, and incubation temperature. However, the pH was increased with temperature in the soils treated with OFPL. The changes in NVAS was higher than in VAS. Soil microbial biomass C content were high in the condition of high temperature and organic fertilizers treatment in VAS. But the contents were gradually decreased with incubation period in both NVAS and VAS. Soil microbial biomass N was high in NVAS treated with organic fertilizers and in VBS treated with PMC and FWC. PLFA content was higher in NVBS than in VBS at 75 days but showed high in VBS at 270 days. Urease activity of NVBS treated with OFPL showed $10^{\circ}C$ (75.0)> $20^{\circ}C$ (16.3)>$30^{\circ}C$ ($4.6ug\;NH{_4-}N\;g^{-1}\;2h^{-1}$) at 150 days. It were decreased gradually high temperature and time passes. And it showed high at $10^{\circ}C$ in VBS. Glucosidase activity was higher in NVBS than in VBS. Correlation coefficient of between soil microbial biomass C and microbial activity indicators showed that PLFA was high significantly at $r^2=0.91$ in NVBS and ${\beta}-glucosidase$ was $r^2=0.83$ in VBS. Soil microbial activities showed differences in the relative sensitivities of soil type and soil temperature.

Fed-Batch Simultaneous Saccharification and Fermentation of Waste Paper to Ethanol (폐지의 유가식 동시당화발효에 의한 에탄올 생산)

  • The fed-bach simultaneous saccharification and fermentation (SSF) of newspaper to ethanol with Brettanomyces custersii was studied. The initial substrate concentration for the effective fed-batch SSF was 8% (w/v). The initial optimum enzyme concentration was 30 FPU/g cellulose for cellulase and the optimum volumetric ratio of $\beta$-glucosidase to cellulase was 0.1. When 4% (w/v) of ball-milled newspaper was supplemented intermittently at time intervals, considering the mixing of newspaper slurry, the fed-batch SSF showed higher ethanol concentration (26.80 g/L) and two times higher ethanol production yield based on enzyme than the batch SSF.

  • Isomaltooligosaccharides in Sikhye were digested with enzyme (30unit/ml) of $\alpha$-amylase, $\alpha$-glucosidase and glucoamylase from Aspergillus awamori, sweet potato $\beta$-amylase and human salivary $\alpha$-amylase at 37$^{\circ}C$ for 1 hour, respectively. These amylases acted on these saccharides to give hydrolysis products with less than 20% of degree of hydrolysis, except the case of glucoamylase with 62% of high degree of hydrolysis. $\alpha$-Glucosidase plus human salivary $\alpha$-amylase hydrolyzed it to attain the hydrolysis value up to 25%, but further increment of hydrolysis was not observed. Rice residue in Sikhye has similar sugar composition and structure, judging from sugar analyses by the enzymatic hydrolysis. These results suggest that isomaltooligosaccharides and rice residue in Sikhye can be a growth factor for Bifidobacterium and dietary fiber which is useful for human health.

Enzyme Activities and Cellulose Degradation of Domestic Softwoods in Shaking Culture of Fomitopsis palustris (국내산 침엽수 목분의 진탕배양에서 나타난 Fomitopsis palustris의 효소 활성 및 셀룰로오스 분해)

  • Activities of the extracellular enzyme from Fomitopsis palustris, a brown-rot fungi, and by which crystallinity changes of cellulose in the various softwoods, such as Larix leptolepsis, Finns rigida, Finns koraiensis and Finns densiflora by liquid culture, were investigated. Activity of Cellobiohydrolase (CBH) from F. palustris was detected in the every test softwoods culture, showing activities of the Endoglucanase (EG), $\beta$-glucosidase (BGL) and $\beta$-1,4-xylosidase (BXL). It was shown high enzyme activities in the sapwood culture than heartwood of the same wood species, However, the enzyme activities in most of test wood cultures increased with longer incubation time, indicating a possibility of intermix sapwood and heartwood for degradation process by enzyme. Also it was shown that protein patterns of the extracellular enzyme from F. palustris in wood particle substrate of the several domestic softwoods were similar with each other wood species, which suggested the possibility of mixing all softwoods in saccharification by enzyme from F. palustris. Crystallinity reduction value of cellulose by F. palustris was 4.2~20.4% in 4 weeks cultivation, 12.9~28.9% in 8 weeks.
