Induction of Melibiase in Yeast

  • Exposing yeast cells with a certain genotype to different inducers, the ability of the yeast cells (Saccharomyces cerevisiae) to obtain enhanced fermentation for carbohydrates was observed. Regardless of the preexposure to any substrate, the inherent character incapable of fermenting a certain carbohydrate was maintained, while utilization of carbohydrates by the cells with a certain gene markers was varied by the previous conditions where they were exposed. Galactose was the best inducer for the cells to elaborate melibiase, even the galactose was not utilized as a substrate. Preexposure to galactose seemed to be necessary for the cells to utilize galactose and melibiose. Galactose fermentation by GA cells was enhanced by the exposure of the cells to galactose, but not to melibiose, raffinose, sucrose or glucose. Delayed fermentation of sucrose by the cells exposed to glucose or melibiose, but not to galactose, was observed. Raffinose fermentation was obtained by the cells with either SU RAF or GA ME genes, but the enhanced fermentation of raffinose seemed to be dependent on which inducer the cells were exposed previously and enzymes induced by the inducer to break either one of the linkages of raffinose molecule, the alpha0galactosidic or the beta-fructo-furanosidic.

  • From the EtOAc fraction of the MeOH extract of Albizzia julibrissin (Leguminosae), a rare 5-deoxyflavone (geraldone, 1), isookanin (2), luteolin (3), an isoflavone (daidzein, 4), five prenylated flavonoids [soph6f1avescenol (5), kurarinone (6), kurarinol (7), kuraridin (8) and kuraridinol (9)], a cerebroside (soya-cerebroside I, 10), and $(-)-syringaresinol-4-O-{\beta}-D-glucopyranoside$ (11) were isolated and characterized on the basis of spectral data. Compounds 2, 3, and 11, showed 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity.

Anti-hyperlipidemic Effect of Phragmites communis and Its Active Principles (갈대의 고지혈증 개선효과와 그 활성성분)

  • Administration of an MeOH extract of Pharagmites communis on hyperlipidemic rats produced a significant decrease in blood triglyceride. The fatty acid mixtures, $\beta$ -sitosterol and its glucoside, p-coumaric acid, vanilic acid, ferulic acid, p-hydroxybenzaldehyde and sucrose were isolated. Sterol and p-coumaric acid were shown to be effective in reducing the elevated level of triglyceride. The wet weight and the level of triglyceride in epididymal adipose tissue increase following administration of the methanol extract in rats fed the purified diets at the 1% level for 4 weeks. This results suggest that the methanol extract may accelerate the accumulation of lipid in adipose tissue.

  • From the MeOH extract of the aerial part of Gastrodia elata Blume (GEB) (Family: Orchidaceae), eight compounds have been isolated on repeated column chromatography, and their structures were elucidated as dotriacontanoic acid (1), beta-sitosterol (2), 4-hydroxybenzaldehyde (3), docosanoic acid oxiranylmethyl ester (4), hentriacotanoic acid (5), octadecanoic acid (6), benzoic acid (7) and gastrodin (8) on the basis of their spectral evidences including EI-Mass and 2D-NMR spectrum. All of them were obtained from the aerial part of Gastrodia elata Blume for the first time, in which compound 4 is a novelty to our best knowledge. It is also known that a phenolic glucoside, gastrodin is a major constituent just like Gastrodia rhizome.

감압증류법에 의한 $^{32}$P 제조

  • $^{32}$P는 순수한 $\beta$$^{-}$ 방출핵종(방출에너지 = 1.71 MeV, 반감기 = 14.3일)이며 의료용, 표지화합물 합성용, 유전공학 실험용 등으로 널리 사용되므로 고품질의 $^{32}$P의 수요에 부응하기 위해 감압증류법을 개발하였는 바 그 방법과 결과는 다음과 같다. 연구로 2호에서 중성자 조사된 황 표적을 감압 증류용기내에서 5~10 mmHg의 감압하에 200~30$0^{\circ}C$로 가열하여 황을 증류해 낸 다음 묽은 염산을 역류시켜 넣고 $^{32}$P 를 울궈냈다. 이 용액을 이온교환 수지로 정제하여 약 60 mCi/batch의 정제 $^{32}$P를 얻었다. 이온교환수지에 흡착되는 $^{32}$P의 방사능은 전체의 3% 미만이었고 여기에 흡착되는 불순 핵종은 $^{131}$ Ba, $^{85}$ Sr, $^{59}$ Fe, $^{65}$ Zn, $^{60}$Co이었다. 이 방법으로 얻은 $^{32}$P 최종제품은 핵종순도 >99%, 방사화학적 순도 >98%, 고형성분 함량 <1.2 mg/mL 이어서 그 품질이 우수함을 알 수 있었다.

  • Dicyclohexyltin(IV) complexes $Cy_2SnX (X = O,\;S,\;(Me_2NCS_2)_2)$ have been prepared and characterized by means of elemental analysis, cryoscopic data, and IR spectroscopy. X-ray structure determination for $Cy_2Sn(S_2CNMe_2)_2$ (monoclinic; P2/c;a = 8.992(1), b = 6.688(1), c = 19.453(2) ${\AA},\;{\beta}$ = 96.556(7) ; R = 0.041) has shown that the molecule is $C_2$-symmetric with both dithiocarbamate ligands bonding in anisobidentate mode and the local geometry around the hexacoordinate Sn(IV) atom is a distorted octahedral arrangement with the two cyclohexyl groups in trans position. The physical and chemical properties measured suggest polymeric and trimeric structures for $Cy_2SnO\;and\;Cy_2SnS$, respectively.

Biodegradation of Kraft Lignins by White-Rot Fungi(I) -Lignin from Pitch Pine- (백색부후균에 의한 크라프트 리그닌의 분해(I) -리기다소나무 리그닌-)

  • This study was carried out to investigate the structural characteristics of kraft lignin and the wood degrading characteristics, the productivity of ligninolytic enzymes and the enzymatic degradation of kraft lignin by white-rot fungi. To purify kraft lignin, precipitation of kraft pulping black liquors of pitch pine meal was done by titration with lN $H_{2}SO_{4}$ reaching to pH 2, and isolation of the precipitates done by centrifugation. The isolated precipitates from pitch pine were redissloved in lN NaOH, reprecipitated by titration with lN $H_{2}SO_{4}$, washed with deionized water, and kept ofr analysis after freeze drying. Fractionation of the precipitates in solution by successive extraction with $CH_{2}Cl_{2}$ and MeOH, and the fractionates were named SwKL, SwKL I, SwKL II, and SwKL III for pitch pine kraft lignin. The more molecular weights of kraft lignin increased, the less phenolic hydroxyl groups and the more aliphatic hydroxyl groups. Because as the molecular weights increased, the ratio of etherified guaiayl/syringyl(G/S ratio) and the percentage were increased. The spectra obtained by 13C NMR and FTIR assigned by comparing the chemical shifts of various signals with shifts of signals from autherized ones reported. The optimal growth temperature and pH of white-rot fungi in medium were $28^{\circ}C$ and 4.5-5.0, respectively. Especially, in temperature and pH range, and mycelial growth, the best white-rot fungus selected was Phanerochaete chrysosporium for biodegradation. For the degradation pathways, the ligninolytic fungus jcultivated with stationary culture using medium of 1% kraft lignin as a substrate for 3 weeks at $28^{\circ}C$. The weight loss of pitch pine kraft lignin was 15.8%. The degraded products extracted successively methoanol, 90% dioxane and diethyl ether. The ether solubles were analyzed by HPLC. Kraft lignin degradation was initiated in $\beta$-O-4 bonds of lignin by the laccase from Phanerochaete chrysosporium and the degraded compounds were produced from the cleavage of $C\alpha$-$C\beta$ linkages at the side chains by oxidation process. After $C\alpha$-$C\beta$ cleavage, $C\alpha$-Carbon was oxidized and changed into aldehyde and acidic compounds such as syringic acid, syringic aldehyde and vanilline. And the other compound as quinonemethide, coumarin, was analyzed. The structural characteristics of kraft lignin were composed of guaiacyl group substituted functional OHs, methoxyl, and carbonyl at C-3, -4, and -5 and these groups were combinated with $\alpha$ aryl ether, $\beta$ aryl ether and biphenyl. Kraft lignin degradation pathways by Phanerochaete chrysosporium were initially accomplished cleavage of $C\alpha$-$C\beta$ linkages and $C\alpha$ oxidation at the propyl side chains and finally cleavage of aromatic ring and oxidation of OHs.

Excessive Levels of Dietary Protein and Energy Induce Lack of Growth Promoting Effects of Clenbuterol in Broilers

  • The present study examined the effects of excessive dietary protein and energy on growth response to clenbuterol in broilers. The chicks were allocated into 6 groups at 14d old, and used for a $3{\times}2$ factorial experiment. Birds were fed six diets, the control diet containing 21% crude protein (CP) and 3,100 kcal of metabolizable energy ME/kg, a high protein (30% CP) or a high energy (3,500 kcal/ ME/kg) diet, with or without 1 ppm clenbuterol, for 18 d. Clenbuterol feeding markedly decreased (p < 0.05) body weight gain by 23% in the high energy group. Feed intake was also decreased (p < 0.05) by clenbuterol administration across diet treatments. Abdominal fat weight was reduced (p < 0.05) by clenbuterol only when chickens were fed the high energy diet. Clenbuterol increased (p < 0.05) leg muscle weight in the control diet group, but decreased (p < 0.05) it in the high energy group. Muscle protein concentration was increased by 11 % in leg muscle only of the birds at the high energy level. In leg muscle, clenbuterol enhanced the protein/DNA ratio by 18%, except for the high protein group. These results indicate that feeding a diet containing excessive amounts of protein and more energy than normal did not necessarily improve growth response to clenbuterol.

Differential expression and in situ localization of a pepper defensin (CADEFl) gene in response to pathogen infection, abiotic elicitors and environmental stresses in Capsium annuum

  • Pepper defensin ( CADEFl) clone was isolated from cDNA library constructed from pepper leaves infected with avirulent strain Bv5-4a of Xanthomonu campestris pv. vesicatoria. The deduced amino acid sequence of CADEFl is 82-64% identical to that of other plant defensins. Putative protein encoded by CADEFl gene consists of 78 amino acids and 8 conserved cysteine residues to form four structure-stabilizing disulfide bridges. Transcription of the CADEF1 gene was earlier and stronger induced by X campestris pv. vesicatoria infection in the incompatible than in the compatible interaction. CADEF1 mRNA was constitutively expressed in stem, root and green fruit of pepper. Transcripts of CADEFl gene drastically accumulated in pepper leaf tissues treated With Salicylic acid (SA), methyl jasmonate (MeJA), abscisic acid (ABA), hydrogen Peroxide (H$_2$O$_2$), benzothiadiazole (BTH) and DL-${\beta}$-amino-n-butyric acid (BABA). In situ hybridization results revealed that CADEF1 mRNA was localized in the phloem areas of vascular bundles in leaf tissues treated with exogenous SA, MeJA and ABA. Strong accumulation of CADEF1 mRNA occurred in pepper leaves in response to wounding, high salinity and drought stress. These results suggest that bacterial pathogen infection, abiotic elicitors and some environmental stresses may play a significant role in signal transduction pathway for CADEF1 gene expression.
