Objective: Progesterone is used to prevent recurrent preterm delivery, however the molecular mechanisms of its effect are incompletely understood. The objective of this study was to determine the effect of progesterone on tumor necrosis factor $(TNF)-{\alpha}$-induced matrix metalloproteinase (MMP)-9 activity in human choriodecidual (CD) membranes. Methods: We collected CD membranes from women with uncomplicated term pregnancies who were scheduled for elective cesarean delivery (n=10). CD membranes ($1{\times}1cm$) were incubated in tissue culture media at $37^{\circ}C$. We pre-treated the CD membranes with progesterone (P4), $17{\alpha}$-hydroxyprogesterone caproate (17P), promegestone (R5020), or vehicle (ethanol) for 24 hours. The CD membranes were subsequently treated with $TNF-{\alpha}$ (with continued progesterone treatment) for 48 hours, then media was harvested for measuring MMP-9 activity by zymography and total protein was isolated from CD membrane tissues for MMP-9 expression by western blot analysis. Results: P4, 17P, and R5020 significantly reduced $TNF-{\alpha}$-induced MMP-9 activity in fetal membrane tissue samples (P=0.0078, P=0.0156, and P=0.0391, respectively) by zymography. Western blot analysis also showed decreased expression of MMP-9 in progesterone pretreated groups (P=0.0313). Conclusion: Progesterone reduces $TNF-{\alpha}$-induced MMP-9 activity in human CD membranes. These findings may provide further support for the role of progesterone in preventing preterm birth.
Background: This study investigated the anti-inflammatory effects of bee venom (BV) through the inhibition of nuclear factor kappa beta ($NF-{\kappa}B$) expression in macrophages and keratinocytes. Methods: Cell viability assays were performed to investigate the cytotoxicity of BV in activated macrophages [lipopolysaccharide (LPS)] and keratinocytes [interferon-gamma/tumor necrosis factor-alpha ($IFN-{\gamma}/TNF-{\alpha}$)]. A luciferase assay was performed to investigate the cellular expression of $NF-{\kappa}B$ in relation to BV dose. The expression of $NF-{\kappa}B$ inhibitors ($p-I{\kappa}B{\alpha}$, $I{\kappa}B{\alpha}$, and p50 and p65) were determined by Western Blot analysis, and the electromobility shift assay. A nitrite quantification assay was performed to investigate the effect of BV, and $NF-{\kappa}B$ inhibitor on nitric oxide (NO) production in macrophages. In addition, Western Blot analysis was performed to investigate the effect of BV on the expression of mitogen-activated protein kinases (MAPK) in activated macrophages and keratinocytes. Results: BV was not cytotoxic to activated macrophages and keratinocytes. Transcriptional activity of $NF-{\kappa}B$, and p50, p65, and $p-I{\kappa}B{\alpha}$ expression was reduced by treatment with BV in activated macrophages and keratinocytes. Treatment with BV and an $NF-{\kappa}B$ inhibitor, reduced the production of NO by activated macrophages, and also reduced $NF-{\kappa}B$ transcriptional activity in activated keratinocytes (compared with either BV, or $NF-{\kappa}B$ inhibitor treatment). Furthermore, BV decreased p38, p-p38, JNK, and p-JNK expression in LPS-activated macrophages and $IFN-{\gamma}/TNF-{\alpha}$-activated keratinocytes. Conclusion: BV blocked the signaling pathway of $NF-{\kappa}B$, which plays an important role in the inflammatory response in macrophages and keratinocytes. These findings provided the possibility of BV in the treatment of atopic dermatitis.
Intercellular adhesion molecule-1 (ICAM-1), which is induced by tumor necrosis factor (TNF)-${\alpha}$, contributes to the entry of immune cells into the site of inflammation in the skin. Here, we show that protein tyrosine phosphatase non-receptor type 21 (PTPN21) negatively regulates ICAM-1 expression in human keratinocytes. PTPN21 expression was transiently induced after stimulation with TNF-${\alpha}$. When overexpressed, PTPN21 inhibited the expression of ICAM-1 in HaCaT cells but PTPN21 C1108S, a phosphatase activity-inactive mutant, failed to inhibit ICAM-1 expression. Nuclear factor-${\kappa}B$ (NF-${\kappa}B$), a key transcription factor of ICAM-1 gene expression, was inhibited by PTPN21, but not by PTPN21 C1108S. PTPN21 directly dephosphorylated phospho-inhibitor of ${\kappa}B$ ($I{\kappa}B$)-kinase ${\beta}$ ($IKK{\beta}$) at Ser177/181. This dephosphorylation led to the stabilization of $I{\kappa}B{\alpha}$ and inhibition of NF-${\kappa}B$ activity. Taken together, our results suggest that PTPN21 could be a valuable molecular target for regulation of inflammation in the skin by dephosphorylating p-$IKK{\beta}$ and inhibiting NF-${\kappa}B$ signaling.

Foreign students in South Korea face important challenges when they try to maintain their health. As a measure of their motivation to actively build skills for overcoming those challenges, we evaluated the 10-item Consumer Health Activation Index (CHAI), testing its feasibility, dimensionality, and reliability. There were no missing data, there was no floor effect, and for the total scores the ceiling effect was trivial (< 2%). Results of the Kaiser-Meyer-Olkin test and Bartlett's test of sphericity indicated that the data were suitable for the detection of structure by factor analysis. The results of parallel analysis and the shape of the scree plot supported a two-factor solution. One factor had 3 items concerning "my doctor" and the other factor had the 7 remaining items. Reliability was high for the 10-item CHAI (alpha = 0.856), for the 3-item subscale (alpha = 0.838), and for the 7-item subscale (alpha = 0.857). Reliability could not be improved by deletion of any items. Use of the CHAI to gather data from these foreign students is feasible, and reliable results can be obtained whether one uses the total score from all 10 items or scores from the proposed 7-item and 3-item subscales.
This project was conducted using a survey method and through interviewing with four persons aged 60 years and over. An interview with an open-ended questionnaire was also used for elderly persons, families, oriental medicine doctor and nurses. topic-related literature review was also done. In total, 382 statements were derived. For content validity, nursing professionals were involved in this study, From that 48 items are developed. The subjects were 340 elderly persons over 60 years old. Data were collected duicing February and March 1997 and analyzed using the SPSS package The result are as follows. 1. Items with low Cronbach Coefficient alpha which means low correlation with total items were removed. 2. Factor analysis was done in order to confirm construct validity and eight factors were obtained from the results. The first factor, 'positive cognition of the aging process', the second factor, 'relationship network' the third factor, 'maintanence of physical functioning, the fourth factor', 'maintanence of peaceful mind' the fifth factor, 'keeping up with daily tasks' the sixth factor, 'continuous adequate body movement' the seventh factor,'involvement of religion in the elderly person's life and the eight factor', 'appropriate resting'. Cronbach Coefficient alpha for the 33 items was .9127 Based on the result, the following is suggested 1. It is anrticipated that the fundamental health of elderly person could be promoted by assessing healthy behaviors of elderly person with this assessment tool. 2. Further studies could be derived from this research. 3. Validity of this assessment tool should be further tested with and a larger sample of elderly person including in-patient elderly persons as well as nursing home residents.
${\alpha}2-macroglobulin$$({\alpha}2-M)$ has been shown to have a variety of activities. One of those activities is the suppression of immune response. Characterization of the immunosuppressive factor secreted by a T cell hybridoma showed that ${\alpha}2-M$ was produced and secreted from the T cell hybridoma. ${\alpha}2-M$ was produced abundantly from the T cell hybridoma when cultured as ascites. The isolation and identification of the ${\alpha}2-M$ were studied using affinity chromatography and N-terminal amino acid sequencing. The extended observations were that the ${\alpha}2-M$ produced by the T cell hybridoma suppresses mixed lymphocyte reaction.
Alpha toxin of S aureus has cytolytic activity respectively. This antigen has been received the most attention since it is a major virulence factor in pathogenesis of staphylococcal mastitis. Thus, alpha toxin has been focused as potential candidate of vaccine to minimize mastitis in cows. The purpose of this study was to develop a simple, efficient production and purification methods of sufficient amount of alpha toxin antigen from S aureus. Alpha toxin production measured by hemolytic activity was the highest at 18 hrs postinoculation in yeast extract culture medium supplemented with thiamine, nicotinic acid and casamino acid. Alpha toxin was purified by ammonium sulfate precipitation (65%) and ultrafiltration. Molecular weight of the toxin was 33 kDa in the analysis with SDS-PAGE. Conclusionally, when alpha toxin was included in the vaccine, the optimal harvest time of alpha toxin was at 18 hrs after inoculation in yeast extract medium supplemented with thiamine and nicotinic acid.
When the reaction rate constant k of phospholipase D on liposome was measured by the ANS fluorometry, k of phospholipase D on DMPC liposome which was made of L-$\alpha$-PI, cholesterol and L-$\alpha$-PS decreased than that of phospholipase D on DMPC liposome with cholesterol or with PI and cholesterol. Optimal $Ca^{2+}$ concentration, the most important factor on effect of phospholipase D, also decreased to 1mM, as compared with 10mM and 60mM respectively when cholesterol and PI were added, and cholesterol only was added. The change of cholesterol Mol% had a great influence on k value of phospholipase D. But in case of addition of L-$\alpha$-PS to cholesterol, the influence was relatively diminished.
In this study, we evaluated the role of apurinic/apyrimidinic endonuclease1/redox factor-1 (Ref-1) on the tumor necrosis factor-$\alpha$ (TNF-$\alpha$) induced cyclooxygenase-2 (COX-2) expression using A549 lung adenocarcinoma cells. TNF-$\alpha$ induced the expression of COX-2 in A549 cells, but did not induce BEAS-2B expression. The expression of COX-2 in A549 cells was TNF-$\alpha$ dose-dependent (5~100 ng/ml). TNF-$\alpha$-stimulated A549 cells evidenced increased Ref-1 expression in a dose-dependent manner. The adenoviral transfection of cells with AdRef-1 inhibited TNF-$\alpha$-induced COX-2 expression relative to that seen in the control cells ($Ad{\beta}gal$). Pretreatment with $10\;{\mu}M$ of SB203580 suppressed TNF-$\alpha$-induced COX-2 expression, thereby suggesting that p38 MAPK might be involved in COX-2 expression in A549 cells. The phosphorylation of p38 MAPK was increased significantly after 5 minutes of treatment with TNF-$\alpha$, reaching a maximum level at 10 min which persisted for up to 60 min. However, p38MAPK phosphorylation was markedly suppressed in the Ref-1-overexpressed A549 cells. Taken together, our results appear to indicate that Ref-1 negatively regulates COX-2 expression in response to cytokine stimulation via the inhibition of p38 MAPK phosphorylation. In the lung cancer cell lines, Ref-1 may be involved as an important negative regulator of inflammatory gene expression.
델피니딘은 양전하를 뛰는 diphenylpropane의 polyphenolic ring 구조를 가진 주요한 안토시아닌 색소 중에 하나이다. 최근 연구에서 델피니딘은 항산화, 항염증 뿐만 아니라 항암 효능을 가진다고 보고되었다. 본 연구에서는 전립샘 암에서 종양의 성장과 신생혈관생성에 관련된 중요한 인자인 VEGF 발현에 대한 델피니딘의 억제 효과를 조사하였다. RT-PCR을 통해 델피니딘을 처리한 PC3M 전립샘 암세포 세포에서 EGF로 유도한 VEGF mRNA 발현 수준이 감소됨을 확인하였다. 또한 델피니딘은 VEGF의 전사인자인 HIF-$1{\alpha}$와 STAT3가 세포 핵으로 전위되는 것을 효과적으로 억제하였다. 한편 luciferase assay을 통해 HRE-promoter 활성을 확인해 본 결과, 델피니딘이 HIF-$1{\alpha}$의 전사 활성을 억제시켜 VEGF 발현을 감소시키는 것을 알 수 있었다. 그리고 델피니딘은 EGFR의 발현에는 영향을 미치지 않고, Akt, p70S6K, 4EBP1의 인산화를 특이적으로 억제하는 것으로 나타났다. 결론적으로 델피니딘이 HIF-$1{\alpha}$와 STAT3, VEGF 발현을 억제를 통하여 암세포 증식억제와 신생혈관형성을 억제하는 역할을 새롭게 확인하였다.
