Effects of Gamma-Tocopherol (GT) Supplementation on Vitamin E Concentration in Cigarette Smoke (CS) Exposed Mice (고농도 감마 토코페롤 보충식이가 흡연에 노출된 쥐의 혈액 및 조직 비타민 E와 대사산물 농도에 미치는 영향)

  • Cigarette smoke (CS) induces oxidative and nitrosative stress to the respiratory tract (RT) via both oxidants contained in CS and by CS-induced activation of RT inflammatory-immune pro-oxidant processes. CS exposure has been associated with reduced levels of plasma micronutrient antioxidants, in part due to an increased utilization and turnover of alpha-tocopherol (AT). It has been suggested that gamma-tocopherol (GT) may have an expanded spectrum of antioxidant activation compared to alpha-tocopherol (AT). In order to investigate effects of high GT supplementation as compared to AT, C57 BL/6 mice were fed control AT (35 mg/kg diet) or high GT (1,000 mg/kg diet) diet for 8-10 weeks and then exposed to 60 $mg/m^3$ CS, 6 hr/day for 3 days. AT and GT levels and their metabolites were measured at endpoints. High GT supplementation significantly reduced AT levels in plasma, liver and lung compared to AT. CS increased levels of AT and GT in plasma and lung of control AT group but decreased GT levels in lung of high GT supplemented group. Moreover, CS significantly decreased GT metabolite, gamma-CEHC. The results suggest that high GT supplementation have selective modulation of concentrations of vitamin E and its metabolite in plasma and lung but not in liver against in vivo CS exposure.

Oxidative Stability of Wheat germ Lipid and Changes in the Concentration of Carotenoid and Tocopherol during Oxidation (밀배아 지방질의 산화 안정성과 카로티노이드 및 토코페롤의 변화)

  • The changes of the lipid composition and of the contents of carotenoid and tocopherol in wheat germ were studied during the storage at $30^{\circ}C$. The contents of triglyceride and free fatty acid were changed from 66% and 7% to 49% and 24% respectively after 30 days. The predominant free fatty acids were lauric acid (29%), palmitic acid (21%) and linoleic acid (20%), however, linoleic acid increased to 30%, lauric acid reduced to 21% after storage of 30 days. The carotenoids in the wheat germ were ${\beta}-carotene,\;{\alpha}-carotene$, lutein and taraxanthin, and the contents of these were 306, 59, 383 and 356 ng/g wheat germ, respectively. Their contents, however, were reduced to 36, 4, 203 and 149 ng respectively after 20 storage days. Especially, degradation rate of ${\beta}-carotene$ was 22.5 ng/day. The tocopherol isomers in wheat germ were ${\alpha}-,\;{\beta}-\;and\;{\gamma}-tocopherol$, and they reduced from $55,\;48\;and\;38\;{\mu}g/g$ wheat germ to 35, 32 and $32\;{\mu}g$ respectively after 20 storage days. The ${\alpha}-tocopherol$ was degraded by $1.26\;{\mu}g/day$ at this storage condition.

Enzymatic Synthesis and Characterization of Structured Lipids from Docosahexaenoic Acid - Enriched Fish oil and Soybean oil (어유와 대두유로부터 기능성 유지의 효소적 합성)

  • Structured lipids(SLs) were synthesized by enzymatic interesterification with DHA-enriched fish oil(containing 27% docosahexaenoic acid) and soybean oil in the hatch-type reactor. The interesterification was performed for 24 hr at $55^{\circ}C$ and TLIM(immobilized lipase from Thermonyces lanuginosa, 10% by weight of total substrates) was mixed with 180 rpm of shaking. The fish oil and soybean oil were interesterifed with several weight ratio(fish oil : soybean oil, 2:8, 3:7, 4:6, 5:5, w:w), Reverse-phase high performance liquid chromatography with an evaporative light-scattering detector separated the triglyceride species of SLs. The products contained the newly synthesized peaks. Especially, one of peaks was distinctively increased with the increasing weight ratio from 2:8 to 5:5 while the peak of trilinolein (LLL) decreased vice versa. The effect of antioxidants such as catechin, BHT(Butylated hydroxytoluene), and their combinations on the oxidative stability in SL were investigated. Oxidative stability was carried out under oven test at $60^{\circ}C$ over 72 hr thereafter SLs were analyzed for total fatty acid content, rancimat, peroxide value, electronic nose and TBARS value. Among all combinations of antioxidant, the highest stability was obtained from 200 ppm of catechin. Besides, total tocopherol ($\alpha$, $\gamma$, and $\delta$-tocopherol), iodine and saponification value were analyzed in which iodine and saponification value of SLs were 151.19 and 182.35.

The Contents of Phytosterols, Squalene, and Vitamin E and the Composition of Fatty Acids of Korean Landrace Setaria italica and Sorghum bicolar Seeds

  • To characterize the nutraceutical property of Italian millet (Setaria italica) and sorghum (Sorghum bicolor), ten Korean landraces of each crop were collected and their vitamin E (tocopherols and tocotrienols), squalene and phytosterols (campesterol, stigmasterol and ${\beta}$-sitosterol) contents as well as fatty acid composition in seeds were evaluated. Italian millet seeds exhibited 5 forms of vitamin E isomers: three (${\alpha}$-, ${\gamma}$- and ${\delta}$-) tocopherols and two (${\alpha}$- and ${\gamma}$-) tocotrienols, while sorghum seeds showed only three forms of vitamin E isomers: ${\alpha}$- and ${\gamma}$-tocopherol and ${\alpha}$-tocotrienol. In both crops, ${\gamma}$-tocopherol was the major constituent of vitamin E in terms of highest quantity. Total vitamin E content in Italian millet and sorghum landraces were 88.3 mg/kg and 44.3 mg/kg, respectively. Among three phytosterols (campesterol, stigmasterol and ${\beta}$-sitosterol) analyzed, ${\beta}$-sitosterol was the major form comprising about 85% and 65% in Italian millet and sorghum landraces, respectively. Total phytosterols content ranged from 443.0 to 568.5 mg/kg and 442.3 to 719.2 mg/kg in Italian millet and sorghum, respectively. Squalene, a precursor of phytosterols biosynthesis, ranged from 6.8 to 10.2 mg/kg in Italian millet and from 62.2 to 115.2 mg/kg in sorghum. Linoleic, oleic and palmitic acids were the major fatty acids in both of the crops and about 80% of the total fatty acids were unsaturated fatty acids. Among the tested landraces, M09 and S10 showed relatively higher proportion of phytonutrients, suggesting their potential as a gene source for further breeding program.

Effect of Vitamin E Supplementation on Plasma Antioxidant Vitamins and Immunity Status of Crossbred Cows

  • Twenty crossbred (HF${\times}$Tharparkar) dry pregnant cows were divided into four equal groups. They were supplemented with 1,000 I.U. $\alpha$-tocopheryl acetate from 0 (group I), 15 (group II), 30 (group III) and 60 (group IV) days before parturition to 1month of lactation. All the cows were kept under similar feeding and management conditions. Blood plasma samples collected on specific days were analyzed for $\alpha$-tocopherol, retinol, total antioxidant activity (FRAP), immunoglobulin and calcium. Plasma $\alpha$-tocopherol concentration at 30 days prepartum averaged 3.5, 4.1, 4.4 and $3.9{\mu}g/ml$ and decreased by 50.0, 41.4, 34.1 and 33.3 percent on the day of parturition in the four respective groups. After calving, plasma vitamin E started to recover earlier in groups II, III and IV as compared to group I. Mean plasma $\alpha$-tocopherol concentration at 21 days postpartum was significantly higher in groups II, III and IV (2.9, 3.5 and $3.1{\mu}g/ml$) compared to group I ($1.9{\mu}g/ml$) cows. Plasma retinol concentration also showed a substantial decrease in all the groups on the day of calving but recovered to its normal value at 3 weeks postpartum. Plasma total antioxidant activity averaged 901, 895, 859 and $875{\mu}mol/l$ in the four respective groups on 30 days prepartum and decreased on the day of calving in all the groups, but the decrease was less in groups III and IV. Plasma immunoglobulin concentration was higher in group IV, followed by groups III, II and I, respectively, showing better immune status of vitamin E supplemented cows due to less oxidative stress. Supplementation of vitamin E resulted in higher plasma calcium concentration. The data showed that vitamin E supplementation should be started at least 30 days prepartum to reduce oxidative stress in periparturient cows.

Effecter of Different Levels of Ethanol Ingestion and Ethanol Withdrawal on Vitamins A and E contents in Rats (에탄올의 섭취수준과 금주 후 회복이 흰쥐의 비타님 A 및 E 함량에 미치는 영향)

  • 본 연구는 에탄올의 급여수준과 금주 후 회복이 체내항산화비타민 상태에 미치는 영향을 조사하고자 평균 체중이 200g인 Dprague-dawley 종 숫쥐 50마리를 사용하여 혈장 및 간조직의 비타민 A 및 E 의 함량을 비교.분석하였다. 실험식이라는 에탄올 액체식이형태로 만들어 공급하였으며, 이때 식이는 mL당 1 kacal의 열량을 공급할 수 있도록 조제하였다. 각 실험군의 식이구성은 에탄올을 첨가하는 대신 dextrin-maltose를 급여한 C군, 전체 칼로리의 10%를 에탄올로 첨가하는 LE 군 , 20%를 첨가하는 ME 군, 30%를 첨가하는 HE군 , 30%의 에탄올을 5주간 공급한 후 다시 2주간 정상식이를 급여한 회복군(HR)으로 설정하여 실험식이를 공급하였다. 혈자에서의 retinol 함량은 에탄올 급여수준에 따른 유의적인 차이는 나타나지 않았다. 간조직 중의 retinol 함량은 20% 이상의 에탄올 군에서 대조군에 비해 유의적으로 감소되었고, 30% 에탄올군인 HE 군에서는 현저하게 저하되었다. 30% 에탄올 급여후 2주간 정상식이를 회복시킨 군에서도 간조직의 retinol의 함량은 그대로 저하된 상태를 유지하였다. 간조직의 retinyl palmitate 함량은 에탄올 급여수준이 높을수록 감소되는 경향이었으며, 30% 에탄올군에서는 대조군에 비해 유의적으로 감소되었다. 회복군의 경우에도 이러한 저하현상은 더욱 심화되었다. 혈장의 $\alpha$-tocopherol 함량은 에탄올 급여량에 따라 별다른 차이를 나타내지 않았다. 간조직에서 10% 에탄올군에서도 대조군에 비해 유의적으로 감소되었으며, 에탄올 급여수준이 증가될수록 현저하게 저하되었다. 그러나 회복군에서는 $\alpha$-tocopherol의 함량이 10% 에탄올군의 수준까지 회복되었다. 이상의 결과에서와 같이 만성적인 에탄올의 섭취는 대체로 열량의 20% 이상을 에탄올로 급여하였을대 섭취수준이 증가함에 따라 흰쥐의 간조직에서 비타민 A와 $\alpha$-tocopherol으 lgkafid이 저하되었다. 이는 에탄올의 섭취를 중단하고 정상식이를 일정기간 급여하여도 특히 비타민 A 함량은 회복되지 않는 경향을 보여 만성적인 에탄올 섭취자들에게 비타민 A영양상태의 문제를 예방할 수 있는 방안이 강구되어야 함을 나타낸다.

Antioxidant Activities of Chrysanthemum coronarium L. Fractions on the Liposomal Phospholipid Membrane (인지질막 liposome에 미치는 쑥갓 분획물의 항산화 효과)

  • This experiment was designed to investigate the antioxidant effects of Chrysanthemum coronarium L. (CC) fractions on the liposomal phospholipid membranes. The sample CC was extracted and fractionated into five different types, methanol (CCMM), hexane (CCMH), ethylacetate (CCMEA), butanol (CCMB), and aqueous (CCMA) fractions. The antioxidant activities of CC fractions in oxidized dilinoleoylphosphatidylcholine (DLPC) liposomes were examined by spectrophotometry measuring conjugated dienes. The oxidation indices of five CC fractions exhibited weaker antioxidant activities than that of BHT in oxidized DLPC liposomes, however, showing much similar antioxidant activities of $\alpha$-tocopherol in the oxidized DLPC liposomes, which is known as a potent antioxidant. Among CC fractions, CCMM and CCMA in oxidized DLPC liposomes showed rather effective than $\alpha$-tocopherol after 2 h. These results strongly indicate that bioactive substances in CC fractions have a kind of function as potent antioxidants against biomembrane oxidation.

Changes in growth performance, carcass characteristics, and meat properties of Hanwoo steers according to supplementation of composite feed additive

  • This study was conducted to evaluate the effects of a composite feed additive (CFA) consisting of vitamin C, vitamin E, selenium, and choline on growth performance, carcass characteristics, and meat properties of Hanwoo steers. Twenty-two late fattening steers were randomly assigned to one of two dietary groups, control (commercial concentrate + rice straw) and treatment (commercial concentrate + rice straw + 50 g of CFA). Average daily gain (ADG) was 4.0% higher in treatment than that in control; however, there was no statistically significant difference. The supplementation of CFA had no effect on overall yield or quality traits of carcass. CFA had no effect on chemical composition, water holding capacity, shear force, cooking loss, or myoglobin values in longissimus muscle; however, the content of ${\alpha}$-tocopherol was increased by supplementation of CFA (p < 0.01). The contents of ${\alpha}$-linolenic acid and n-3 fatty acids were higher in treatment group than in control (p < 0.01). The ratio of n-6/n-3 fatty acids was decreased by supplementation of CFA (p < 0.01). The supplementation of CFA had no effect on pH or thiobarbituric acid reactive substances (TBARS) values in longissimus muscle (raw); however, TBARS (cooked) values were lower in treatment group than in control during refrigerated storage (p < 0.01). Thus, the results indicate that the supplementation of CFA (vitamin C + vitamin E + selenium + choline) had some positive effects on the increase in ${\alpha}$-tocopherol and n-3 fatty in longissimus muscle, without any negative effect on growth performance or carcass characteristics of late fattening Hanwoo steers.

Characterization of Lipoxygenase in AOT/Isooctane Reversed Micelles (AOT/isooctane 역미셀계내에서의 lipoxygenase 반응 특성)

  • An investigation was carried out to study the characteristics of lipoxygenase in dioctyl sulfosuccinate (aerosol-OT, AOT)/isooctane revered micelles of microaqueous system containing infinitesimal water. ${\alpha}-Linoleic$ acid as a substrate could be analyzed by the colorimetric methodology using 5%(w/v) cupric acetate-pyridine solution and the activity of lipoxygenase was able to be assayed by the degree of ${\alpha}-linoleic$ acid consumption per minute. Optimal pH, temperature, and R-value ([water]/[AOT]) were determined as the value of 5.0, $25^{\circ}C$, and 10.0, respectively. Kinetic analysis of the enzyme reaction under the optimal conditions showed that the values of $K_m$ and $V_{max}$ were 0.31 mM of ${\alpha}-linoleic$ acid and $384.16{\mu}mol$ of ${\alpha}-linoleic$ acid decomposed/min, respectively. The results indicate the reaction to be lipoxygenase-catalyzed oxidation of ${\alpha}-linoleic$ acid in AOT/isooctane reversed micellar system. The inhibitory effect of natural antioxidants on lipoxygenase showed little inhibitory effect of L-ascrobic acid while ${\alpha}-tocopherol$ showed 72% of inhibitory effect.
