비타민 E 보충섭취가 폐경기 여성의 혈장 항산화 영양상태 및 DNA 손상 개선에 미치는 영향 (The Effect of Alpha-tocopherol Supplementation on the Improvement of Antioxidant Status and Lymphocyte DNA Damage in Postmenopausal Women)

  • 본 연구는 비타민 E의 보충섭취가 폐경기 여성의 항산화 영양상태 및 임파구 DNA 손상을 개선할 수 있는지를 알아보고자 수행되었으며, 이중맹검법으로 실시되었고 플라시보를 위약군으로 하였다. 폐경기 여성 35명을 대상으로 6주 동안 비타민 E 보충섭취군에게는 비타민 E 캡슐($\alpha$-tocopherol 성분 400 IU/capsule)을, 위약군에게는 대두유로 만든 위약 캡슐(400 mg/capsule)을 하루 2회 섭취하도록 하였으며, 이 때 두 군의 분류는 무작위로 하였다. 항산화 영양상태를 알아보기 위해 혈장 vitamin C, $\alpha$-tocopherol, $\gamma$-tocopherol, $\alpha$-carotenoid, $\beta$-carotenoid, lycopene 농도를 측정하였고, 임파구 DNA 손상정도를 알아보기 위해서는 Comet assay를 이용하여 tail length, %DNA in tail, tail moment를 측정하였다. 비타민 E 보충섭취군에서는 혈장 vitamin C(p<0.05)와 $\alpha$-tocopherol(p<0.000)농도가 유의적으로 증가하였고, $\gamma$-tocopherol(p<0.000) 농도와 tail length(p<0.05)는 유의적으로 감소하였다. 반면, 위약군에서는 혈장 vitamin C(p<0.05)의 농도만이 유의적으로 증가하였다. 결론적으로, 본 연구는 비타민 E의 보충섭취가 혈장 항산화 상태 및 DNA 손상에 대해 부분적인 개선 효과가 있음을 보여주었다.

  • High PUFA식이와 vitamin E 함량이 흰쥐 간장내의 지질과산화에 미치는 영향을 알아보기 위하여, 45Kcal% 고불포화 옥수수유의 정상적 vitamin E함량을 가한 식이군(PF군)과 45Kcal% 고불포화 옥수수유의 가한 PF군에 vitamin E를 5배 첨가한 식이군(PFE군), 45Kcal% 고불포화 옥수수유 식이에 vitamin E를 가하지 않은 식이군 (PFO군)을 실험군으로 하여 3주, 6주 및 9주간 사육한 후 간장에서의 vitamin E 함량과 과산화지질량 및 SOD와 GPX활성을 조사하였다. High PUFA식이에 vitamin E를 정상적으로 가한 PF군은 대조군과 차이가 나타나지 않았으나, vitamin E를 충분히 첨가한 식이 PFE군은 식이내 vitamin E 공급이 부족한 PFO군보다 vitamin E 함량과 GPX활성이 증가하였고, SOD활성과 과산화지질향은 현저하게 감소하였다. 식이 투여기간별로 PF군과 PFE군에 비해 PFO군이 실험기간이 경과할수록 vitamin E 함량과 GPX활성이 감소하였으나, SOD활성과 과산화지질향은 현저히 증가하였다.

Effects of n-3 Polyunsaturated Fatty Acids-enriched Diet Supplemented with Different Levels of α-Tocopherol on Lipid Metabolism in Laying Tsaiya Ducks

  • The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.

베타-카로틴과 알파-토코페롤의 산화안정성에 대한 리폭시게나아제의 영향 (Effect of Lipoxygenase on Oxidative Stability of ${\beta}-Carotene$ and ${\alpha}-Tocopherol$)

  • 고형상의 모델시스템에서 베타-카로틴과 알파-토코폐롤의 산화에 미치는 리폭시게나아제, 리놀레산, 수분활성도의 영향에 대해 검토하였다. 리놀레산 존재하에서 리폭시게나아제에 의한 베타-카로틴과 알파-토코페롤의 산화반응은 높은 수분활성도에서 잘 일어났으며 반응 48시간 경과 후에는 베타-카로틴의 경우 초기함량에 대해 19% 잔존량만을 보였고 알파-토코페롤은 5%를 나타내어 현저히 감소되는 경향을 보였다. 이와같이 동일 조건에서 알파-토코페롤은 베타-카로틴보다 더 많은 감소량을 보여 peroxy radical과의 반응성이 베타-카로틴보다 뛰어난 것을 알 수 있었다. 또한 베타-카로틴의 농도가 높을수록 베타-카로틴의 산화속도도 비례적으로 빨라지는 양상을 보였다. 그러나 스펙트럼상으로 볼 때 베타카로틴의 산화로 인한 최대파장의 이동은 일어나지 않았다. 알파-토코페롤의 산화생성물로는 ${\alpha}-tocopheryl\;quinone$${\alpha}-tocopheryl\;dimer$가 검출되었으며 주요 산화생성물은 ${\alpha}-tocopheryl\;quinone$으로서 반응경과에 따라 크게 증가하였다. 또한 이들 산화생성물은 리폭시게나아제와의 반응에 의해 그 함량이 훨씬 많음을 나타내었다.

Effects of d-${\alpha}$-tocopherol supplements on lipid metabolism in a high-fat diet-fed animal model

  • High-fat diet up-regulates either insulin resistance or triglycerides, which is assumed to be related to the expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$. The beneficial effects of vitamin E on insulin resistance are well known; however, it is not clear if vitamin E with a high-fat diet alters the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$. We investigated the effects of d-${\alpha}$-tocopherol supplementation on insulin sensitivity, blood lipid profiles, lipid peroxidation, and the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat (HF) diet-fed male C57BL/6J model of insulin resistance. The animals were given a regular diet (CON; 10% fat), a HF diet containing 45% fat, or a HF diet plus d-${\alpha}$-tocopherol (HF-E) for a period of 20 weeks. The results showed that the HF diet induced insulin resistance and altered the lipid profile, specifically the triglyceride (TG) and total cholesterol (TC) levels (P < 0.05). In this animal model, supplementation with d-${\alpha}$-tocopherol improved insulin resistance as well as the serum levels of TG and very-low-density lipoprotein-cholesterol (VLDL-C) (P < 0.05). Moreover, the treatment decreased the levels of malondialdehyde (MDA) in the serum and liver while increasing hepatic PPAR-${\alpha}$ expression and decreasing PPAR-${\gamma}$ expression. In conclusion, the oral administration of d-${\alpha}$-tocopherol with a high-fat diet had positive effects on insulin resistance, lipid profiles, and oxidative stress through the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat diet-fed male mice.

Changes in Meat Color and α-Tocopherol Concentrations in Plasma and Tissues from Japanese Beef Cattle Fed by Two Methods of Vitamin E Supplementation

  • The influence of dietary vitamin E supplementation on meat color and ${\alpha}$-tocopherol concentrations in plasma, longissimus thoracis muscle and subcutaneous fat was investigated. Ten Japanese $Black{\times}Holstein$ crossbred steers were placed in three experimental groups receiving different supplements of vitamin E. Four steers (control) were fed no supplemental vitamin E. Two groups of three steers each, were supplemented with 500 mg dl-${\alpha}$-tocopherol acetate per animal daily for 175 days and 1,000 mg for 100 days, respectively, before slaughter. The ${\alpha}$-tocopherol concentration in plasma increased, as vitamin E were fed, and were related to the length of time and the amount of supplement. The ${\alpha}$-tocopherol concentrations in the muscle and the fat from the two supplemental groups did not differ significantly and were three or more times greater than those in the control group. Vitamin E supplementation did not affect the quantity of marbling of beef. Supplemental vitamin E stabilized the color of displayed beef around wavelengths of 500 nm and 640 nm. The two methods of vitamin E supplementation had similar effects on meat color. The effect of supplemental vitamin E on the color of beef with marbling was observed 2-3 days after slaughter and was followed for another two weeks.

랏트에 대한 Polychlorinated Biphenyl의 독성에 미치는 $\alpha$-Tocopherol과 Perilla oil의 효과 (Effects of $\alpha$-Tocopherol and Perilla oil on the Toxicity of Polychlorinated biphenyl in Rat)

  • Effects of ${\alpha}$-tocopherol and perilla oil on the toxicity of polychlorinated biphenyls (PCB) in male rat were studied. Rats were fed ad libitum for 6 weeks with the animal diet which contains PCB 30 ppm and 100 ppm. Perilla oil (0.5 g/kg body weight) and ${\alpha}$-tocopherol (30 mg/kg body weight) were administered intraperitoneally twice a week for 6 weeks. Rats fed with PCB showed enlargement of liver and spleen, increase in aspartate aminotransferase, alkaline phosphatase, sereum lipid and cytochrome P 450 and decrease in body weight and glutathione. When perilla oil was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were significantly augmented, compared to rats fed with PCB alone. This means that perilla oil potentiates the toxicity of PCB. On the other hand when ${\alpha}$-tocopherol was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were signigicantly reduced, compared to rats fed with PCB alone. This means that u-tocopherol reduces the toxicity of PCB. From the above results, it may be concluded that PCB is metabolized by microsomal mixed function oxidase and the metabolite causes the toxicity and microsomal glutathione plays a role of protection on the toxicity of PCB.

Tocopherol and Tocotrienol Contents of Vegetable Oils, Margarines, Butters, and Peanut Butters Consumed in the Korean Diet

  • The tocopherol and tocotrienol contents of 39 vegetable oils, margarines, butters, and peanut butters typically consumed in the Korea diet were determined by direct solvent extraction method followed by normal phase liquid chromatography. All samples were locally obtained in the Cheongju area in 2003 and 2004. The study examined data on 19 vegetable oils, 10 margarines, 6 butters, and 4 peanut butters. ${\gamma}$-Tocopherol (${\gamma}$-T) and ${\alpha}$-tocopherol (${\alpha}$-T) were detectable in all products except butters and were usually present in the vitamin E form in greater quantity than the other isomers. Usually, ${\gamma}$-T content was higher than ${\alpha}$-T content in many samples. Tocotrienols were measurable in some samples but usually at levels less than the corresponding tocopherols. Three analytical method validation parameters, accuracy, precision, and specificity, were calculated to ensure the method's validity and were all confirmed to be reliable and satisfactory.

산란노계를 이용한 훈제닭 개발에 있어서의 $\alpha$-토코페롤과 오징어간유의 급여 효과 (The Effect of Feeding $\alpha$-Tocopherol and Squid Liver Oil on the Development of Smoked Chicken using Old Laying Hens)

  • 본 연구는 산란노계를 이용해 훈제닭을 만들기 위해서 실행되었다. 70∼72주령된 산란노계를 이용하여 시판용 일반사료 (대조구)에 100 IU의 $\alpha$-토코페롤과 10%오징어 간유를 썩은 사료 (처리구)를 15일간 급여시킨 후 도계하였다. 그리고 소우스액 침지, 삶기 및 시판용 훈연 소우스를 살포하는 훈연 과정을 거쳐서 새로운 제품을 만들었다. 훈제닭 (처리구) 중의 수분 및 조회분 함량은 대조구에 비해서 각각 증가하는 경향 및 유의하게 증가하였다 (P<0.05). 훈제닭의 근육중의 EPA 및 DHA 함량은 100 IU $\alpha$-토코페롤과 10% 오징어 간유를 섭취 할 때 유의하게 높았다 (P<0.01). 훈연닭 중의 아미노산의 함량은 100 IU $\alpha$-토코페롤과 10% 오징어 간유를 섭취하는 것에 의한 영향은 인정되지 않았다. 훈제닭에 대해서 500명 이상의 설문조사도 받았다. 이상의 결과를 고려할 때 산란노계를 이용한 훈제닭을 만드는 것이 가능해진 것으로 사료된다.

천년초 선인장 추출물의 항산화 효과 (Antioxidative Effect of the Fractions Extracted from a Cactus Cheonnyuncho (Opuntia humifusa))

  • 본 연구에서는 천년초 손바닥 선인장 추출분획물의 항산화 활성을 비교, 검토하고자 하였다. 추출용매별 수율 및 항산화 활성을 알아본 결과 70% ethanol로 추출 하였을 경우 추출 수율 및 항산화 활성이 우수하게 나타났다. 70% ethanol 추출물을 hexane, chloroform, ethyl acetate, butanol, water로 순차 분획하여 BHA, ${\alpha}$-tocopherol과 항산화 활성을 비교하여 보았다. DPPH를 측정하여 free radical 소거활성을 검토한 결과 ethyl acetate 분획물이 가장 우수한 항산화 활성을 나타내었고 BHA와 ${\alpha}$-tocopherol 활성과 견줄만한 효과를 보여주었다. 항산화 활성이 높게 나타난 ethyl acetate 분획물을 취하여 촉진된 자동산화조건에서 AV, POV, TBA 등을 측정하여 항산화 활성을 비교하였다. BHA, ${\alpha}$-tocopherol과 비교하여 AV와 POV 측정결과 BHA가 가장 우수한 활성을 보여주었으며 ${\alpha}$-tocopherol과 ethyl acetate 분획물은 비슷한 정도의 항산화 활성이 나타났다. TBA가에 의한 지질 과산화를 측정한 결과 BHA가 가장 우수한 활성을 보여주었으며 ethyl acetate 분획물은 ${\alpha}$-tocopherol 보다 우수한 활성을 보여주었다. 전체적으로 천년초의 ethyl acetate 분획물은 ${\alpha}$-tocopherol과 비슷한 항산화 활성을 보여주었다.
