Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

제조방법에 따른 제호탕의 항산화 및 항당뇨 활성 (Antioxidant and antidiabetic Activity of Jehotang according to Different Cooking Methods)

  • 제호탕은 오매, 사인, 백단향, 초과 등의 한약재와 꿀을 넣어 만든 여름철 전통음료이다. 본 연구에서는 제호탕의 재료인 오매, 사인, 초과, 백단향의 항산화 활성, Total phenolic content, ${\alpha}$-glucosidase, ${\alpha}$-amylase inhibitory activity를 생리활성을 측정하였다. 또한, 전통적인 조리법(연밀)과 현대적인 조리법(중탕법, 모두 끓이는 방법)으로 제호탕을 만들었을 때 pH, 색도, 당도의 차이를 비교분석하였다. Total phenolic content는 사인이 120.45 mg GAE/g, 오매분말 연밀미비법이 152.66 mg GAE/g를 나타냈으며, DPPH free radical 소거활성은 사인이 93.13% 그리고 오매분말 연밀미비법이 56.44%를 나타내었다. ${\rho}$-nitrophenyl-${\alpha}$-Dglucopyranoside를 기질로 사용한 ${\alpha}$-glucosidase 저해효과는 $100{\mu}g/mL$ 농도에서 사인이 89.51% 저해율을 보였으며, 끓이는 방법이 ${\alpha}$-glucosidase 저해효과 52.38%, ${\alpha}$-amylase 저해활성 72.52%로 높은 저해능을 보였다. 이와 같은 결과, 제호탕 소재로는 사인이 항산화, 항당뇨 효과에 뛰어남을 확인할 수 있었다. 조리방법에 서는 오매분말 연밀미비법이 높은 항산화 활성을 나타내었으며, 항당뇨 효과는 끓이는 방법이 뛰어남을 확인할 수 있었다.

Bacillus sp. SKU31-1가 생산하는 α-Glucosidase 저해제 분리 및 특성 조사 (Isolation and Characterization of α-Glucosidase Inhibitor Produced by Bacillus sp. SKU31-1 Strain)

  • 미생물 기원의 ${\alpha}$-glucosidase 저해제를 탐색하는 과정에서 토양에서 분리한 균주인 Bacillus sp. SKU31-1 배양액에서 강력한 저해제 compound K을 일련의 크로마토그래피 방법에 의해 분리 정제하였고 $^1H$ NMR, $^{13}C$ NMR, $^1H-^1H$ COSY spectra 분석과 문헌조사를 통해 5-amino-1-hydroxymethyl-1,2,3,4-cyclohexanetetrol로 동정되었다. Compound K의 ${\alpha}$-glucosidase 저해 활성은 $IC_{50}$값이 maltose 기질에서는 $1.9{\mu}M$이고, sucrose 기질 사용시 4.9 mM이었다. Lineweaver Burk plot에 의해 $K_i$값이 0.15 mM 로 강력한 경쟁적 저해제 임이 밝혀졌다.

김치에서 분리한 Lactobacillus plantarum K6의 생리적 특성 및 비만억제효과 (Physiological Characteristics and Anti-Obesity Effect of Lactobacillus plantarum K6 isolated from Kimchi)

  • 본 연구는 김치로부터 비만 억제능력이 있는 젖산균을 분리 및 동정하고, 이 균주의 생리적 특성을 규명하여 상업적으로의 이용가능성을 검토하고자 실시하였다. 이를 위해 Modified MRS 분별배지를 사용하여 노란색 집락을 형성하는 균주를 대상으로 각각 ${\alpha}-amylase$ inhibitory activity, ${\alpha}-glucosidase$ inhibitory activity와 lipase inhibitory activity가 우수한 균주를 선발한 결과 K6 균주가 최종 선발되었다. K6 균주는 ${\alpha}-amylase$ 억제활성 $96.78{\pm}3.29%$, ${\alpha}-glucosidase$ 억제활성 $92.55{\pm}9.62%$, lipase 억제활성 $85.17{\pm}0.79%$, 지방분화 억제활성 $27.4{\pm}1.4%$로 나타났으며, 동정결과 Lactobacillus plantarum으로 판명되었고, Lactobacillus plantarum K6으로 명명하였다. L. plantarum K6은 답즙산과 산성의 pH에서 모두 우수한 생존력을 나타내었고, 효소활성은 전반적으로 낮았으나 arylamidase와 ${\beta}-galactosidase$에 대해 비교적 높은 효소 활성을 나타내었다. 항생제 내성 실험 결과 vancomycin, ampicillin, polymyxin B에 내성이 있는 반면 erythromycin에 감수성을 나타냈으며, Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes와 Staphyloccous aureus에 대해 각각 51.8%, 42.4%, 61.6%와 54.9%의 억제 효과를 지니고 있는 것으로 나타났다. 또한, 아민을 생성하지 않으며, 장부착성은 대조구인 L. rhamnosus GG보다 우수하였다.

노루궁뎅이버섯 물 추출물 및 유기용매 분획물의 생리활성 효과 (Biological Activities of Water Extract and Solvent Fractions of an Edible Mushroom, Hericium erinaceus)

  • 노루궁뎅이버섯을 이용하여 성인병 치료와 예방을 위한 기능성식품 개발을 위한 기초자료를 얻기 위해 노루궁뎅이버섯 물 추출물과 유기용매 분획물을 이용하여 성인병관련 생리활성을 확인하였다. 노루궁뎅이버섯 물 추출물은 87.78%의 항산화활성을 나타냈으며, 부탄올 분획물과물 분획물은 87.91%와 92.27%의 높은 활성을 나타냈다. 에틸 아세테이트 분획물은 2.96 plasmin unit의 높은 혈전용해 활성을 보였다. 핵산 분획물로부터 77.67%와 90.53%의 높은 트롬빈 저해활성과 ${\alpha}$-glucosidase 저해활성을 확인하였다. 실험 결과로부터 노루궁뎅이버섯 에틸아세테이트 분획물의 높은 혈전용해 활성과 핵산 분획물의 높은 트롬빈 저해활성과 ${\alpha}$-glucosidase 저해활성은 혈관계질환 치료나 예방을 위한 기능성식품 개발에 이용 가능할 것으로 기대된다.

Hypoglycemic Effect of Fermented Soymilk Extract in STZ-induced Diabetic Mice

  • This study investigated the hypoglycemic effect of fermented soymilk extract (FSE) in STZ-induced diabetic mice. FSE was prepared via fermentation of soymilk with Bacillus subtilis followed by methanol extraction. The hypoglycemic effect was determined by inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase as well as the alleviation of postprandial glucose level. The non-fermented soymilk extract (SE) was used as control in this experiment. FSE showed higher (p<0.05) inhibitory activities than SE against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of FSE for ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.77 ancd 0.94 mg/mL, respectively, which were comparable or even superior to those of acarbose (0.79 and 0.68 mg/mL, respectively). In addition, a further suppression on the postprandial blood glucose levels were observed in the FSE than SE group for both STZ-induced diabetic mice and normal mice. Furthermore, FSE significantly lowered the incremental area under the curve (AUC) in the diabetic mice and the AUC in normal mice corroborated the hypoglycemic effect of FSE (p<0.05). Results from this study suggest that FSE may help decrease the postprandial blood glucose level via inhibiting ${\alpha}$-glucosidase and ${\alpha}$-amylase and the usefulness of FSE was proven to be better than SE.

Inhibitory Effects of Paeonia suffruticosa Extracts on Maltase and Sucrase

  • This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa on ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the crude drug were determined for the inhibitory activities against maltase, sucrase and ${\alpha}-amylase$. The methanol extract from the crude drug strongly inhibited maltase (72%) and sucrase (76%) at the concentration of $100\;{\mu}g/ml$. Among the fractions examined, the ethyl acetate fraction from the natural plant drug showed potent inhibitory effects on maltase (85%) and sucrase (81%) at the concentration of $100\;{\mu}g/ml$. The ethyl acetate fraction from root cortex of Paeonia suffruticosa also exhibited significant reductions (21%) of blood glucose elevation in mice loaded with maltose.

Antioxidant and Antidiabetic Activities of Eucommia ulmoides Bark

  • Eucommia ulmoides bark extracts by cold water, boiling water, 100% EtOH, 70% EtOH, 100% MeOH, 70% MeOH and $CHCl_3$ were assayed for their medicinal effects. The antioxidant activity of the extracts ranged from $IC_{50}$ 125.2 to $IC_{50}\;872.7{\mu}g/ml$ in the 1,1-diphenyl-2-picrylhydrazyl (DDPH) free radical-scavenging assay, and cold water extracts had the highest antioxidant activity. $CHCl_3$ extracts had the highest inhibitory effect on angiotensin I-converting enzyme (ACE) giving inhibition of up to 56.4% at a concentration of 1 mg/ml. Extracts in 100% EtOH had the greatest inhibitory effect on $\acute{a}-amylase$ activity ($IC_{50}=174.6{\mu}g/ml$), and 70% MeOH extracts had the greatest inhibitory effect on ${\alpha}-glucosidase$ activity ($IC_{50}=14.0{\mu}g/ml$). Taken together, these results provided the in vitro evidence on the ACE, amylase and glucosidase inhibitory actions of E. ulmoides bark that form the pharmacological basis for its antihypertensive and antidiabetic action.

꽃치자나무 추출물의 HIV-1 효소 억제 활성과 QSAR에 의한 활성인자 예측 (Inhibitory Effects of Gardenia jasminoides var. radicans Makino on HIV-1 Enzymes and Prediction of Inhibitory Factor by QSAR)

  • 꽃치자나무 추출물의 HIV-1 reverse transcriptase, protease 및 alpha-glucosidase에 대한 억제활성실험을 실시하였다. ELOSA 방법으로 실험한 역전사효소 억제활성 실험에서는 꽃치자나무 잎 MeOH 추출물 $100{\mu}g/ml$ 농도에서 13.4%의 약한 억제활성이 관찰되었고, 줄기의 메탄올추출물에서는 32.5%의 높은 HIV-1 protease 억제활성과 줄기의 메탄올 추출물에서는 26.1%의 alpha-glucosidase 억제활성이 관찰되었다. 그리고 HIV-1 복제 억제활성은 MT-4 세포에 대한 HIV-1 유도 세포변성억제를 광학현미경으로 관찰하여 살펴본 결과 잎과 줄기의 모든 추출물에서 HIV-1 바이러스 증식억제에 억제활성이 관찰되지 않았다. In silico 실험결과 주요성분인 crocetin이 81.8%의 높은 역전사 효소활성이 예측되었으며, genipin이 55.39%, geniposidic acid aglycone이 64.5% 역전사효소 활성이 예측되었다. 주로 aglycone의 활성이 높게 예측되었으며 배당체의 경우 활성이 현저하게 저하되는 것으로 나타났다.

β-Glucuronidase 저해 활성이 우수한 결명자를 첨가한 상황 균사체 배양액의 생리활성 (Biological Activities of Phellinus linteus Mycelium Culture with Cassiae Semen Extract on β-Glucuronidase Inhibitory Activity)

  • We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.
