약물 및 유전자 전달체로 응용하기 위한 Poly(L-Lysine)이 결합된 O-Carboxymethyl Chitosan PEG의 제조와 특성 (Preparation and Characterization of O-Carboxymethyl Chitosan Ion-complexed Poly(L-Lysine) for Drug and Gene Delivery System)

  • 키토산의 응용성을 높이기 위해 제조된 O-carboxymethyl water soluble chitosan (OCMCh)의 구조에 인체 내 순환시간을 증가시키기 위하여 PEG를 도입하였으며, 약물 및 유전자 전달체로 응용하기 위하여 PEG가 결합된 OCMCh-PEG를 Poly(L-Lysine) (PLL)과 이온복합체를 형성함으로써 OCMCh-PEG-PLL를 제조하였다. 제조된 OCMCh-PEG-PLL의 물리화학적특성은 적외선 분광광도계와 핵자기공명장치를 이용하여 분석하였으며, 성공적으로 PLL이 결합되었음을 확인하였다. 또한 동적광산란장치와 투과전자현미경을 통하여 PLL의 양을 고정하였을 때, PEG의 양이 증가함에 따라 입자의 크기가 감소하는 것을 볼 수 있었으며, 구형의 입자형태를 가지는 것을 확인할 수 있었다. 이상의 결과는 OCMCh-PEG-PLL이 약물 및 유전자 전달체 등과 같은 생체재료로의 응용 가능성을 가지는 것을 볼 수 있다.

  • 새로운 poly(tetramethylene 2,6-(naphthaloyldioxy)dibenzoates)(TLCP) 블록과 poly (butylene 2,6-naphthalate)(PBN) 블록으로 구성된 열방성 블록공중합체(TLCP-b-PBN)를 용액중합에 의하여 합성하였고, in-situ 복합재료를 제조하기 위해서 poly(ethylene 2,6-naphthalate) (PEN)과 용응블렌드하였다. TLCP domain은 용융상태에서 네마틱 상을 보여 주었다. 블록공중합체는 DSC 열곡선에서 PBN과 TLCP domain에 해당되는 두 개의 뚜렷한 용응전이점을 보여 주었다. 블렌드내의 PEN의 유리전이온도 ($T_g$)는 TLCP-b-PBN의 함량에 따라 감소하였으며, TLCP-b-PBN은 매트릭스 고분자에 대한 기핵제로 작용하였다. 편광현미경 관찰결과 20% TLCP-b-PBN 블렌드 경우 PEN의 용융점이상 온도에서 잘 배향된 TLCP fibril을 볼 수 있었다. 압출된 블렌드를 액체질소내에서 절단하여 전자현미경을 이용하여 모폴로지를 관찰한 결과 TLCP domain은 $0.15{\mu}m$에서 $0.2{\mu}m$ 크기로 균일하게 분포되어 있음을 확인하였다. 매트릭스 곡분자와 TLCP와의 계면접착력은 비교적 좋았으며, 매트릭스 고분자내의 TLCP domain은 중앙에서는 구형의 모양을, 표면에서는 가늘게 배향된 섬유 모양을 보였다.

인삼(Panax ginseng) 열매로부터 분리한 ginsenoside의 동정 및 암세포독성 효과 (Ginsenosides from the fruits of Panax ginseng and their cytotoxic effects on human cancer cell lines)

  • 인삼(Panax. ginseng) 열매를 80% MeOH 수용액으로 3회 반복 추출한 뒤, 감압 농축한 추출물을 EtOAc, n-BuOH과 $H_2O$ 층으로 계통 분획을 실시하였다. EtOAc분획에 대하여 $SiO_2$ 및 ODS column chromatography를 반복실시하여 5종의 ginsenoside 화합물을 분리 및 정제하였다. NMR, IR, FAB/MS 데이터를 해석하여, 각각 ginsenoside F1 (1), ginsenoside F2 (2), ginsenpside F3 (3), ginsenoside Ia (4) 및 notoginsenoside Fe (5)로 구조 동정 하였다. 화합물 2-5는 인삼열매에서는 이번에 처음 분리 보고되었다. 분리한 5종의 화합물을 인체 암세포주(HCT-116, SK-OV-3, HeLa, HepG2, SK-MEL-5)에 처리하여 세포독성을 측정하였다. 이 중 화합물 2, 4, 및 5가 인체 암세포주에 대해 세포독성을 저해시키는 것을 알 수 있었다. 화합물 2는 SK-MEL-5, HepG2, HeLa세포에서 $IC_{50}$값이 82.8, 86.8, $78.3{\mu}M$로 확인되었다. 화합물 4는 HCT-116, SK-MEL-5, SK-OV-3, HepG2, HeLa 세포에서 $IC_{50}$ 값이 24.5, 25.4, 26.3, 22.0, $24.9{\mu}M$로 확인되었다. 화합물 5는 SK-MEL-5 세포에서 $IC_{50}$ 값이 $81.7{\mu}M$로 확인되었다. 인삼 열매에서 분리한 화합물2, 4, 및 5가 암세포주에 대해 강한 세포독성을 나타내는 것을 확인하였으며, 이 화합물들은 공통적으로 3번 수산기에 glucopyranose를 가지고 있음을 확인하였다.

LPS로 유도된 RAW264.7 대식세포에 대한 헛개나무(Hovenia dulcis) 추출물의 항염증 효과 (Anti-inflammatory Activity of Extracts of Hovenia dulcis on Lipopolysaccharides-stimulated RAW264.7 Cells)

  • 본 연구는 헛개나무 잎, 줄기, 뿌리로부터 얻어진 메탄올 추출물의 항염증 효과를 구명하기 위해서 수행되었다. LPS로 염증이 유도된 RAW264.7 세포 내 부위별 추출물을 동시에 처리하여 염증 매개성 물질인 NO 생성량 분석 결과 $40{\mu}g/m{\ell}$농도에서 뿌리(94.7%)와 줄기(42.6%)에서는 효과가 있는 반면 잎에서는 효과가 없는 것으로 확인되었다. 이 중 탁월한 효과를 보인 뿌리 추출물(RE)의 항염증 효과 및 관련 분자적 기전을 확인하였다. 그 결과, 염증 반응의 주요 경로인 NF-kB 및 MAPK 신호전달경로에서 RE가 LPS로 유도된 NF-kB의 핵 이동을 억제하고, ERK, JNK, p38의 인산화를 억제함으로써 iNOS, COX-2의 발현이 감소되고, NO와 pro-inflammatory cytokine (IL-6, $IL-1{\beta}$, $TNF-{\alpha}$)의 생성이 억제됨을 확인하였다. 또한 RE는 대식세포에서 Nrf2를 활성화시켜 항염증성 단백질인 HO-1 발현을 유도하여 항염증 효과를 나타내었다. 또한, RE로부터 주요 성분을 분리한 후 NMR과 MS 기기를 이용하여 구조 동정된 27-O-protocatechuoylbetulinic acid 화합물에서도 높은 항염증 효과를 확인하였다. 이러한 연구결과는 헛개나무뿌리와 그 주요성분은 의약품 소재 및 기능성 식품 등의 기능성 소재로 활용될 수 있는 기초적인 정보를 제공할 것으로 생각된다.

핵의학 영상을 이용한 chitosan의 galactosylation 효과에 대한 평가 (Nuclear Imaging Evaluation of Galactosylation of Chitosan)

  • 목적: 약물이나 유전자 전달에 이용되는 생체적합성이 높은 키토산의 간세포 지향성을 위해서 갈락토스를 수식하는 방법이 널리 이용되어지고 있다. 이번 연구에서는 갈락토스 수식 키토산의 간세포지향성 획득을 평가하는데 있어서 핵의학 영상법의 유용성에 대해 알아보고자 하였다. 대상 및 방법: 키토산에 NHS와 EDC를 이용하여 30 mol%의 lactobionic acid를 결합시켜 갈락토스 수식 키토산을 제조하였다. 얻어진 갈락토스 수식 키토산을 투석 시킨 후 동결 건조하여 얻어 낸 다음 키토산의 기본구조인 glucoseamine에 methyl기를 첨가시키는 반응을 하여 최종화합물을 합성하였다. GMC의 세포내 독성은 MTT assay를 통하여 확인하였다. 제조된 GMC에 $SnCl_2{\cdot}2H_2O$를 이용하여 $^{99m}Tc$을 표지하였다. 표지 후 안정성은 아세톤과 생리식염수를 이용하여 1시간까지 확인하였다. $^{99m}Tc$-GMC와 $^{99m}Tc$-MC 55.5 MBq (1.5 mCi)를 토끼의 외이정맥으로 주사 후, 감마카메라를 이용하여 10분, 30분, 60분, 90분 간격으로 전면 영상을 얻었다. 결과: 키토산에 lactobionic acid 30 mol%를 반응시켜 7.4 mol%의 galactose group이 키토산에 결합한 것을 확인하였다. 갈락토스 수식 키토산을 메틸화한 결과는 tri, di, mono가 각각 8.8%, 46%, 35.2%인 것을 확인하였다. MTT assay결과를 통해 GMC의 세포에 미치는 독성은 거의 없는 것을 확인할 수 있었다. 표지 효율은 메틸화시키지 않은 $^{99m}Tc$-GC가 아세톤과 생리식염수에서 각각 88%, 72%를 보인 반면에 $^{99m}Tc$-GMC는 96%정도를 보여 보다 높은 표지효율을 가지는 것을 확인하였다. $^{99m}Tc$-MC를 주사후 얻은 토끼 영상에서 키토산은 일반적으로 대부분 신장을 통해 배출되며, 간과 비장, 골격계에는 매우 적은 분포를 보이는데 비해. 갈락토스가 수식된 $^{99m}Tc$-GMC에서는 분포에 변화가 생겨 간, 신장, 그리고 방광에서 높은 방사능이 관찰되는 소견을 보였다. 결론: 핵의학 영상법은 갈락토스 리간드 수식 키토산의 간세포 지향성 여부를 평가하기 위한 생체내 평가법으로 이용될 수 있을 것으로 사료되었다.

몰약(Commiphora molmol Engl.)의 식중독 미생물 증식 억제 물질의 구조동정 및 식품적용 (Identification of Growth Inhibitory Substance on Food-borne Microorganisms from Commiphora molmol Engl. and Its Application to Food Products)

  • 몰약 에탄올 추출물은 50 ppm에서, 몰약 헥산 분획물의 경우 25 ppm 첨가 수준에서 Listeria monocytogenes 5 균주에 대해 72시간까지 완전 증식 억제 효과가 있었다. 몰약 헥산 분획물의 항균활성 물질을 분리, 정제하여 얻은 C3-3-2 소획분의 경우는 최소증식저해농도가 10 ppm이었다. 항균활성이 인정된 C3-3-2 소획분의 항균력을 생균수로 측정하였을 때 초기 접종 균수 보다 $5{\sim}6\;log\;cycle$ 정도 감소하는 것을 확인하여 C3-3-2 소획분에 강력한 살균효과가 있음이 입증되었다. 몰약 헥산 분획물은 Bacillus cereus와 Staphylococcus aureus에 대해서 25 ppm, Salmonella enteritidis에 대해서는 50 ppm, Vibrio parahaemolyticus에 대해서는 500 ppm을 액체 배지에 첨가할 때 72시간 동안 완전 증식억제 효과를 나타내었다. 몰약의 항균활성 물질은 m-nonylphenol로 동정되었다. 액체배양법에서 Bioscreen C를 이용하여 optical density를 측정하여 그 항균활성을 확인하였고, 표준한천배양법으로 살균효과를 확인하였던 몰약 단일물질(C3-3-2) 100 ppm을 식품에 적용하였다. 쇠고기와 명태육 마쇄물에 몰약 항균활성 물질을 첨가하여 $32^{\circ}C$$5^{\circ}C$에서 저장실험을 실시한 결과, $32^{\circ}C$ 저장온도에서는 쇠고기와 명태육에 첨가한 몰약 C3-3-2 획분의 항균효과는 나타내지 못함을 알 수 있었고, $5^{\circ}C$ 저장온도에서는 쇠고기와 명태육에 첨가된 몰약 C3-3-2 획분은 항균효과가 있었다. 그 중에서도 쇠고기에 처리된 몰약 C3-3-2 획분의 항균효과가 더 우수하였다. 그러나 몰약 C3-3-2 획분의 항균효과가 배양액 상태보다 감소하였는데, 이 결과를 통하여 천연 항균활성 물질을 식품에 적용할 때 그 효과가 감소하는 이유와 그 해결방법에 대한 연구가 필요한 것으로 판단된다.

고내후성 활엽수종의 추출성분을 이용한 신기능성 물질의 분리 및 생리활성 (Bioactivities and Isolation of Functional Compounds from Decay-Resistant Hardwood Species)

  • 아까시나무의 목질부와 현사시나무, 물푸레나무 및 느릅나무의 수피를 채취하여 아세톤-물 혼합용액(7:3, v/v) 으로 추출한 후 hexane, chloroform, ethylacetate 및 수용성으로 분획하고 동결건조하여 분말로 조제한 후 메탄올-물 등의 용리용매로 Sephadex LH-20 칼럼에서 크로마토그래피를 수행하였다. 물푸레나무에서는 aesculitin 및 그 파생물인 fraxetin 등 다량의 쿠마린 화합물과 에스테르화합물을 단리하였으며, 느릅나무로부터 C-7에 xylopyranose와 apiofuranose와 같은 5탄당이 결합된(+)-catechin 배당체 화합물과 procyanidn B-3를 단리하였다. 아까시나무에서는 leucorobinetinidin의 C-4에 ethoxyl 기가 결합된 flavan 유도체 화합물과 robinetin 등의 flavanonol 화합물을 단리하였다. 현사시나무에서는 taxifolin 등의 후라보노이드 화합물과 배당체인 sakuranetin-5-O-glucopyranoside를 단리하였으며 살리신 유도체인 salireposide 등을 단리하였다. 내후성 시험에서는 목재블록에 부후균을 접종하여 배양한 후 중량감소를 측정하는 방법과 목분-agar 배지에 부후균을 접종한 후 균사의 생장 직경을 측정하는 방법을 적용하였다. 아까시나무가 다른 시룓르보다 우수한 활성을 나타내었으며 특히 메탄올 추출머리를 하지 않은 시료가 처리한 시료보다 좋은 균사생장 저해효과를 나타냈다. 항산화 활성 시험에서는 물푸레나무의 에틸아세테이트 분획이 가장 높은 활성을 보였으며, 아까시나무의 에틸아세테이트 분획도 비교적 높은 효과를 나타내었고, 이 두 분획으로부터 단리된 주요 단리화합물에 대해서는 물푸레나무의 aesculetin이 가장 높았으며 아까시나무의 robinetinidin도 비교적 좋은 효과를 나타냈다.)나 틈새시장(niche market) 마케팅 등에 적용 가능하리라 여겨진다.된다.다.산물로 판단되었다.징하며 WLWQ에 적용되는 몇 가지 제약을 관찰하고 이를 일반적인 언어원리로 설명한다. 첫째, XP는 주어로만 해석되는데 그 이유는 XP가 목적어 혹은 부가어 등 다른 기능을 할 경우 생략 부위가 생략의 복원 가능선 원리 (the deletion-up-to recoverability principle)를 위배하기 때문이다. 둘째, WLWQ가 내용 의문문으로만 해석되는데 그 이유는 양의 공리(the maxim of quantity: Grice 1975) 때문이다. 평서문으로 해석될 경우 WP에 들어갈 부분이 XP의 자질의 부분집합에 불과하므로 명제가 아무런 정보제공을 하지 못한다. 반면 의문문 자체는 정보제공을 추구하지 않으므로 앞에서 언급한 양의 공리로부터 자유롭다. 셋째, WLWQ의 XP는 주제어 표지 ‘는/-은’을 취하나 주어표지 ‘가/-이’는 취하지 못한다(XP-는/-은 vs. XP-가/-이). 이는 IP내부 에 비공범주의 존재 여부에 따라 C의 음운형태(PF)가 시성이 정해진다는 가설로 설명하고자 했다. WLWQ에 대한 우리의 논의가 옳다면, 본 논문은 다음과 같은 이론적 함의를 기닌다. 첫째, WLWQ의 존재는 생략에 대한 두 이론 즉 LF 복사 이론과 PF 삭제 이론 중 전자의 입장을 지지한다. 둘째, WP를 XP로부터 복원할 때 부분 자질만 복사된다. 이는 어휘가 통사층위로 들어온 이후에도 어휘 자질들이 완전히 동결되는 것이 아니라 계속 지시될 수 있다는 가설을 지지한다.ance and stress, and high threshold voltage.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6~13 times higher than that of the microspheres synthesized by conventional seeded batch copolymerization.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
