Nitrogen Isotope Labeled Tetraheme Cytochrome c3 on a Defined Medium

  • To obtain cytochrome $c_3$ labeled with a stable isotope, the conditions of cultivation and the composition of medium for DvMF were examined. The growth of DvMF was steady and reproducible under purging with $N_2$ and under pH control. DvMF was able to go on a defined medium without natural products. The composition of medium containing a small amount of $NH_4C$l as sole nitrogen source was established. Then, uniformly $^{15}N$labeled cytochrome $c_3$ was obtained during the culture of DvMF in a defined medium with $^{15}NH_4$Cl; it was confirmed by $^1H-^{15}N$ HMQC.

  • In the spectrum of uniformly 15N labeled cytochrome c3, the relative linewidths of the doublet peaks of the 15N-coupled imido proton of the coordinated imidazole group were reversed on oxidation. This inversion was explained by the interference relaxation process between the electron-proton dipolar and 15N-1H dipolear interactions. The inversion can be used to assign the imido protons of the coordinated imidazole groups in heme proteins.

  • 해면동물 P. incrustans 시료를 거제도 해금강 연안에서 SCUBA 다이빙에 의하여 채집하였으며, 채집한 시료를 메탄올로 반복해서 추출한 후에 다시 dichloromethane으로 반복 추출하였다 얻어진 유기 조추출물을 n-butanol과 물을 이용하여 분배한 후 n-butanol 층은 다시 15% 메탄올 수용액층과 n-hexane으로 분배하였다. n-Hexane 분배 분획에 대한 silica flash chromatography와 이렇게 얻어진 크로마토그래피 분획에 대한 반복적인 HPLC에 의해서 saringosterol을 분리할 수 있었으며, 이차원적 핵자기 공명 분광기법에 의하여 이 화합물을 구성하는 탄소원자들의 위치를 정확히 지정할 수 있었다. 이 스테로이드는 아직까지 우리나라에서 채집한 해양생물에서는 분리된 적이 없으며 이 화합물의 탄소 위치에 대한 정확한 지정도 처음으로 이루어졌다.
