Gamnamoside, a Phenylpropanoid Glycoside from Persimmon Leaves (Diospyros kaki) with an Inhibitory Effect against an Alcohol Metabolizing Enzyme

  • Phytochemical investigation of the methanolic extract of Diospyros kaki leaves led to the isolation of osmanthuside H (1) and a new phenol glycoside, named gamnamoside [4-(3-hydroxypropyl)-2-methoxyphenol $\beta$-D-apiofuranosyl( 1 $\rightarrow$ 6)$\beta$-D-glucopyranoside] (2) along with (-) catechin (3) through a series of reversed phase column chromatography and preparative C18 HPLC. The structures of the isolates were determined by spectroscopic methods including IR, UV, HRTOFMS, and 2D NMR. Compounds 1, 2, and 3, showed good inhibitory activities ($IC_{50}$) of 175.4, 94.4, and 126.6 ${\mu}g/mL$ respectively, whereas a reversible ADH inhibitor, 4-methylpyrazole, showed the $IC_{50}$ of 326.6 ${\mu}g/mL$ against alcohol dehydrogenase (ADH).

  • Poly(ethylene glycol) end-capped with pentafluorophenyl group(s) in ABA (FP-PEG-FP) and AB (mPEG-FP) types were prepared. Even though they were similar in composition, the lower critical solution temperature (LCST) of FP-PEG-FP was observed at $23^{\circ}C$, whereas that of mPEG-FP was observed at $65^{\circ}C$. To understand the large difference in solution behaviour of the two polymers, UV-VIS spectroscopy, microcalorimetry, 1H-NMR spectroscopy, and dynamic light scattering were used. FP-PEG-FP has two hydrophobic pentafluorophenyl groups at the ends of hydrophilic PEG (1000 Daltons), whereas mPEG-PF has a highly dynamic PEG (550 Daltons) block that are anchored to a hydrophobic pentafluorophenyl group. PF-PEG-PF not only has a smaller conformational degree of freedom than mPEG-PF but also can form extensive intermolecular aggregates, therefore, PF-PEG-PF exhibits a significantly lower LCST than mPEG-PF. This paper suggests that topological control is very important in designing a temperature-sensitive polymer.

Synthesis and Characterization of Perylene-based Pyrrolopyrone Derivative for Organic Thin Film Transistor

  • Perylene-based pyrrolopyrone derivative (PPD) was synthesized via condensation reaction with perylenetetracarboxylic dianhydride and 1,2-phenylenediarnine as n-type channel material. The structure of PPD was characterized by spectroscopic methods such FT-IR and $^1H$-NMR. PPD exhibited high thermal stability ($T_{d5wt%}: 560^{\circ}C$) and was found to be soluble only in protonic solvents with high acidity such as methane sulfonic acid and trifluoroacetic acid. The PPD solution showed maximum absorption and emission at 601 and 628 nm, respectively. Thin film transistors were fabricated by vacuum deposition and solution casting method. The electron mobilities of the devices were achieved as high as $0.17{\times}10^{-6}cm^2/Vs$ for vacuum deposited device and $0.4{\times}10^{-6}cm^2/Vs$ for spin coated device, respectively.

Binding Aspect of Cyclic AMP Receptor Protein to Symmetrically Synthetic 22-, 28- and 30-Base-Pair lac Promoters

  • The effect of the binding of CRP to the symmetrically synthetic 22, 28, and 30 bp lac promoter was investigated by 1H NMR. The binding of cAMP*CRP to the 22 bp DNA did not bring about any changes in the chemical shift values, but did cause selective line broadening of imino proton resonances of specific base pairs. However, The binding of cAMP*CRP to the 28 and 30 bp DNA brought about large changes on the imino proton resonances that seems to be induced by DNA bending. We studied also the role of cAMP as an activator of DNA/CRP complex formation by gel mobility shift assay. Gel mobility shift assay revealed that the cAMP*CRP complex was not able to bind to the 22 bp DNA fragment, but was able to bind to the 28 bp DNA fragment of lac promoter region.

Synthesis and Properties of Gemini Type's Diethylene Glycol Dicarboxylates (제미니형 디에틸렌글리콜 디카르복실레이트 류의 합성 및 특성)

  • This study concerned about "Gemini type co-surfactant" which has very interesting properties with new components. They were synthesized by reaction of diethylene glycol monoethyl ether and dicarboxylic acid. The structure could be comfirmed with FT-IR and $^1H$-NMR. Surface active properties such as surface tension, evaluated cmc, cloud point, emulsing power were measured respectively at given conditions. Their surface tensions in the aqueous solution were decreased to 33~35 dyne/cm, which was lower than 39 dyne/cm of SDS, and their cmc values evaluated by surface tension method were $5.0{\times}10^{-1}\sim7.5{\times}10^{-1}$ mol/L. And the emulsifying power was excellent in jojoba oil. All of the synthesized Gemini surfactants possessed good water solubility and their cloud point were $48\sim58^{\circ}C$. As results, Gemini surfactants which were synthesized are expected to be applied as O/W emulsifiers.

  • The stability constants, log K of the 1 : 1 complexation for IA ions, $Ag^+$, and $Tl^+$ with a series of podands having different aromatic end-groups (I-IV) have been determined conductometrically in methanol at 25.0 $^{\circ}$C. Exceptionally the equivalent conductivity, ${\lambda}_{eq}\;of\;Li^+\;and\;Na^+$ were increased by the addition of I, because the complexed ions are less mobile than solvated ions. The order of log K values for I was $Ag^+{\gg}Tl^+>K^+>Na^+>Rb^+>Cs6+>Li^+$. The log K sequence of the podands for the certain cations was I>II>III${\geq}$IV. And every podands except IV showed the maximum selectivity for $Ag^+$ among the cations. These results were discussed in terms of the aromatic end-group effects, such as hetero-donor atoms or conformational changes by ${\pi}-{\pi}$ stacking interactions. The detailed conformations of ${\pi}-{\pi}$ stacking were also discussed by the observations of upfield shifts of some aromatic protons upon complexation from $^1H$ NMR spectra.

  • Aiphatic diester monomer, 3-[(benzyloxycarbonylamino)butyl]-1,4-dioxane-2,5-dione (BABD), was synthesized with the N-$\varepsilon$-benzyloxy-carbonyl-L-lysine as starting material. This monomer was synthesized to add the functionality to poly(lactic acid)s. BABD unit was successfully incorporated into the PLLA chain which was confirmed by $^1H$ NMR. The copolymer composition could be controlled by the feed ratios of monomer. The $M_n$ of this resultant polymer is expected to reach high molecular weight after the purification of monomer and optimization of polymerization time, though the polymer showed relatively low degree of polymerization ($M_n$=3300). The copolymer is expected to possess the enhanced hydrophilicity and the possibility of chemical modification on amino group.

  • Ferrocene and ferrocene derivatives have been widely used as a burning rate catalyst for composite solid propellants. However, its tendency to migrate through the propellant grain and to crystallize at the surface changes the composition of propellant which results in unpredictable burning rate. To overcome the weakness of ferrocene catalyst, we designed a polymer containing ferrocene, poly(glycidyl azide-co-glycidyl ferrocenyl ether) (GAFP). GAFPs were synthesized from poly(epichlorohydrin-co-glycidyl ferrocenyl ether) (PEGF) which has ferrocenyl ethers in its pendant groups. The structures of GAFPs were confirmed by FT-IR, $^1H$ and $^{13}C$ NMR spectral analyses. Thermal properties of the GAFPs were evaluated using differential scanning calorimeter (DSC). As the contents of ferrocene increased, the glass transition temperature ($T_g$) of the GAFPs shifted to a higher temperature, and the decomposition temperature ($T_d$) decreased because the ferrocene worked as a burning rate catalyst.

Reaction and Theoretical Study of the Coordination of an N2O-Donor Amino Alcoholic Ligand Toward Group 12 Metals Mixtures

  • A series of reactions between an amino alcoholic ligand, cis-2-((2-((2-hydroxyethyl)amino)ethyl)amino)cyclohexan-1-ol (HEAC), with the mixtures of group 12 metals including, $HgCl_2/CdCl_2$, $HgCl_2/CdI_2$, $ZnCl_2/CdCl_2$ and $ZnCl_2/CdCl_2/HgCl_2$ was experimentally and theoretically studied to determine the most stable product of these reactions. Furthermore, the Cambridge Structural Database (CSD) studies were done to evaluate the theoretical results. The products were characterized by elemental analysis, FT-IR, Raman, $^1H$ NMR spectroscopy and single-crystal X-ray diffraction. Based on these investigations a binuclear structure of cadmium, [$Cd_2(HEAC)_2({\mu}-Cl)_2Cl_2$] (1), is the most stable product that was formed in all studied reactions between HEAC and metals mixtures. In this structure, the cadmium atom has a $CdN_2O({\mu}-Cl)_2Cl$ environment and distorted octahedral geometry.

NMR Relaxation Study of Segmental Motions in Polymer-n-Alkanes

  • $^{13}C$ spin-lattice relaxation times were measured for n-alkanes of moderate chain length, ranging from n-octane to n-dodecane, under the condition of proton broad-band decoupling within the temperature range of 248-318 K in order to gain some insight into basic features of segmental motions occurring in long chain ploymeric molecules. The NOE data showed that except for methyl carbon-13 dipole-dipole interactions between $^{13}C$ and directly bonded $^1H$ provide the major relaxation pathway, and we have analyzed the observed $T_1data$ on the basis of the internal rotational diffusion theory by Wallach and the conformational jump theory by London and Avitabile. The results show that the internal rotational diffusion constants about C-C bonds in the alkane backbone are all within the range of $10^9\;-10^10\;sec^{-1}$ in magnitude while the mean lifetimes for rotational isomers are all of the order of $10^{-11}\;-10^{-10}$ sec. Analysis by the L-A theory predicts that activation energies for conformational interconversion between gauche and trans form gradually increase as we move from the chain end toward the central C-C bond and they are within the range of 2-4 kcal/mol for all the compounds investigated.
