Antineoplastic Natural Productx and Analogues VIII Synthesis of some Coumarins and Their cytotoxic Activities on L1210 Cell

  • Some coumarins were sythesized for the screening of their cytotoxic activities against L1210 cell. Of the conmarins sythesized, 6, 7-dihydroxycoumarin (esculetin) and 7, 8-dihydroxycoumain (dephnetin) as coumarins with dioxygenated A-ring, and 6-acetoxy-5, 7-dimethoxycoumarin and5, 7-dimethoxy-6-hydroxycoumarin as trioxygenated ones, show considerable cytotoxic activities, ED 50 being 4. 3, 8. 8, 17.2 and 5.5 $\mu$g/ml in the same other as the substances. THe extent oxygenation of the A-ring and the positions of the oxygen functions eventually play an important role for the cytotoxic activity.

  • 2-Arylchromones, also known as flavones, are among the most ubiquitous classes of natural products occurring in the plant kingdom. On the other hand, 2-styrylchromones are relatively scarce in nature and only a few compounds has been isolated from the blue-green algae species. Therefore, new 2-arylethenylchromone derivatives ( 4a∼n, 5a∼f) were synthesized by the aldol condensation of 2-methylchromone ( 3) with several aromatic aldehydes in order to evaluate their cytotoxicities using a MTT assay on three tumor cells. 2-Arylethenylchromone derivative 4a showed the significant cytotoxic activities on KB, HL-60 and P-388 cell lines with $IC_{50}$/ values of 25.2, 63.59 and 49.51 $\mu$M, respectively, indicating that 2-arylethenylchromone skeleton has a potential anti-tumor application.

  • A series of 5-hydroxy-4-quinolone (3) and 5-methoxy-4-quinolone (4) derivatives were synthesized as truncated acridone analogues and evaluated for antitumor, antiheroes and antituberculosis activities. Among them 5-hydroxy-8-methoxy-quinolone showed potent antitumor activity ($IC_{50}$=17.7 $\mu\textrm{M}$ for HL60) which was greater than that of acronycine. However, these compounds didn't show any significant antiheroes or antituberculosis activity.

  • Tolerance of Mycobacterium sp. against organic solvents has been induced for the cholesterol side chain degradation by adding chemicals associated with synthesis of fatty acids or alcohols. Biotin of 300 .mu.g/1 and 0.5% aqueous ethanol solution were optima for the enhancement of ethanol tolerance of the microorganism. The induction of ethanol tolerance by biotin was found to be due to increase of degree of unsaturation of the fatty acids in membranous phospholipid of the cell, especially due to increase of oleic acid content. However when 0.5% of ethanol was added for the ethanol tolerance induction, there was an ambiguous correlation between ethanol tolerance and degree of unsaturation of the fatty acids, in spite of the fact that the induction increased the content of unsaturated fatty acids. Addition of 0.5% of ethanol induced several ethanol shock proteins having molecular weight similar to that of heat shock proteins.

Changes of Thiols and Oxidative Stress in Tomato Seedlings Exposed to Cadmium

  • Tomato (Lycopersicon esculentum Mill) seedlings exposed to various concentrations of $CdCl_2(0{\sim}100{\mu}M)$ in a nutrient solution for up to 9 days were analyzed with respect to the thiol changes and oxidative stress. The Cd exposure increased total non-protein thiols (NPT) and cysteine in both leaves and roots, total glutathione in leaves, and the ratios of oxidized glutathione (GSSG)/reduced glutathione (GSH) in both leaves and roots, but decreased the ratio of dehydroascorbate (DASA)/ascorbate(ASA) in leaves. Our results suggest that the Cd-induced GSH depletion due to thiol synthesis and oxidation alters the antioxidant activity of seedlings for $H_2O_2$, and the subsequent $H_2O_2$ accumulationand oxidative stress result in phytotoxicity.

  • Zr0.94Y0.06O1.97 powder was synthesized by the ultrasonic spray pyrolysis with various concentrations of starting solution and the influence of powder characteristics on sintering behavior was investigated. Powders prepared at 75$0^{\circ}C$ were characterized as narrowly distributed submicron spherical particles, which were crystalline, nonagglomerated, and compositionally homogeneous. The changes in concentration from 0.01 to 01. mol/ι increased mean particle size from 0.24 to 0.38${\mu}{\textrm}{m}$ and decreased the specific surface area from 14.2 to 2.9$m^2$/g. The relative density of the specimen from the powders, prepared with the solution concentration of 0.01 mol/ι, was 98% after sintering for 2 hr at 1,45$0^{\circ}C$ and the monoclinic phase was observed after sintering at 1,55$0^{\circ}C$. As the concentration of starting solution was increased, the formation temperature of monoclinic phase was lowered.

  • Mullite powders were synthesized from the common solution of aluminum sulfate and sodium silicate solutions by the emulsion-hot kerosene technique. The reaction temperature and mechanism for mullitization and the characteristics of synthesized mullite powders were investigated. The effect of Na components introduced from sodium silicate solution on the physical property and microstructure of sintered mullite was also examined. It was proved that mullites were formed at 75$0^{\circ}C$ through the reaction mechanism of Na2O.2.2SiO2+3.3Al2(SO4)3longrightarrow1.1(3Al2O3.2SiO2)+Na2SO4+8.9SO3. Synthetic mullite powders consisted of the compositiion of 3Al2O3.2SiO2 and showed highly agglomeration of hollow spherical particles of 1${\mu}{\textrm}{m}$ diameter. The density and fracture toughness of sintered mullites were somewhat reduced because of the effect of a very small amount of residual Na components.

  • In order to synthesize fine AlN powder by the direct nitridation of Aluminum metal power added LiF and BaF2 as additives was heated at 150$0^{\circ}C$ for 3 hrs. in nitrogen gas with flow rate of 20 mι/sec. Additives are promoted the nitridation by prevented the aggromerate of powders when 3% LiF and 2% BaF2 were added to Al metal powder. Rate of nitridation was about 100% and average size of AlN powders were very fine such as 0.3 ${\mu}{\textrm}{m}$. Specific surface area of synthesized AlN powder was 3.95$m^2$/g and also O2 and N2 contents were 2.595% and 33.25%, respectively.

  • This paper designs a digital controller by making use of the $H^{\infty}$ control algorithm and $\mu$-synthesis in order to keep a balance of the VTOL(Vertical Take Off and Landing Plane) vehicle with 4 fans. A identification of the actual model is acquired by the vehicle rolling, pitching and yawing angles for a pseude-random signal input and various identification theories. In spite of parameter variations and existing disturbances, the designed controller showes its robust performance through simulations and experiments.

  • A suitable simple model tested by experiments is required to address complex biological reactions like esterase synthesis by Saccharomyces cerevisiae. Such an approach might be the answer to a proper bioprocessing strategy. In this regard, a logistic model for esterase production from Saccharomyces cerevisiae has been developed, which predicts well the cell mass, the carbon source (glucose) consumption, and the esterase activity. The accuracy of the model has been statistically examined by using the Student's t-test. The parameter sensitivity analysis showed that all five parameters (${\mu}_m$, $K_a$, $X_m$, $Y_{x/s}$, and $Y_{p/x}$) have significant influence on the predicted values of esterase activity.
