Role of Endogenous Nitric Oxide in the Control of Renin Release

  • The present study was undertaken to investigate the role of endogenous nitric oxide in renin release under different physiological conditions. In the first series of experiments, renin release was either inhibited by acute volume-expansion (VE) or stimulated by clipping one renal artery in the rat. VE was induced by intravenous infusion of saline (0.9% NaCl) up to 5% of the body weight over 45 min under thiopental (50 mg/kg, IP) anesthesia. VE caused a decrease of plasma renin concentration (PRC). With $N^G-nitro-L-arginine$ methyl ester $(L-NAME,\;5\;{\mu}g/kg\;per\;min)$ superadded to VE, PRC decreased further. The magnitude of increase in plasma atrial natriuretic peptide levels following VE was not affected by the L-NAME. In two-kidney, one clip rats, L-NAME-supplementation resulted in a decrease, and L-arginine-supplementation an increase of PRC. Plasma atrial natriuretic peptide levels were significantly lower in the L-arginine group than in the control. Blood pressure did not differ among the L-NAME, L-arginine, and control groups. In another series of experiments, the renin response to a blockade of NO synthesis was examined using in vitro preparations from isolated renal cortex. L-NAME significantly increased basal renin release, although it was without effect on the isoproterenol-stimulated release. These findings suggest that endogenous nitric oxide significantly contributes to the renin release. Since many factors may affect the renin release in vivo, an interaction between NO and renin under various pathophysiological states is to be further defined.

  • Metabolic inhibitors which act in the process of pyrimidine salvage influenced on the uracil incorporation into nucleic acids of Toxoplasma. Inhibitors of dihydrofolate reductase, pyrimethamine and methotrexate, and inhibitors of thymidylate synthase, fluoro-uridine, fluoro·dUMP and fluoro-uracil, diminished isotopic uracil uptake in dose-dependent manners. Azauridine which suppresses do novo pyrimidine biosynthesis did not affect the salvage even in a relatively high dose. These results suggested that the activation of uracil salvage should be closely related with the function of TMP biosynthetic enzymes. The pattern of thymidine uptake had no differences between control HL-60 cells and Toxoplasma infected cells, which did not reject the specific proliferation of Texoplasma. It can be exploited to characterize the elects of various compounds related with the proliferation of Toxoplasma, especially its DNA synthesis.   • The antiulcer effects of newly synthesized prostaglandin derivatives were investigated in various experimental ulcer models and on gastric secretion in rats. HK-3 and HK-4, PG $E_2$derivatives, prevented the formation of acute gastric ulcer induced by ethanol or aspirin in pylorus-ligated rats. The ulcer formation was moderately inhibited by HK-1 and HK-2, PG $F_{2{\alpha}}$ derivatives, and aggravated by SK-1, SK-2 and SK-3, PG $F_{2{\alpha}}$ derivatives. HK-3 and HK-4 reduced the volume, acid output and pepsin output of gastric juice in pylorus-ligated rats. The gastric perfusion with physiologic saline(pH 6.0) showed relatively constant acid secretion and indomethacin increased the acid secretion. The acid secretion was markedly decreased by PG $E_2$but PG $F_{2{\alpha}}$ caused little change. Prostaglandin derivatives, especially HK-3 arid HK-4, significantly inhibited the acid secretion induced by indomethacin. The results show that, PG $E_2$ derivatives, HK-3 and HK-4, inhibit acid secretion and also have protective effects on gastric ulceration induced by ethanol or aspirin.

Synthesis and Luminescence Properties of Tb3+-Doped K2BaW2O8 Phosphors (Tb3+ 이온이 첨가된 K2BaW2O8 형광체의 합성 및 형광특성)

  • Green phosphors $K_2BaW_2O_8:Tb^{3+}$(1.0 mol%) were synthesized by solid state reaction method. Differential thermal analysis was applied to trace the reaction processes. Three endothermic values of 95, 706, and $1055^{\circ}C$ correspond to the loss of absorbed water, the release of carbon dioxide, and the beginning of the melting point, respectively. The phase purity of the powders was examined using powder X-ray diffraction(XRD). Two strong excitation bands in the wavelength region of 200-310 nm were found to be due to the ${WO_4}^{2-}$ exciton transition and the 4f-5d transition of $Tb^{3+}$ in $K_2BaW_2O_8$. The excitation spectrum presents several lines in the range of 310-380 nm; these are assigned to the 4f-4f transitions of the $Tb^{3+}$ ion. The strong emission line at around 550 nm, due to the $^5D_4{\rightarrow}^7F_5$ transition, is observed together with weak lines of the $^5D_4{\rightarrow}^7F_J$(J = 3, 4, and 6) transitions. A broad emission band peaking at 530 nm is observed at 10 K, while it disappears at room temperature. The decay times of $Tb^{3+}$ $^5D_4{\rightarrow}^7F_5$ emission are estimated to be 4.8 and 1.4 ms, respectively, at 10 and 295 K; those of the ${WO_4}^{2-}$ exciton emissions are 22 and 0.92 ${\mu}s$ at 10 and 200 K, respectively.

GaAs-Carbon Nanotubes Nanocomposite: Synthesis and Field-Emission Property (갈륨비소-탄소나노튜브 복합체 제작과 전계방출특성)

  • Hybridization of semiconductor materials with carbon nanotubes (CNTs) is a recent field of interest in which new nanodevice fabrication and applications are expected. In this work, nanowire type GaAs structures are synthesized on porous single-wall carbon nanotubes (SWCNTs) as templates using the molecular beam epitaxy (MBE) technique. The field emission properties of the as-synthesized products were investigated to suggest their potential applications as cold electron sources, as well. The SWCNT template was synthesized by the arc-discharge method. SWCNT samples were heat-treated at $400^{\circ}C$ under an $N_2/O_2$ atmosphere to remove amorphous carbon. After heat treatment, GaAs was grown on the SWCNT template. The growth conditions of the GaAs in the MBE system were set by changing the growth temperatures from $400^{\circ}C$ to $600^{\circ}C$. The morphology of the GaAs synthesized on the SWCNTs strongly depends on the substrate temperature. Namely, nano-crystalline beads of GaAs are formed on the CNTs under $500^{\circ}C$, while nanowire structures begin to form on the beads above $600^{\circ}C$. The crystal qualities of GaAs and SWCNT were examined by X-ray diffraction and Raman spectra. The field emission properties of the synthesized GaAs nanowires were also investigated and a low turn-on field of $2.0\;V/{\mu}m$ was achieved. But, the turn-on field was increased in the second and third measurements. It is thought that arsenic atoms were evaporated during the measurement of the field emission.

  • In this paper, we present the architecture design of CDMA/TDMA modem chipset for wireless telemetry system. The wireless telemetry system a measuring data collecting system from many RTs(Remote Terminal) installed at the specific area using wireless communication technology. It consists of CU single CU (Central Unit) for collecting data and a large amount of RTs for transmitting the measuring data. We propose the hardware architecture of the modem for RT and CU. We also design those modem using Verilog HDL and synthesis them using Synopsys$^{TM}$ CAD tool. The modem of RT is implemented with 27K gates and that of CU is implemented around 220k gates using 0.6${\mu}{\textrm}{m}$ CMOS standard cell. The proposed system is implemented and tested using Altera$^{TM}$ FPGA.PGA.

An Efficient Bit Loading Algorithm for OFDM-based Wireless LAN systems and Hardware Architecture Design (OFDM 기반의 무선 LAN 시스템을 위한 효율적인 비트 로딩 알고리즘 및 하드웨어 구조 설계)

  • In this paper, we propose an efficient bit loading algorithm for IEEE 802.11a wireless LAN systems. While a conventional bit loading algorithm uses the SNR value of each subcarrier, it is very difficult to estimate the exact SNR value in wireless LAN systems due to randomness of AWGN. Therefore, in order to solve this problem our proposed algorithm uses the channel frequency response instead of the SNR of each subcarrier. Through simulation results, we can obtain the performance gain of 3.5∼8㏈ at PER of 10-2 with the proposed bit loading algorithm while the conventional one obtains the performance gain of 0.5∼5㏈ at the same conditions. Also, the increased data rate can be confirmed 63Mbps. After the logic synthesis using 0.3${\mu}{\textrm}{m}$ CMOS technology, the logic gate count for the processor with proposed algorithm can be reduced by 34% in comparison with the conventional one.

Regulation of the Gene Encoding Glutathione Synthetase from the Fission Yeast

  • The fission yeast cells that contained the cloned glutathione synthetase (GS) gene showed 1.4-fold higher glutathione (GSB) content and 1.9-fold higher GS activity than the cells without the cloned GS gene. Interestingly, $\gamma$-glutamylcysteine synthetase activity increased 2.1-fold in the S. pombe cells that contained the cloned GS gene. The S. pombe cells that harbored the multi copy-number plasmid pRGS49 (containing the cloned GS gene) showed a higher level of survival on solid media with cadmium chloride (1 mM) or mercuric chloride ($10\;{\mu}M$) than the cells that harbored the YEp357R vector. The 506 bp upstream sequence from the translational initiation point and N-terminal8 amino acid-coding region were fused into the promoteriess $\beta$-galactosidase gene of the shuttle vector YEp367R to generate the fusion plasmid pUGS39. Synthesis of $\beta$-galactosidase from the fusion plasmid pUGS39 was significantly enhanced by cadmium chloride and NO-generating S-nitroso-N-acetylpenicillamine (SNAP) and sodium nitroprusside (SN). It was also induced by L-buthionine-(S,R)-sulfoximine, a specific inhibitor of $\gamma$-glutamylcysteine synthetase (GCS). We also found that the expression of the S. pombe GS gene is regulated by the Atf1-Spc1-Wis1 signal pathway.

The Effect of Bee Venom on Scopolamine Induced Memorial Impairment (봉약침액(蜂藥鍼液)이 Scopolamine으로 유발(誘發)된 기억(記憶) 장애(障碍)에 미치는 영향(影響))

  • Alzheimer's disease (AD) is the most prevalent form of neurodegenerative disease associated with aging in the human population. This disease is characterized by the following 4 structural changes : Atrophy of the Cortex, Parasympathetic, and other neural cells, the existence of Neurofibrillary tangles (NFTs), and the accumulation of Senile plaques. NFTs and Senile plaques is known to be the index of this disease. Senile plaques disturbs the neutro transmission and depletes of Acetylcholine. So, Recovery of Acetylcholine is the primal objective for treating Alzheimer's disease. So, Inhibiting the activity of Acetylcholine Esterase (AChE), which causes the hydrolysus of acetylcholine into choline and acetate, can be seen as a key role for treating Alzheimer's disease. Increasing body of evidence has been demonstrated that Bee Venom Acupuncture (BV) could compete with complex protein involving in multiple step of $NF-_{\kappa}B$ activation and exert the anti -inflammatory potential of combined inhibition of the prostanoid and nitric oxide synthesis systems by inhibition of IKK and $NF-_{\kappa}B$. The effect of BV through behavioral tests after memory impairment induced by Scopolamine. We examined the improving effect of BV on the Scopolamine (1 mg/Kg, i.p.)-induced memorial impairment using passive avoidance response and water maze tests in the mice. BV (0.84, $1.67\;{\mu}g/ml$) reversed the Scopolamine-induced memorial impairment in dose dependent manner. This study therefore suggests that BV acupuncture method may be useful for prevention of development or progression of AD.

Synthesis of Kaolinitic Clay Mineral from Amorphous Alumino-Silicate by Hydrothermal Process (비정질 Alumino-Silicate로부터 수열반응에 의한 Kaolinite질 인공점토의 합성에 관한 연구)

  • This study covers synthetic effect of the various hydrothermal treatments on formation of artificially made kaolinite mineral. The hydrothermal treatment includes the temperature treatment with time duration, addition of seeds, particle size of the starting material used, pH variation and the different types of organic acids. A colloidal silica and alumina sol which are commercially available are used for this study. A colloidal silica and alumina sol are mixed by the atomic ratio of Al/Si = 1, based on the theoretical kaolinite composition and calcined at $600^{\circ}C$ for 8 hours duration. It was found that the kaolinitic clay mineral was well developed; thereby, the different patterns of crystalline mineral are appeared. Spherical type as a crystal form was distinctively formed at the temperature of 20$0^{\circ}C$ to 25$0^{\circ}C$ with short duration time, while platy type as a crystal was highly yielded at 300~35$0^{\circ}C$. Moreover, by adding more than 20 wt% of seed as the natural kaolinitic clay to the starting material is widely distributed and developed when 2 ${\mu}{\textrm}{m}$ or less particle size of the starting material is used; also, when they are heat-treated at the temperature of 25$0^{\circ}C$ with 5 hours duration. With respect of the effect of pH variation on formation of the synthetic kaolinite minerals, the crystalline minerals are highly yielded at less than pH 2 and gradually diminished at more than pH9. Regarding to the effect of different acids on development of the kaolinite mineral, the organic acids with high chelating capacity produces good formation of crystalline minerals; whereas, amine radical-(NH2) is not an effective agent to generate the crystalline minerals.
