Effects of ${\gamma}-Irradiation$ on Meat Proteins (감마선 조사가 육단백질에 미치는 영향)

  • The proteins extracted from beef, pork and chicken meats were irradiated with up to 100 kGy at room temperature. The extracted proteins were evaluated on their in vitro digestibility by incubating successively with pepsin and pancreatin conjugate. Amino acid compositions and SDS-PAGE pattern were also analyzedin for these proteins. Gamma irradiation within the applied dose range (up to 100 kGy) produced negligible in in vitro digestibility and amino acid composition. Analysis of gamma-irradiated proteins by SDS-PAGE revealed radiolysis of ovalbumin to proteins or peptides with lower molecular weight. On the other hand, the proteins directly extracted from irradiated meats containing moisture were also evaluated for their in vitro digestibility, amino acid compositions and SDS-PAGE pattern. However, the results obtained from this experiment were similar to those of irradiated proteins after extraction from the meats.

Plasma, Tissue Thiobarbituric Acid Reactive Substance and Lymphocyte Oxidative DNA Damage in Mouse Fed Gamma Irradiated Diet (방사선 조사 사료를 섭취한 Mouse의 혈장, 간, 소장 점막의 과산화지질과 림프구 DNA의 산화적 손상)

  • Food irradiation has been steadily increasing in many countries in line with increasing international trade and concerns about naturally occurring harmful contaminants in food. Although irradiation provides an excellent safeguard for the consumer by destroying almost 100% of harmful bacteria, it is necessary to ensure the safety of irradiated foods. This study was performed to investigate the effect of an irradiated diet on lipid peroxidation in the plasma, liver, small intestinal mucosa, and lymphocyte DNA damage in mice. Eight-week old ICR mice were assigned to two groups to receive either non-irradiated or irradiated (10 kGy) diets containing 20.38% fish powder and 6.06% sesame seeds for 4 weeks. The resulting changes in the degrees of lipid peroxidation were evaluated based on the level of plasma and liver thiobarbituric acid reactive substance (TBARS), transmission electron micrograph of jejunal mucosa, and free radical-induced oxidative DNA damage in lymphocytes, as measured by alkaline comet assay (single cell gel electrophoresis). The peroxide values of the gamma irradiated diet were measured every week, and the sample for comet assay was taken at the end of the four week experimental period. There was no significant difference in food efficiency ratio between the two groups. The peroxide values of the diet were immediately increased to 35.5% after gamma irradiation and kept on increasing during storage. After 4 weeks, no differences in tissue or plasma TBARS value were observed between the two groups, but epithelial cells of jejumum showed osmiophillic laminated membranous structures, considered as myelin figures,. The oxidative DNA damage expressed as tail moment (TM) increased 30% in the blood lymphocytes of the mice fed the irradiated diet. In conclusion, the comet assay sensitively detected differences in lymphocyte DNA damage after feeding with the irradiated diet for 4 weeks. However, in order to ensure the safety of irradiated foods, it would be more useful to conduct a long-term feeding regimen using an irradiated diet and examine the level of lipid peroxidation and the state of oxidative stress in a greater range of organs.

Effects of Gamma Irradiation on Biogenic Amines Levels in Doenjang during Fermentation (감마선 조사가 된장 숙성중의 Biogenic Amine 함량에 미치는 영향)

  • Biogenic amine levels in irradiated doenjang, Korean fermented soybean paste, were investigated during fermentation at $25^{\circ}C$ for 12 weeks. Biogenic amines detected in doenjang were putrescine, cadaverine, tryptamine, $\beta$-phenylethylamine, spermidine, spermine, histamne, tyramine and agmatine. Agmatine showed the most highest level among biogenic amines detected in doenjang during fermentation. Most biogenic amines, such as putrescine, cadaverine, tryptamine, $\beta$-phenylethylamine, spermidine, histamne and tyramine, significantly decreased 20~60% by gamma irradiation during fermentation, while spermine and agmatine did not. Therefore, gamma irradiation can be applied to inhibit the biogenic amines formation in doenjang during fermentation.

Comparative Effects of Gamma Irradiation and Ethylene Oxide Fumigation on Sorption Properties and Microbiological Quality of White Ginseng Powder (백삼분말의 흡습특성 및 미생물학적 품질 안정성에 대한 감마선 및 에틸렌옥시드 처리의 영향)

  • The microbial populations of exportable and domestic white ginseng powders, which have been problems in quality control, were higher than the legally-permissive level of microorganisms as $5{\times}10^{4}$/g in total bacteria and as negative coliforms. Various microorganisms contaminated in the sample were effectively decontaminated by gamma irradiation at below 10 kGy as well as ethylene oxide (EO) fumigation. The radiosens.tivity of microorganisms was the highest in coliforms, followed by molds and aerobic bacteria ($D_{10}$: 1.25 kGy). The good microbiological quality could be retained in white ginseng powders for more than 7 months of storage at $30{\pm}2^{\circ}C$ irrespective of relative humidities if products are prepared with a lower moisture content below 10% and treated by gamma irradiation at 5 to 10 kGy in an airtight packaging.

Improvement of Hygienic Quality and Long-term Storage of Dried Red Pepper by Gamma Irradiation (감마선 조사에 의한 건고추의 위생화와 장기 안전저장)

  • Dried-red pepper, whole and powdered types, was subjected to a storage-study by investigation the effects of packaging methods (polycloth & polyethylene/polycloth, whole dried-red pepper; nylon/polyethylene-lam-inated film, red pepper powder), temperature (ambient, $5{\sim}10^{\circ}C$) and gamma irradiation doses (0-10 kGy). After 6 months storage in polyclith sack at ambient temperature, all whole dried-red pepper showed quality deterioration, such as weight change, insect infestation, discoloration and chemical changes, After 2 years storage in combined packaging with polyethylene/polycloth sack of 5-7.5 kGy irradiated whole dried-red pepper at ambient temperature, however, quality deterioration was not observed. Gamma-irradiated red pepper powder (7.5-10kGy) showed a good quality in hygienic, physicochemical and organoleptic evaluation after 2 years of storage at ambient temperature.

Inhibitory Effect on Tyrosinase and Xanthine Oxidase, and Nitrite Scavenging Activities of Schizandrae Fructus Extract by Gamma Irradiation (감마선 조사한 오미자 추출물의 아질산염 소거능, Tyrosinase와 Xanthine Oxidase 저해 효과)

  • This study was performed to investigate the effects of tyrosinase inhibition, xanthine oxidase inhibition and nitrite-scavenging effects of gamma-irradiated Schizandrae Fructus extracts from different solvents. Schizandrae Fructus was extracted by hot water, ethanol, acetone and methanol, and the extracts were irradiated 10, 20 and 30 kGy with gamma rays. All extracts from Schizandrae Fructus showed inhibition effect against tyrosinase. Tyrosinase inhibition effect of Schizandrae Fructus were higher in solvent extracts than hot water extracts by irradiation. The Schizandrae Fructus extracts had a higher inhibitory effect against xanthine oxidase, and the effect was not changed by irradiation. Nitrite scavenging activity, which was measured at various pH conditions (1.2, 3.0, 6.0), was the highest in Schizandrae Fructus extracts at pH 1.2 and 3.0. Hot water extracts provided higher nitrite scavenging effect than those of the methanol, ethanol and acetone extracts. Gamma-Irradiation may not influence on biological activites of the extracts when irradiated up to 30 kGy.

Evaluation of the Sensory Quality Characteristics during Storage and Mutagenicity of Gamma-irradiated Sujeonggwa Powder (Korean Traditional Cinnamon Beverage) (감마선 조사 분말수정과의 저장 중 관능적 품질특성 및 유전독성학적 안전성 평가)

  • This study evaluated the sensory quality characteristics of gamma-irradiated Sujeonggwa a Korean traditional cinnamon beverage during storage, and the mutagenicity of the beverage. The minimum irradiation dose required to ensure the microbiological safety of powdered Sujeonggwa was 4.5 kGy. Sensory characteristics, and the Hunter's color value, of Sujeonggwa, deteriorated with increased irradiation doses, but no significant changes in sensory qualities were found during storage. The Ames test yielded no evidence that gamma irradiation induced mutagenicity in Sujeonggwa powder.

Effects of Extracts from Acanthopanax sessiliflorus SEEM Following Gamma-ray Irradiation on Solid Tumor and Immune Cells in Mice (방사선이 조사된 오갈피 나무의 추출물이 생쥐의 복강암 및 면역세포에 미치는 영향)

  • Acanthopanax sessiliflorus SEEM extracts(AS) have been used to treat patient with diseases including cancer in Oriental countries. Recently, AS was known to have anti-cancer and immuno-stimulating activites. For these reasons, we investigated the effects of AS following gamma-ray irradiation on cytotoxicity for solid tumor cell line (S-180) and immune-potentiating ability such as proliferation of thymocytes and splenocytes. Finally we also investigated tumor weight and survival rate in tumor bearing mice. In our results, Treatment with AS suppressed proliferation of solid tumor cells (S-180) effectively. Treatment with AS accelerated thymocyte and splenocyte proliferation in tumor bearing mice. In addition, Treatment with AS reduced tumor weight and prolonged life of tumor bearing mice. In conclusion, we demonstrate that AS following gamma-ray irradiation is useful to treat patients with cancer, and also demonstrate that AS have both direct cytotoxic ability for cancer cells and indirect immune-stimulating action for thymocytes and splenocytes.

  • Cooking drips which were obtained as by-product after seafood processing in the food industries, still contain lots of proteins, carbohydrates, and other functional materials. This study was conducted to investigate the effect of gamma irradiation on the biological activities of seafood cooking drips. When the cooking drips of Hizikia fusiformis, Enteroctopus dofleini and Thunnus thynnus were irradiated, the antioxidant activities, whitening effect, and angiotensin I converting enzyme inhibition activity of the ethanol extract from seafood cooking drips were all increased by gamma irradiation. This was because of the increased extraction efficiency of available compounds by irradiation. These results suggested that the seafood cooking drips, wasted as by-products, can be used as functional compounds with gamma irradiation treatment.

Quality Characteristics of Gamma-Irradiated Chickens (감마선 조사 계육의 품질특성)

  • This research was conducted to investigate changes in quality characteristics of gamma-irradiated chickens during storage at different temperature and periods. In heme pigment(myoglobin) content, metmyoglobin content of chicken stored at 5$^{\circ}C$ was not affected by gamma irradiation but slightly increased with the increase in storage period. All samples stored at -20$^{\circ}C$ were no different in heme pigment content between nonirradiated and irradiated samples and slightly decreased as the freezing storage period increased. The SDS electrophoresis patterns were not significantly different between nonirradiated and irradiated samples. All samples stored in at 5$^{\circ}C$ showed a prominent breakdown of molecular weights ranging from 97,000 to 116,000 Daltons after 8 weeks' storage. TBA values increased according to the increment of irradiation dose level and storage period at both temperatures, 5$^{\circ}C$and -20$^{\circ}C$. However, The acid value decreased with increasing irradiation dose level. In the VBN value, nonirradiated chickens were four times higher than that of 7 kGy-irradiated one.
