사과푸른곰팡이병에 관여하는 Penicillium의 계통분석, 형태 및 병원성 (Phylogenetic Analysis, Morphology and Pathogenicity of Penicillium spp. associated with Blue Mold of Apple in Korea)

  • Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.

Transformation of the Edible Basidiomycete, Pleurotus ostreatus to Phleomycin Resistance

  • For transformation of Pleurotus ostreatus, two novel vectors, pPhKM1 and pPhKM2, were constructed, using the regulatory sequences of the P. sajor-caju $\beta$-tubulin gene(TUB1) and the ble gene encoding phleomycin binding protein. pPhKM1 contains ble fused to the TUB1 promoter and the Schizophyllum commune GPD terminator. pPhKM2 contains ble fused to the promoter and terminator regions of P. sajor-caju TUB1. To confirm phleomycin-resistance activity, each vector was cotrans-formed with pTRura3-2 into the P. ostreatus homokaryotic $ura^-$ strain. The transforming DNA was stably integrated into the genomic DNA. Subsequently, phleomycin resistance was conferred on wild-type dikaryotic P. ostreatus by transformation with pPhKM1 or pPhKM2. This transformation system generated stable phleomycin-resistant transformants.

Six Species of Penicillium Associated with Blue Mold of Grape

  • Grape fruits with blue mold symptoms were collected from house storages in different locations in Korea and were investigated for their association with Penicillium species. A total of 12 isolates of Penicillium were isolated from the collected fruits. Based on morphological and cultural characteristics and ${\beta}-tublin$ gene sequence data analysis, they were identified as P. bialowiezense, P. citrinum, P. echinulatum, P. expansum, P. solitum and unidentified Penicillium species. P. solitum was the predominant followed by P. expansum. P. bialowiezense and P. echinulatum were newly recorded in Korea. ${\beta}-Tubulin$ gene sequences could be used to distinguish each species of Penicillium and the molecular groups were correlated well with the morphological species. The unidentified species was supposed to be a new species, not previously reported in literature.

눈향나무의 잎에서 분리된 4종의 국내 미기록 내생균 (First Report on Four Novel Endophytic Fungal Species Isolated from Leaves of Juniperus chinensis var. sargentii in Korea)

  • 한라산에 서식하는 눈향나무의 침엽에서 내생균을 분리하였다. 분리된 균주들은 형태적 특성과 internal transcribed spacer 영역, large subunit rDNA 영역, translation elongation factor 영역 및 ${\beta}$-tublin 영역 염기서열의 계통분석을 이용하여 동정하였다. 그 결과 국내 미기록 내생균인 Allantophomopsis lycopodina, Epicoleosporium ramularioides, Kabatina juniperi, Seiridium podocarpi 4종의 균주를 확인하였다. 4종의 미기록 균주에 대한 형태적 특성 및 계통분석의 결과에 대해 기술하였다.

CD Gene Microarray Profiles of Bambusae Caulis in Liquamen in Human Mast Cell

  • Bambusae Caulis in Liquamen(BCL) has been used to relieve the cough and asthma, and remove the phlegm in traditional Oriental medicine. In recent years, it was studied for its antiinflammatory, antiallergenic, immune-modulating, and anticarcinogenic capabilities. This experiment was performed to evaluate the microarray profiles of CD genes in human mast cells before and after BCL treatment. The results are as follows: The expression of 51 of the genes studied was up-regulated in the Bel-treated group; they include the genes coding L apoferritin, beta-2-microglobulin, ferritin light polypeptide, CD63, monocyte chemotactic and activating fact, heme oxygenase 1, CD140a, integrin alpha M, colony stimulating factor 2 receptor, eukaryotic translation elongation factor, CD37, interleukin 18, NADH dehydrogenase 1 beta, CD48, 5-lipoxygenase activating protein, interleukin 4, ribosomal protein L5, GABA(A) receptor-associated protein, beta-tubulin, integrin beta 1, CD162, CD32, lymphotoxin beta, alpha-tublin, integrin alpha L, CD2, CD151, CD331, 90 kDa heat shock protein, CD59, CD3Z, microsomal glutathione S-transferase 2, CD33, CD162R, cyclophilinA, CD84, interleukin 9 receptor, interleukin 11, CD117, CD39-Like 2, and so forth. The expression of 7 of the genes studied was down-regulated in the BCL-treated group; they include the genes coding con, CD238, SCF, CD160, CD231, CD24, and CD130. Consequently, the treatment of BCL on the human mast cells increased the expression of 51 genes and decreased the expression of 7 genes. These data would provide a fundamental basis to the traditional applications of Bambusae Caulis in Liquamen.

  • 꼬마초롱이끼의 헛뿌리와 가는털깃털이끼의 잎에서 내생균을 분리하였다. 분리된 균주는 형태적 특성과 internal transcribed spacer, large subunit rDNA, Beta-tublin 영역 및 RNA polymerase II second largest subunit gene의 분자적 분석을 토대로 동정하였다. 연구 과정에서 2종의 국내 미기록종 내생균인 Cercophora thailandica와 Biscogniauxia petrensis를 확인하였다. 분리된 미기록종 내생균 균주의 형태적 특성 확인 및 계통 분석 결과에 대해 기술하였다.

오미자 70% 에탄올 추출물의 신경줄기세포 증식과 분화에 미치는 영향 (Effects of Schisandrae Fructus 70% Ethanol Extract on Proliferation and Differentiation of Human Embryonic Neural Stem Cells)

  • Neural stem cells (NSCs), with self-renewal and neuronal differentiation capacity, are a feasible resource in cell-based therapies for various neurodegenerative diseases and neural tissue injuries. In this study, we investigated the effects of Schisandrae Fructus (SF) on proliferation and differentiation of human embryonic NSCs. Treatment with 70% ethanol extract of SF increased the viability of NSCs derived from human embryonic stem cells, which was accompanied by increased mRNA expression of cyclin D1. Whereas 70% ethanol extract of SF also decreased the mRNA expression of nestin, it increased class III ${\beta}$-tublin (Tuj-1) and MAP2 in both growth and differentiation media. Lastly, we found increased mRNA expression of BDNF in SF-treated NSCs. In conclusion, our study demonstrates for the first time that SF induced proliferation and neuronal differentiation of NSCs and increased mRNA expression of BDNF, suggesting its potential as a regulator of NSC fate in NSC-based therapy for neuronal injuries from various diseases.

파밤나방의 미생물적 방제를 위한 병원성 곰팡이 Nomuraea rileyi의 특성 및 병원성 검정 (Characteristics and Virulence Assay of Entomopathogenic Fungus Nomuraea rileyi for the Microbial Control of Spodoptera exigua (Lepidoptera: Noctuidae))

  • 파밤나방(Spodoptera exigua)은 유기합성 농약을 이용한 화학 살충제 외에 뚜렷한 방제방법이 알려져 있지 않으나, 그에 대한 저항성과 더불어 식물을 가해하는 방법에 있어 그들만의 독특한 숙주 가해습성으로 인해 난방제 해충으로 알려져 있다. 화학살충제에 대한 저항성의 해결을 위하여 곤충병원성 바이러스 및 세균을 이용한 살충제가 개발되어 있으나, 이들 역시 파밤나방 유충이 줄기 속으로 들어가는 가해습성으로 인하여 효과적이지 못한 실정이다. 그러므로 본 연구에서는 그들의 가해습성과 저항성을 극복할 수 있는 새로운 방제원으로써, 체벽과의 일시적인 접촉을 통해 살충성을 발휘할 수 있는 파밤나방에 대한 병원성 곰팡이를 분리하고 그 특성 및 살충성을 조사하였다. 파밤나방 병원성 곰팡이는 파밤나방 누대사육 중 곰팡이병 증상을 보이며 이병된 사충으로부터 분리하였으며, 분리 곰팡이는 배지에서의 증식상 및 현미경적 관찰을 통한 형태학적 동정과 더불어 ITS, ${\beta}$-tublin 및 EF1-${\alpha}$ 부분의 염기서열 분석을 통해 Nomuraea rileyi로 최종 동정하고 N. rileyi SDSe로 명명하였다. 분리 균주의 파밤나방에 대한 살충력 검정은 $1{\times}10^4-10^8conidia/ml$까지의 다양한 포자현탁액에 파밤나방 3령 유충을 침지하는 조건으로 수행한 결과, 20-54%의 살충률을 나타냈으며 포자현탁액의 농도가 증가할수록 살충력도 증가하는 양상을 나타냈다. 본 연구결과 분리된 N. rileyi 균주가 비록 높은 살충력을 보이지 않았으나, 파밤나방이 난방제 해충임을 감안할 때 다른 방제방법과 함께 종합적 해충 방제 방법에 있어 효과적인 방제수단의 일환이 될 수 있을 것으로 기대된다.
