A microorganism capable of producing high level of extracellular cyclodextrin glucanotransferase(EC 2.4.1.19 ; CGTase) was isolated from Kimchi. 2-O-$\alpha$-D-glucopyranosyl L-ascorbic acid(AA-2G) was synthesized by transglycosylation reaction of CGTase using starch as a donor and L-ascorbic acid as an acceptor. The isolated strain S-6 was identified as Bacillus sp. S-6. The maximal CGTase production was observed in a medium containing 0.5% soluble starch, 1% yeast extract, 1% NaCO3, 0.1% K2HPO4, and 0.02% MgSO4 with initial pH 8.0. The strain was cultured at 37$^{\circ}C$ for 40 hr with reciprocal shaking. Using the culture supernatant as crude enzyme, the optimal pH and temperature of the CGTase activity of this strain were 7.0 and 4$0^{\circ}C$. In the effects of pH and temperature on the stability of the enzyme, the enzyme was stable in the range of pH 6.0~10.0 and up to 45$^{\circ}C$, respectively.

The capacity factor of fifteen phenol derivatives was determined with respect to the concentration of ${\alpha}$- or ${\beta}$-cyclodextrin [CD], the type as well as the content of organic solvent in the mobile phase, and the temperature. The effect of the inclusion complex formation between solutes and ${\alpha}$- or ${\beta}$-cyclodextrin on their retention and selectivity has been investigated. The inclusion effect of ${\beta}$-cyclodextrin was the most effective in aqueous methanol, whereas only a poor effect was observed in aqueous tetrahydrofuran and aqueous acetonitrile. A plot of the reciprocal of the capacity factor against $[CD]_T$ gives a straight line and the dissociation constant, $K_D$ of the inclusion complex can be calculated from the slope. It was possible to estimate the $k_D$ values in 100% water from a linear plot of $pK_D$ vs. water content in the solution by extrapolation. The separation factor, ${\alpha}$, of two compounds has been found to be affected not only by the $[CD]_T$ but also by their $K_D$ values. Under optimum conditions, some mixtures of phenol derivatives were able to separate successfully.

Triamino ${\alpha}$-cyclodextrin (CD) was synthesized and the inclusion complex with Minoxidil (MXD) was prepared. ${\alpha}$-CD was azidated by modifying the 6-hydroxylmethyl CD rim with sodium azide. Then, mono-, di-, tri-, and tetra-azidocyclodextrins were separated by a flash column chromatography and reduced to the corresponding amines by hydrogenation with Pd/C. The substantivities of MXD included in either 2-hydroxypropyl ${\alpha}$-CD (HP ${\alpha}$-CD) or triamino ${\alpha}$-CD were evaluated in vitro using hairless mice skins. After applying the preparations onto the skin and rinsing it, the amount of the drug left on the skin was determined using high-performance liquid chromatography (HPLC). It was the highest when the drug was included in triamino ${\alpha}$-CD. The electrostatic interaction between the protonated amino CD and the negatively charged skin would be responsible for the relatively high substantivity. The in vivo hair growth promotion effect of each preparation was investigated, where the sample application onto the clipped backs of female mice (C57BL6) and the subsequent rinsing of the backs were done once a day for 30 days. Only MXD in triamino ${\alpha}$-CD had hair growth promotion effect, possibly due to the significant substantivity.

Enantioselective production of levofloxacin from ofloxacin butyl ester by porcine liver esterase was enhanced with the addition of ${\alpha}$-cyclodextrin(${\alpha}-CD$) and pH change. The conversion yield was increased from 27 to 64% and 100% with 150 mM substrate when the molar ratio of ${\alpha}-CD$ to substrate was 1 and 2, respectively. When 100 mM of substrate was added with the same molar ratio of ${\alpha}-CD$ at pH 5.6, the solubility was increased 3.8 times and the conversion yield was increased 4.4 times.

An enzyme reactor installed with ultrafiltration membrane was developed to produce ${\alpha}-,\;{\beta}-$, and ${\gamma}$-cyclodextrins (CDs) from soluble starch by Bacillus macerans cyclodextrin glycosyltransferase (CGTase) tagged with 10 lysines at its C-terminus (CGTKIOase). Ultrafiltration membrane YM10 with 10,000 of molecular cutoff was chosen for membrane modification and CD production. A repeated-batch type of the enzyme reaction with free CGTK10ase resulted in a ${\alpha}$-CD yield of 24.0 (${\pm}1.5$)% and a productivity of 4.68 (${\pm}0.88$) g/l-h, which were 7 times higher that those for CGTK10ase immobilized on modified YM10 membrane. Addition of 1-nonanol increased CD yields by 30% relative to the control, which might be due to prevention of the reversible hydrolysis of CDs.

The cyclodextrin glycosyltransferase(EC 3.2.1.19) from Bacillus firmus was purified by precipitating with ammonium sulfate followed by, DEAE-Sephadex A-50 column chromatography and Sephadex G-100 column chromatography. In this way, we were able to obtain the single band protein on SDS-PAGE with a yield of 12%, whose purity was 49 fold. The purified CGTase was identified as a protein having molecular weight of approximately 80,000 dalton and isoelectric point of 9.6. The optimum pH and temperature for the enzyme activity were 8.0 and $65^{\circ}C$, respectively. The enzyme was stable at between pH 5.5 and 9.0 and up to $50^{\circ}C$. After 24hr of enzyme reaction using soluble starch as substrate, the ratio of ${\alpha}-$, ${\beta}-$ and ${\gamma}-cyclodextrin$ production was 0.01 : 2.90 : 1.00, respectively. And this CGTase pro-duced mainly ${\beta}-$ and ${\gamma}-cyclodextrin$.

In this research, an extractive production system for alkaloids, where production and some degree of separation occur simultaneously, was developed in a way that the fast removal of alkaloid produced from the suspension cultures was done by capturing alkaloid with cyclodextrins. The alkaloid production was substantially enhanced up to 40 fold when the solid cultures of E. califonica cells treated with ${\beta}$-cyclodextrin compared to the control. The enhancement of alkaloid production was also observed in the suspension cultures. Interestingly, the production pattern seemed to change when the cultures were treated with ${\beta}$-cyclodextrin so that the major part of the alkaloids in the treated cultures was present in the medium, while the non-treated cultures produced the alkaloids intracellularly. ${\beta}$-cyclodextrin was the most effective one in terms of the alkaloid production among the cyclodextrilns(${\alpha}$-cylodextrin, ${\beta}$-cyclodextrin and ${\gamma}$-cyclodextrin) tested in the suspension cultures. ${\beta}$-cyclodextrin showed no adverse effect on the cell growth. The most effective concentration of ${\beta}$-cyclodextrin was observed around 1.5% (w/v) in the suspension cultures. The formation of the inclusion complex of the alkaloids with ${\beta}$-cyclodextrin in the suspension cultures was confirmed by detecting the shift of UV absorbance from 274 nm to 282 nm with a UV spectrophotometer.

The inclusion of ${\alpha}-cyclodextrin$ and ${\beta}-cyclodextrin$ with flurbiprofen in aqueous phase was investigated by UV absorption and circular dichroism spectroscopies. The inclusion complex in solid powder form were made by the freeze-drying and coprecipitation methods in molar ratio 1:1. The inclusion complex formation was confirmed by infrared absorption spectroscopy. The freeze-drying method was successful in obtaining the inclusion compounds compared with the coprecipitation method. The dissolution of solid flurbiprofen inclusion complex was examined in comparison with those of flurbiprofen alone. The inclusion complex obtained by freeze-drying method increased the dissolution rate.

Cyclodextrin Glucanotransferase(EC 2.4.1.19 : 1,4-${\alpha}$-glucano) transferase, cyclizing; CGTase) can be separated and recovered in an aqueous two-phase system composed of poly(ethylene glycol)(PEG)/dextran and PEG/salt. In an aqueous two-phase system consisting of PEG 35000 (5%) and dextran T2000 (7%), all cell and debris were collected at the interphase. CGTase partitioned to the denser dextran phase at an yield of 83.4%. On the other hand, in an aqueous two-phase system consisting of PEG 35000 (10%) and sodium phosphate (15%), CGTase partitioned to the denser salt phase at an yield of 95.5%. In order to recover CGTase using an aqueous two-phase system, the PEG/salt system proved to be more efficient than the PEG/dextran system in terms of yield and cost.

${\beta}$-CD and CM- ${\beta}$-CD as chiral NMR shift agents were used to resolve the enantiomers of noradrenaline (NA). The stoichiometry of each complex formed between the CDs and the enantiomers of NA was found to be 1 : 1 through the continuous variation plots. The binding constants (K) of the complexes were determined from $^1H$ NMR titration curves. This result indicated that both ${\beta}$-CD and CM- ${\beta}$-CD formed the complexes with the S(+)-NA more preferentially than its R(-)-enantiomer. The K values for the complexes with ${\beta}$-CD ($K_{S(+)}$ = 537 $M^{-1}$ and $K_{R(-)}$ = 516 $M^{-1}$ was larger than those with CM- ${\beta}$-CD ($K_{S(+)}$ = 435 $M^{-1}$ and $K_{R(-)}$ = 313 $M^{-1}$), however, enantioselectivity (${\alpha}$) of S(+)- and R(-)-NA to CM- ${\beta}$-CD ( ${\alpha}$ = 1.38) was larger than that to ${\beta}$-CD ( ${\alpha}$ = 1.04), indicating that CM- ${\beta}$-CD was the better chiral NMR solvating agents for the recognition of the enantiomers of NA. Two dimensional rotating frame nuclear Overhauser enhancement spectroscopy (ROESY) experiments were also performed to explain the binding properties in terms of spatial fitting of the NA molecule into the macrocyclic cavities.
