The Experimental Effect of Haedongpibokhap-bang (Hǎitóngpífùhé-fāng) on Collagen-induced Arthritis (해동피복합방(海桐皮複合方)이 Collagen II 유발 관절염에 미치는 실험적 연구)

  • Objectives : The aim of this study was to know the immunity response of Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) to rheumatoid arthritis in collagen-induced arthritis(CIA) mice. Methods : For this purpose, Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) was orally administerd to mice with arthritis induced by collagen II and then value of immunocyte in spleen, draining lymph node and paw joint and cytokine(IL-6, $TNF-{\alpha}$), rheumatoid factor (IgG and IgM) in serum were measured. Results : 1. The arthritis index was significantly decreased. 2. In total cell counts of spleen, DLN and paw joint, the cells in spleen decreased while there was a significant increase in DLN and significant decrease in paw joint. 3. In lymph nodes, CD3+, CD3+/CD69+, CD4+, CD8+ cells increased significantly. 4. In joints, CD3+ and CD11+b/Gr-1+ cells decreased significantly. 5. Serum IL-6 and $TNF-{\alpha}$ were decreased significantly. 6. Production of serum IgG and IgM decreased significantly. Conclusions : The results present that Haedongpibokhap-bang($H{\check{a}}it{\acute{o}}ngp{\acute{i}}f{\grave{u}}h{\acute{e}}-f{\bar{a}}ng$) controls abnormal activity of immune system, inhibitig collagen-induced arthritis(CIA).

Up-regulation of CD11c Expression on Human Acute Myelogenous Leukemia Cells by Flt-3 Ligand (인간 골수성 백혈병 세포에서 Flt-3 수용체 리간드에 의한 CD11c 발현의 증가)

  • CD11c and costimulatory molecules such as CD80 and CD86 express mainly in dendritic cells (DCs). In this study, we investigated the biologic effects of recombinant Fms-like tyrosine kinase-3 (Flt-3) ligand on the expression of DC surface markers, including CD11c in leukemia cell lines, such as KG-1, HL-60, NB4, and THP-1 cells. The expression of the Flt-3 receptor was found in NB4 and HL-60 cells, as well as KG-1 cells, but not in THP-1 cells. When KG-1 cells were cultured in a medium containing Flt-3 ligand or granulocyte macrophage-colony stimulating factor (GM-CSF) plus tumor necrosis factor (TNF)-$\alpha$, cell proliferation was inhibited and the expression levels of CD11c, major histocompatibility complex (MHC)-I, and MHC-II were increased in the cells. Flt-3 ligand also increased the expression level of CD11c on HL-60 and NB4 cells, but not on THP-1 cells. In comparison with CD11c expression, the expression level of CD11b on KG-1 cells, but not on NB4 and HL-60 cells, was slightly increased by Flt-3 ligand. Flt-3 ligand induced phosphorylation of extracellular signal-regulated kinase-1/2 (ERK-1/2) and p38-mitogen-activated protein kinase (p38-MAPK) in KG-1 cells, and the up-regulation of CD11c expression by Flt-3 ligand in the cells was abrogated by PD98059, an inhibitor of MEK. The results suggest that Flt-3 ligand up-regulates DC surface markers on $CD34^+$ myelomonocytic KG-1 cells, as well as promyelocytic leukemia cells, and that the differentiation of the leukemia cells into DC-like cells by Flt-3 ligand is mediated by ERK-1/2 activity.

Purification and Characterization of Cyclodextrin Glucanotransferase from Paenibacillus sp. JK-12

  • Extracellular cyclodextrin glucanotransferase (CGTase) from Paenibacillus sp. JK-12 was purified through sev-eral purification steps consisting of ammonium sulfate precipitation and chromatographies on DEAE-sephadex A-50 and Mono QIM HR5/5. The purified CGTase exhibited a single band on SDS-PAGE and was estimated to be approximately 82 kDa. The isoelectric point of the enzyme was 7.2 as determined by isoelectric focusing. The CGTase from Paenibacillus sp. JK-12 had a transglucosylation activity at the C-2 position of L-ascorbic acid. The optimum pH and temperature for the CGTase activity were 8.0 and 5$0^{\circ}C$, respectively. The enzyme activity was stable from pH 6.0 to 9.() and at temperatures up to 55$^{\circ}C$ at pB 8.0, having 80% residual activity. The activity of the CGTase was strongly resistant to metals such as A $g^{+}$ and $Ba^{2+}$ but slightly inhibited by H $g^{+}$, N $i^{2+}$ and $Mg^{2+}$. The enzymeproduced $\alpha$ -cyclodextrin ($\alpha$-CD) and $\beta$-CD as the main products from starch, but not ${\gamma}$-CD.X>-CD.

Effects of JaUmJeSeupTangKaKam (JUJSTK) on Atopic Dermatitis-like Skin Lesions in NC/Nga Mouse (아토피양(樣)피부염 NC/Nga생쥐에서 자음제습탕가감(滋陰除濕湯加減)의 투여가 피부염에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of JUJSTK on atopic dermatitis in an in-vitro experiment using an NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to the humans in terms of health condition. Methods : We evaluated IL-1$\beta$, IL-6, IL-10, TNF-$\alpha$ mRNA, TGF-$\beta$ mRNA, CD4+/IFN-$\gamma$+ and IL-17+CD4+Th17 cells of NC/Nga atopic dermatitis mouse by real-time PCR and intracellular staining in vitro. Results : JUJSTK medicines supressed the activities of IL-1$\beta$, IL-6, TNF-$\alpha$, TGF-$\beta$ mRNA and IL-17+CD4+Th17 cells and it incresed the activities of IL-10 mRNA in B cells. The level of CD4+/IFN-$\gamma$+ in T cells were increased by JUSSTK. Conclusions : JUJSTK on atopic dermatitis might be incredibly effective to the atopic dermatitis treatment.

Characterization of Bacillus stearothermophilue Cyclodextrin Glucanotransferase that Expressed by Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 발현된 Bacillus stearothermophilus Cyclodextrin Glucanotransferase의 특성)

  • The cyclodextrin glucanotransferase (CGTase) gene from Bacillus stearothermophilus NO2 was expressed in Saccharomyces cerevisiae 2805 under the adhl promoter. The CGTase was purified from S. cerevisiae 2805/pVT-CGTS. The purified enzyme exhibited a optima of activity around pH 7.0 and $65^{\circ}C$. Thermal stability of the enzyme was increased fairly as compared with the CGTase of B. stearothermophilus NO2. The conversion yield of cyclodextrin (CD) and the production ratio of $\alpha$-, $\beta$,-, ${\gamma}$-CD from starch were showed similarly aspect to the CGTase of B. stearothermophilus NO2.

Effect of Dietary Fibers Isolated from Tangerine Peels on Lipid and Cadmium Metabolism in the Rat (감귤과피로부터 분리한 식이섬유가 흰쥐의 지방 및 Cadmium 대사에 미치는 영향)

  • This study was performed to investigate the effects of dietary fibers from tangerine peels on lipid and cadmium metabolism. And effects were compared with those of commercial dietary fibers($\alpha$-cellulose, citrus pectin). Sixty male rats of Sprague-Dawley strain weighing 186.7$\pm$2.6g were blocked into 12 groups according to body weight, and were raised for 2 weeks. Cadmium chloride was given at the levels of 0 of 400 ppm in diet. Various dietary fibers were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. In lipid metabolism, insoluble fibers[insoluble dietary fibers from tangerine peels(IDE), $\alpha$-celluolse] increased fecal excretion of lipids by inhcreasing feces weight, and decreased the concentrations of serum triglyceride and liver lipids. Soluble dietary fibers from tangerine peels(SDF) decreased the concentrations of serum cholesterol and liver lipids by increasing fecal lipids, too. In cadmium metabolism, soluble fivers(SDF, pectin) inhibited Cd absorption by increasing fecal Cd excretion and decreased Cd concentrations of intestion, liver and kidney. In conclusion, among the extracted fibers, SDF were more effective on lipid and Cd lowering activity and IDF had effect of increasing fecal lipid excretion. This result is useful to reduce food waste and utilize waste products.

  • Objective : The purpose of this study is to observe the effect of Herbal-cupuncture with Acanthopanacis cortex Herbal-Acupuncture Solution (AC-HAS) on rheumatoid arthritis. Methods : The author performed several experimental items : it is paw thickness, weight of spleen, expression of $CD4^+,\;CD8^+,\;B220^+$ in the spleen, production of IL-6, $TNF-{\alpha}$, and examination of histology. Results : The obstain results are summarized s follows. 1. Treatment group were significantly inhibited the incidence of arthritis, hind paw edema and index of arthritis of CIA mice. 2. Treatment group were significantly decreased splenetic weight 3. The number of $CD4^+,\;CD8^+,\;B220^+$ activated cells were significantly inhibited in treatment group. 4. Treatment group were significantly decreased the amout of IL-4, $TNF-{\alpha}$ of CIA mice.. 5. Treatment group were expressed form of new bone, synoviumin and new margine in histology imperison to control group. Conclusions : Taking all these observations into account, AC-HAS injection is considered to be effective in treating rheumatoid arthritis and put to practical use in future rheumatoid arthritis clinic.

Effects of GHX02 on Chronic Obstructive Pulmonary Disease Mouse Model

  • Objectives: Chronic obstructive pulmonary disease (COPD) is characterized by chronic inflammation and irreversible airflow. This study aimed to evaluate the effects of GHX02 in a COPD-induced mouse model. Methods: The COPD mouse model was established by exposure to cigarette smoke extract and lipopolysaccharide which were administered by intratracheal injection three times with a 7 day interval. GHX02 (100, 200, 400 mg/kg) and all other drugs were orally administrated for 14 days from Day 7 to Day 21. Results: GHX02 significantly decreased the neutrophil counts in bronchoalveolar lavage fluid (BALF) and the number of $CD4^+$, $CD8^+$, $CD69^+$, and $CD11b^+/GR1^+$ cells in BALF and lung cells. GHX02 also suppressed the secretion of tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin-17A, macrophage inflammatory protein 2 (MIP2), and chemokine (C-X-C motif) ligand 1 (CXCL-1) in BALF and ameliorated the lung pathological changes. Conclusions: Thus, GHX02 effectively inhibited airway inflammation by inhibiting migration of inflammatory cells and expression of pro-inflammatory cytokines. Therefore, GHX02 may be a promising therapeutic agent for COPD.

The Effects of Bogimakseong-bang(補氣膜性方) Treatment on cBSA-induced Membranous Nephropathy in Mouse Model (보기막성방(補氣膜性方)이 Cationized Bovine Serum Albumin투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향)

  • Objective : We aimed to identify the effects of Bogimakseong-bang(BGMSB) treatment on cBSA-induced MN in a mouse model. Methods : We divided 20 mice into 4 groups. The normal group (NR) had no treatment. We used cBSA to induced MN to the other 3 groups. One group (CT) was treated with cBSA (7mg/kg i.p) only. The second (BG-250) was treated with cBSA (7mg/kg i.p) and BGMSB extract (250mg/kg, p.o). The third group (BG-500) was treated with cBSA(7mg/kg i.p) and BGMSB extract (500mg/kg, p.o). After cBSA and BGMSB extract treatment for 4 weeks, proteinuria, serum albumin, total cholesterol, serum creatinine, BUN, total nucleated spleen cell number and total infiltrated kidney cell number of all groups were measured. CD3e+/CD19+ and CD4+/CD8 cells ratio of peripheral blood, kidney and spleen of all groups were analyzed. $IL-1{\beta}$ and $TNF-{\alpha}$, IL-6, IgG, IgM, and $IFN-{\gamma}$ levels of all groups were gauged. Histological analysis of kidney tissue and immunohistochemical staining (CD4 CD8) of kidneys were observed. Results : Proteinuria significantly decreased and serum albumin increased in groups treated with cBSA and BGMSB extract compared with the control. Total cholesterol decreased but not significantly. CD3e+/CD19cells ratio of peripheral blood decreased. CD3e+/CD19+ and CD4+/CD8 cells percentage of kidney and spleen showed no significant change. Level of $IL-1{\beta}$, $TNF-{\alpha}$ and IL-6 significantly decreased. and $IFN-{\gamma}$ increased but has not significantly. Concentration of IgG and IgM significantly decreased compared with control. Thickness of GBM decreased on histological analysis of kidney. Deposition of CD4 and CD8 decreased on immunohistochemical staining of kidney. Conclusions : According to the above result, BGMSB had a significant effect for treating MN which is cBSA-induced.

Optical Properties of Cdlnsub 2Ssub 4 and Cdlnsub 2Ssub 4 : $CdIn_2S_4$$CdIn_2S_4 : Co^{2+}$Single Crystals ($CdIn_2S_4$$CdIn_2S_4 : Co^{2+}$ 단결정의 광학적 특성)

  • $CdIn_2S_4 and CdIn_2S_4 : Co^{2+}$ singlecrystals of thenormal spinel structure were grown by the C.T.R. method. The optical energy band structure of these compounds had a indirect band gap at the fundamental optical absorption band edge. The direct and the indirect energy gaps are found to be 2.325 and2.179eV for $Cdln_2S_4$ , and 2.303 and 2.169eV for $CdIn_2S_4 and CdIn_2S_4 : Co^{2+}$ at 5K, respectivly. The fundamental absorption band edge of these single crystals shift to a shorter wavelength region with decreasing temperature, and the temperature dependence of the optical energy gaps in these compounds satisfy Varshni equation. The Varshni constants$\alpha and \beta$ of the direct energy gap are given by $13.39{\times}10_{-4}eV/K$ and 509 K for $Cdln_2S_4$ and $29.73{\times}10_{-4} eV/K$ and 1398K for $CdIn_2S_4 and CdIn_2S_4 : Co^{2+}$. The Varshni constants ${\alpha}and {\beta}$ of the indirect energy gap are given by 9.68${\times}10^{-4}$ eV/K 308K for $Cdln_2S_4$ and $13.33{\times}10_{-4}eV/K$ and 440K for $CdIn_2S_4 : Co^{2+}$ respectivly. The impurity optical absorption peaks due to cobalt dopant are observed in $CdIn_2S_4 : Co^{2+}$ single crystal. These impurity optical absorption peaks can be attributed to the electronic transitions between the split energy levels of $Co_{2+}$ ions located at $T_d$ symmetry site of $Cdln_2S_4$ host lattece.
