Phosphorus Significance in Alleviating Oxidative Stress Induced by Drought in Kentucky Bluegrass (Poa pratensis L.)

  • The objective of this study was to determine phosphorus effects on drought stress-induced oxidative stress in Kentucky bluegrass. Drought stress was induced by reducing of water to plants in pots. Two types of phosphorus were applied as potassium phosphate (P) or potassium phosphonate (PA). Application of phosphorus was efficient to ameliorate the adverse effects of drought. Osmotic potential, total chlorophyll and carotenoid content were significantly decreased by drought stress, but was relieved by P or PA application. Superoxide (O2•-) concentration was significantly increased more than 14-fold under drought-stressed plants, was accompanied with increase of hydrogen peroxide (H2O2) and lipid peroxidation (MDA). However, malondialdehyde (MDA) was much less in P or PA applied plants under drought stress condition. Activities of catalase (CAT), ascorbate peroxidase (APX) and guaiacol-peroxidase (GPX) were largely increased by drought stress and its increase rate was much higher in P or PA applied plants except APX. These results indicate that drought stress-induced oxidative stress is alleviated by P or PA application due to the increase of activities of antioxidant enzymes.

  • Cadmium (Cd) is non-essential heavy metal that negatively affects plant metabolism. Nitric oxide (NO) is an increasingly important molecule for plant metabolism that makes signaling. In this study, it was aimed to investigate the alleviating effect of sodium nitroprusside (SNP) application as NO donor in white poplar (Populus alba) under Cd stress conditions. SNP and without SNP treatments increased the Cd accumulation in root tissue. While photosynthetic pigments (Chl a, Chl b, Chl a+b, and carotenoid) content decreased by only Cd application, SNP+Cd application decreased the rate of photosynthetic pigments reduction. When the results of Cd and Cd+SNP applications were evaluated for mineral (Fe, Zn, Mn and Cu) uptake, it was found that the positive effect of SNP was heterogeneously affected. Depending on SNP application, it was found that malondialdehyde (MDA) amount decreased in leaf in 100 µM Cd applications while hydrogen peroxide (H2O2) amount decreased in 100 and 500 µM Cd applications. When antioxidant enzyme activities were examined, it was found that catalase (CAT) and ascorbate peroxidase (APX) enzyme activities increased with 100 µM SNP applications under all Cd applications. As a result, it was found that SNP application under Cd stress generally supports physiological processes positively in white poplar, suggesting that NO molecule plays important alleviating roles in plant metabolism.

  • Amylose contents and amylopectin chain architecture of sorghmn and waxy sorghum starches were determined and compared with those of other common cereal and tuber starches. Also, in vitro digestibility of sorghum starch was estimated using a novel methodology. The absolute amylose content of sorghum starch was similar to that of com and wheat starches. The side chain length distribution patterns for sorghum and waxy sorghum amylopectin were very similar to those of com and waxy com, respectively. The $k_{cat}/K_m$ values for sorghum and potato amylopectin did not show a significant difference. The kinetic parameters could be used as novel indicators for starch digestibility.

  • A biochemical genetic study on blood enzyme/protein systems in some breeds/crosses of sheep in Malaysia was carried out using horizontal starch gel electrophoresis. Blood samples were collected from 435 sheep, representing 8 breeds/crosses. These included 5 wool sheep breeds (Thai Longtail, wiltshire, Suffolk, Dorsimal and cMBLx), 1 hair sheep breed (Barbados Blackbelly) and 2 hybrids between wool sheep and hair sheep (Cameroon ${\times}$ Thai Longtail and Bali Bali ${\times}$ Malin). Twenty loci systems were examined. Of these, ten ($HB{\beta}$, ALB, TF, XP, CAT, DIA1, EsA, GPI, ME and NP) exhibited genetic variation whereas the other ten (AAT, CA, DIA2, ${\alpha}GLO$, ${\alpha}GLU$, LDH, MDH, PEP[leu-gly-gly], 6PGD and SOD) were monomorphic. The allelic frequencies which were obtained in 10 polymorphic markers are assessed and compared with the results obtained by previous workers. The estimations of inbreeding coefficient, intrabreed variation and breed relationships have been critically discussed and are used to reveal some important recommendations.

  • A Persian chinchila (2 years old, intact female) and a Korean domestic shorthaired cat (3 months, intact male) were referred to the Veterinary Medical Teaching Hospital of Chungnam National University with tachypnea. The two cats were diagnosed as feline infectious peritonitis (FIP) by blood and blood chemical examination, radiographic examination, RT-PCR and electrophoresis analysis of pleural effusion. Thromboxane synthetase inhibitor (Ozagrel HCl, 5 mg/kg, twice a day) was administered to the Persian chinchila and Korean domestic shorthair for 13 days and 16 days, respectively. Pleural effusion disappeared after treatment with Ozagrel HCl. Further study is needed to establish a new application protocol of Ozagrel HCl for FIP cases.

  • Low temperature deposition of silicon and silicon nitride films by catalytic CVD technique was studied for application to thin film transistors on plastic substrates for flexible AMOLEDs. The substrate temperature initially held at room temperature, and was controlled successfully below $150^{\circ}C$ during the entire deposition process. Amorphous silicon films having good adhesion, good surface morphology and sufficiently low content of atomic hydrogen were obtained and could be successfully crystallized using excimer laser without a prior dehydrogenation step. $SiN_x$ films showed a good refractive index, a high deposition rate, a moderate breakdown field and a dielectric constant. The Cat-CVD silicon and silicon nitride films can be good candidates for fabricating thin films transistors on plastic substrates to drive active-matrix organic light emitting display.

Characterization of Cellobiohydrolase from a Newly Isolated Strain of Agaricus arvencis

  • A highly efficient cellobiohydrolase (CBH)-secreting basidiomycetous fungus, Agaricus arvensis KMJ623, was isolated and identified based on its morphological features and sequence analysis of internal transcribed spacer rDNA. An extracellular CBH was purified to homogeneity from A. arvencis culture supernatant using sequential chromatography. The relative molecular mass of A. arvencis CBH was determined to be 65 kDa by SDSPAGE and 130 kDa by size-exclusion chromatography, indicating that the enzyme is a dimer. A. arvencis CBH showed a catalytic efficiency ($k_{cat}/K_m$) of 31.8 $mM^{-1}\;s^{-1}$ for p-nitrophenyl-${\beta}$-D-cellobioside, the highest level seen for CBH-producing microorganisms. Its internal amino acid sequences showed significant homology with CBHs from glycoside hydrolase family 7. Although CBHs have been purified and characterized from other sources, A. arvencis CBH is distinguished from other CBHs by its high catalytic efficiency.

  • A 1-year-old, 2.35 kg spayed female American short hair cat was referred with episodic signs of heat at 3 months after ovariohysterectomy. Through the screening tests, bilateral cystic, ovary like masses were shown at the caudal to both kidneys with high serum estradiol concentration. It was considered that the patient was suffered from ovarian remnant syndrome. Laparoscopic exploration was performed, and each of mass lesions was resected by ultrasonic scalpel. Patient was recovered favorably and has been doing well without recurrence of estrus signs until 2 years after surgery.

  • Monogeneans(Phylum Platyhelminthes) have been known as common parasites onto the skin and gills of cultured freshwater fishes. Infestation with these parasites result in a great loss in aquaculture industry. Some classification studies on these parasites have mostly been conducted in Japan but rarely in Korea. For the purpose of classifying monogenean parasites infesting two Korean freshwater fishes, eel (Anguilla japonica) and catfish(Parasilurus asotus), samples captured from March 1998 to April 2000 were examined. Here we report for the first time in Korea that four different species of monogeneans are identified: Pseudodactylogyrus bini and P. anguillae in eels; Ancylodiscoides infundibulovagina and Ancylodiscoides sp. in catfish.

Guided Bone Regeneration in Comminuted Long-Bone Fractures Using Recombinant Human Bone Morphogenetic Protein-2 and a Collagen Membrane

  • A dog aged two years and seven months and a cat aged seven years were referred owing to fractures of long bones. Preoperative radiographs revealed comminuted bone fractures close to joints. Conventionally, long-bone fractures are treated using intramedullary pins, plate and screw systems, or an external fixator system. In cases of non-reducible fractures, various graft materials have been used in fracture treatments to stimulate bone repair. Here, recombinant human bone morphogenetic protein-2 (rhBMP-2) and a collagen membrane were applied. Four weeks after surgery, fractured bone fragments began to unite and the bone union was observed using radiography four months after surgery. No complications occurred related to grafted materials. We successfully applied rhBMP-2 and collagen membranes in two different species to support the healing process of comminuted fractures, according to the concept of guided bone regeneration.
