Discovery of Larval Gnathostoma nipponicum in Frogs and Snakes from Jeju-do (Province), Republic of Korea

  • A survey was performed to find out the intermediate hosts of Gnathostoma nipponicum in Jeju-do (Province), the Republic of Korea. In August 2009 and 2010, a total of 82 tadpoles, 23 black-spotted pond frogs (Rana nigromaculata), 7 tiger keelback snakes (Rhabdophis tigrinus tigrinus), 6 red-tongue viper snakes (Agkistrodon ussuriensis), and 2 cat snakes (Elaphe dione) were collected in Jeju-do and examined by the pepsin-HCl digestion method. Total 5 gnathostome larvae were detected in 3 (50%) of 6 A. ussuriensis, 70 larvae in 3 of 7 (42.9%) R. tigrinus tigrinus, and 2 larvae in 2 of 82 (8.7%) frogs. No gnathostome larvae were detected in tadpoles and cat snakes. The larvae detected were a single species, and $2.17{\times}0.22mm$ in average size. They had characteristic head bulbs, muscular esophagus, and 4 cervical sacs. Three rows of hooklets were arranged in the head bulbs, and the number of hooklets in each row was 29, 33, and 36 posteriorly. All these characters were consistent with the advanced third-stage larvae of G. nipponicum. It has been first confirmed in Jeju-do that R. nigromaculata, A. ussuriensis, and R. tigrinus tigrinus play a role for intermediate and/or paratenic hosts for G. nipponicum.

Adaptive Garbage Collection Policy based on Analysis of Page Ratio for Flash Memory (플래시 메모리를 위한 페이지 비율 분석 기반의 적응적 가비지 컬렉션 정책)

  • NAND flash memory is widely used in embedded systems because of many attractive features, such as small size, light weight, low power consumption and fast access speed. However, it requires garbage collection, which includes erase operations. Erase operation is slower than other operations. Further, a block has a limited erase lifetime (typically 100,000) after which a block becomes unusable. The proposed garbage collection policy focuses on minimizing the total number of erase operations, the deviation value of each block and the garbage collection time. NAND flash memory consists of pages of three types, such as valid pages, invalid pages and free pages. In order to achieve above goals, we use a page ratio to decide when to do garbage collection and to select the target victimblock. Additionally, we implement allocating method and group management method. Simulation results show that the proposed policy performs better than Greedy or CAT with the maximum rate 85% of reduction in the deviation value of the erase operations and 6% reduction in garbage collection time.

Western blot analysis of stray cat sera against Toxoplasma gondii and the diagnostic availability of monoclonal antibodies in sandwich-ELISA

  • A total of 198 sera from stray cats was assayed against Toxoplasma gondii antigen by western blot. Out of 198 sera assayed, 26 sera (13.1%) showed typical blot patterns against T gondii. When spotted by ELISA absorbance and indirect latex agglutination lest (ILAT) titer, all 26 cases were distributed over the cut-off value of ELISA whereas 24 cases (92.3%) were in the positive range of 1:32 or higher and 2 cases in negative range by ILAT. Among western blot negative 172 sera, 162 cases were negative in both ILAT and ELISA while 10 cases were reactive falsely such that three cases were ILAT positive with 1:32 titer and 9 cases were ELISA positive (2 cases overlapped). These 10 cases reacted peculiarly without typical binding pattern in Western blot. Sandwich-ELISA was performed with monoclonal antibodies (mAbs) of Tg563 (30 kDa, SAG 1), Tg505 (22 kDa, SAG2), Tg605 (43 kDa, SAG3), Tg556 (28 kDa, GRA2), Tg737 (32 kDa, GRA6). Tg695 (66 kDa, ROP2), Tg786 (42 kDa, ROP6), and Tg621 (32 kDa, anonymous but cytosolic) clone, respectively. All western blot-positive cases were in the positive range and negative cases in the negative range clearly. Among the 10 false reactive cases, 3 cases were in the positive range with one or more mAbs. All mAbs used in this study were confirmed to be specific to T. gondii infection as a standardized sandwich-ELISA to differentiate it from other pathogens.

Protective Effect of Galgeun-Tang Against $CCl_4$ Induced Hepatotoxicity (갈근탕의 사염화탄소에 의한 간세포 독성 억제효과)

  • Galgeun-tang (GGT) has been a great source for treating cold diseases in the folk medicine recipe. Carbon tetrachloride ($CCl_4$) is one type of hepatotoxin that can eventually cause liver injury. During the experiment, we first studied the protective effects of GGT against $CC_4$-induced hepatotoxicity. GGT was pretreated for 3 h, and 1% $CCl_4$ was added to mouse primary liver cells. After 4 h, ROS generation and expression of antioxidant enzymes (catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx)) were analyzed by FACS and real time PCR. Also, the activities of ALT and LDH were measured using cultured medium. The hepatic levels of TNF-alpha and iNOS, which are related to inflammation and stress response gene, HSP72 and HO-1 were analyzed by PCR or real time PCR. Liver tissues were analyzed by HE stain. From the observation, we discovered that GGT treatment protects $CCl_4$-induced hepatotoxicity, and that GGT pretreatment decreases ROS generation, TNF-alpha and iNOS expression. However, gene expression of CAT, SOD, GPx, HSP72 and HO-1 were increased by GGT. These results lead to the conclusion that GGT has protective effects against $CCl_4$-induced hepatotoxicity.

Purification and Characterization of Xylanase from Fomitopsis palustris in Rice Straw Culture (볏짚분해과정 중에 생산하는 Fomitopsis palustris 균체 외 Xylanase의 분리정제 및 효소특성)

  • An extracellular xylanase from the brown-rot fungus Fomitopsis palustris grown on rice straw culture was purified to a single protein band. On SDS-PAGE, the molecular mass of purified enzyme was estimated to be about 43 kDa. The amino acid sequence of the proteolytic fragments showed high homology with fungal glycoside hydrolase family 10 xylanases. The $K_m$, $K_{cat}$ and $V_{max}$ for birch xylan were $31mg/m{\ell}$, $2.3{\times}10^4/min$ and 252.3 U/mg, respectively. The optimal activity of the purified xylanase from F palustris was observed at pH 4.0~5.0 and $70^{\circ}C$.

Effect of Dioscorea Rhizoma on Gastritis by Acute Gastric Ulcer in Rats (산약이 흰쥐의 급성 위궤양에 의한 위장 장애에 미치는 효과)

  • In order to verify the effects of Dioscorea Rhizoma on gastrointestinal damages, we investigated the protective role of the crude extracts on induced gastric ulceration in rats. Rats administered of Dioscorea Rhizoma extracts showed gradually decreased congestion and hemorrhage, but control group did not show any symptom in gastric tissue. Moreover, Dioscorea Rhizoma extract had a role in lowering gastrin and histamine levels in gastric ulcer rats, thereby inhibiting the gastric tissue damages. Excessive production of malondialdehyde shown in gastric ulcer rats was declined in all rats administered with Dioscorea Rhizoma extract as well as the levels of SOD and GPX surged by acute gastric ulcer. Also, the increased activity of CAT showed an effect in activation of antioxidant enzyme to normal state. All data suggest that Dioscorea Rhizoma extract was verified to be highly effective resource in improving the gastrointestinal function of rats by preventing from gastric tissue damage in acute gastric ulcer and restoring the activities of plasma substances and antioxidant enzymes.

Effect of Natural Functional Mixture on the Descent of Blood Glucose Level in Streptozotocin-Induced Diabetic(type I) Rats(II) (Streptozotocin 유발 제1형 당뇨 쥐의 혈당강하에 대한 천연 기능성 소재 혼합물의 효과(II))

  • The aim of this study was designed to evaluate the effects of natural functional mixture(FM) on plasma BUN and lipid levels, hepatic lipid levels, hepatic antioxidant enzyme activities and plama aminotransferase activity in streptozotocin (STZ) induced diabe1ic rats. Total cholesterol (TC) level in the diabe1ic rats supplemented with FM(70.69 mg/dL) was reduced comparing to groups without FM(87.12 mg/dL). This results caused the increase of the ratio of HDL-cholesterol to TC (42.60 to 51.49 %). However, superoxide dismutase (SOD), cytosol catalase (CAT), glutathione peroxidase (GSH-Px), GSH and lipoperoxide (LPO) activities were not significantly changed, which indicated the supplementation with FM could not reduce the oxidative stress in diabetic rats. In addition, asperate aminotransferase (AST) activity in FM-diabetic rat was lower than that in diabetic group. This results showed supplementation with FM in rats could improve the hepatic function damaged by STZ-induced diabetes.

  • Skin is constantly exposed to air, solar radiation, ozone and other air pollutants formulating free radicals. The reactive oxygen species(ROS), formed under these conditions, are associated with skin cancers, cutaneous photoaging, and cutaneous inflammatory disorders. In this study, we sought to establish an animal model for UVB-induced skin alteration using BALB/c mice. The level of UVB irradiation used in this model was within physiological dose. BALB/c mice were exposed to a single dose of UVB ($200mJ/cm^2$ and were sacrificed at 3, 6, 24, and 48 hours following the irradiation. The effect of a single exposure to UVB irradiation on skin catalase(CAT), superoxide dismutase(SOD), and glutathione peroxidase(GPx) activities were examined. Significant decrease in the activity of all enzymes were observed at 6 hours after irradiation(p<.05). The activity of CAT decreased more sharply than those of SOD and GPx, and then remained depressed until 48 hours after UVB irradiation, whereas the activity of GPx recovered to basal level at 48 h after UVB irradiation. Our results indicate that BALB/c mouse could be an adequate animal model of UVB irradiation experiment. These results will also provide fundamental knowledge for the effective nursing strategies in reducing UV-induced skin disorders.

Comparison of limited- and large-volume cone-beam computed tomography using a small voxel size for detecting isthmuses in mandibular molars

  • Purpose: This study was performed to compare the ability of limited- and large-volume cone-beam computed tomography (CBCT) to display isthmuses in the apical root canals of mandibular molars. Materials and Methods: Forty human mandibular first molars with isthmuses in the apical 3 mm of mesial roots were scanned by micro-computed tomography (micro-CT), and their thickness, area, and length were recorded. The samples were examined using 2 CBCT systems, using the smallest voxels and field of view available for each device. The Mann-Whitney, Friedman, and Dunn multiple comparison tests were performed (α=0.05). Results: The 3D Accuitomo 170 and i-Cat devices detected 77.5% and 75.0% of isthmuses, respectively (P>0.05). For length measurements, there were significant differences between micro-CT and both 3D Accuitomo 170 and i-Cat(P<0.05). Conclusion: Both CBCT systems performed similarly and did not detect isthmuses in the apical third in some cases. CBCT still does not equal the performance of micro-CT in isthmus detection, but it is nonetheless a valuable tool in endodontic practice.

Isolation and molecular characterization of feline panleukopenia viruses from Korean cats

  • Feline panleukopenia virus (FPV) causes fatal leukopenia and severe hemorrhagic diarrhea in cats. Although FPV isolates have been reported worldwide from several animals, the biological and genetic features of South Korean FPVs remain unclear. We characterized molecularly South Korean FPV isolates. Crandell-Rees feline kidney (CRFK) cells were used to isolate FPV from 60 organ homogenates. The isolates were confirmed to be FPVs via analyses of cytopathic effects, immunofluorescence studies, electron microscopy, and polymerase chain reaction. Viral genetic analyses used the full VP2 sequences. Eight isolates propagated in CRFK cells were confirmed to be FPVs. All isolates yielded viral titers ranging from 104.5 to 106.0 TCID50/mL 5 days after inoculation into CRFK cells and exhibited hemagglutination titers ranging from 27 to 212 (using pig erythrocytes). The Korean FPV isolates grew well in cat cells such as CRFK and Fcwf-4 cells. The FPV isolates were most similar to the KS42 strain isolated from a Korean cat in 2008. The FPV isolates will serve as useful antigens in future sero-epidemiological studies and will aid in the development of diagnostic tools.
