During an investigation into the indigenous prokaryotic species diversity in Korea, a total of five bacterial strains were isolated from various environments in Korea. The isolated bacterial strains were identified by analyzing their 16S rRNA gene sequences, and those with a minimum of 98.7% sequence similarity with known bacterial species but not reported in Korea were designated as unrecorded species. These isolates were assigned to three phyla, five orders, five families, and five different genera. The isolates were identified as Cumulibacter manganitolerans (99.1%) and Myolicibacterium tusciae (98.7%) of the class Actinomycetes; Bacillus marasmi (99.9%) of the class Bacilli; and Novosphingobium mathurense (100%) and Microvirga ossetica (98.8%) of the class Alphaproteobacteria. Gram reaction, colony and cellular morphology, basic biochemical characteristics, and phylogenetic position of theses isolates are also described.

Several genera belonging to the family Acetobacteraceae were generally considered to be acetic acid bacteria (AAB) which produce acetic acid from alcohols using an oxidation pathway. Some species of the family Acetobacteraceae have been of interest to the industry due to their capability to produce vinegar. In 2018-2020, several bacterial strains were isolated from plants, fruits and vinegar in various regions of the Republic of Korea. Based on the 16S rRNA gene sequences, seven species classified into the family Acetobacteraceae were found to be ones unrecorded in the Republic of Korea, including two Asaia species, one Gluconacetobacter species, three Gluconobacter species and one Komagataeibacter species. As a result, we report Asaia lannensis AF11C3 (=KACC 22050) isolated from plant Chrysanthemum zawadskii, Asaia platycodi AF15C2 (=KACC 22051) isolated from plant Isodon inflexus, Gluconacetobacter liquefaciens C23-3 (=KACC 22064) isolated from fruit of Pyrus pyrifolia, Gluconobacter cerinus BGF2-R2 (=KACC 22053) isolated from fruit of Prunus armeniaca, Gluconobacter kondonii FR39A4 (=KACC 22388) isolated from persimmon fruit, Gluconobacter thailandicus FR36C4 (=KACC 22057) isolated from fruit of Pseudocydonia sinensis and Komagataeibacter melaceti SPV-2 (=KACC 22058) isolated from potato vinegar.

During a comprehensive investigation of indigenous prokaryotic species in Korea, 20 and one bacterial strains assigned to the phyla Bacillota and Verrucomicrobiota, respectively, were isolated from diverse environmental habitats, including soil, mud, tidal flat, seawater, sand, sediment, brackish water, and healthy human urine. Based on their high 16S rRNA gene sequence similarities (>98.7%) and the formation of robust phylogenetic clades with their closest related reported species, each strain was assigned to an independent and predefined bacterial species. Since there were no published or official reports regarding these 21 isolates in Korea, they - 20 species of four families in two orders of the phylum Bacillota and one species of the phylum Verrucomicrobiota - have been reported as unrecorded species in Korea. In addition, Gram staining, colony and cell morphology, basic biochemical characteristic, isolation source, and strain ID of each species are also described in the species description sections.

During screening for indigenous prokaryotic species in Republic of Korea in 2021, a total of 21 bacterial strains assigned to the phylum Bacteroidota were isolated from a variety of environmental habitats including pine cone, seaweed, soil, sea sediment, brackish water and moss. Based on the 16S rRNA gene sequence similarity value of more than 98.7% and formation of a robust phylogenetic clade with the type strain of the closest bacterial species, it was found that the 21 strains belong to independent and recognized bacterial species. There has been no official report that the identified 21 species have been isolated in Republic of Korea up to date. Therefore, 16 species in six genera of two families in the order Flavobacteriales, two species in two genera of two families in the order Cytophagales, one species in one genus of one family in the order Chitinophagales and two species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidota isolated in Republic of Korea. Their Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.

In 2021, a total of 27 bacterial strains were isolated from soil, tree bark, moss, wetland, sea sediment, tidal flat, seawater and seaweed within Republic of Korea. Based on the analysis of 16S rRNA gene sequence (>98.7% sequence similarity), these isolates were assigned to the class Alphaproteobacteria as unrecorded species in Korea. The 27 strains were classified into the 10 families: Maricaulaceae of the order Caulobacterales; Brucellaceae, Methylobacteriaceae, Nitrobacteraceae and Rhizobiaceae of the order Hyphomicrobiales; Micropepsaceae of the order Micropepsales; Rhodobacteraceae of the order Rhodobacterales; Azospirillaceae of the order Rhodospirillales; and Erythrobacteraceae and Sphingomonadales of the order Sphingomonadaceae. There is no official report of these 27 species in Korea. Therefore, we report 27 isolates as unrecorded species, and described isolation sources, Gram-stain reactions, physiological and biochemical properties and morphologies of these strains.

In July 2018, solar saltern samples were collected from Siheung, Gyeonggi-do Province to obtain unrecorded haloarchaea in Korea. The samples were suspended in a 20% NaCl (w/v) solution, and serial dilution was performed in fresh DB Characterization media No. 2. The strains isolated in this study showed at least 98.7% sequence similarity or more compared to the previously reported. Finally, 16 haloarchaeal strains, which were not reported in Korea but validly published under the International Code of Nomenclature of Prokaryotes (ICNP), were obtained from a solar saltern in Siheung. These 16 isolates were allocated to the orders Halobacteriales and Haloferacales. The 10 Halobacteriales strains were classified into the family Halobacteriaceae and Haloarculaceae. Each family belonged to three genera, respectively. The other six Haloferacales belonged to the families Haloferacaceae and Halorubraceae. Each family belonged to genus genus, respectively. Collectively, the unrecorded haloarchaeal strains belonged to two orders, four families, and eight genera. During the research, the possibility of discovering previously unknown species in domestic solar saltern was established. Gram-staining, cell morphology, physiological and basic biochemical parameters, and phylogenetic analysis were all performed in this study and are described in detail for each strain.

To obtain unrecorded bacterial species from Korean islands, various samples were collected from the islands in 2022. After plating the samples on marine agar or Reasoner's 2A, and incubating aerobically, approximately 1,200 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 10 strains showed ≥98.7% 16S rRNA gene sequence similarity with the bacterial species that were validly published but not reported in Korea. The unrecorded bacterial strains belong to three phyla, five classes, 10 orders, 10 families, and 10 genera, which are assigned to Sphingomonas, Falsirhodobacter and Asticcacaulis of the class Alphaproteobacteria; Colwellia and Halomonas of the class Gammaproteobacteria; Chitinophaga of the class Chitinophagia; Chryseobacterium of the class Flavobacteriia; Microlunatus, Zhihengliuella, and Streptomyces of the class Actinomycetia.
