90 wild Agaricus strains from China, including 44 Agaricus bisporus strains identified preliminarily by isozyme electrophoresis, were studied by the techniques of SRAP and ISSR. 18 special SRAP bands and 12 special ISSR bands were analyzed, the strains were clustered and a demdrogram was obtained. The results showed that the strains were divided into 2 groups, wild A. bisporus group and the other Agaricus group. It is similar to the result of isozyme electrophoresis. 41 wild A. bisporus strains from Sichuan and Tibet were divided into 4 groups based on their growing places, suggesting the regionally difference of the strains to be quite obvious. Some white wild A. bisporus strains from Xinjiang and Tibet had special patterns, resulting in lower coefficient values with other wild A. bisporus strains. The biological characteristics of three wild A. bisporus strains were analyzed, and the results showed: 1. The wild strains grew slowly on PDA medium with weak appressed mycelia, and grew normally in kernel or fermented cottonseed shell substrate. 2. They grew faster than control strain As2796 under lower temperature of
$16^{\circ}C$ , and higher temperature of$32^{\circ}C$ , with optimum growing temperature of$20-24^{\circ}C$ , which was$4^{\circ}C$ lower than that of control strain. 3. In the cultivation with manure compost via twice fermentation, the mycelia grew normally in compost and quite slowly in casing soil, and the fruitbodies occurred less and late with easily opening and low production. 4. The fruitbody was off-white with flat and scaled cap, long stipe and dark gill. The bisporus basidia occupied 70-80% and trisporus basidia 20-30% of the total basidia. 5. Heterokaryotic monospore isolates could fruit in cultivation, and the homokaryotic isolates could cross with those derived from overseas wild A.bisporus strains. 6. The electrophoresis phenotype of isozymes such as esterase etc. belonged to high production type (H type). 7. The RAPD patterns made much difference from those of high production, good quality or hybrid strains, which indicated that the wild strains produce a new kind of RAPD type. -
Poria cocos is an importantant medical macrofungus,the sclerotium of Poria cocos has specific value as the drug material. There are few papers about its breeding and spawn rejuvenation. In this project, the protoplasts of cultivated strain T and wild strain L were prepared and treated separately by ultraviolet and heating, then fused with the PEG6000. The tural fusants were selected and identified by the affinity and ISSR analysis. 71 incompatibility strains between parents and reg regenerations were obtained from 118 regenerations by the affinity analysis. Five incompatibility strains were amplified with different primers, the results were showed that they had specific bands of both parents in the profile amplified with 3 primers, which proved these 5 strains were fusants by means of molecular biology marker. On the other hand, 25 strain were selected from 168 protoplast regenerations of cultivated strain T for cultivation experiment. The fresh sclerotium weight of these protoplast regenerations were better than the original strain.significantil 3 strains (T-1, T-4, T-7) increased respectively 118%, 73% and 73% than original strain. This method could be the effective in the rejuvenation Poria cocos.

$Epichlo\ddot{e}$ endophytes, including Neotyphodium spp. and$Epichlo\ddot{e}$ spp., enhance plant growth, mediate more plant tolerance or resistance to biotic and abiotic stresses, and also synthesis various biologically active compounds in their host plants, and important in many areas. In early stages, most of$epichlo\ddot{e}$ endophytes were described during surveys practiced by American, European and Oceania scientists, while fungal endophytes within native Asian plants were poorly investigated. In recent years, an$Epichlo\ddot{e}$ sp. and 4 Neotyphodium spp. were described in cool season Chinese native gramineous plants. Most of Chinese native Neotyphodium spp. were presumed as hybrids originated from members of ETC and EBY. Investigation on NRPS genes shows lack of toxic ergopeptines and potential production of peramine. Biological and ecological roles of Chinese native$epichlo\ddot{e}$ endophytes should be investigated in future, and it will be very valuable if we can have some joint projects with Korean scientists for Asian native$epichlo\ddot{e}$ endophytes. -

In the fungal genus Cordyceps, the type species Cordyceps militaris produces bioactive ingredients and exhibits medicinal effects as a Traditional Chinese Medicine (TCM), The fruiting bodies of C.militaris have now been mass-produced artificially and used as functional food and medicine in China. The unstable variation in forming fruiting body is however a key restrictive factor in industrial production. The genetic study on in vitro stromata formation of C. militaris has rarely been carried out. Here, we report the effects of genetic variation including the mating system on perithecial stromata formation of C. militaris. Monoconidial isolates which have both MAT1-1-1 and MAT1-2-1(genotype MAT1-1/2) could produce stromata. While the isolates only have either MAT1-1-1 or MAT1-2-1 (genotype MAT1-1 or MAT1-2) failed to produce stromata. Despite obvious heterothallism, homothallism was occasionally observed in a few isolates of C. militaris. High genetic variation was observed amongst the different monoconidial isolates of C. militaris. The unstable variation or lose of fruiting body formation was found to be caused by the inner-species high genetic variation of C. militaris. These results also indicated that C. militaris sexually behaved as both heterothallic and homothallic and required two mating type compatible in the same culture in order to produce regular clubshaped perithecial stromata.
