Aim & Scope
The Food Science and Biotechnology (Food Sci. Biotechnol.; FSB) was launched in 1992 as the Food Biotechnology and changed to the present name in 1998. It is an international peer-reviewed journal published monthly by the Korean Society of Food Science and Technology (KoSFoST). The FSB journal covers; Food chemistry/food component analysis Food microbiology and biotechnology Food processing and engineering Food hygiene and toxicology Biological activity and nutrition in foods Sensory and consumer science s Consumer perception and sensory evaluation on processed foods are accepted only when they are relevant to the laboratory research work. As a general rule, manuscripts dealing with analysis and efficacy of extracts from natural resources prior to the processing or without any related food processing may not be considered within the scope of the journal. The FSB journal does not deal with only local interest and a lack of significant scientific merit. The main scope of our journal is seeking for human health and wellness through constructive works and new findings in food science and biotechnology field.

The effect of alginate coating in combination with an anti-browning agent on increasing the post-cutting shelf life and improving the quality of minimally processed apple cubes was studied during storage at room temperature for 5 days. A simple coating technique involving the chemical cross-linkage of alginate by calcium was used. Statistical-based experimental designs were applied to improve the quality of the alginate-coated apple cubes (ACAC). Plackett-Burman design was first used to determine the main factors influencing the preservation of the original weight, color, and texture of ACAC. Among these variables, alginate concentration (
$X_1$ ), dipping time ($X_2$ ), and dipping temperature ($X_3$ ) significantly influenced the ACAC weight and color (confidence levels above 90%). Subsequently, the effects of the 3 main factors were further investigated by a central composite design. The polynomial models developed by response surface methodology were adequate to describe the relationships between the studied factors and the responses. Overall optimization conducted by superimposing the curves of the responses enabled the determination of an optimal range of the independent variables in which the five responses were simultaneously optimized. The point chosen as representative of this optimal area corresponded to$X_1=2.98%$ ,$X_2=0.85\;min$ , and$X_3=55^{\circ}C$ and under these conditions the model predicted weight loss=0.522%, relative hardness=1.517,${\Delta}E=1.423$ , browning inhibition=93.403%, and${\Delta}L=0.158$ . -
To determine whether alcohol-treated rat liver cells can be protected by a static magnetic field (SMF), we analyzed the blood chemistry and histology of hepatic tissue removed from alcohol-exposed rats that had been exposed to a static magnetic field. The rats were exposed to a 0.3 tesla (3,000 gauss) magnetic field (MF) for 24 hr daily for 5 weeks with appropriate controls. Glutamic pyruvic transaminase activity and the triglyceride levels in animals exposed to the north (N) or south (S) pole of the MF decreased significantly (p<0.01 and p<0.05, respectively) compared with negative control animals with alcohol exposure. A histological examination of hepatic tissue revealed a moderate to severe accumulation of fat vacuoles of various sizes in the cytoplasm of the hepatocytes of animals in the negative control group throughout the study; whereas in groups exposed to the MF poles, fewer fat vacuoles were seen compared with the negative control group. Electron microscopic observations showed that exposure to the N or S pole protected organelles, including the nucleus, from damage during exposure to this toxic agent, as indicated by the fact that the nucleus and the mithochondria virtually retained their shape throughout this study. These results suggest that exposure to a SMF could be an excellent way of protecting against alcohol-induced damage to the rat liver cell.

The chemical processes involved in the formation of green pigment in crushed garlic cloves were investigated based on the principle of pink pigmentation in macerated onions. Intact greening and non-greening garlic cloves were either left untreated or heated at
$90^{\circ}C$ for 3 min to inactivate enzyme activities. First, a colorless ether soluble compound referred to as color developer reacted with glycine (among all free amino acids) in garlic to form a second compound insoluble in ether. The latter compound then reacted with formaldehyde to yield the green colored pigment. Alliinase activity was necessary for the production of color developer and for the development of green pigment. In greening garlic that had been heat treated, green pigmentation did not proceed due to the heat-inactivation of alliinase, but the addition of alliinase solution into the garlic homogenates restored the pigmentation. However, this phenomenon was not observed in non-greening garlic with or without heat treatment. Finally, the mechanism of green pigment formation in crushed garlicis similar to that of pink pigment formation in macerated onions. -
This study was designed to compare various quality characteristics, such as nutrient composition and physicochemical and sensory properties of freeze-concentrated milk made by a newly developed continuous multi-stage process with those of evaporated milk. The freeze concentration process reduced the water content up to 73%. Most of the physicochemical properties of evaporated milk were different from raw milk; however the freeze-concentrated milk showed little difference from the raw milk. The thiobarbituric acid value and free fatty acid concentrations were significantly greater in the evaporated milk than in the freeze-concentrated milk. Several effects on sensory characteristics, such as off-taste, were significantly stronger in the evaporated milk. Overall, this study indicates that the newly developed freeze concentration technique results in improved physicochemical and sensory properties, and has little effect on most nutrient levels when compared with the evaporation process. Further research is necessary to further elucidate the chemical and sensory properties of freeze-concentrated milk.

Acarviosine-glucose (AcvGlc) is an${\alpha}$ -glucosidase inhibitor and has similar inhibitory activity to acarbose in vitro. We synthesized AcvGlc by treating acarbose with Bacillus stearothermophilus maltogenic amylase and fed C57BL/6J and db/db mice with diets containing purified AcvGlc and acarbose for 1 week. AcvGlc (50 and 100 mg/100 g diet) significantly reduced plasma glucose and triglyceride levels in db/db mice by 42 and 51 %, respectively (p<0.0001). The hypoglycemic and hypotriglyceridemic effects of AcvGlc were slightly, but significantly, greater than those seen with acarbose treatment (p<0.0001) in C57BL/6J mice. In an oral glucose tolerance test, glucose tolerance was significantly improved at all time points (p<0.01). The expression of two novel glucose transporters (GLUTs), GLUT10 and GLUT12, were examined by Western blot analysis. GLUT10 was markedly increased in the db/db livers. After AcvGlc treatment, the expression of hepatic GLUT10 was decreased whereas intestinal GLUT12 was significantly increased in both strains of mice. Our results show that AcvGlc improves plasma lipid and glucose metabolism slightly more than acarbose. Regulation of hepatic GLUT10 and intestinal GLUT12 may be important in controlling blood glucose levels. -
The effects of moisture content on non-fracture dynamic properties and fracture gel quality of Pacific whiting surimi were investigated to determine their relationships. Surimi samples were tested at various moisture contents (75, 78, and 81 %). Torsion test showed that shear stress decreased rapidly and strain values decreased gradually as moisture concentration increased. Dynamic storage modulus (G') also decreased as moisture content increased. A strong positive correlation (
$R^2=0.90$ to 0.99) was found between the G' measured at temperatures between 10 and$45^{\circ}C$ and fracture stress values. The results indicate that dynamic rheological measurements could be used as a tool for early gel quality assessment. -
The quality parameters of sponge cake supplemented with soy protein concentrate (SPC) were evaluated. The addition of SPC to wheat flour increased the protein content and alkaline water retention value, but decreased the sedimentation value. Protein content had a positive correlation with the alkaline water retention value, and a negative correlation with the sedintentation value. The higher the concentration of SPC, the higher the RVA pasting temperature and the lower the viscosity. Increasing the level of SPC in flour led to a decrease in mixogram peak time, whereas peak height, width at peak, and width at 8 min progressively increased. As the amount of SPC increased in the formulation, the pH and specific gravity of cake batter increased, whereas the volume and specific volume of sponge cake decreased. The total isoflavones content in SPC increased after heat treatment. The hardness, gumminess, and chewiness increased progressively in accordance with increasing level of SPC.

The maintenance of physiological activity during dilution is very critical for the accurate enumeration of Vibrio spp. in marine samples. We investigated the effect of various diluents on the recovery of Vibrio parahaemolyticus using the direct plate counting and most probable number (MPN) methods. The effects of NaCl (0.85 and 3%) and pH (from 6.6 to 7.4) in diluents based on distilled water or phosphate buffered saline (PBS) were evaluated with three V. parahaemolyticus strains. PBS-3% NaCl (pH 6.6), as opposed to PBS, was the most effective diluent at maintaining viable cell numbers up to 2 log CFU/g during dilution for direct plate counting using on thiosulfate citrate bile salts sucrose (TCBS) selective agar, as well as minimizing the difference in cell numbers between TCBS and non-selective nutrient agar. It also increased counts of V parahaemolyticus inoculated into oysters relative to PBS (p<0.01), suggesting that PBS-3% NaCl (PH 6.6) can reduce the problem of underestimating V. parhaemolyticus counts using PBS alone.

Rheological behavior of commercial hot pepper-soybean paste (HPSP) was evaluated in small amplitude oscillatory and steady shear tests. Storage modulus (G'), loss modulus (G"), and complex viscosity (
${\eta}^*$ ) as a function of angular frequency (${\omega}$ ), and shear stress (${\sigma}$ ) as a function of shear rate (${\gamma}$ ) data were obtained for 5 commercial HPSP samples. HPSP samples at$25^{\circ}C$ exhibited a non-Newtonian, shear-thinning flow behavior with high yield stresses and their flow behaviors were described by power law, Casson, and Herschel-Bulkley models. Time-dependent flow properties were also described by the Weltman, Hahn, and Figoni & Shoemaker models. Apparent viscosity over the temperature range of$5-35^{\circ}C$ obeyed the Arrhenius temperature relationship with activation energies (Ea) ranging 18.3-20.1 kJ/mol. Magnitudes of G' and G" increased with an increase in${\omega}$ , while${\eta}^*$ decreased. G' values were higher than G" over the most of the frequency range (0.63-63 rad/sec), showing that they were frequency dependent. Steady shear viscosity and complex viscosity of the commercial HPSP did not fit the Cox-Merz rule. -
The immunostimulatory activities of Lactobacillus plantarum, the major microorganism in kimchi fermentations were investigated. Five strains of L. plantarum exhibited weak immunopotentiating activity, but L. plantarum PS-21 showed as strong a mitogenic activity as Bifidobacterium adolescentis M101-4, a known positive strain. It is of interest that, L. plantarum PS-21 stimulated proliferation of Peyer's patch cells, one of the most important tissues in the gut-associated lymphoreticular system. Cell' wall fractions from L. plantarum PS-21 also showed strong mitogenic activity compared with the soluble cytoplasmic fraction. A peptidoglycan fraction (PG) extracted from the cell wall of L. plantarum PS-21 was identified as an active mitogenic component when used in murine lymph node and spleen cell test systems. PG showed dose-dependent mitogenic activity and significantly enhanced antibody production in lymph node cells when studied in vitro. The lysosomal enzyme activity of murine peritoneal macrophages was increased when analyzed following injection of PG to the host animal. Furthermore, PG enhanced the production of cytokines such (
$TNF-{\alpha}$ and IL-6) in the in vitro culture of RAW 264.7 macrophage cells. -
In this study, we have attempted to characterize the effects of encapsulated mustard oil with regard to the extension of the shelf life of kimchi. The quantity of mustard oil or encapsulated mustard oil added to the brined cabbage in this study was 0.05%(w/w). Overall, the fermentation processes in the encapsulated mustard oil-added kimchi (EMO) and mustard oil-added kimchi (MO) occurred at a slower rate than in the controls. The periods during which the MO and EMO kimchi samples were edible were prolonged for more than two weeks, according to measurements of pH, total acidity, and microbial changes. The overall acceptability of EMO was superior to MO throughout the entirety of the fermentation period. Sensory evaluation verified that EMO yielded a more favorable product than the MO and control varieties. Encapsulated mustard oil can be employed as a natural food additive to prolong the shelf life of kimchi, via an induced delay of the fermentation process.

Cytotoxic effects of extracts from Tremella fuciformis strain FB001 were evaluated on the DLD-1 human colon adenocarcinoma cell line and the content of polyphenolic compounds in the extracts were analyzed. Hexane, chloroform, and ethyl acetate subfractions (experimental setting I) exhibited cytotoxic effects on the human colon adenocarcinoma DLD-1 cell line with
$IC_{50}$ values of 350, 400, and 450 ppm, respectively. When T. fuciformis was extracted sequentially with ether, ethyl acetate, chloroform, and ethanol (experimental setting II), the ether extract demonstrated potent cytotoxicity with an$IC_{50}$ value of 150 ppm, followed by ethyl acetate and chloroform fractions. If the first extraction solvent was chloroform instead of ether (experimental setting III), exposure of the cell line to chloroform, ethyl acetate, and ether extracts at 1,000 ppm led to cell death. High levels of phenolic compounds were estimated for all hydrophobic extracts, which exhibited cytotoxic effects. We propose that this useful information gives additional support to our understanding of the biology and utility of this particular mushroom. -

Patulin is a food mycotoxin which induces genotoxicity and acute intestinal disease in infants. Patulin mainly originates from fruit putrefactive moulds, especially in apples, which necessitates the maintenance of strong safety standards against patulin for fresh and processed apples. To investigate the patulin producing moulds in Korean apples, 16 morphological types of fungi were isolated from Korean apples and a patulin producing fungus was identified based on a sequence analysis of the region of internal transcribed spacers (ITS5-5.8S-ITS4 region, 505 base pair) and the 26 rRNA D1/D2 region (527 base pair). Morphological analyses were also performed. The isolated patulin producing fungus was found to a representative species of Penicillium crustosum. The maximal patulin production ability of the isolated fungus (P. crustosum) and the patulin producing standard strain (P. griseofulvum, ATCC 46037) in an SY broth medium were 0.32 and 2.46 mg/L, respectively. -
Freeze dried crude bacteriocin was encapsulated within an acid-soluble coating material, Eudragit EPO, using a surface modification technique through a hybridization system. The pH and titratable acidity of control yoghurt were 3.92 and 1.56%, respectively, after 24 hr of fermentation at
$42^{\circ}C$ , whereas yoghurt containing 500 AU/mL encapsulated bacteriocin exhibited a higher pH (4.37) and lower titratable acidity (1.2%). Yoghurt containing encapsulated bacteriocin had significantly lower titratable acidity when the duration of fermentation (to pH 4.5) and subsequent refrigerated storage ($4^{\circ}C$ ) was longer than 20 days. There were no significant differences in the viability of lactic acid bacteria after 15 hr of fermentation. This suggests that microencapsulated bacteriocin has the potential to control the excessive growth of yoghurt starters caused by temperature abuse or post-acidification. -
Recombinant
${\beta}$ -galactosidase from Bifidobacterium longum LL04 was expressed in Escherichia coli and partially purified by ammonium sulphate precipitation and anion-exchange chromatography (Mono-Q). The optimum temperature and pH of the partially purified enzyme were$50^{\circ}C$ and pH 7.0-8.0, respectively, when o-nitrophenyl-${\beta}$ -D-galactopyranoside was used as a substrate. The enzyme was stable over the pH range of 5.0-9.0, and was active at$40^{\circ}C$ for more than 60 min at pH 7.0. The enzyme was significantly activated by$Na^+$ and$K^+$ . Maximal activity was observed at the concentration of 10 mM for both$Na^+$ and$K^+$ . The enzyme activity was strongly inhibited by most bivalent metal ions. The Km and Vmax on ONPG at 37 and$50^{\circ}C$ were 0.72, 167.9, and 0.507 mM, 310.9 U/mL, respectively. -

We investigated the catechin content in green tea leaf (GTL) and green tea seed (GTS), the antiproliferative and detoxifying phase II enzyme-inducing activities of the methanolic (80%, v/v) extracts from GTL and GTS. GTL and GTS contained$8,685{\pm}1,061$ and$108{\pm}32\;{\mu}g/g$ epigallocatechin gallate (EGCG),$11,486{\pm}506$ and$116{\pm}72\;{\mu}g/g$ epigallocatechin (EGC),$3,535{\pm}308$ and$821{\pm}95\;{\mu}g/g$ epicatechin gallate (ECG), and$1,429{\pm}177$ and$37{\pm}44\;{\mu}g/g$ epicatechin (EC), respectively. The methanolic extract of GTS showed a greater increase in quinone reductase activity and antiproliferation potential against mouse hepatoma cells than GTL extract did. GTS treatment resulted in the accumulation at sub-G1 phase of mouse hepatoma hepa1c1c7 cells as assessed by flow cytometry. Enhancement of phase II enzyme activity by GTS extract was shown to be mediated, directly or indirectly, via interaction with the antioxidant response element (ARE) sequence in the genes encoding the phase enzymes. As the catechin content in GTS was significantly lower than that in GTL, components other than catechins appear to be responsible for the anticarcinogenic activity of the seed. In summary, these results suggest that the 80% methanolic extract of GTS deserves further study to evaluate its potential as an anticarcinogenic agent and to investigate its mechanism of action. -
The microbiological safety of foodservice facilities and side dishes in 6 elementary schools and 6 universities in the Jeolla-do region was evaluated. The micro-aerosol evaluation of foodservice facilities including the kitchen, refrigerators, and freezers showed a comparatively high level of microbial contamination. In the microbial safety evaluation of cooking utensils and appliances, the total plate counts of serving tables, cutting boards, and food plates were comparatively high, but did not reveal significant counts of coliforms, Staphylococcus aureus, and Escherichia coli were both below the general limit of microbial contamination. The microbiological safety of the cooking utensils and appliances were satisfactory. In the microbiological safety evaluation of side dishes served at these foodservice facilities, microbial counts were generally below the limit of microbial contamination.

Chitosan-based nanocomposite films were prepared using a solution intercalation method incorporating varying amounts of organically modified montmorillonite (Cloisite 30B) from 0 to 30 wt%. The nanocomposite films prepared were optically clear despite a slight decrease in the transmittance due to the spatial distribution of nanoclay. X-ray diffraction patterns indicated that a certain degree of intercalation or exfoliation formed when the amount of clay in the film was low and that microscale tactoids formed when the clay content in the sample was high (more than 10 wt%). The tensile strength (TS) of the chitosan film increased when the clay was incorporated up to 10 wt% and then decreased with further increases in the clay content of the film. The elongation at break (E) increased slightly upon the addition of low levels of clay up to 5 wt% and then decreased with further increases in the amount of the clay in the film. The water vapor permeability (WVP) decreased exponentially with increasing clay content. The water solubility (WS) and swelling ratio (SR) of the nanocomposite films decreased slightly, indicating that the water resistance of the chitosan film increased due to the incorporation of the nanoclay.

This study sought to investigate any additive or interactive effects that soy isoflavones may have on the ecosystem of the gut, which is influenced by fructooligosaccharide (FOS) in colon-cancer model rats. Male Sprague-Dawley rats treated with 1,2-dimethylhydrazine were given experimental diets containing 0, 3, 6, or 9% FOS with or without 0.1% soy isoflavone for 12 weeks. In addition to the effects of FOS dosage on the gut ecosystem, dietary supplementation with soy isoflavone reduced the number of colonic aberrant crypts (ACs). The fecal weight, fecal pH, and gut transit time significantly decreased in a dose-dependent manner in rats fed FOS and the fecal concentration of bifidobacteria was higher in rats fed FOS than in control rats. The fecal output of total short-chain fatty acids, acetate, and propionate was significantly increased by the presence of FOS and was negatively correlated with the number of ACs, whereas the fecal output of butyrate showed no significant correlation with FOS dosage. The addition of soy isoflavone to the diet did not result in any significant differences in gut ecosystem parameters. Therefore, we conclude that the suppressive effect of soy isoflavone on ACs was not associated with the intestinal ecosystem, which was significantly altered by the dosage of FOS.

Chitosan has a wide range of applications in biomedical materials as well as in dietary supplements. Chitosan oligosaccharide with free-amine group (COFa) is an improvement over traditional chitosan that lacks the usual impurities and materials detrimental to the body. Based on a previous study of water soluble chitosan (WSC, chitosan lactate), we investigated the molecular weight (Mw) - dependent absorption phenomena of COFa in vitro and in vivo with various Mws. The absorption of CO Fa was significantly influenced by its molecular weight. As Mw increases, the absorption decreases. The absorption profiles for 5 K COFa (Mw=5 kDa) were observed to be more than 10 times higher than those of high molecular weight chitosan (100 K HWSC Mw=100 kDa) in both in vitro and in vivo transport experiments. Furthermore, the in vitro transport experiment suggested that transcellular transport of the COFa (Mw <10 kDa) through Caco-2 cell layer could occur with a negligible cytotoxic effect. The COFas showed a cytotoxic effect on Caco-2 cells that was dependent on dose and Mw. COFa could be transported transcellularly through the Caco-2 cell layer.

Preventive Effects of Oat Bran Extracts on Rat Colon Carcinogenesis Induced by 1,2-DimethylhydrazinePark, Hee-Joeng;Kang, Tae-Su;Kim, Kwang-Yup;Woo, Koan-Sik;Kim, Tae-Myoung;Kim, Yun-Bae;Kim, Dae-Joong;Jeong, Heon-Sang 942
The effect of oat bran extracts on the formation of aberrant crypt foci (ACF) in the colon induced by 1,2-dimethylhydrazine (DMH) was studied in F344 male rats. Extracts were prepared using various combinations of temperature (40, 45, 50, 55, or60$^{\circ}C:\;X_1$ ), ethanol concentration (0,5, 10, 15, or 20%:$X_2$ ), and pH (5, 6, 7, 8, or 9:$X_3$ ). Among the various extracts tested, one ethanol extract (EE;$45^{\circ}C$ , 15% ethanol at pH 6) and one water extract (WE;$50^{\circ}C$ at pH 5) were selected based on their in vitro antitumor activity. The animals were fed with basal diet alone or basal diet supplemented with 0.25 or 0.5% of EE or WE for 6 weeks. During the initial 2 weeks of the 6-week test period, the rats were subcutaneously injected with DMH (30 mg/kg) 4 times for the induction of ACF. DMH induced an average of 322.7 and 142.9 aberrant crypts (AC) and ACF, respectively. A low dose (0.25%) of EE (containing 38.3%${\beta}$ -glucan) and WE (containing 22.8%${\beta}$ -glucan) greatly reduced the numbers of DMH-induced AC and ACF. Significantly, ACF consisting of more than 3 AC were reduced by half in which the effect of EE, containing a higher concentration of${\beta}$ -glucan, was superior to that of WE. These results demonstrate that oat bran extracts may confer protection against colon carcinogenesis. -
The D-form of lactic acid is frequently detected in fermented foods, and an excessive dietary intake of D-lactic acid may induce metabolic stress in both infants and patients. This work was carried out to determine the prevailing microorganisms relevant to the accumulation of D-lactic acid in kimchi. Leuconostoc (Leuc.) mesenteroides and Leuc. citreum primarily synthesized D-lactate with a small quantity of L-form. Leuc. gelidum and Leuc. inhae evidenced patterns similar to this. Lactobacillus (Lb.) plantarum and Lb. brevis were shown to convert glucose into a balanced mixture of D-/L-lactic acid, whereas Lb. casei principally synthesized L-lactic acid and a very small quantity of D-lactic acid. When kimchi was incubated at 8 or
$22^{\circ}C$ , D-lactic acid was over-produced than L-form. Leuconostoc was determined as the primary producer between the initial to mid-phase of fermentation and Lb. plantarum or Lb. brevis seemed to boost D-lactic acid content during later stage of acid accumulation. -

A comprehensive safety evaluation was conducted to assess the potential allergenicity of newly introduced proteins in genetically modified (GM) crops. We assessed the allergenicity of CP4 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) in GM soybeans. This assessment was performed by IgE immunoblotting with soy-allergic children's sera, amino acid sequence homology with known allergens, and the digestibility of CP4 EPSPS. No differences in IgE-antigen binding by immunoblotting were found between GM soy samples and the corresponding non-GM samples. Based on the comparison of EPSPS amino acid sequence homology with current allergen databases, no known allergen was found. In addition, CP4 EPSPS protein was rapidly digested by simulated gastric fluid (SGF). Taken together, these results indicate that GM soybeans have no allergenicity in children and are as safe as conventional soybeans. -
The electron-capture detector (ECD) of gas chromatographs (GC) has been used widely in pesticide analysis. However, as ECD relies on radioactive material, it is troublesome to purchase and maintain. Therefore, potent replacements for ECD were investigated. A Pulsed-discharge detector (PDD) for ECD was tested and the analytical results of PDD (ECD mode),
${\mu}ECD$ , and nitrogen-phosphorus detector (NPD) were compared for 107 pesticides including organochroline, organophosphorus, pyrethroids etc. The number of pesticides identified at the lowest limit of detection (LOD) was 36, 29, and 2 for PDD,${\mu}ECD$ , and NPD, respectively. The remaining pesticides showed same response to PDD and${\mu}ECD$ . The GC-PDD analysis of pesticides spiked into representative agricultural products (brown rice, spinach, and mandarin oranges) also showed good and/or equivalent recoveries using$GC-{\mu}ECD$ . -

Cyclomaltodextrinases (CDases), maitogenic amylases, and neopullulanases share highly conserved primary structures and similar characteristics, and are thus classified into the same family. BLAST search has showed that a variety of bacterial strains harbor putative CDase family genes with several well-conserved motif amino acid sequences. In this study, four degenerate polymerase chain reaction (PCR) primer sets were designed for the detection of CDase genes, on the basis of their highly conserved amino acid blocks (WYQIFP, DGWRLD, LGSHDT, and KCMVW). The PCR detection conditions were optimized and the detection specificity of each for the primer sets was tested against the genomic DNAs isolated from 23 different Bacillus-associated species. Consequently, all tested primer sets evidenced successful amplification of specific PCR products in length, which share 55-98% amino acid sequence identity with known and putative CDases. The primers developed herein, therefore, can be applied for the easy and efficient detection and isolation of CDase family genes for the modification of functional food carbohydrates. -

The extracts of Oenanthe javanica were evaluated for their effects on the expression of cyclooxygenase-2 (COX-2), which is mediated by the translocation of the p65 subunit into the nucleus. Fractions of ethyl acetate and chloroform from 80% ethanol extracts of O. javanica exhibited inhibitory effects on the secretion of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$ ) from lipopolysaccharide (LPS)-stimulated peritoneal macrophages; however, the aqueous- and hexane-fractions showed no significant effect. The ethyl acetate- and chloroform-fractions also reduced the COX-2 enzyme levels after 24-hr treatment. RT-PCR showed that the mRNA levels of COX-2 decreased following treatment with these fractions, suggesting that COX-2 expression is transcriptionally regulated by these extracts. We examined the effects of the chloroform- and ethyl acetate-fractions on the cytosolic activation of nuclear factor-${\kappa}B$ ($NF-{\kappa}B$ , p65 subunit) and on the degradation of inhibitor-${\kappa}B{\alpha}$ ($I-{\kappa}B{\alpha}$ ) in order to determine the mechanism of COX-2 regulation. The LPS-stimulated activation of the p65 subunit was significantly blocked upon the addition of$50\;{\mu}g/mL$ of these fractions, and the cytosolic$I-{\kappa}B{\alpha}$ degradation process was simultaneously inhibited. These findings suggest that the inhibition of COX-2 expression by the ethyl acetate-and chloroform-fractions may result from the inhibition of p65 translocation by blocking the degradation of$I-{\kappa}B{\alpha}$ ; this may be the mechanistic basis for the anti-inflammatory effects of O. javanica. -
The goal of this study was to investigate the effects of a newly developed functional food containing Bacillus polyfermenticus (BP) and other physiologically active materials on the antioxidant system and the process of colon carcinogenesis in male F344 rats. Following a one-week adaptation period, the rats were divided into 3 groups and fed either a high-fat, low-fiber diet (control and DMH groups), or a high-fat, low-fiber diet supplemented with B. polyfermenticus (
$3.1{\times}10^8\;CFU/day$ ) and other physiologically active materials (chitosan, chicory,${\alpha}$ -tocopherol, and flavonoids) (DMH+BP group). One week after the initiation of the diets, 2 groups of rats were subjected to six weeks of treatment with 1,2-dimethylhydrazine (DMH, 180 mg/kg BW, s.c.). The dietary treatments remained consistent throughout the entire experimental period. Nine weeks after the initial DMH injection, the rats supplemented with B. polyfermenticus had significantly lower numbers of aberrant crypt foci than those in the DMH group. Injections with DMH resulted in significantly higher leukocytic DNA damage and plasma lipid peroxidation levels, as well as in a lower plasma total antioxidant potential. These effects were reversed following supplementation with B. polyfermenticus and other physiological materials. Our results indicate that a functional food containing B. polyfermenticus exerts a protective effect on the antioxidant system and on the process of colon carcinogenesis, thereby suppressing the development of preneoplastic lesions. -
Sweet potato starch noodles prepared without freezing exhibited higher cooking loss and water uptake during cooking and usually resulted in noodles with a softer and stickier texture compared to commercial sweet potato starch noodles manufactured using a freezing process. By utilizing the starches of different plant sources (potato, cowpea, and sago in an equivalent mixture with sweet potato starch), however, the cooking properties and texture of the starch noodles could be improved. Among the starches tested, cowpea starch was most effective in providing cooking and textural properties similar to those of commercial noodles. As an alternative approach, the addition of a minor amount (0.1 % based on total solid weight) of various gums (xanthan, gellan, locust bean gum, curdlan, and carboxymethyl cellulose) was also examined. The addition of curdlan to noodles was effective in increasing the gumminess and hardness, and reducing the stickiness of noodles. Utilizing different starches and gums can improve the overall texture and quality of sweet potato starch noodles produced without freezing.

In a previous study, we obtained polyclonal antibodies against the organophosphorus insecticide fenitrothion and developed an enzyme-linked immunosorbent assay (ELISA) for this pesticide. Using these antibodies and an enzyme tracer, a direct competitive ELISA method specific for fenitrothion using a dipstick format was developed. Dipstick ELISA using antibodies to fenitrothion immobilized on an Immunodyne membrane allowed the quick visual detection of fenitrothion at concentrations above
$10\;{\mu}g/L$ . The$IC_{50}$ value of dipstick ELISA using reflectance detection was$27\;{\mu}g/L$ with a detection limit of$2\;{\mu}g/L$ . The recovery of fenitrothion from spiked lettuce and rice samples using the dipstick ELISA ranged from 87-107%. -

Cyanidin 3-glucoside (C3G) isolated from black rice was investigated for hepatoprotective effects in HepG2 cells under oxidative stress. When an increase in the production of reactive oxygen species (ROS) was induced by gramoxone, cell viability was drastically decreased by 42%. However, in the presence of C3G, no hepatocytic damage was observed in HepG2 cells treated with gramoxone. C3G was found to manifest a stronger scavenging effect (91%) on superoxide anion radical ($O_2\;^{.-}$ ) than any of the other natural and synthetic antioxidants. Results suggest that C3G from black rice possesses hepatoprotective effects in vitro, which may be, at least in part, due to$O_2\;^{.-}$ scavenging.
