BDF 20을 사용하는 디젤기관들의 연소 및 내구특성 (Combustion Characteristics and Durability of Diesel Engines Burning BDF 20)

  • Three diesel engines were fueled with BDF 20, a blend of 80% diesel fuel and 20% biodiesel fuel by volume, and run in excess of 200 h to evaluate their combustion characteristics and durability. The engines used for this study were a 4-cylinder 2476-cc displacement IDI diesel engine(Engine 1), a 4-cylinder l732-cc displacement IDI diesel engine(Engine 2), and a single cylinder 673-cc displacement DI diesel engine(Engine 3). Engine dynamometer testing was performed on each engine at regularly scheduled intervals to monitor the performance and exhaust emissions, which were sampled at 1h intervals for analysis, The peak combustion pressure with BDF 20 increased in Engines 1 and 3 over that measured when burning pure diesel fuel, but that in Engine 2 remained constant. Combustion parameters, such as the maximum combustion pressure and corresponding crank angle, did not change over the long-term dynamometer testing. The BSFC with BDF 20 in Engine 1 was less than that measured with pure diesel fuel. The amount of smoke produced with BDF 20 was less for all engines ; the greatest reduction was observed for Engine 3. The NOx emissions were lower in the IDI engines than the DI engine. The traditional trade-off between smoke and NOx emissions was maintained for BDF 20 fuel for Engines 1 and 3. There was not a big difference in the $CO_2\;and\;O_2$ emissions for BDF 20, as compared to pure diesel fuel, but more $CO_2$ was exhausted by Engine 1 than by Engines 2 or 3 and less $O_2$ was exhausted by Engine 1 than by Engines 2 or 3. The engine parts remained clean, except for some carbon attached to the area surrounding the nozzle hole of the DI diesel engine.

Higher Biomass Productivity of Microalgae in an Attached Growth System, Using Wastewater

  • Although most algae cultivation systems are operated in suspended culture, an attached growth system can offer several advantages over suspended systems. Algal cultivation becomes light-limited as the microalgal concentration increases in the suspended system; on the other hand, sunlight penetrates deeper and stronger in attached systems owing to the more transparent water. Such higher availability of sunlight makes it possible to operate a raceway pond deeper than usual, resulting in a higher areal productivity. The attached system achieved 2.8-times higher biomass productivity and total lipid productivity of $9.1g\;m^{-2}day^{-1}$ and $1.9g\;m^{-2}day^{-1}$, respectively, than the suspended system. Biomass productivity can be further increased by optimization of the culture conditions. Moreover, algal biomass harvesting and dewatering were made simpler and cheaper in attached systems, because mesh-type substrates with attached microalgae were easily removed from the culture and the remaining treated wastewater could be discharged directly. When the algal biomass was dewatered using natural sunlight, the palmitic acid (C16:0) content increased by 16% compared with the freeze-drying method. There was no great difference in other fatty acid composition. Therefore, the attached system for algal cultivation is a promising cultivation system for mass biodiesel production.

Lipid and Citric Acid Production by Wild Yeasts Grown in Glycerol

  • In this study, crude glycerol was used as a carbon source in the cultivation of wild yeasts, aiming at the production of microbial lipids and citric acid. Forty yeasts of different sources were tested concerning their growth in crude and commercial glycerol. Four yeasts (Lindnera saturnus UFLA CES-Y677, Yarrowia lipolytica UFLA CM-Y9.4, Rhodotorula glutinis NCYC 2439, and Cryptococcus curvatus NCYC 476) were then selected owing to their ability to grow in pure ($OD_{600}$ 2.133, 1.633, 2.055, and 2.049, respectively) and crude ($OD_{600}$ 2.354, 1.753, 2.316, and 2.281, respectively) glycerol (10%, 20%, and 30%). Y. lipolytica UFLA CM-Y9.4 was selected for its ability to maintain cell viability in concentrations of 30% of crude glycerol, and high glycerol intake (18.907 g/l). This yeast was submitted to lipid production in 30 g/l of crude glycerol, and therefore obtained 63.4% of microbial lipids. In the fatty acid profile, there was a predominance of stearic (C18:0) and palmitic (C16:0) acids in the concentrations of 87.64% and 74.67%, respectively. We also performed optimization of the parameters for the production of citric acid, which yielded a production of 0.19 g/l of citric acid in optimum conditions (38.4 g/l of crude glycerol, agitation of 184 rpm, and temperature of $30^{\circ}C$). Yarrowia lipolytica UFLA CM-Y9.4 presented good lipid production when in the concentration of 30 g/l of glycerol. These data may be used for production in large quantities for the application of industrial biodiesel.

  • Pretreatment of eliminating FFA is needed to make biodiesel from animal fat recovered from leather wastes because its acid value is high. This study was carried out to investigate the influence of 4 different pretreatment methods, which are heterogeneous catalyst method, ion exchange resin method, low pressure.high temperature method, and alkali method on the eliminating FFA and fatty acid composition. The results showed that the rate of eliminating FFA increased in the order of alkali method > catalyst method > low pressure high temperature method > ion exchange method. In the case of pretreatment of alkali method using NaOH, the rate of eliminating FFA appeared more than 86% regardless of acid value. Therefore, it was considered that alkali method using NaOH was the most effective in the view of economical and productive aspects, taking it into account that the acid value of animal fat recovered from fleshing scrap generated during leather making processes was 7 to 8.

바이오연료의 엔진 적용을 위한 실험적 기초연구 (Basic Experimental Study on the Application of Biofuel to a Diesel Engine)

  • 압축착화 방식의 디젤엔진은 스파크점화 방식의 가솔린 엔진에 비하여 열효율이 높아 연비가 향상되고 그 결과 $CO_2$ 저감효과도 높다. 또한 디젤엔진은 점화계통 장치의 불필요 등 기존 엔진의 개조비용이 적어 세탄가가 높은 바이오연료의 적용엔진으로서 적합한 장점이 있다. 따라서 본 연구에서는 식물성 자트로파유, 대두유 2종의 바이오연료와 경유연료의 분무특성을 비교 분석하였다. 실험변수로서는 분사압력과 자트로파 연료의 경우는 혼합비율(BD3, BD5, BD20)을 달리하였다. 분사압력은 500bar, 1000bar, 1500bar 및 1600bar로 설정하고 분사기간은 500ms로 동일하게 하였다. 본 연구의 결과로서, 사용한 바이오디젤 연료의 종류 및 분사압력 변화에 대한 분무거동특성(분무각)의 변화는 뚜렷하지 않으나, 고압분사의 경우가 분무각이 약간 감소하는 결과를 얻을 수 있었다.

Increased Microalgae Growth and Nutrient Removal Using Balanced N:P Ratio in Wastewater

  • Microalgal cultivation using wastewater is now regarded as essential for biodiesel production, as two goals can be achieved simultaneously; that is, nutrient removal efficiency and biomass production. Therefore, this study examined the effects of carbon sources, the N:P ratio, and the hydraulic retention time (HRT) to identify the optimal conditions for nutrient removal efficiency and biomass production. The effluent from a 2nd lagoon was used to cultivate microalgae. Whereas the algal species diversity and lipid content increased with a longer HRT, the algal biomass productivity decreased. Different carbon sources also affected the algal species composition. Diatoms were dominant with an increased pH when bicarbonate was supplied. However, 2% $CO_2$ gas led to a lower pH and the dominance of filamentous green algae with a much lower biomass productivity. Among the experiments, the highest chlorophyll-a concentration and lipid productivity were obtained with the addition of phosphate up to 0.5 mg/l P, since phosphorus was in short supply compared with nitrogen. The N and P removal efficiencies were also higher with a balanced N:P ratio, based on the addition of phosphate. Thus, optimizing the N:P ratio for the dominant algae could be critical in attaining higher algal growth, lipid productivity, and nutrient removal efficiency.

High-frequency plant regeneration from leaf-disc cultures of Jatropha curcas L.: an important biodiesel plant

  • A simple, high-frequency and reproducible protocol for induction of adventitious shoot buds and plant regeneration from leaf-disc cultures of Jatropha curcas L. has been developed. Adventitious shoot buds were induced from very young leaf explants of in vitro germinated seedlings as well as mature field-grown plants cultured on Murashige and Skoog's (MS) medium supplemented with thidiazuron (TDZ) ($2.27{\mu}M$), 6-benzylaminopurine (BA) ($2.22{\mu}M$) and indole-3-butyric acid (IBA) ($0.49{\mu}M$). The presence of TDZ in the induction medium has greater influence on the induction of adventitious shoot buds, whereas BA in the absence of TDZ promoted callus induction rather than shoot buds. Induced shoot buds were multiplied and elongated into shoots following transfer to the MS medium supplemented with BA ($4.44{\mu}M$), kinetin (Kn) ($2.33{\mu}M$), indole-3-acetic acid (IAA) ($1.43{\mu}M$), and gibberellic acid ($GA_3$) ($0.72{\mu}M$). Well-developed shoots were rooted on MS medium supplemented with IBA ($0.5{\mu}M$) after 30 days. Regenerated plants after 2 months of acclimatization were successfully transferred to the field without visible morphological variation. This protocol might find use in mass production of true-to-type plants and in production of transgenic plants through Agrobacterium/biolistic-mediated transformation.

Identification of Green Alga Chlorella vulgaris Isolated from Freshwater and Improvement Biodiesel Productivity via UV Irradiation

  • Chlorella vulgaris was isolated from the Nile River, Qalubia Governorate, Egypt, for possible use in biodiesel production. BG-II nutrient growth media was used for isolation and laboratory growth. Identification was performed via 18S rRNA gene amplification, followed by sequencing. The alga was exposed to UV-C (254 nm) for 15, 30, and 45 s to improve dry weight accumulation and to increase the oil production. Daily measurements of dry weight ($g{\cdot}l^{-1}$) were performed; oil content and volumetric lipid productivity were also determined. UV-C exposure led to an increase in the volumetric lipid productivity by 27, 27.3, and $32.4mg{\cdot}l^{-1}{\cdot}d^{-1}$ with 15, 30, and 45 s, respectively, as compared with the control, which resulted in $18mg{\cdot}l^{-1}{\cdot}d^{-1}$. Of the examined mutants, the one with the highest productivity was re-irradiated by UV-C (254 nm) for 15, 30, 45, and 60 s. For 15 s of exposure time, the oil content increased to 34%, while it was 31% at 30 s; further, it decreased to 22% at 45 and 60 s exposures. The fatty acid methyl ester profile was 82.22% in the first mutant at 45 s, compared with the wild strain that contained a total of 66.01% of FAs. Furthermore, the highest levels of polyunsaturated fatty acid methyl ester were observed in the mutant exposed for 45 s, and it reached 11.41%, which reduced the cetane number to 71.3.

Characterization of the Growth, Total Lipid and Fatty Acid Profiles in Microalga, Nannochloropsis oceanica under Different Nitrogen Sources

  • The properties of microalgae as bioresources for biodiesel production can be improved by adding nitrogen sources into the culture medium. Thus, Nannochloropsis oceanica CCAP 849/10 was cultured in f/2 media supplemented with five different forms of nitrogen at $0.88mmol-N\;l^{-1}$ each: ammonium bicarbonate ($NH_4HCO_3$), ammonium sulfate ($(NH_4)_2SO_4$), sodium nitrate ($NaNO_3$), ammonium nitrate ($NH_4NO_3$), and urea. The cell density, lipid content, and fatty acid profile of the microalga were determined after 15 days of cultivation. The growth of N. oceanica based on cell number was lowest in the medium with $NH_4NO_3$, and increased significantly in the medium with $NH_4HCO_3$. Cells treated with $(NH_4)_2SO_4$, and $NH_4NO_3$ produced the highest total lipid contents (i.e., 65% and 62% by dry weight, respectively). The fatty acid profiles of the microalga were significantly different in the various nitrogen sources. The major fatty acids detected in cultures supplemented with $NH_4HCO_3$, $(NH_4)_2SO_4$, $NH_4NO_3$, or urea were C14:0, C16:0, C16:1, C18:0, C18:1, C18:2, C20:5, and C22:6. However, the C16:1 content in the $NaNO_3$-supplemented culture was very low. This study highlights that the nitrogen source can strongly influence lipid production in N. oceanica and its fatty acid composition.

Purification, Characterization and Immobilization of Lipase from Proteus vulgaris OR34 for Synthesis of Methyl Oleate

  • A newly isolated strain, Proteus vulgaris OR34, from olive mill waste was found to secrete an alkaline extracellular lipase at 11 U·ml-1 when cultivated on an optimized liquid medium. This lipase was purified 94.64-fold with a total yield of 9.11% and its maximal specific activity was shown to be 3232.58 and 1777.92 U·mg-1 when evaluated using the pH-stat technique at 55℃ and pH 9 and Tributyrin TC4 or olive oil as the substrate. The molecular mass of the pure OR34 lipase was estimated to be around 31 kDa, as revealed by SDS-PAGE and its substrate specificity was investigated using a variety of triglycerides. This assay revealed that OR34 lipase preferred short and medium chain fatty acids. In addition, this lipase was stable in the presence of high concentrations of bile salt (NaDC) and calcium ions appear not to be necessary for its activity. This lipase was inhibited by THL (Orlistat) which confirmed its identity as a serine enzyme. In addition, the immobilization of OR34 lipase by adsorption onto calcium carbonate increased its stability at higher temperatures and within a larger pH range. The immobilized lipase exhibited a high tolerance to organic solvents and retained 60% of its activity after 10 months of storage at 4℃. Finally, the OR34 lipase was applied in biodiesel synthesis via oleic acid mediated esterification of methanol when using hexane as solvent. The best conversion yield (67%) was obtained at 12 h and 40℃ using the immobilized enzyme and this enzyme could be reused for six cycles with the same efficiency.
