High-Level Expression of Pseudomonas sp. LBC505 Endoglucanase Gene in Escherichia coli

  • Endoglucanase gene of Pseudomonas sp. LBC505 was previously cloned in pUC19 to yield plasmid pLCl. The Pseudomonas sp. LBC505 endoglucanase gene was subcloned in a temperature-regulated Es-cherichia coli expression vector, pAS1, containing the leftward promoter $P_L$ of bacteriophage lambda. The level of gene expression was controlled by the thermal inactivation of the heat-sensitive lambda cI857 repressor. Best yield of endoglucanase was obtained by lowering the incubation temperature to $37^{\circ}C$ after induction at $42^{\circ}C$ for 1h. Under these conditions enzyme production continued for about 5h at a gradually decreasing rate. Ecoli harboring recombinant plasmid pASC10 expressed 4.3 times as much CMCase activity as E.coli containing pLCl. To enhance the expression level of endogl, ucanase gene, we have also changed the presumptive Shine-Dalgamo sequence (AGAGGT) of the gene to consensus sequence (AGGAGGT) by site-directed mutagenesis. The genes mutated were subcloned in pASl resulting in the formation of recombinant plasmid pASS50. E.coli harboring the plasmid pASS50 expressed 6.2-fold higher levels of CMCase activity than that of E.coli harboring pLC1.

Effect of Solidification Conditions and Heat Treatment on the Mechanical Properties of the $Al-CuAl_2$ Eutectic Composite (Al-$CuAl_2$ 공정복합재료의 기계적 성질에 미치는 응고조건과 열처리의 영향)

  • The structure and tensile properties of the unidirectionally solidified Al-33wt.%Cu alloy have been investigated. Casted Al-33wt.%Cu alloy was unidirectionally solidified with rates (R) between 1㎝/hr and 24cm/hr maintaining the thermal gradient(G) at solid-liquid interface, $32^{\circ}C/cm$ and $21^{\circ}C/cm$. The entectic struture was varied according to the growth condition(G/R radio). When G/R ratio was larger than $8.5{\times}10^3$ $^{\circ}C/cm^2/sec$ the lamellar structure was formed, and colony structure was formed when G/R ratio was smaller than $8.5{\times}10^3$ $^{\circ}C/cm^2/sec$. The interlamellar spacing(${\gamma}$) in the above alloy system was vaired with the growth rate(R) According to "${\gamma}^2{\cdot}R=8.8{\times}10^{-11}cm^2/sec$" relationship. The yield stress (${\sigma}$0.001) and UTS for samples in the as-grown condition increased with the interlamellar spacing decrease and the values corresponding to colony structure are lower than those corresponding to amellar structure with the same lamellar spacing. The yield stress for samples in aged condition did not change with the interlamellar spacing.

SCP Production from Mandarin Orange Peel Press Liquor (감귤과피 압착액을 기질로 한 SCP 생산)

  • The bioconversion of mandarin orange peel press liquor to single cell protein (SCP) by two yeast strains, F-60, and C-7, which were isolated from mandarin orange peel was carried out and compared with that of using Candida utilis IFO 0598. Experiments were directed toward the high yield of biomass and high protein in cultures of the strains mentioned above. Candida utilis IFO 0598, F-60 and C-7 strains were cultivated at 3$0^{\circ}C$, pH 5.2 for 3 days in shaking flasks. The effects of some nutrients on cell growth were studied. Cell mass and protein content per cell mass were increased by addition of urea 1%, KH$_2$PO$_4$ 0.1% and MgSO$_4$ㆍ7$H_2O$ 0.05%, When the F-60 strain cultured under the optimal conditions, cell mass, growth yield and protein content were 41.2g/l, 53.9%, 59.7%, respectively. Cell mass was also increased up to 15% by modifying the fermentation condition on the bench type 20l jar fermentor. Crude fat content (10.3%) of dried C-7 cell was higher than those of C. utilis and F-60, 4.9% and 5.6% respectively. Total protein content of the F-60 strain was 59.7% per dry weight. And we compared their amino acid compositions with that of FAO provisional pattern. In the case of the F-60 strains, amino acid contents such as lysine, leucine and isoleucine were much higher than those of methionine, cystine and tryptophan.

Production of Ethanol from D-Xylose by Fusarium sp. (Fusarium sp.에 의한 D-Xylose로부터 Ethanol 생산)

  • Microorganisms capable of utilizing D-xylose as a sole carbon and energy source were isolated to ferment D-xylose directly to ethanol. Among them, the strain, which showed the best ability to pro-duce ethanol, was selected and was identified as Fusarium sp. The optimal conditions for the pro-duction of ethanol were 8.0 of initial pH, 33$^{\circ}C$ of temperature, and 2% of substrate concentration. Under this optimal condition, the following results were obtained : maximum ethanol concentration, 7.0g/$\ell$; ethanol yield, 0.35g of ethanol per g of D-xylose (68.6% of theoretical); biomass yield, 0.27g of dry biomass per g of D-xylose.

  • Background: Angelica gigas Nakai, that belong to the Umbelliferae family, is one of the traditional medicinal plants in Korea. Its roots have been used to treat gynecological diseases. In this study, growth characteristics and index components were compared with the forest microclimate at several forest sites. Methods and Results: A. gigas was planted in three climatic zones according to the temperature (southern temperature zone - Hamyang, central temperature zone - Bonghwa, and northern temperature zone - Jeongseon) and growth characteristics were investigated in comparison with the forest microclimate. Our results indicated that the root diameter and length, and fresh and dry weight were the highest in Jeongseon. The total content of decursin was the highest in Jeongseon (9.52%), followed by those in Hamyang (8.07%) and Bonghwa (7.48%), respectively. Additionally, the yield of decursin (1.39 g) was the highest in Jeongseon. Conclusions: The yield and index components were influenced by the microclimate in the forests, and it was assumed that high altitude and low temperature affected the increase in growth and index components. These results will be useful as basic data to study the correlation among environmental conditions, growth, and index components.

Conversion of Cellulose over Ni Loaded Mesoporous MSU-F Catalysts via Air Gasification

  • Catalytic gasification of cellulose was carried out in a U-type fixed reactor with Ni loaded MSU-F catalyst (Ni/MSU-F) and Ni loaded ${\gamma}-Al_2O_3$ (Ni/${\gamma}-Al_2O_3$). The characteristics of the catalysts were analyzed by using X-ray diffraction, $H_2$-temperature programmed reduction, and Brunauer-Emmett-Teller analyses. The operation conditions of catalytic gasification reactions were $750^{\circ}$ and 0.2 equivalence ratio. Air was used as gasification agent. Catalytic gasification characteristics, such as gas yield and gas composition ($H_2$, CO, $CO_2$, $C_1-C_4$), were measured and calculated. The gas yield of Ni/MSU-F was much higher than that of Ni/${\gamma}-Al_2O_3$. Especially high amount of hydrogen was produced by Ni/MSU-F.

Preparation of High Molecular Weight Poly(methyl methacrylate) with High Yield by Room Temperature Suspension Polymerization of Methyl Methacrylate

  • To obtain high molecular weight (HMW) poly(methyl methacrylate) (PMMA) with high conversion, methyl methacrylate (MMA) was polymerized in suspension using a room temperature initiator, 2,2'-azobis(2,4-dimethylvaleronitrile) (ADMVN), and the effects of polymerization conditions on the polymerization behavior of MMA and the molecular parameters of PMMA were investigated. On the whole, the experimental results well corresponded to the theoretically predicted tendencies. These effects could be explained by a kinetic order of ADMVN concentration calculated by an initial rate method and an activation energy difference of polymerization obtained from the Arrhenius plot. Suspension polymerization at 25℃ by adopting ADMVN proved to be successful in obtaining PMMA of HMW (number-average degree of polymerization (P/sub n/): 30,900-36,100) and of high yield (ultimate conversion of MMA into PMMA: 83-93 %) with diminishing heat generated during polymerization. The P/sub n/ and lightness were higher and polydispersity index was lower with PMMA polymerized at lower temperatures.

Study on the Production of L-Latic Acid from Soluble Starch by Streptococcus sp. JEJ-6 (Streptococcus sp. JEJ-6에 의한 가용성 전분으로부터 L-Lactic Acid 생성에 관한 연구)

  • The strain producing L-lactic acid from starch was isolated from kimchi. The isolated strain was identified as a homofermentative Streptococcus sp. through its morphological, cultural, biochemical characteristics, and named Streptococcus sp. JEJ-6. Lactic acids are of two types, one L-specific and the other D-specific form in a stereospecific form. Streptococcus sp. JEJ-6 produced selectively L-lactic acid from all of the tested carbon sources. The optimum conditions for the L-lactic acid production from the isolated microorganism were determined. For the maximum yield of L-lactic acid from Streptococcus sp. JEJ-6, the cell should be harvested at the early stationary phase, and the growth temperature, pH, and NaCl concentration should be 37$^{\circ}C$, pH 7.0 and 0.1%, respectively. 4% Soluble starch as substrate and organic nitrogen sources such as peptone and yeast extract should be used for the best yield. The optimum pH of the nicotinamide adenine dinucleotide(NAD)-dependent and NAD-independent lactate dehydrogenase(LDH) activities was pH 8.5 and pH 7.0, respectively.

  • Recently the ozone generation system is well used for cleaning the contaminated water by using the strong oxidization effects of ozone. In this paper the concentration, yield and generation quantity of the ozone by the Surface-Silent Discharge According to the Length of Coaxial Electrode is described. The electrodes composed of 3 electrodes-2 gaps are coaxial type and the ozone generation tube were designed and fabricated from the point of view of the energy efficiency, stability and easy control. To investigate the ozone generation dependency on electrode length, a few discharge tubes with different lengths were fabricated. The experimental equipments were provided with cooling system and dehumidifier for the many testing conditions. The main results show that the concentration, yield and generation quantity of ozone are improved by decreasing gas temperatures and increasing electrode lengths.

Effects of elevated CO2 concentration and temperature on growth and production of Oryza sativa L. cv. Ilmi, one of the main rice varieties in Korea

  • This research was conducted to examine the changes in growth and production of Oryza sativa L. cv. Ilmi, which was developed to cultivate high yielding rice variety in the Southern plains of Korea. The seedlings of the rice were cultivated from May to October in 2012 under three different conditions: control, AC-AT, ambient $CO_2$ + ambient temperature; AC-ET, ambient $CO_2$ + elevated temperature; EC-ET, elevated $CO_2$ + elevated temperature. The aboveground biomass, belowground biomass, the total biomass of the rice, and panicle weight per individual were the heaviest in the EC-ET. But, the number of grains per panicle and the weight of one grain was higher at the condition of AC-ET and EC-ET than that of AC-AT. The number of tiller was higher at the condition of AC-AT and AC-ET than that of EC-ET. However, there was no significant difference in the number of panicles per individual and the ripened grain rate among the control and global warming treatments. Crop yield was the highest in the EC-ET. This result means that the global warming condition should be considered in the selection of suitable paddy field for the limibyeo in the future.
