Production of Streptomyces albus KSM-35 Amylase from Bacillus subtilis LKS88 Haboring the Recombinant Plasmid pASA240 (재조합 균주 Bacillus subtilis LKS88에 의한 Streptomyces albus KSM-35 Amylase의 생산조건)

  • The effects of culture conditions on the production of amylase expressed by Bacillus subtilis LKS88 with a cloned gene from Streptomyces albus KSM-35 were investigated. The production of amylase was increased significantly by using sodium citrate and rice hull as a carbon source. In addition, the use of a mixture of sodium citrate and rice hull (1:1) resulted in increase of enzyme production by 20-fold when compared to that of soluble starch. The soybean meal as the nitrogen source could be partially replaced with yeast extract without changing the enzyme production yield. The amylase production was also increased by adjusting initial pH to 6.0 or by adding 0.01% SDS. Maximum amylase production was observed in the medium containing 1.5% sodium cirtate, 1.5% rice hull, 0.7% soybean meal, 0.3% yeast extract, 0.66% K2HPO4, 0.05% MgSO4$.$7H2O, 0.008% CaCl2$.$2H2O, 0.01% SDS with initial pH of 6.0. The maximum yield of amylase reached 56.6 U/ml when B. subtilits LKS88 (pASA 240) was cultured at 37$^{\circ}C$ for 36 hr.

Triboelectrostatic Separation System for Separation of PVC and PS Materials Using Fluidized Bed Tribocharger

  • A triboelectrostatic separation system using a fluidized bed tribocharger for the removal of PVC material in the mixture of PVC/PS plastics is designed and evaluated as a function of electric field strength, air flow rate, and the mixing ratio of two-component mixed plastics. It consists of a fluidized-bed tribocharger, a separation chamber, a collection chamber and a controller. PVC and PS particles can be imparted negative and positive surface charges, respectively, due to the difference in the work function values of plastics suspended in the fluidized-bed tribocharger, and can be separated by passing them through an external electric field. Experimental results show that separation efficiency is strongly dependent on the electric Deld strength and particle mixing ratio. In the optimum conditions of 150 Ipm air flow rate and 2.6 kV/cm electric field strength a highly concentrated PVC (99.1%) can be recovered with a yield of more than 99.2% from the mixture of PVC and PS materials for a single stage of processing.

Effect of Potassium Silicate Amendments in Hydroponic Nutrient Solution on the Suppressing of Phytophthora Blight (Phytophthora capsici) in Pepper

  • Amendments of a recirculating nutrient solution with potassium silicate were evaluated as a means to control Phytophthora capsici infections on pepper plant(Capsicum annuum L.). Supplying the solutions with 100 or 200 ppm of silicate significantly reduced motility, root decay, and yield losses attributed to infection of P. capsici. Treating inoculated plants with potassium silicate increased root dry weights and number of fruit, especially high-grade fruit. Results were slightly superior to non-inoculated controls. The two varieties, PBC 137 and PBC 602, responded similarly to the treatments. No significant differences were observed between the 100- and 200 ppm silicate treatments. Results were better when greenhouse conditions favored the spread of P. capsici. Silicon alone did not increase pepper yield, suggesting that it acts as a disease suppression agent rather than as a fertilizer, The phenomena by which silicon confers protection against P. capsici infection and disease development are not fully understood, but our results indicate that mechanisms other than a mechanical barrier to fungal penetration are involved.

  • The synthesis of N-(benzyloxycarbonyl)-L-aspartyl-L-phenylalanine methylester (Z-APM), a precursor of aspartame, from N-(benzyloxycarbonyl)-L-aspartic acid (Z-Asp) and L-phenylalanine methylester hydrochlolide($L-PM\cdot HCI$) was investigated in a saturated-ethylacetate single phase system using immobilized thermolysin. Among the various supports tested, glyceryl-CPG was found to be most efficient for retaining enzyme activity. The enzyme immobilized onto glyceryl-CPG also showed the highest activity for Z-APM synthesis in saturated ethyl acetate. Z-APM conversion yield in saturated ethylacetate was half of that obtained in an ethyl acetate-buffer two-phase system under the same reaction conditions. However, as the mole ratio of $L-PM \cdot HCI$ to Z-Asp was increased to 4.0, the conversion yield reached 95 %. When continuous synthesis of Z-APM was canied out in a plug flow reactor (PFR) with 80 mM of L-PMㆍHCI and 20 mM of Z-Asp in saturated ethylacetate (pH 5.5), more than 95 % of Z-Asp was converted to Z-APM with a space velocity of 1.16 $hr^{-1} at 40^{\circ}C$. Although the operational stability in PFR was reduced rapidly, more than 80% of initial activity was maintained in CSTR even after a week of operation.

High-Temperature Tensile Strengths of Alloy 617 Diffusion Weldment (Alloy 617 확산용접재의 고온 인장강도)

  • A compact heat exchanger is one of critical components in a very high temperature gas-cooled reactor (VHTR). Alloy 617 (Ni-Cr-Co-Mo) is considered as one of leading candidates for this application due to its excellent thermal stability and strengths in anticipated operating conditions. On the basis of current ASME code requirements, sixty sheets of this alloy are prepared for diffusion welding, which is the key technology to have a reliable compact heat exchanger. Optical microscopic analysis show that there are no cracks, incomplete bond, and porosity at/near the interface of diffusion weldment, but Cr-rich carbides and Al-rich oxides are identified through high resolution electron microscopic analysis. In high-temperature tensile testing, superior yield strengths of the diffusion weldment compared to the code requirement are obtained up to 1223 K ($950^{\circ}C$). However, both tensile strength and ductility drop rapidly at higher temperature due to the insufficient grain boundary migration across the interface of diffusion weldment. Best fit curves for minimum yield strength and average tensile strength are drawn from the experimental tensile results of this study.

Effects of Low Dose Gamma Radiation on the Root Growth of Soybean Cultivars

  • ${\gamma}-Radiation$ at very low doses frequently has a stimulating or hormetic effect on the growth of organism. Effects of low dose ${\gamma}-ray$ irradiation on the root growth of soybean cultivars were investigated and hormetic effects by environmental conditions were compared with the occurrence of increased economic yield, seeds of cultivars were irradiated with the dose of $0.5{\sim}20Gy$ and cultivated in growth chamber controling temperature, humidity, light, greenhouse and field respectively. To understand hormetic effect on root growth of cultivars and the difference of hormetic effect by cultivation environment, harvested root of soybean cultivars were scanned with image file, and root surface area, root length, root average diameter etc. were examined by WinRhizo program. Also, dry weight of cultivars was examined. Root growth and dry weight of soybean cultivars showed apparently hormetic effect at cultivation of growth chamber condition. In field experiment executed for whole life cycle, yields of pea were not different significantly in each ${\gamma}-ray$ irradiated cultivars but weight of one hundred peas increased in whole ${\gamma}-ray$ irradiated cultivars. Increment of yield was assumed to be induced through shortening of maturing stage caused by ${\gamma}-ray$ hormesis in early growth stage.

Effects of Tempering on Tensile Properties of Medium-Carbon Low-Alloy Steels (중탄소 저합금강의 인장성질에 미치는 템퍼링의 영향)

  • A series of Ni-Cr-Mo alloy steels were austenitized, quenched to martensite, and tempered at various temperature and time conditions. Tensile testing was conducted at room temperature with cylindrical specimens, and hardness was measured using Rockwell hardness tester. In the tempering stage I, high strain hardening and yield strength accounted for the high ultimate strength and hardness. In the tempering stage II, strengths and hardness linearly decreased with increasing tempering temperature. Specimens tempered in the temperin stage III showed incipient discontinuous yielding and tensile strengths only slightly higher than yield strengths. Ductilities decreased slightly in specimens tempered in the tempered martensite embrittlement range, and severely decreased in specimens tempered for 10 hours at $500^{\circ}C$ in the temper embrittlement range. Specimens tempered at $600^{\circ}C$ for 10 hours showed recrystallized microstructures, a number of fine dimples, and increased strain hardening, probably due to the precipitation of alloy carbides. The simple formulae for the mechanical properties of these steels were suggested as a function of carbon content and Hollomon-Jaffe tempering parameter.

Novel Suspension-Phase Enzyme Reaction System Using Insoluble Extrusion Starch as Glycosyl Donor for Intermolecular Transglycosylation of L-Ascorbic Acid

  • A novel suspension-phase enzyme reaction system for the intermolecular transglycosylation of L-ascorbic acid into 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid supplementing extrusion starch as the glycosyl donor was developed using cyclodextrin glucanotransferase from Thermoanaerobacter sp. A high conversion yield compared to the conventional soluble-phase enzyme reaction system using cyclodextrins and soluble starch was achieved. The optimal reaction conditions were 2,000 units of cycIodextrin glucanotransferase, 20 g/l of L-ascorbic acid, and 50 g/l of extrusion starch at $50^{\circ}C$ for 24 h. The new suspension-phase enzyme reaction system also exhibited several distinct advantages other than a high conversion yield, including a lower accumulation of oligosaccharides and easily separable residual extrusion starch by centrifugation or filtration in the reaction mixture, which will facilitate the purification of 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid. The new suspension-phase enzyme reaction system seems to be potentially applicable as the industrial process for the production of thermally and oxidatively stable 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid.

Finite Element Analysis of Mechanical Behavior of Bolt Tightened in Plastic Region (소성역 체결 볼트의 기계적 거동 유한요소해석)

  • Plastic region tightening is widely used in critical bolted joints in internal combustion engines in order to reduce the engine weight by maximizing the use of load-carrying capacity of bolt. Mechanical behavior of bolt tightened in plastic region under external axial tensile load is investigated for various friction conditions using three dimensional finite element analysis. The behavior of bolt tightened in elastic region as well as that in tensile test are investigated for comparison. Tightening process is simulated by rotating the bolt in order to examine the friction effect realistically. It is revealed that the bolt tightened in plastic region can carry more external load until the joint is opened, and yields at lower bolt load than the bolt tightened in elastic region. The friction coefficient has effect on the yield load, but not on the load-carrying capacity. Moreover, the scatter in the bolt preload due to friction begins with plastic deformation of bolt in the angle tightening control, whereas it begins with the onset of tightening in the torque tightening control. The observations are interpreted with the residual torsional stress in the bolt generated during the tightening.

  • We have developed the methodology for the quantitative analysis of microencapsulation yield and optimized the conditions for the microencapsulation of ${\beta}$-galactosidase by butter oil. The degree of ${\beta}$-galactosidase deactivation by the microencapsulation process was the value of 5.2% of initial activity. And the yield for the microencapsulation of ${\beta}$-galactosidase by the indirect, heat treatment, and enzymatic methodology were 92.6%, 88.6%, and 94.1%, respectively.
