Influence of time-dependency on elastic rock properties under constant load and its effect on tunnel stability

  • In structures excavated in rock mass, load progressively increases to a level and remains constant during the construction. Rocks display different elastic properties such as Ei and ʋ under different loading conditions and this requires to use the true values of elastic properties for the design of safe structures in rock. Also, rocks will undergo horizontal and vertical deformations depending on the amount of load applied. However, under constant loads, values of Ei and ʋ will vary in time and induce variations in the behavior of the rock mass. In some empirical equations in which deformation modulus of the rock mass is taken into consideration, elastic parameters of intact rock become functions in the equation. Hence, the use of time dependent elastic properties determined under constant loading will yield more reliable results than when only constant elastic properties are used. As well known, rock material will play an important role in the deformation mechanism since the discontinuities will be closed due to the load. In this study, Ei and ʋ values of intact rocks were investigated under different constant loads for certain rocks with high deformation capabilities. The results indicated significant time dependent variations in elastic properties under constant loading conditions. Ei value obtained from deformability test was found to be higher than the Ei value obtained from the constant loading test. This implies that when static values of elastic properties are used, the material is defined as more elastic than the rock material itself. In fact, Ei and ʋ values embedded in empirical equations are not static. Hence, this workattempts to emerge a new understanding in designing of safer structures in rock mass by numerical methods. The use of time-dependent values of Ei and ʋ under different constant loads will yield more accurate results in numerical modeling analysis.

Characteristics of Carbonization Residue from Cow and Chicken Manure (우분과 계분에 대한 탄화물의 에너지 특성)

  • The basic properties of cow and chicken manure such as proximate analysis and element analysis were estimated and the comparison to energy characteristics of carbonization residue between cow and chicken manure was evaluated. The optimum carbonization condition of cow and chicken manure was decided by total heating value of carbonization residue which was expressed by multiplying low heating value by yield. The optimum carbonization conditions for carbonization time and temperature can be decided by 15 min, and $350^{\circ}C$ for chicken manure, and 20 min, and $300^{\circ}C$ for cow manure. At the optimum carbonization conditions, low heating values for the carbonization residue of cow and chicken manure are evaluated by 4,378kcal/kg, and 3,462kcal/kg, respectively. The residues of cow manure were satisfied with the standard of solid fuel product. However, the residue of chicken should be improved by materials changes to be used as a renewable energy source.

Studies on the Production of Roughages from Hyun-aspen(Populus Alba × P. Glandulosa) by Steaming-Defibration and Steaming-Explosion (열해섬(熱解纖) 및 폭쇄처리에 의한 현사시 나무의 조사료화(粗飼料化) 연구(硏究))

  • Roughage feeds were produced from Hyun-aspen (Populus alba $\times$ p. glandulosa) by steaming-defibration and steaming-explosion. The objectives of this work were to find proper conditions for the treatment of Hyun-aspen by analyzing the compositional change and digestibility and to investigate the change of physical properties of exploded woods. The results of this work were as follows; 1. The method of steaming-de fibration gave the best producing rate of feedstuffs when the chips were steamed (9kg/$cm^2$ under the pressure) for 10 minutes. The yield and the digestibility of feedstuffs were 84.2% and 38.1%, respectively. It is the merit of this method that feedstuffs manufactured by this method was uniformity in particle size, and facilities of fiberboard factory could be used directly, 2. For defibration of the chip by explosion, the proper condition was steamed under the pressure (20kg/$cm^2$) for 4 minutes. The yield and the digestibility of feedstuffs were 93.4% and 68.1%, respectively. The feedstuffs produced under these conditions had higher nutritional quality than rice straw and this method was considered as the best for making feedstuffs from Hyun-aspen chip. But it is defect that exploded feedstuffs was ununiformity in particle size and had unique odor. The physical properties of the feedstuffs were investigated by a light microscope and a TEM. The feedstuffs produced under the low pressure (20 kg/$cm^2$) still maintained the structure of fibers. However, the feedstuffs produced under the high pressure (28 kg/$cm^2$) resulted in higher de fib ration than these prepared under the low pressure. The highly defibrated feedstuffs recombined with solublized lignin. The crystallinity of feedstuffs was increased by 10% and micelle width increased double after treatment.

Effect of light-emitting diodes on cordycepin production in submerged Cordyceps militaris cultures

  • Light is an important factor for cordycepin production in Cordyceps militaris. We investigated the effects of different light-emitting diode (LED) conditions including various LED wavelengths and their combinations on cordycepin production in Cordyceps militaris cultivated in submerged culture. The results of our study showed that the combinations of LED wavelengths were more beneficial than single LED sources for cordycepin production. Among the three tested wavelength combinations, the greatest effects for cordycepin production were observed for the red:blue light combination at the wavelength ratio of 5:5 or 3:7. The optimal culture conditions were 19.2278 h/day of illumination time; 9.19497 g/50 mL of glucose content in the media; and 53.112 h of cultivation time. Our model predicted a maximum yield of 2860.01 ㎍/mL cordycepin. Finally, to verify the calculated maximum, we performed experiments in the culture media representing the obtained optimum combination and the cordycepin yield of 2412.5 ㎍/mL.

Historical Changes of Sediment Accumulation in Lake Shirarutoro Due to Land Use Development in the Forest Catchment, Kushiro Mire in Northern Japan

  • Eleven lake sediment core samples were obtained and analyzed to develop a chronology using $^{137}Cs$ (in 1963) and two tephra layers (Ko-c2 in 1694 and Ta-a in 1739). Sedimentation rates estimated for the past ca 300 years in Lake Shirarutoro indicated that catchment development has influenced the shallowing process in the lake by increasing sediment production. The sediment yield under initial land-use development conditions for the first two periods was estimated as 514 tons $yr^{-1}$ from 1694 to 1739 and 542 tons $yr^{-1}$ from 1739~1963. The development of the Shirarutoro catchment intensified in the 1960s with deforestation and agriculture activity leading to an increased sediment yield of 1261 tons $yr^{-1}$ after 1963. The sediment yields after intensified land use development, such as forestry and agricultural development, were about 2 times higher than that under initial development conditions, leading to accelerated lake shallowing over the last ca 50 years. Sedimentation rates differed with location in the lake because of spatial variation in the sediment flux from the contributing rivers and their catchments. The sedimentation rates before 1963 were low in all sites except for one site close to the Shirarutoroetoro River. The sedimentation rate in 1739~1963 was accumulated mostly at the inflow of the Shirarutoroetoro River by sediment production associated with forestry for charcoal production and initial agricultural development. The sedimentation rate after 1963 increased. In particular, the southern zone of the lake near the conjunction with the Kushiro River had a high sedimentation rate, which is attributable to sediment inflow back from the Kushiro River during floods.

Comparison of Yield and Growth Characteristics of Platycodon grandiflorum According to the Ploidy Levels and Growing Conditions

  • The present study aimed to compare and investigate the morphological characteristics and yield components according to ploidy level of diploid and tetraploid Platycodon grandiflorum under vinyl-greenhouse and open field conditions. Plant height of diploid and tetraploid P. grandiflorum was 51.3 ㎝, 54.0 ㎝, respectively. The results revealed that the plants grown in the vinyl-greenhouse showed significantly higher growth compared to those grown in the open field. Regardless of the growing place, diploid and tetraploid of P. grandiflorum showed the rapid elongation of internodes after 4 and 3 internodes respectively and elongation tends to be decreased as entering the flower-bud differentiation period. The starting day of flowering in vinyl-greenhouse cultivation was found to be faster than that of the open field cultivation by 2∼3 days and tended to be delayed by about 5∼6 days in tetraploid P. grandiflorum compared to diploid. Fresh weight of roots from the vinyl-greenhouse cultivation showed a high quantity as 34.2g and 49.4g in diploid and tetraploid P. grandiflorum, respectively and especially tetraploid P. grandiflorum was found to be increased by approximately 44.4% compared to other plots.

  • Crude papain extracted at optimum condition was purified with an ethanol precipitation method. Four factors of protein recovery method were optimized by response surface methodology (RSM) and the function was expressed in terms of a quadratic polynomial equation. Adequacy of the model equation for optimum response values was tested and optimum conditions of protein recovery were 38.2 mg/mL of protein, ethanol concentration of 40% and precipitation temperature of $-8^{\circ}C$. The experimental value (68.97%) for recovery yield was closed to the predicted value (77.28%) under these conditions.

Emergence Rate and Growth Characteristics of Ginseng Affected by Different Types of Organic Matters in Greenhouse of Direct-Sowing Culture (비닐하우스에서 인삼 직파재배 시 유기물 처리에 따른 연차간 입모율 및 생육특성)

  • Shading and soil environment are the main factors of growth and yield in ginseng (Panax ginseng C. A. Meyer). Ginseng yield is directly related to survival rate because of increased missing plant for their growing period. Under field conditions, diseases and pests significantly affect plant survival rate. We evaluated the seedling establishment, growth and ginsenoside of the ginseng plants, under controlled management conditions in a plastic greenhouse, when their treated with different types of organic matter. Ginseng seeds were sown at a rate of three seeds per hole, and the seeding space measured $10cm{\times}15cm$. Compared to the control, treatment of cattle manure vermicompost (CMV) was shown to increase seedling establishment and decrease ginsenoside content. Root weights of plants treated with CMV were higher than those of plants treated with other types of organic matter. In addition, seedling establishment of 2-year-old ginseng plants was decreased when it was compared to that of 1-year-old ginseng plants. Our results indicated that organic matter type and rate were associated with seedling establishment, growth characteristic and ginsenoside content in greenhouse of ginseng direct-sowing culture.

Optimization the Xylose Fractionation Conditions of Pepper Stem with Dilute Sulfuric Acid (농업부산물 고추대 (Pepper Stem)을 이용한 묽은 황산 자일로즈 분별공정의 최적화)

  • Response surface methodology (RSM) was used for optimization the fraction conditions of xylose from pepper stem with dilute sulfuric acid. The independent variables were acid concentration in the range of 1.134 to 2.866%, reaction temperatures in the range of 142.68 to $177.32^{\circ}C$, and hydrolysis time in the range of 6.34 to 23.66 min. were studied. The dependent variables were xylose yield from pepper stem, and the production of by-products, for example, furfural, acetic aicd, HMF etc. Experimental results had a good match with statistical result. The maximum xylose yield obtained in this experiment was 71% concentration.

Optimization for Enzymatic Hydrolysis of Mannitol (만니톨의 효소 가수분해 반응 조건 최적화)

  • This study aimed to investigate the enzymatic hydrolysis of mannitol using Viscozyme$^{(R)}$ L, Celluclast$^{(R)}$ 1.5 L, Saczyme$^{(R)}$, Novozym$^{(R)}$, Fungamyl$^{(R)}$ 800 L, Driselase$^{(R)}$ Basidiomycetes sp., and Alginate Lyase, and to optimize of reaction conditions for production of reducing sugar. Response surface methodology (RSM) based on central composite rotatable design was used to study effects of the independent variables such as enzyme (1-9% v/w), reaction time (10-30 h), pH (3.0-7.0) and reaction temperature ($30-70^{\circ}C$) on production of reducing sugar from mannitol. The coefficient of determination ($R^2$) of $Y_1$ (yield of reducing sugar by Viscozyme$^{(R)}$ L) and $Y_3$ (yield of reducing sugar by Saczyme$^{(R)}$) for the dependent variable regression equation was analyzed as 0.985 and 0.814. And the p-value of $Y_1$ and $Y_3$ showing 0.000 and 0.001 within 1% (p < 0.01), respectively, was very significant. The optimum conditions for production of reducing sugar with Viscozyme$^{(R)}$ L were 9.0 % (v/w) amount of enzyme, 30.0 hours of reaction time, pH 4.5 and $30.0^{\circ}C$ of reaction temperature, and those with Saczyme$^{(R)}$ were 9.0% (v/w) of amount of enzyme dosage, 30.0 h of reaction time, pH 7.0 and $30.0^{\circ}C$ of reaction temperature, consequently, the predicted reducing sugar yields were 22.5 and 27.9 mg/g-mannitol, respectively.
