Development of high yield rice of long grain type adaptable to South-East Asia tropical region

  • The long grain rice varieties adaptable to South-east Asia tropical regions were tried to develop in Cambodian Agriculture Research and Development Institute (CARDI), Cambodia. The final goal is to develop rice varieties which can culture in diverse environmental conditions of tropical regions of South-east Asia under climate change. We collected and evaluated for agronomic traits of 131 rice germplasm from Cambodia, China, India, Indonesia, Myanmar, Philippines and Vietnam in CARDI. We selected core germplasm including leading varieties of target countries and made 813 F1 cross combinations between leading varieties of each country and promising germplasm of high yield potential, resistance to biotic/abiotic stresses, aromatic rice, and so on. Out of 607 F1s evaluated to heading date, plant type, agronomic traits, and grain type, 106 F1s selected and advanced to F2 populations. 106 F2 populations were evaluated to major agronomic traits, grain type and yield-component traits, and selected 2,560 plants in 62 F2 populations. During six seasons in 2014~2016, the lines of F3 subsequent-generation were cultured a total of 6,256 lines. In yield trial for promising lines in F5 generation, the growth duration from sowing to harvesting was 97~114 days. These lines were 88~129 in number of grain per panicle, an average of 84.6% in the range of 79.3~91.9% in the percentage of ripened grain and 17.5~22.8g in 1000-brown rice weight. The rough rice yields were in the range of 4.33~6.06 ton/ha with an average of 5.23 ton/ha. The yield was increased to 5~47% than Chulsa and 12~41% than IR66. Five lines, KR54-28-1, KR55-14-2, KR57-5-2, KR67-57-2 and KR128-19-1 were 5.33~6.06 ton/ha in rough rice yield. These high yield potential lines would be evaluated to adaptability in Cambodia, Laos, Myanmar and Vietnam during 2017.

Application of Factorial Experimental Designs for Optimization of Cyclosporin A Production by Tolypocladium inflatum in Submerged Culture

  • A sequential optimization strategy based on statistical experimental designs was employed to enhance the production of cyclosporin A (CyA) by Tolypocladium inflatum DSMZ 915 in a submerged culture. A 2-level Plackett-Burman design was used to screen the bioprocess parameters significantly influencing CyA production. Among the 11 variables tested, sucrose, ammonium sulfate, and soluble starch were selected, owing to their significant positive effect on CyA production. A response surface methodology (RSM) involving a 3-level Box-Behnken design was adopted to acquire the best process conditions. Thus, a polynomial model was created to correlate the relationship between the three variables and the CyA yield, and the optimal combination of the major media constituents for cyclosporin A production, evaluated using the nonlinear optimization algorithm of EXCEL-Solver, was as follows (g/l): sucrose, 20; starch, 20; and ammonium sulfate, 10. The predicted optimum CyA yield was 113 mg/l, which was 2-fold the amount obtained with the basal medium. Experimental verification of the predicted model resulted in a CyA yield of 110 mg/l, representing 97% of the theoretically calculated yield.

Somatic Cells Count and Its Genetic Association with Milk Yield in Dairy Cattle Raised under Thai Tropical Environmental Conditions

  • Somatic cells count (SCC), milk yield (MY) and pedigree information of 2,791 first lactation cows that calved between 1990 and 2010 on 259 Thai farms were used to estimate genetic parameters and trends for SCC and its genetic association with MY. The SCC were log-transformed (lnSCC) to make them normally distributed. An average information-restricted maximum likelihood procedure was used to estimate variance components. A bivariate animal model that considered herd-yr-season, calving age, and regression additive genetic group as fixed effects, and animal and residual as random effects was used for genetic evaluation. Heritability estimates were 0.12 (SE = 0.19) for lnSCC, and 0.31 (SE = 0.06) for MY. The genetic correlation estimate between lnSCC and MY was 0.26 (SE = 0.59). Mean yearly estimated breeding values during the last 20 years increased for SCC (49.02 cells/ml/yr, SE = 26.81 cells/ml/yr; p = 0.08), but not for MY (0.37 kg/yr, SE = 0.87 kg/yr; p = 0.68). Sire average breeding values for SCC and MY were higher than those of cows and dams (p<0.01). Heritability estimates for lnSCC and MY and their low but positive genetic correlation suggested that selection for low SCC may be feasible in this population as it is in other populations of dairy cows. Thus, selection for high MY and low SCC should be encouraged in Thai dairy improvement programs to increase profitability by improving both cow health and milk yield.

Photoproduction of Hydrogen from Acetate by Rhodopseudomonas: Effect of Culture Conditions and Sequential Dark/Light Fermentation

  • Rhodopseudomonas palustris P4 can produce $H_2$ either from CO by water-gas shift reaction or from various sugars by anaerobic fermentation. Fermentative $H_2$ production by P4 is fast, but its yield is relatively low due to the formation of various organic acids. In order to increase $H_2$ production yield from glucose, P4 was investigated for the photo-fermentation of acetate which is a major by-product of fermentative $H_2$ production. Experiments were performed in batch modes using both light-grown and dark-grown cells. When the dark-grown P4 was challenged with light and acetate, $H_2$ was produced with the consumption of acetate after a lag period of 25 h. $H_2$ production was inhibited when a nitrogen source, especially ammonium, is present. When the dark-fermentation broth containing acetate was adopted for photo-fermentation with light-grown cells, $H_2$ production and concomitant acetate consumption occurred without a lag period. The $H_2$ yield was estimated as 2.4 - 2.8 mol $H_2/mol$ acetate and the specific $H_2$ production rate was as 9.8 ml $H_2/g$ cell${\cdot}$h, The fact that a single strain can perform both dark- and light-fermentation gives a great advantage in process development Compared to a one-step dark-fermentation, the combined dark- and light-fermentation can increase the $H_2$ production yield on glucose by two-fold.

Responses of Capsicum annum (red pepper) to Fertilization Rates at Various Soil Moisture Conditions

  • This research was performed to test the hypothesis that the optimal fertilization rate for red pepper is changed by soil moisture condition. The experiment was conducted in rainfall-intercepted fields in Suwon, South Korea from 2002 to 2003. Soil was irrigated at 30, 50, or 80 kPa of soil moisture tension at 20 cm soil depth in 2002 and 30, 50, 100, or 150 kPa in 2003. For both years, fertilization was performed with four levels: none, 0.5, 1, and 1.5 times of the recommended N, P, and K fertilization rate. The irrigation amount was the greatest at 30 kPa irrigation while the water use efficiency increased with decrease of irrigation amount. The Irrigation amount was 508 mm at 30 kPa irrigation and ranged from 355 mm to 435 mm at 50 kPa irrigation. The maximum yield was found at 30 kPa irrigation and 1.5 times of the recommend fertilization rate in 2002 and 2003. The yield index of red pepper increased linearly with the fertilization rate at 30 kPa which implied that excess irrigation induced nutrient leaching and reduced nutrient availability. The maximum yield in 50 kPa and 80 kPa was found at the recommend fertilization rate while the yield decreased by fertilization at 100 kPa and 150 kPa irrigation. It implies that reduction of fertilization is the feasible practice to mitigate drought stress in fields without stable irrigation resources.

Spray Drying of Lignocellulose Nanofibril (LCNF) and Characterization of Spray-dried LCNF (리그노셀룰로오스 나노피브릴의 분무건조 및 건조물의 특성)

  • In this study, the effect of spray-drying conditions and surfactant addition on the spray-drying yield, morphological characterization, size distribution and re-dispersity in water of spray-dried lignocellulose nanofibril (LCNF) were investigated. The freeze-dried LCNF after solvent exchange had linear fiber morphology with a diameter of 70-300 nm, and the spray-dried LCNF showed rod-like particle morphology. The spray-drying yield and particle size of spary-dried LCNF at $140^{\circ}C$ was highest and smallest, respectively. As LCNF concentration and blowing rate decreased and increased, respectively, the spray-drying yield and particle size were increased. The highest spray-drying yield was found at distearyl dimethyl ammonium chloride (DDAC) addition of 10 phr at $140^{\circ}C$. As the particle size decreased and the DDAC content increased, filtration time of spray-dried LCNF in water was decreased and increased, respectively.

Amino Acids Recovery from Fish Entrails by Hydrolysis in Sub- and Supercritical Water (생선내장의 아임계 및 초임계 가수분해에 의한 아미노산의 회수)

  • A resource recovery technique using sub- and supercritical water hydrolysis was applied to recover amino aicds from waste fish entrails. The effect of reaction parameters such as temperature and time necessary for the control of reaction towards optimum yield of amino acids was investigated using semi-batch and batch reactors. Results showed a maximum yield of total amino acids (137 mg/g-dry entrails) from waste fish entrails at T=$250^{\circ}C$ (P=4 MPa) and reaction time of 60 min in a batch reactor. Under supercritical conditions (e.g., T=$400^{\circ}C$, P=45 MPa), the yield decreased due to rapid decomposition compared to production rate of amino acids. As a result, the low temperature and the short reaction time were needed to produce a maximum yield of amino acids.

Effects of Feeding Methylthio Butyric Acid Isopropyl Ester on Postpartum Performance and Metabolism in Dairy Cows

  • The present experiment aimed to evaluate the effect of HMBi on the production performance and metabolism in dairy cows. Thirty multiparous Holstein dairy cows under similar conditions were randomly assigned to three dietary treatments; i) Control, a basal diet; ii) T1, a basal diet plus HMBi (0 g prepartum and 18 g postpartum); and iii) T2, a basal diet plus HMBi (10 g prepartum and 18 g postpartum). Treatments were initiated 21 d before expected calving and continued through 91 d postpartum. HMBi was top-dressed onto the total mixed ration of each cow. Treatments did not affect dry matter intake, plasma urea nitrogen, peak milk yield, days to peak milk yield, nonesterified fatty acid, glutamate pyruvate transaminase, glutamic oxalaetic transaminase, milk fat content, milk protein content, milk lactose content, and milk solid non-fat content. The milk composition yields were increased by the HMBi-supplemented treatment. The T1 and T2 treatments increased the yields of 4% fat-corrected milk yield, milk fat, milk protein, and milk lactose compared with the control. Although there was no difference in the milk composition of the control and T2-treated cows, the T2-treated cows exhibited higher milk fat yield (increased by 74 g/d), lower milk urea nitrogen (reduced by 3.41%), and plasma ${\beta}$-hydroxy butyrate than the control cows. The results indicate that HMBi supplementation to diet has beneficial effects, and that there is no difference between supplementation at prepartum and starting only at parturition.

A Study on the Kinetics and the Biogas Formation for Organic Wastewater Treatment in Anaerobic Fluidized-Bed Bioreactor and New Model AFPBBR (혐기성 유동층 생물 반응기와 새로운 모델의 AFPBBR에서 유기성폐수 처리시 Biogas 생성과 반응상수에 관한 연구)

  • The anaerobic digestion of organic synthetic wastewater in anaerobic fluidized bed bioreactor (AFBBR) and anaerobic fluidized packed bed bioreactor (AFPBBR) was studied. This study was conducted to evaluate efficiency and reliability of two reactor. Experiment was performed to find the effect of upflow rate with AFBBR and the height of packed bed with AFPBBR. As a result, this program obtained several conclusion. These are given as follows: As applied the upflow rate increased in AFBBR the produced volume of biogas increased, while the gas production and COD removal decreased at above 0.3 m$^3$/h. When a upflow rate is 0.4 m$^3$/h in AFBBR the volatile suspended solid (VSS) became significantly increased. At an organic loading rate from 0.1 to 0.4 of upflow rate in AFBBR, the methane yield was 1.5584 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0933 gVSS/gCOD. In case of AFPBBR, the results showed also that 20 cm of height of packed bed was superior to other in the aspect ot biogas production, the content of methane and COD removal. At 20 cm of height, the profile of microorganisms was stable, while at 30 cm the VSS of effluent became higher than AFBBR. Though COD removal of AFPBBR increased with packed bed, COD removal deteriorate with over packing because the loss of pressure became higher in the reactor. At an organic loading rate from 20 to 40 cm of packed bed in-AFPBBR, the methane yield was 2.5649 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0506 gVSS/gCOD. Based upon the results obtained, it is suggested that AFBBR and AFPBBR is the most effective conditions at 0.3 m3/h of upflow rate, the 20cm of packed bed, respectively.
