A Constitutive Model on the Behavior Under $K_0$ Condition for Cohesionless Soils and Optimization Method of Parameter Evaluation Based on Genetic Algorithm (사질토의 $K_0$ 조건하 거동에 대한 구성모델 및 유전자 알고리즘을 적용한 계수의 최적화 산정기법)

  • This study is focused on the constitutive model in order to represent brittleness and dilatancy for cohesionless soils. The constitutive model was based on an anisotropic hardening rule derived from generalized isotropic hardening nile, which includes an appropriate hardening equation for the overall strain behavior at small to large strains. The yield surface is a simple cylinder type in stress space and it makes the model practically useful. Hence dilatancy behavior in cohesionless soils could be modeled reasonably. A peak stress ratio was defined in order to model brittle stress-strain relationships. An optimized design methodology was proposed on the basis of real-coded genetic algorithm in order to determine parameters for the proposed model systematically. The material parameters were then determined by that algorithm. In order to verify the proposed model, triaxial tests were performed under $K_0$ conditions far weathered soils. In comparison with the triaxial test results under $K_0$ conditions, the proposed model could calculate appropriately the actual effective stress behavior on brittle stress-strain relationships and dilatancy.

Production and Characterization of Keratinase from Paracoccus sp. WJ-98

  • A bacterial strain WJ-98 found to produce active extracellular keratinase was isolated from the soil of a poultry factory. It was identified as Paracoccus sp. based on its 16S rRNA sequence analysis, morphological and physiological characteristics. The optimal culture conditions for the production of keratinase by Paracoccus sp. WJ-98 were investigated. The optimal medium composition for keratinase production was determined to be 1.0% keratin, 0.05% urea and NaCl, 0.03% K$_2$HPO$_4$, 0.04% KH$_2$PO$_4$, and 0.01% MgCl$_2$$.$6H$_2$O. Optimal initial pH and temperature for the production of keratinase were 7.5 and 37$^{\circ}C$, respectively. The maximum keratinase production of 90 U/mL was reached after 84 h of cultivation under the optimal culturing conditions. The keratinase from Paracoccus sp. WJ-98 was partially purified from a culture broth by using ammonium sulfate precipitation, ion-exchange chromatography on DEAE-cellulose, followed by gel filtration chromatography on Sephadex G-75. Optimum pH and temperature for the enzyme reaction were pH 6.8 and 50$^{\circ}C$, respectively and the enzymes were stable in the pH range from 6.0 to 8.0 and below 50$^{\circ}C$. The enzyme activity was significantly inhibited by EDTA, Zn$\^$2+/ and Hg$\^$2+/. Inquiry into the characteristics of keratinase production from these bacteria may yield useful agricultural feed processing applications.

  • Optimization of the medium and fermentation conditions for erythritol production has been studied. We have found that the optimal carbon source was glucose at the concentration of 400 g/L. The optimal temperature was 3$0^{\circ}C$ with excessive aeration. Improved erythritol productivity was achieved by reducing the yeast extract from 5 g/L to 3g/L while adding 2.7 g/L urea, 1.79g/L $K_2HPO_4, and 0.18g/L MgSO$_4$. 7$H_2O. The erythritol productivity increased from 0.747 g/L/h to 1.071 g/L/h and the yield increased from 31.4% to 45.2%. The byproduct glycerol was reduced from 96.6g/L to 45.7g/L as well. We have performed 5L fermentation with and without the pH control. The erythritol productivity with the pH control was about 30% lower than that without pH control. Excessive foaming of 5L fermentation has been observed during fermentation.

Impact of Shading Net Color on Phytochemical Contents in Two Chili Pepper Hybrids Cultivated Under Greenhouse Conditions

  • The objective of this study was to understand the combined effect of shading treatment and harvest time on pungency, ascorbic acid content, market-quality pepper yield, and photosynthetic activity in a yellow 'Star Flame' and 'Fire Flame' F1 hybrid pepper cultivar (Capsicum annuum). Plants were cultivated in a greenhouse under white, red, and green shading nets and compared to those grown under unshaded control conditions. The ascorbic acid and pungency parameters were analyzed by high-performance liquid chromatographic methods. Ascorbic acid, a compound of high nutritive value in pepper, was highest in fruits gathered during the first summer harvest in both hybrids. The ascorbic acid content was found to be further increased in fruits from plants grown under white net shading in both 'Star Flame' and 'Fire Flame'. Total capsaicinoid contents in 'Star Flame' were influenced by the interaction of shading treatment and harvest time (p = 0.004), with the lowest amounts ($241.2-251.5mg{\cdot}kg^{-1}$ fresh weight) resulting from white and red shading treatments at the early autumn harvest. In 'Fire Flame', both shading treatment (p = 0.009) and harvest time (p < 0.001) affected total capsaicinoid contents. We observed significantly higher total capsaicinoid contents in control fruits compared to that resulting from red and green shading treatments at the second harvest, and that resulting from red shading treatment at the third harvest. Analysis of photosynthetic activity revealed a significant and negative correlation between Fv/Fm values and total capsaicinoid contents in both hybrids (p < 0.001). Based on the obtained results, the harvest of peppers during summer should be promoted to produce fruits with high pungency.

Determination of Boxthorn Drying conditions and using Agricultural Dryer (구기자 품질향상을 위한 적정 건조조건 구명)

  • This study was carried out to define the optimum drying conditions for Lycium chinense Miller as a useful healthy food, because recently the cultivation area and yield of this fruit are increased. The experiments of two varieties were performed at the temperature of $45^{\circ}C$, $50^{\circ}C$, $55^{\circ}C$ and $60^{\circ}C$. The drying ratio was the slowest and quality was the best at the drying temperature of $45^{\circ}C$. The drying temperature was higher, drying ratio was more faster and the quality became worse. The difference of drying ratios between the varieties was insignificant. The energy consumption per hour was the minimum at the drying temperature of $45^{\circ}C$, but the total energy consumption was the maximum for the long drying time. Also, the energy consumption at the drying temperature $50^{\circ}C$, $55^{\circ}C$ and $60^{\circ}C$ was not very different from others. Considering the drying ratio, quality and energy consumption, the drying time of 36 hours at the drying temperature of $50^{\circ}C$ was the most optimum condition.

Optimization of Transglucosylation Reaction of Stevioside (스테비오시드 당전이 반응의 최적화)

  • Stevioside was transglucosylated to improve its sweetness. Eighteen conditions were tested using the tables of orthogonal arrays of $L_{18}\;(2^{1}{\times}3^{7})$. Statistical analysis showed that the transglucosylation rate was significantly affected by temperature, the ratio of co-substrate (maltodextrin) to stevioside, pH, DE of maltodextrin and concentration of stevioside, in their order. Optimum conditions selected for temperature, ratio of co-substrate to stevioside, pH and DE of maltodextrin were $80^{\circ}C$, 1.0, 6.0 and 15, respectively. Glycosyl-stevioside with 68% yield of transglucosylation was produced at the optimum condition and found to have better quality in sweetness than stevioside and rebaudioside A.

Evaluation and Verification of Barley Genotypes with Known Genes for Resistance to Barley yellow mosaic virus and Barley mild mosaic virus Under Field Conditions in South Korea

  • Soil-borne barley yellow mosaic disease caused by Barley yellow mosaic virus (BaYMV) or Barley mild mosaic virus (BaMMV) gives a serious threat to the winter barley cultivated in the southern regions in Korea. It is important to develop resistant varieties for stable and high-yield production. The objectives of this study were to evaluate 22 genotypes of exotic barley germplasms carrying the resistance genes rym1 through rym12, with the exception of rym10, and to determine the genes that confer resistance to BaYMV or BaMMV in Korea. Using the traditional visual scoring of symptoms at 4 locations over 3 years, average disease rate values differed (P < 0.001) among the genotypes. ELISA test revealed the presence of both BaYMV and BaMMV in all of the field sites but Jinju and significantly different rates of infection among genotypes and years. Barley genotypes differed in how virus quantities and pathogen-induced symptoms were correlated, especially in response to BaYMV. Disease incidence was affected by the climatic conditions present during the early growing stage before overwintering. A Chinese landrace, 'Mokusekko 3', carrying rym1 and rym5 was comparatively resistant at all locations studied. The barley genotypes carrying either rym6 or rym9 were susceptible to the viral strains. The genotypes carrying rym5 were resistant in Jinju and Milyang but susceptible in Iksan and Naju. The resistance genes rym2 and rym3 were effective in local strains and would be potent contributors to disease resistance.

  • Adverse environmental conditions such as drought, high salt and cold/freezing are major factors that reduces crop productivity worldwide. According to a survey, $50-80\%$ of the maximum potential yield is lost by these 'environmental or abiotic stresses', which is approximately ten times higher than the loss by biotic stresses. Thus, Improving stress-tolerance of crop plants is an important way to improve agricultural productivity. In order to develop such stress-tolerant crop plants, we set out to identify key stress signaling components that can be used to develop commercially viable crop varieties with enhanced stress tolerance. Our primary focus so far has been on the identification of transcription factors that regulate stress responsive gene expression, especially those involved in ABA-mediated stress response. Be sessile, plants have the unique capability to adapt themselves to the abiotic stresses. This adaptive capability is largely dependent on the plant hormone abscisic acid (ABA), whose level increases under various stress conditions, triggering adaptive response. Central to the response is ABA-regulated gene expression, which ultimately leads to physiological changes at the whole plant level. Thus, once identified, it would be possible to enhance stress tolerance of crop plants by manipulating the expression of the factors that mediate ABA-dependent stress response. Here, we present our work on the isolation and functional characterization of the transcription factors.

A Comparative Study on Responses to Korean Version Questionaires on Respiratory Symptoms (호흡기계(系) 질환의 역학적(疫學的) 조사방법 개발에 관한 연구(I) -번역설문서 응답양상(應答樣相)에 대한 비교평가-)

  • Korean versions of British Medical Research Council (MRC), Cornell Medical Index(CMI), and American Thoracic Society 78 (ATS-DLD-78) respiratory questionaires were compared with each other, and were tested the stability, in terms of test-retest reliability, of each questions by self-administration of those to 156 medical students. The results obtained and conclusions drawn are as follow: 1. The degree of agreements between responses to the comparable questions of CMI vs MRC, and of CMI vs ATS-DLD-78 were not satisfactory. There were, however, $71{\sim}100$ per cent of agreement between responses to the questions on Cough, Wheezing, Phlegm, Breathlessness, and Chest illness of ATS-DLD-78 vs MRC questionaire. And the ATS-DLD-78 tended to yield greater number of positive responses than MRC (See Table 4). 2. All of the coefficient of stability of each questions in 3 questionaires were statistically significant, ranged $77{\sim}100$ per cent, except that of the question on episode of cough and phlegm in ATS-DLD-78 questionaire (See Table 5-1). The question is composed of two collateral conditions, 'lasting for 3 weeks or more' and 'each year'. 3. It can be insisted that the Section-B questions of CMI is not proper for use in epidemiologic survey on respiratory illness. And rather than MRC, the ATS-DLD-78 questionaire deserves to prefer to be used in epidemiologic studies on respiratory illness. 4. In question-wording, especially, of inquiring past experience, it is possible to lessen the reliability of the question that including collateral conditions such as 'the duration lasted of symptoms', and moreover, of which words are not common usage. For example, for Korean '10days' or 'half a month' is more familiar time unit rather than 'week'.

  • The effect of dissolved oxygen(DO) concentration on the growth of Pseudomonas aeruginosa and it's mutant M-10 was studied the growth kinetics and the possibility of waste treatment for reducing the amount of excess sludge. Different DO concentrations on the growth of wild type Pseudomonas aeruginosa affected to cellular yields, decreasing with increasing DO concentrations. Under these conditions, the maximum 14 folds decrease of cellular yield was achieved at 90 ppm DO levels by screened, being able to reduce their cellular yields under low DO concentration. The optimum mutation conditions were obtained by the treatment of NTG mutagen at 30${\circ}C$ for 1 hr. The growth characteristics of the selected mutant M-10 showed the same as the wild type growth kinetics. However, cellular yields are significantly decreased to 55% compared with those of wild type under DO concentrations increased. We, therefore, expect the application of the mutants to waste treatment for reducing excess sludge.
