Edible Culture Media from Cereals and Soybeans for Pre-cultivation of Lactic Acid Bacteria (곡류 및 두류를 이용한 젖산균 전배양용 식용 배지의 제조)

  • This study was conducted to develop an edible culture media with various types of cereals and soybeans for the pre-cultivation of lactic acid bacteria (LAB). To manufacture the edible culture media, LAB enrichment media were prepared using cereals such as brown rice (including germinated brown rice, glutinous brown rice, and germinated glutinous brown rice), yellow soybeans (including yellow soybeans, hulled yellow soybeans, germinated yellow soybeans, hulled and germinated yellow soybeans), and black soybeans (black soybeans, hulled black soybeans, germinated black soybeans, hulled and germinated black soybeans). Seven species of LAB were used in the experiment: Lactobacillus (Lb.) farciminis, Lb. homohiochii, Lb. pentosus, Lb. plantarum, Leuconostoc (Leu.) paramesenteroides, Leu. citreum, and Leu. lactis. For edible culture media from cereals, the average viable cell count of the seven starter cultures was 7.6~8.0 log CFU/mL, while that of the MRS culture medium, a synthetic medium, was 9.2 log CFU/mL; thus proliferation was lower by about 1~2 log CFU/mL in starter cultures from cereals compared to the synthetic medium. In the case of the edible culture media from soybeans, most bacteria showed higher proliferation in the hulled and germinated soybean media. In particular, Lb. plantarum showed the highest cell count at 10.08 log CFU/mL. In the case of edible culture media from black soybeans, the proliferation rate was higher in the hulled and germinated black soybean medium. Lb. homohiochii showed the highest proliferation in the hulled and germinated black soybean medium at 9.90 log CFU/mL. All results show that edible culture media using cereals and soybeans are generally good for LAB. Especially, hulled and germinated black soybeans are optimal for the pre-cultivation of LAB medium.

Effect of Raw Soy Flour(yellow and black) on Serum Glucose and Lipid Concentration in Streptozotocin-induced Diabetic Rats (대두(생콩)급여가 당뇨쥐의 혈당 및 지질농도에 미치는 영향)

  • This study investigated the effect of raw soybean yellow(RSY) and raw soybean black(RSB) on body weights, pancreas weights, serum glucose and lipid concentrations of boty normal and diabetic male rats(Sprague-Dawley, body weight 338.8$\pm$19.2g). For each experimental, some rats were injected with streptozotocin intraperitoneal(i.P.) to induce diabets, and other rats were injected with buffer i.P. as a control group. Body weight gain(BWG) was lower in rats fed both RSY and RSB diet(3.4~5.0g BWG) than in those fed control diet(58.1g BWG). The diabetic rats showed significant decrease of bo요 weight (-50~-67g) compared with the control rats. RSY and RSB feeding resulted in greater pancreatic weight relative to body weight, and diabetic rats fed RSY and RSB had heavier pancreatic weights relative to body weight than control rats. The levels of fasting serum glucose in diabetic rats fed both RSY and RSB decreased by 42 and 31%, respectively, compared with the diabetic control rats, but they increased by 80 and 110% compared with the normal control rats. The concentrations of total lipid, triglyceride, phospholipid and total cholesterol in serum of diabetic rats fed RSB diet were lower than those in control rats, but serum HDL-cholesterol, HDL-cholesterol/total cholesterol ratio were not affected by raw soybeans feeding. Atherogenic index wa lower in diabetic rats fed RSY and RSB diet than those in control rats. The concentrations of total lipid, triglyceride and phospholipid in liver were lower in raw soybean groups than those in control group. The concentrations of cholesterol in liver of groups treated raw soybean were decreased compared with the control group.

Antioxidant and Anticancer Activities of Glycine Semen Germinatum Fermented with Germinated Black Soybean and Some Bacteria (발효콩 대두황권의 항산화 및 항암효과)

  • This study was conducted to evaluate the antioxidant (ABTS, DPPH radical scavenging and reducing power), nitric oxide (NO)) production and anticancer activities against human cancer cells (HeLa HepG2, HT-29 and MCF-7) for methanol extracts of Glycine Semen Germinatum (Daedoowhangkeun in Korean) fermented with germinated black soybean and some bacteria. Antioxidant activities were increased by increasing the concentration of the extract at dose-dependent manner, Their activities of black soybean were higher than those of yellow soybean. Non fermented sample was slightly higher than Glycine Semen Germinatum fermented with Bacillus subtilis and lactic acid bacteria. In 1 mg/mL of the extract NO production levels were $0.374\;{\mu}M$ for yellow soybean, $0.368\;{\mu}M$ for black soybean, and $0.367\;{\mu}M$ and $0.358\;{\mu}M$ for Glycine Semen Germinatum fermented with B. subtilis and lactic acid bacteria, respectively. Methanol extract of Glycine Semen Germinatum fermented with mixture broth of lactic acid bacteria was shown to be the highest activity for anticancer activities against human cancer cells tested and their activities were exhibited in the order of HeLa > HT-29 > HepG2 > MCF-7 cell.

Evaluation of External Quality of Brand Soybeans (콩 시판 브랜드 제품의 외관 품질 평가)

  • Although high nutritional values and continuous identification of important functional substances of soybean [Glycine max (L.) Merrill.] promote consumption of soybean products worldwide, informations on quality of brand soybean is not enough for consumers. Total of 100 brand soybeans [32 for soypaste and source, 45 black testa (lage), and 17 black testa (small) or medicinal soybean and beansprout soybean] were collected at supermarkets and several external quality factors were analyzed. Brand soybeans were marked with the environmental friendly and intimating words along with soybean (white or yellow), black soybean (black-, frost-, late frost-, green or inner-green-), medicinal soybean and beansprout soybean. Among 100 brand soybeans 30% was 1 kg package and 59% was 500 g package, difference between printed and actual weights of 70% brand soybeans was ${\pm}1%$ and weights of 2/3 of brand soybeans were higher than printed weight. Range of 100 seed weights of soypaste and source, black testa (large) and black testa (small) and beansprout soybeans were $23.7{\sim}47.8g$, $21.9{\sim}44.5g$ and $9.5{\sim}15.0g$, respectively. Although ranges of 100 seed weights of soypaste and source and black testa (large) soybeans were similar, 63% of soypaste and source were less than 29 g, while 78% of black testa (large) soybeans were higher than 30 g. Although average and highest percentages of seeds separated with 6.7 mm sieve were similar with 87.4% and 99.9% for soypaste and source soybean and 86.5% and 99.5% for black testa (large) soybean, respectively, the lowest percentages were 70.7% for soypaste and source soybean and 14.4% for black testa (large) soybean. When 100 seed weight was greater than 35 g, 90% of seeds were remained on 6.7 mm sieve. On the other hand 100 g weight and percentage of seeds remained on 6.7 mm sieve showed significantly positive correlations [r=0.7488** for soypaste and source soybean and r=0.7874** for black testa (large) soybean when 100 seed weight was $20{\sim}30g$. Based on hilum color and/or appearance, 76% of brand soybeans collected (more than 90% in yellow testa soybeans) were found to be mixed more than 10% with other cultivars or landraces. Foreign materials such as sand, piece of clothe, wood piece, dead insects, other soybeans were found in 20% of brand soybeans. Average test weight of brand soybeans was 762g $L^{-1}$ with a range of $645{\sim}820g\;L^{-1}$. Soybeans from local markets were as good as brand soybeans in 100 seed weight, uniformity of seeds, weight of foreign materials and test weight.

  • Soybean has been cultivated in China for 5,000 years. The soybean cyst neamtode (SCN), Heterodra glycines, was recongnized in Northeastern China in 1899. Currently, it is known to occur in 12 provinces. The biology of SCN was investigated in several provinces. Six races of SCN were identified (race 1, 2, 3, 4, 5 and 7). About f10,000 soybean germplasm lines were evaluated for their resistance to race 1, 3, 4 and 5 of SCN. At least two black-seeded cultivars are resistant to all four races. Several tolerant soybean cultivars with yellow seed coat were released and are in production. Additional resistant cultivars are being developed. Nematicides were not applied in production Potential biocontrol agents and related aspects are being investigated.

Effect of Mixed Soybeans Materials on Qulity Characteristics of Traditional Soybean Paste(Doenjang) during Aging (혼합콩이 전통된장의 숙성 중 품질에 미치는 영향)

  • The effects of five colored soybean (blue, red, yellow, white, and black) varieties on the quality characteristics of soybean paste (Doenjang) were investigated for proximate composition, salt content, pH, color, organic acid content, amino acid content, and mineral content after 80 days of aging. Soybean paste (Doenjang) samples were divided into TDS (traditional Doenjang prepared with soybean) and TDM (traditional Doenjang prepared with mixed soybeans, kidney bean, black soybean, kind of small bean, sword bean, green bean, red bean, at a radio of 1:1:1:1:1(w/w)). Moisture content was 49.66% for TDM and 49.52% for TDM. Protein and salt contents of the TDS sample were higher than those of TDM. The "L" values of the TDB sample increased the aging period was decreased. In contrast, the "L" and "$\alpha$" values of TDM did not show any significant differences. The main organic acids detected were malic acid > lactic acid > tartaric acid in the TDS and TDM samples. Glutamic acid contents were 2.83 g/100 g for the TDS sample and 2.15 g/100 g for the TDM. The levels of K, Mg, and Ca of the TDS sample were higher than those of TDM. No significant difference occurred between TDS and TDM regarding their quality characteristics after 80 days of aging. Further studies needed include sensory evaluation and functionality of soybean paste made from mixed soybean materials.

Soybean Milk Production (대두유생산(大豆乳生産))

  • The utilization of soybean milk has lasted for past several thousand years, mostly in Orient as a Precious food or as a substitute for cow's milk. There are several hundred varieties in soybean that is called Glycine max Merryl There are three typical varieties, Yellow, Black and Green. However, these are due to pigment in soybean cuticle while their chemical constituents are similar (Table 2). Soybean milk is constituted of protein, fat and carbohydrate of soybean, and these constituents are extracted by hot water. Thus becomes emulsion which is highly digestible and nutritious for human diet. However, it was not found any report on the water extractability comparison between two different varieties Black cuticle soybean which is common in India and Yellow cuticle soybean which is common in Korea. Also there was no report on the comparison on the yield and organoleptic quality derived by varied process. Also several attempts were carried out to improve nutritional value as well as acceptability by use of food additives. A model of continuous soybean milk plant was introduced.

A New Black Seed Coat Soybean Cultivar, 'Daeheug' with Lodging Tolerance, and Large Seed Size (검정콩 대립 내도복 신품종 '대흑')

  • A new black seed coat soybean cultivar, 'Daeheug', was developed at the National Institute of Crop Science (NICS) in 2007. The breeding goal for black seed coat soybean is to develop the cultivar with large seed size, high yield, lodging tolerance, and resistance to disease such as soybean mosaic virus (SMV), and bacterial pustule. 'Daeheug' was selected from the cross between 'Daehwang' which had yellow seed coat, large seed size, and late maturity, and 'Milyang 79' which was lodging tolerant, and SMV resistant with medium seed size, black seed coat, and yellow cotyledon. The preliminary, advanced and regional yield trials for evaluation and selection of this cultivar were carried out from 2003 to 2007. It has determinate growth habit, purple flower, brown pubescence, brown pod color, black seed coat, yellow cotyledon, spherical flattened seed shape, oval leaf shape and large seed size (34.3 g/100 seeds). In maturity, 'Daeheug' was 2 days later than the check cultivar 'Ilpumgeomjeongkong'. In the seed quality such as isoflavone contents ($635{\mu}g/g$), and anthocyanin contents ($11.2m{\ell}/g$ of seed coat), 'Daeheug' was better than the check cultivar. It has good adaptability for cooking with rice in ratio of water absorption and seed hardness. Specially, it has good processability for soybean tea with 2.13 absorbance at 530 nm, scavenge activity of DPPH radical was high, and contents of total phenolics was $1,263{\mu}g/m{\ell}$. The average yield of 'Daeheug' was 2.26 ton per hectare in the regional yield trials (RYT) carried out in six locations of Korea from 2005 to 2007.

Taxonomy of Yellow koji mold (Aspergillus flavus/oryzae) in Korea

  • Koji molds are comprised of yellow, black and white. Black and white koji molds were recently re-visited by this author and it is concluded that they consists of Aspergillus luchuesnsis, A. niger and A. tubingensis, and the most important species for alcoholic beverage production is A. luchuensis. In the case of yellow koji mold, it is comprised of Aspergillus oryzae, A. sojae and A. tamari. In the case of A. sojae, the species is scarcely isolated from nature and rarely used for industry in Korea. Aspergillus tamari is often isolated from traditional Korean Meju, a fermented soybean product, and the classification of the species is clear. However, in the case of A. oryzae, differentiation between A. oryzae and A. flavus is still in controversy. In this study, we collected 415 strains of Aspergillus flavus/oryzae complex from air, rice straw, soybean, corn, peanut, arable soil and Meju in Korea and we examined the aflatoxin producing capacity of the strains. The norB-cypA, omtA and aflR genes in the aflatoxin biosynthesis gene cluster were analyzed. We found that 367 strains (88.4%) belonged to non-aflatoxigenic group (Type I of norB-cypA, IB-L-B-, IC-AO, or IA-L-B- of omtA, and AO type of aflR), and only 48 strains (11.6%) belonged to aflatoxin-producible group (Type II of norB-cypA, IC-L-B+/B- or IC-L-B+ of omtA, and AF type of aflR). In the case of A. flavus/oryzae strains from Meju, almost strains (178/192, 92.7%) belonged to non-aflatoxigenic group and only 14 strains (7.3 %) belonged to aflatoxin-producible group. It is proposed in this study that non-aflatoxigenic strain from Meju is classified as A. oryzae, considering that Meju is food material.

Effects of Raw Soy Flour(yellow and black) on Serum Protein Concentrations and Enzyme Activity in Streptozotocin-Diabetic Rats (대두급여가 당뇨쥐의 혈청과 간의 단백질농도 및 효소활성에 미치는 영향)

  • Effects of raw soy flour(RSY) and black(RSB) feeding on protein concentration of liver and serum, and GOT, GPT and alkaline phosphatase activities of serum in diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley), mean weight of (338.4$\pm$19.2g) were assigned to six dietary groups and fed with the assigned diet for 28 days. For each experimental, some rats were injected with streptozotocin intraperitoneally(L.P.) to induce diabets, and other rats were injected with buffer L.P. as a control group. The liver, kidney and spleen weights relative to bo요 weigth were higher in raw yellow soy flour diet diabetes(D-RSY) and black soy flour diet diabetes(D-RSB) groups than control, but the body weights were lower than control. The protein and albumin concentrations of liver and serum were lower in D-RSY and D-RSB groups than control. The albumin concentration of serum in D-RSB group was lower than control. The GOT activities of serum in RSY and RSB groups were increased compared with control, but the GPT activities were lower in diabetic control, D-RSY and D-RSB groups than control. The alkaline phosphatase activities of serum in RSY and RSB groups were higher than control, but those in D-RSY and D-RSB groups were lower than diabetic control.
