The 117 soybean cultivars were collected from nine provinces in Korea, and various seed quality traits along with isoflavone contents were evaluated to elucidate their relationship. The 100-seed weight of the black soybean (31.2 g) was significantly higher (p<0.05) than yellow soybeans (28.6 g). The composition of genistein, daidzein, and glycitein accounted for 75.8, 22.8, and 1.4 % of total isoflavone in yellow soybean cultivars, while their compositions in black soybeans were 58.5, 39.7, and 1.8%, respectively. The mean contents of total isoflavone in yellow and black soybean were $l,561.6{\mu}g\;g^{-1}\;and\;l,018.3{\mu}g\;g^{-1}$. The isofalvone content showed significant variation among cultivars when classified by the seed size. In the yellow soybeans, total isoflavone content was higher in small size soybean cultivars $(1,776.0{\mu}g\;g^{-1})$ and medium size soybean cultivars $(1,714.3{\mu}g\;g^{-1})$ compared to large size ones $(1,518.5{\mu}g\;g^{-1})$. Genistein content was proved as the major factor determining the relationship between isoflavone content and 100-seed weights (r =-0.206*). Daidzein and glycitein, however, showed no significant relationship with the 100-seed weights. Isoflavone content was not significantly correlated with color parameters L (lightness) and a (redness) values, but color parameter b (yellowness) was positively correlated with glycitein (r=0.264*) in the yellow soybeans, while its negative correlation between daidzein (r=-0.245*) and total isoflavone (r=-0.256*) were observed in black soybeans. However, these findings suggested that the seed color value may not serve as an effective parameter for estimating the isoflavone intensity of the soybeans. Variation of protein and lipid contents between yellow soybeans (n=58) and black soybeans (n=59) was relatively stable, however, protein and lipid contents have no significant relationship with isoflavone content.

Analysis of black soybeans [Glycine max (L.) Merr.; 59 Korean varieties] revealed that 100-seed weights of green cotyledon seeds (33.5 g, n=31) were higher than those of yellow ones (28.9 g, n=28). Contents of delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), petunidin-3-glucoside (P3G), and total anthocyanins in seed coats of black soybeans were 0.03-4.15, 0.74-18.36, 0.02-1.60, and 0.87-23.52 mg/g, respectively, among which most prominent anthocyanin was C3G (80.9% of total content), followed by D3G (13.6%) and P3G (5.5%). No significant differences were observed in color parameters $a^*$ and $b^*$ between black soybeans with yellow cotyledon (BYC) and green cotyledon (BGC). Total isoflavone content of BGC was higher than that of BYC, and negative correlation was found between total anthocyanin and isoflavone contents.
The effects of adding hull powders of black soybean on quality characteristics of tobu were studied. Proximate composition of the black soybean hull powders was crude ash, crude protein, crude fat, and carbohydrate at 2.60, 11.35, 2.18, and 73.97%, respectively. The addition of 5% black soybean hull powder decreased the production of whey and consequently increased the yield of tobu. Texture properties of evaluation hardness, adhesiveness, chewiness were the best in tobu which was made with 5% black soybean hull powders. Sensory evaluation showed texture, firmness were not significantly different among tobu with yellow soybean, black soybean, and added to 0.5% black soybean hull powders. At the beginning of the period of storage, there were not differences of microbial cell count as increasing black soybean hull powders, but observed after passing 5 days. The results of total microbial cell count showed that adding 5% black soybean hull powders in processing tobu extend storage time.

The 117 soybeans seeds were collected from the nine provinces of Korea, and protein and lipid contents, and fatty acid composition levels were evaluated to investigate their relationship. The 100-seed weights of the black soybeans were varied $27.7{\sim}33.1g$, while the 100-seed weight of yellow soybeans were varied $24.6{\sim}36.6g$. Protein and lipid content of the 117 soybean seeds was 38.3% and 17.8%, respectively Protein contents of the 59 black soybean seeds (38.6%) were significantly higher than those of the 58 yellow soybean seeds (37.9%). However, lipid contents of the black soybean seeds (17.6%) were lower than those of the yellow soybean seeds (18.1%). Linoleic and oleic acid composition levels of the 117 soybean seeds were 53.75% and 22.08%. Unsaturated fatty acid levels of soybean seeds showed a statistically significant variability among the nine provinces of Korea, however, the differences were not found in the linoleic (18:2) and oleic acid (18:1) levels. Therefore, it was considered that the significant variability of unsaturated fatty acid were mainly due to the variations of linolenic acid (18:3) level. The composition levels of linoleic, oleic, palmitic (16:0), and linolenic acid in the yellow soybean seeds were 53.43%, 22.73%, 12.23%, and 8.24%, while those of the black soybean seeds were 54.13%, 21.48%, 12.47%, and 8.31%. Obtained results suggested that fatty acid composition levels were varied and possibly influence by the phenotype of seed coat colors. Oleic acid, mono-saturated fatty acid, showed the most remarkable variability between yellow and black soybean seeds, and the composition levels were higher in the yellow soybean seeds. Relationship between unsaturated fatty acid levels and 100-seed weights in the yellow soybean seeds showed a negative correlation (r=-0.513, P<0.01), but no relationship $(r=0.154^{ns})$ was observed in the black soybean seeds.
We investigated the effect of black soybean and doenjang with black soybean on production of cytokines including interleukin-2 (IL-2), interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) compared with yellow soybean and doenjang with yellow soybean. We also determined inhibitory effect of two types of soybeans and doenjang on tumor metastasis produced by colon 26-M3.1 carcinoma cells. The cytokine productions of mouse splenocytes increased by the exposure of lipopolysaccharide (LPS, 1 ${\mu}g$/ml) compared to without treatment of LPS. The LPS-induced IL-2 production was highest in the black soybean methanol extract, while the methanol extract from black soybean doenjang fermented for 2 mo showed the higher levels of IL-2 without LPS (p<0.05). In case of LPS-induced IL-6 production, the methanol extracts from control, yellow soybean, black soybean doenjang fermented for 2 and 7 mo showed higher levels of IL-6 compared to those of black soybean, and yellow soybean doenjang fermented for 2 mo (p<0.05). The methanol extract of black soybean doenjang fermented for 2 mo showed significantly higher levels of TNF-${\alpha}$ in both with and without LPS (p<0.05). In experiment of tumor metastasis, the treatment (1 mg/mouse) of methanol extract from black soybean doenjang fermented for 7 mo inhibited tumor metastasis by 50% and had the highest inhibitory effect among other samples (p<0.05). From these results, doenjang manufactured with black soybean modulated the production of cytokine and showed anticancer effect, suggesting that this effect was increased with increased periods of fermentation.

To investigate the antioxidative effect of Korean soybeans, four kinds of soybeans(yellow, brown, black and small black bean) were selected and determined their antioxidative activities in vitro by POV method. Dark colored soybeans such as brown, black and small black soybean showed stronger antioxidative effect than pale yellow one. When compared the antioxidative effect of these soybeans in hull and dehulled condition, in dehulled soybeans, they showed the same antioxidative effect in each soybeans, but in soybean hull, dark colored soybeans showed stronger antioxidative effect than yellow soybean hull, which means the pigments of soybeans play important roles in their antioxidative activities. To separate the main components showing antioxidative effect in soybeans, these soybeans were extracted with hexane, methanol and water. The methanol extract showed the strongest antioxidative effect among them, which means the major antioxidative effect materials contained in methanol extract.

Anthocyanins are naturally occuring phytochemicals and the main components of the coloring of plants, flowers and fruits. They are known to elicit antioxidative, anti-inflammatory and cancer preventive activity. In this study, we investigated anthocyanins in black / yellow soybean seedcoats using different methods of detection - thin layer chromatography (TLC), capillary zone electrophoresis (CZE) and HPLC analysis. The anthocyanins in soybean seedcoats were extracted by five independent methods of extraction and the aglycons (anthocyanidins) of the corresponding anthocyanins were prepared by acid mediated hydrolysis. The anthocyanin / anthocyanidin in black soybean seedcoat showed characteristic TLC mobility, CZE electrophoretic retention and HPLC migration time while little of anthocyanins were detected from yellow soybean seedcoat. The extracted anthocyanins showed pH dependent retention time in CZE and spectral change in UV-Vis spectrum. HPLC analysis of the hydrolyzed extract of black soybean seedcoat identified the presence of four anthocyanidins. The major anthocyanin in black soybean seedcoat was cyanin (cyanidin-3-O-glucoside), with the relative order of anthocyanidin in cyanidin > delphinidin > petunidin > pelargonidin.
The effects of cooking on the dietary fiber content in rice, brown rice, yellow soybean and black soybean were investigated. The dietary fiber contents were analyzed by Prosky's method(AOAC method) after boiling, microwave heating and autoclaving of the sample. It was showed that the different cooking methods resulted in different effects on the insoluble dietary fiber contents. Except yellow soybean, cooking time had little effect on insoluble dietary fiber contets in the other samples. The contents of soluble dietary fiber were generally increased by cooking. Increased cooking time reduced the content of soluble dietary fiber in brown rice but increased in rice. However, no significant differences caused by cooking time were observed for soluble dietary fiber in black soybean. The effects of cooking method on the total dietary fiber contents were similar to those of insoluble dietary fiber. The reasons for this might be that the main fraction of total dietary fiber was insoluble forms and the content of total dietary fiber was calculated as the sum of insoluble and soluble dietary fiber content.

The improvement of superior black soybeans could be accomplished through maintenance and use of black soybean collections. A collection of 965 black soybean lines currently grown by farmers was made in 197 locations on spring, 1991. This study was carried out to evaluate them as useful germplasm. Seed characteristics of black soybean collections were evaluated for black soybean breeding for seed quality. The 100-seeds weight of 929 black soybean genotypes were distributed from 6.9 to 48.1 gram. The maximum and minimum 100-seeds weight were derived from Chungnam and Jeonbuk province, respectively. l00-seeds weight was heavier in middle and late maturity group, as maturity was later, whereas no tendency was shown in early maturity group. Length, width and thickness of the collected black soybean seeds were 9.5$\pm$1.5mm, 7.3$\pm1.3mm and 7.3$\pm1.2mm, respectively. Black soybean collections with yellow seed embryo was 72% and the others were green seed embryo.
Isoflavone contents in soybean seeds are affected by both genetic and environmental factors. Correlation analysis between these factors and isoflavone contents are considered as valuable inputs when breeding improved soybean cultivars. In this study, the seeds of 49 soybean accessions grown in Korea were grouped as black, yellow, green, yellowish-green, pale yellow, and green with a black spot based on their seed coat colors. The contents of 12 isoflavones were analyzed and the association between isoflavone content and seed coat color was determined. The accessions were also grouped as early, intermediate, and late-maturing based on their days to maturity. Out of the 12 isoflavones, 11 were found in 2 accessions, 9 in 18 accessions, 8 in 11 accessions, 7 in another 11 accessions, and 6 in 7 accessions. The total isoflavone content (TIC) in black, yellow, green, yellowish-green, pale yellow, and green with black spot soybeans was in the ranges 2.110 ~ 5.777, 2.487 ~ 4.733, 2.185 ~ 4.413, 2.681 ~ 4.065, 1.827 ~ 4.085, and 3.376 ~ 4.133 mg/g, respectively. The average TIC was highest in green with black spot soybeans (3.616 mg/g), and lowest in pale yellow soybeans (2.875 mg/g). Besides, the average TIC was lowest in early maturing accessions compared to late- and intermediate-maturing accessions. TIC was strongly correlated to malonylgenistin (r = 0.91) and malonyldaidzin (r = 0.78) contents, and poorly correlated to glycitein (r = 0.04) and malonylglycitin (r = 0.18) contents. Also, days to maturity showed strong correlation with malonylgenistin (r = 0.47) content and TIC (r = 0.38). The principal component analysis outlined accessions with high TIC and diverse isoflavones along the first and second components, respectively. The results of the present study depicted that green soybeans with a black spot could be sources of high TIC. Furthermore, late-maturing accessions with diverse isoflavones in their seeds could be useful in future agricultural systems in Korea.
