오미자(Schizandra chjinensis) 추출물의 항균활성 (Antimicrobial Activity of Omija (Schizandra cheinensis) Extracts)

  • 오미자 에탄올 추출물의 항미생물 활성을 측정하기 위하여 세균, 효모, 곰팡이가 포함되는 12종류의 미생물에 대한 항미생물 활성을 측정하였다. 추출물은 세균의 생육을 억제하지만 효모와 곰팡이의 생육은 억제하지 못하였다. B. subtilis와 S. aureus에 대한 최소치사농도는 1.6~3.2 mg/mL이었으며, E. coli를 비롯한 gram음성 세균의 최소치사농도는 6.3~12.5 mg/mL이였다. B. subtilis와 S. aureus의 생육은 각 각 900 ppm과 300 ppm에서 억제되었으며 gram음성 세균의 생육은 2,000 ppm 첨가시 4시간 정도 생육이 억제되었다. 오미자 추출물의 항균활성은 열처리에 안정하였으며 부위에 따른 추출물의 항균활성은 오미자 과육의 추출물이 주로 항균력을 나타내는 것으로 나타났다.

감귤 진피 추출물을 이용한 발효액 제조 및 Campylobacter jejuni 에 대한 항균 활성 (Preparation of Fermented Citrus Peels Extracts for Their Antimicrobial Activity against Campylobacter jejuni)

  • Jeju citrus, which contains an abundance of calcium and vitamin, was used to develop fermented citrus peel extract. A total of seven probiotic strains were applied to tangerine dermis to select the best growing bacteria in citrus peel extracts. B. longum, B. bifidum, and L. mesenteroides were found to grow best in citrus peel extract culture containing glucose, yeast extracts, peptone, and potassium phosphate. Citrus peel extract culture consisting of 1% yeast extract, 5% peptone, and 0.1% phosphate was the best environment for growth of probiotics. The pH, acidity, and viable cell numbers of these fermented extracts were measured. The initial pH level of fermented extracts with nutrients was 5.25 and dropped rapidly to 3.39 after 72 hours of fermentation. The acidity of fermented extracts increased to 4.08 % after 72 hours of fermentation, and the viable cell number in fermented extracts after refrigeration for 2 weeks was $1.3{\times}10^{10}CFU/mL$. The antimicrobial activity of citrus peel fermented extracts against Campylobacter jejuni was determined, and concentrations more than 25,000 ppm showed antimicrobial activity.

쌀단백질 잔사발효물이 효모추출물의 맛특성에 미치는 영향 (Effect of a Fermented Rice Protein Residue on the Taste Property of Yeast Extract)

  • 쌀단백질을 이용한 고부가가치 천연 조미소재를 개발하기 위하여 쌀단백질을 프로테아제로 효소분해한 배지에서 Saccharomyces cerevisiae를 배양하여 제조한 효모 추출물(Yx)에, 효소분해 후 남은 쌀단백질 잔사를 Bacillus licheniformis 혹은 Bacillus subtilis로 발효하여 얻은 발효물(Rfl, Rfs)을 각각 첨가하였다. 쌀단백질 잔사의 발효물이 첨가된 효모추출물(YxRfl, YxRfs)의 전체적 선호도는 첨가전의 효모추출물(Yx)에 비하여 높았으며, 특히 쌀단백질 발효물의 보충에 의해서 감칠맛과 같은 풍미가 증가함을 미각센서 분석 및 관능검사에 의해서 확인할 수 있었다. 쌀단백질 잔사발효물에 의한 감칠맛의 상승은 감칠맛을 내는 아미노산 이 외에도 쌀단백질의 발효에 따라 유리된 다양한 펩타이드 분획의 영향이 있었을 것으로 예상된다. 이와 같이 감칠맛 아미노산 및 펩타이드가 함유된 쌀단백질 발효물이 보충된 효모추출물은 감칠맛과 풍미의 상승작용으로 전체적인 기호도가 높아짐에 따라 고부가가치 천연조미소재의 제조에 응용될 수 있을 것으로 기대되었다.

  • Yeast extract is a product prepared mainly from waste brewer's yeast, which is rich in nucleotides, proteins, amino acids, sugars and a variety of trace elements, and has the advantages of low production cost and abundant supply of raw material. Consequently, yeast extracts are widely used in various fields as animal feed additives, food flavoring agents and additives, cosmetic supplements, and microbial fermentation media; however, their full potential has not yet been realized. To improve understanding of current research knowledge, this review summarizes the ingredients, production technology, and applications of yeast extracts, and discusses the relationship between their properties and applications. Developmental trends and future prospects of yeast extract are also previewed, with the aim of providing a theoretical basis for the development and expansion of future applications.

Detection of estrogenic hormone 17β-estradiol in soil samples by a recombinant yeast bioassay and supercritical fluid extraction

  • Recombinant yeast estrogenicity (YES) assay was used as a bioanalytical tool in order to screen $17{\beta}$-estradiol in the soil samples collected from different sites of South Korea. Solvent extraction and supercritical fluid extraction (SFE) methods were compared for the extraction of the estradiol from the soils. Most high detection of the estradiol based on YES assay was observed in the soils extracted with methanol. Different types of estrogenic hormones including $17{\beta}$-estradiol were suggested to be possibly exiting in the soils, since the methanol extracts of the soils showed an estrogenic activity that was not observed in the hexane extracts of the soil. SFE extracts showed estrogenic activity in some of the samples but methanol extract showed best activity.

Rhodosporidium toruloides를 활용한 비트 열수추출물의 중성지방 억제효과 (Inhibitory Effects of Hot Water Extract of Beta vulgaris L. on Triglyceride Biosynthesis Using Rhodosporidium toruloides)

  • 본 연구에서는 Rhodosporidium toruloides를 활용하여 비트 열수추출물의 중성지방 억제효과를 확인하고자 하였다. 대조구, 실험대조구, 비트 열수추출물, 30% 및 60% 에탄올 추출물을 첨가한 YPD 배지에서 배양한 유지효모의 중성지방의 함량은 각각 8.76, 3.43, 2.87, 3.16 및 3.64 mg/dL로 나타났으며, 유지 효모의 세포수는 각각 10.48, 8.46, 12.40, 12.80 및 $8.24{\times}10^3cell/mL$로 나타났다. 대조구, 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물을 첨가한 유지효모의 총 지질 함량은 각각 112.29, 38.90, 147.87, 211.36 및 291.89 mg/g로 나타났으며, 유지효모의 중성지방의 함량은 각각 6.02, 2.83, 2.38, 1.37 및 0.73 mg/dL로 나타났다. 대조구, 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물의 첨가 농도가 높아짐에 따라서 유지효모내 소량의 형광이 발현됨을 확인하였다. 실험대조구, 0.02, 0.1 및 0.5% 비트 열수추출물을 첨가한 유지효모의 세포수 증가율은 대조구에 비해 각각 -4, 21, 82 및 103%로 증가하였다. 유리지방산과 총 카로티노이드 함량은 비트 열수추출물의 첨가 농도에 따라 농도의존적으로 증가하였다.

블루베리 천연 발효액종을 첨가한 우리밀 식빵의 품질 특성 (Quality Characteristics of Korean Wheat Bread prepared with Substitutions of Naturally Fermented Blueberry Starters)

  • In this study, a natural fermentation starter formulation was developed for manufacturing Korean bread products by substituting baker's yeast with naturally fermented blueberry starters. As the incubation time of the blueberry extracts increased, the pH and total titratable acidity increased. The sweetness (brix%) of blueberry extracts containing various amounts of sugar were higher than the other sample. The result of alcoholicity for naturally fermented blueberry extracts, the fermented blueberry extract containing 20% sugar was highest. Lactic acid bacteria counts increased until the 4th day; however, it decreased from the 5th day, and viable yeast counts increased consistently until the 5th day. The volume for naturally fermented blueberry extracts increased as the incubation time increased. As the fermentation time of blueberry starters increased, the pH of bread dough decreased. The RVA analysis conveyed that wheat flour retrogradation was retarded by increasing the blueberry starter content. The weight of pan breads containing blueberry starters were higher than that of the control, while the volume, specific volume and baking loss rate were lower than those of the control. The moisture content of pan breads containing blueberry starter decreased as storage time increased. In analyzing the visible mold colony during 7 days of storage at $28^{\circ}C$, mold growth in pan breads containing the blueberry starter was retarded. The hardness of breads containing blueberry starters were significantly increased as storage time increased. The breads containing 50% naturally fermented blueberry starter have acceptable sensory properties. In conclusion, these results indicated that 50% of natural fermentation blueberry starter could be very useful as a substitute for yeast when making naturally fermented bread.

Antimicrobial Activity and Components of Extracts from Agastache rugosa during Growth Period

  • Antimicrobial activities of volatile flavor, water and methanol extracts from Agastache rugosa were investigated. The volatile flavor extract was obtained from A. rugosa by simulataneous distillation-extraction (SDE) method. Antimicrobial activity was investigated by disc diffusion method against several microorganisms, four species of Gram positive, three species of Gram negative and tow species of yeast. The volatile flavor extracts had strong antimicrobial activity againstc. utilisand S. cerevisiae. During the growth period, a difference in antimicrobial activity among volatile flavor extracts from A. rugosa was not shown. The water extract of above 10 mg/disc showed antimicrobial activity. Methanol extracts from A. rugosa harvested in June showed antimicrobial activity against tested Gram positive and Gram negative bacteria, showed weak antimicrobial activity against the bacteria from those harvested in July and August. In particular, antimicrobial activity against V. parahaemolyticus was stronger than that against other bacteria. Water and methanol extracts did not inhibit yeast. C. utilis and S. cerevisiae. To further elucidate the effective components, volatile flavor extracts was analyzed by GC/MS. harvested in June, the components included 8 phenols (93.031%), 18 terpenes (5.230%), 12 alcohols (1.300%) 8 alkanes (0.181%), 1 ester (0.056%), 2 ketones (0.033%), 2 aldehydes (0.011%) and 1 pyrrole (0.007%). In July, the components included 6 phenols (94.366%), 19 terpenes (3.394%), 11 alcohols (2.045%), 1 ester (0.039%), 2 ketones (0.028%), 1 furan (0.005%) and 1 aldehyde (0.005%). And in August, the components included 7 phenols (95.270%), 19 terpenes (2.951%), 13 alcohols (1.399%), 1 ester (0.063%), 2 aldehydes (0.016%), 2 ketones (0.011%), 1 alkane (0.006%), 1 acid (0.005%) and 1 pyrrole (0.005%). The major component of volatile flavors was estragole, a phenolic compound.

Production of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor-Enriched Edible Yeast Using Gugija (Lycium chinesis Mill)

  • To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

인삼성분이 효모의 생육 및 대사에 미치는 영향 (The Effects of Korean Ginseng (Panax ginseng C.A. Meyer) Extracts and Their Fractions on the Growth and Metabolism of Saccharomyces cerevisiae and Saccharomyces uvarum)

  • This study was conducted to investigate the effects of Korean ginseng extracts and their fractions on the growth of Saccharomyces cerevsiae and Saccharomyces uvamm, their glucose consumption and alcohol production. The growth of both yeasts were stimulated by ginseng extracts and their water soluble fractions, but were supressed by ether extracts and an n-butanol extracts. Their growth were enhanced considerably by low molecular weight fractions (< 1,000) in water solubles. Similar results were also obtained with glucose consumption by yeasts. Substances increasing the growth and glucose consumption by yeasts proved to be a low molecular weight fractions (<1,000) in water solubles not saponins. The production of n-propyl alcohol by yeast was enhanced by adding ginseng extracts into the media, but that of ism-butyl alcohol was suppressed at same condition.
