Impact of Sulphur and Nitrogen Application on Seed and Xanthotoxin Yield in Ammi majus L.

  • Field experiments were conducted to determine the physiological and biochemical basis of the interactive effect of sulphur (S) and nitrogen (N) application on seed and xanthotoxin yield of Ammi majus L. Six treatments were tested ($T_1$ = control-without manure and fertilizers, $T_2$ = manure @ 9 kg $plot^{-1}-10\;t\;ha^{-1},\;T_3=A_0N_{50}K_{25}P_{25},\;T_4=S_{40}N_{50}K_{25}P_{25},\;T_5=S_{40}N_{100}K_{25}P_{25}\;T_6=S_{20+20}N_{50+50}K_{25}P_{25})$). Nitrate reductase (NR) activity and ATP-sulphurylase activity in the leaves were measured at various phonological stages, as the two enzymes catalyze rate-limiting steps of the assimilatory pathways of nitrate and sulphate, respectively. The activities of these two enzymes were strongly correlated with seed and xanthotoxin yield. The highest nitrate reductase activity, ATP-sulphurylase activity and xanthotoxin yield were achieved with the treatment $T_4$. Any variation from this treatment decreased the activity of these enzymes, resulting in a reduction of the seed and xanthotoxin yield in Ammi majus L. The higher seed and xanthotoxin yield achieved in Ammi majus L. at treatment $T_4$ could be due to optimization of leaf soluble protein and photosynthetic rate, as these parameters are Influenced by S and N assimilation.

The Comparison of Constituents Distributed in the Root of Angelica dahurica (구릿대 뿌리의 부위별 성분 비교)

  • In this study, we analyzed and quantified the amounts of bioactive phenolic constituents, xanthotoxin (1), oxypeucedanin (2), and imperatorin (3) in each part of the root of Angelica dahurica by HPLC, which validated by ICH guide lines comparing the linearity, intra day precision, inter-day precision. As a result, the amount of imperatorin 2.96% in the rootlet was two fold higher than that of the main root 1.32%. On the other hand, the amounts of xanthotoxin and oxypeucedanin in the rootlet showed similar to those of main root. In addition, the cortex of root is more plentiful of three consitituents (0.66%, 0.53%, and 1.85%) than those of xylem (0.29%, 0.05%, and 0.07%). These results show that the rootlet and cortex contain a large amount of bioactive phenolic constituent including xanthotoxin, oxypeucedanin, and imperatorin than other parts of the root.

  • The root of Heracleum moellendorffii was extracted with methanol and extract was fractionated with n-hexane, $CHCI_3$ and n-BuOH. Repaeated column chromatography of silica gel, sephadex LH 20 and ODS led to the isolation of ten compounds from n-hexane fraction and n-BuOH fraction. On the basis of spectroscopic evidences, the structures of isolated compounds were identified as isobergapten, psoralen, angelicin, sphondin, xanthotoxin, skimmin, cichoriin, $heratomol-6-O-{\beta}-D-glucopyranoside$, scopolin and apterin.

  • Hot water extracts of Angelica keiskei aerial parts were solvent-fractionated with ethyl acetate (EtOAc) and buffers (5% $NaHCO_3$, pH 8.0; 1.0 N HCl, pH 3.0) to obtain EtOAc-soluble acidic and neutral fractions. EtOAc-soluble neutral fraction was purified by Sephadex LH-20 column chromatography and reverse phase HPLC. Assay for purification of antioxidative compounds was performed by spraying DPPH solution on thin layer chromatography. Two isolated substances were identified as pseudoisopsoralen and 8-methoxypsoralen(xanthotoxin) by FAB-MS and NMR analyses.

  • The mechanism of skin-sensitizing photoreactions of coumarins and furocoumarins are studied by spectroscopic, triplet quenching, and fluorescence techniques. The excited singlet mechanism is suggested for xanthotoxin-thymine/or DNA photoreactions from the results of triplet quenching studies utilizing ${\beta}$-carotene as a quencher.

  • Three coumarins, one chromone, and a phytosterol were isolated from the stem of Ostericum koreanum. Basis on the spectral data, we determined to be the structures of isolated compounds as bergapten, xanthotoxin, auraptenol, hamaudol, and mixture of phytosterols. Auraptenol was isolated for the first time from this plant.

  • Four coumarins were isolated from chloroform extract of the root of Peucedanum japonicum and identified as praeruptorin A (1), xanthotoxin (2), psoralen (3) and bergapten (4) on the basis of spectroscopic methods. The inhibitory activities of these coumarins on monoamine oxidase prepared by mouse brian were tested. The IC50 values of them were shown to be $27.4{\mu}M$ (1), $40.7{\mu}M$ (2), $35.8{\mu}M$ (3), and $13.8{\mu}M$ (4), in vitro.

Constituents of the Fruits and Leaves of Euodia daniellii

  • Four flavonoid glycosides, flavaprin (7), evodioside B (8), vitexin (11), and hesperidin (12), as well as the coumarins bergapten (1), xanthotoxin (2), and isopimpinellin (3), the lignan simplexoside (10), the steroids ${\beta}-sitosterol$ (4) and daucosterol (5), the limonoids isolimonexic acid (6) and limonin (9), and uracil (13) and myo-inositol (14) have been isolated from Euodia daniellii. The structures of these compounds were established from spectral data. Among the isolates, bergapten showed cyclooxygenase-2 inhibitory activity with an $IC_{50}$ value of $6.2{\;}{\mu\textrm{g}}/ml. Flavonoids isolated from this plant exhibited no cytotoxic activity against the human tumor cell lines, A549, SKOV-3, SKMEL-2, XF498, and HCT15.

  • Two polyacetylenes (1 and 2), four coumarins (3-6), and one sesquiterpene (7) were isolated from the halophyte Glehnia littoralis. Particularly, compound 6 and 7 were isolated for the first time from Glehnia littoralis. Their chemical structures have been determined by extensive 2-D NMR experiments such as $^1H$, COSY, HMQC and HMBC and by comparison with the reported data in the literature.
