Objectives : To compare the anti-oxidant effects of Ligusticum chuanxiang and Cnidium officinale extracts and their mixture with Angelica gigas, we investigated the anti-oxidative activities using rat liver tissues. Methods : We investigated the anti-oxidative activities by analysis of lipid peroxidation, xanthine oxidase activity, aldehyde oxidase activity, hydroxyl radical scavenging activity, superoxide dismutase activity and catalase activity in rat liver tissues. Results : Both Ligusticum chuanxing and Cnidium offieinale inhibited the lipid peroxidation compared to the control, there is no significant differences between two groups. Cnidium officinale showed strong inhibitory effects on xanthine oxidase and aldehyde oxidase activities compared with that of Ligusticum chuanxing. Ligusticum chuanxing and Cnidium officinale were scavenged the hydroxy radicals, and increased SOD and catalase activities. These results suggested that Ligusticum chuanxing and Cnidium officinale were showed antioxidant activity, especially Cnidium officinale showed higher activity than Ligusticum chuanxing. Conclusions: These results suggest that Ligusticum chuanxing could be replaced by Cmdium afficinale in clinical trial.
The essential oil of Acorus gramineus(Aracease), which has been used as an antioconvulsant in Korean folk medicine, was evaluated for its effects on antioxidative system in vitro and in vivo. This mixture of terpenes showed inhibitory effects on xanthine oxidase activity with 13.3% at 10 $\mu\textrm{g}$/ml and on aldehyde oxidase activity with 5.0% at 1$\mu\textrm{g}$/ml. Lipid perosidation was inhibited by 49.4% at 1.0 mg/ml of the essential oil in vitro and by 16.7% after 7 days inhablation of an oil as compared to PTZ-treated control group. DPPH radical scavenging activity of this essential oil was ralatively werak.
Steaming is a method that has traditionally been used for medicinal plant extraction. This study investigated nitrite oxide production, ferrous ion chelating activity, α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities of ethanol, acetone and hot-water extracts of Codonopsis lanceolata prepared by steaming seven times. MTT assay showed that each extract was non-toxic up to a concentration of 700 ㎍/mL confirming that there was no cytotoxicity in all extracts. The α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities exhibited by the hot-water extract obtained from steaming seven times were higher (83.1%) than the other extracts. Higher production of nitrite oxide and better ferrous chelating activity was recorded with hot-water extract compared to ethanol and acetone extracts. These results indicated that more steaming of Codonopsis lanceolata extracts would be required to validate the possibility of developing antioxidants. Also, further study is needed to determine if the components present in the tested extracts might be useful in the prevention of Alzheimer's disease. These results showed that hot-water extracts may be useful for their antioxidant and the production inhibitory activity of nitrite oxide. It will be helpful in the investigation of the constituent analysis of the steam-processed product of Codonopsis lanceolata.
Opuntia humifusa is a member of the Cactaceae family. In the present study, the antioxidant, nitric oxide(NO) inhibitory activities and potential irritation response of the fermented Opuntia humifusa cladodes(FOH) were investigated for cosmetic use. Antioxidant activities were tested using 1,1-diphenyl-2-picrylhydrazyl(DPPH) and xanthine oxidase assay, we found that FOH could scavenge DPPH free radicals and inhibit xanthine oxidase activity in a dose dependent manner, with $IC_{50}$ of 2599.46${\mu}g/ml$ and 721.38${\mu}g/ml$, respectively. To investigate the possible anti-inflammatory effects of FOH, RAW 264.7 macrophages were pretreated with FOH($0{\sim}400{\mu}g/ml$) for 30 min and then treated with LPS for 24 h. We found that cell number did not vary significantly with the treatment of FOH, and FOH did not show any inhibitory effects on LPS-induced NO production. After application of FOH to rabbits for skin and eye irritation test, the experimental sites did not show any response compared to the control. FOH were considered to be a non-irritant to the skin and eye. Based on the above information, we suggest that FOH can be considered to be a non-irritant base cosmetic material for safely use.

Uric acid is the end product of purine metabolism in human body, originating from hypoxanthine after enzyme catalysis by Xanthine oxidase (XOD). Hyperuricemia results as a result of either over-generation of uric acid or a reduction in its excretion. In silico modelling methods such as Absorption, Distribution, Metabolism, Excretion and Toxicity (ADMET) prediction, Autodock 4.2.6 program were used to study the potential inhibitory compounds of XOD. Also we investigated the inhibition of XOD activity by using the extracts of Dendropanax morbifera and Rubus coreanus Miq spectrophotometrically. According to ADMET data, several compounds from D. morbifera and R. coreanus plants, were found to be more potent in inhibiting the XOD activity than allopurinol. XOD inhibitory activity is evaluated by quantifying the formation of uric acid by measuring the absorbance at 290 m ($A_{290}$).D. morbifera extract inhibited XOD activity at $250{\mu}g/ml$, however the extracts from R. coreanus has inhibited XOD activity at $25{\mu}g/ml$. The major compound of R. coreanus, ellagic acid significantly increased the inhibition rate from $9{\mu}g/ml$ and showed a 71% suppression rate at $15{\mu}g/ml$. Finally, these results suggested a potential inhibitory activities of the extracts from D. morbifera and R. coreanus Miq, but further research is needed to validate to ensure their safe usage as drug.
Pine trees (Pinus densiflora Sieb. et Zacc.) have been used as a traditional health-promoting medicinal food in Korea. This research was performed to determine the antioxidative and antibacterial activities, tyrosinase, nitric oxide synthesis, angiotensin converting enzyme (ACE), and xanthine oxidase inhibition effects of the pine bud ethanol extract (PBE). Antioxidative activities of PBE were measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) free radical scavenging activity and superoxide dismutase-like activity (SODA). DPPH radical scavenging and SOD-like activities of PBE were remarkably increased in a dose-dependent manner, and were about 88.9% and 47.9% at 1 mg/ml and 10 mg/ml, respectively. The xanthine oxidase and angiotensin converting enzyme activities were inhibited about 71.9% and 60.8% at 1 mg/ml and $100{\mu}g/ml$ of PBE, respectively. The tyrosinase inhibitory activities of PBE were slightly increased in a dose-dependent manner. The PBE showed strong antimicrobial activities on Escherichia coli (E. coli) and Vibrio paraheamolyticus. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, NO synthesis was reduced up to 54% by addition of PBE at $200{\mu}g/ml$. These results revealed that pine buds have a strong antioxidative and anti-inflammatory activity, and exhibit angiotensin converting enzyme and xanthine oxidase inhibitory activities. This suggests that pine buds have the greatest property as a source for natural health products.
The methanolic extracts of some marine natural products were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzymes. The methanolic extracts of Styela clava, Ecklonia stolonifera, Pachymeniopsis elliptica and Hypnea charoides which decreased the formation of lipid peroxide inhibited the activity of xanthine oxidase about 41, 20, 20 and 21% by adding $100\;{\mu}g/ml$ of each methanolic extracts, respectively. However, the four extracts didn't inhibit the activity of aldehyde oxidase.

This research was performed to determine the antioxidant activity, nitrite scavenging activity, and its inhibitory activity on angiotensin converting enzyme (ACE), xanthine oxidase, $\alpha$-glucosidase, and elastase of hot water extract from pine bud (WPB). Antioxidant activity of WPB was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity. DPPH radical scavenging activity and SOD-like activity of WPB were remarkably increased in a dose-dependent manner, and were about 71.4 and 85.4% at 2 mg/mL, respectively. The xanthine oxidase and ACE inhibitory activities were about 70.9 and 51.9% at 2 mg/mL of WPB, respectively. Nitrite scavenging activity of WPB was about 59.1, 53.8, and 39.5% on pH 1.2, 3.0, and 6.0 at 2 mg/mL, respectively. The WPB also showed elastase and $\alpha$-glucosidase inhibitory effects. These results revealed that pine bud have strong antioxidant activity and positive effects on the inhibition of xanthine oxidase, ACE, and elastase.
This study prepared extracts of Angelica dahurica leaves using reflux water extraction (RW), reflux ethanol extraction (RE) and pressure heating water extraction (PW). The extracts were extraction for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for xanthine oxidase and tyrosinase. The PW extraction method yielded the highest content of polyphenol compounds (95.23 mg/g). The electron donating abilities (EDAs) of RE and PW extracts were 76.02% and 70.08% respectively. The superoxide dismutase (SOD)-like activities were 13.45 19.00%, when extracts were assayed at 1 mg/mL. The nitrite scavenging ability (pH 1.2) of the PW extract was 54.33% higher than levels shown (44.24%) by the RE and RW extracts. The inhibition of xanthine oxidase by the RW extract was highest (99.71% at 5 mg/mL) while that of the PW extract was over 97% at 500 g/mL. Tyrosinase inhibition was highest in the RE extract (46.25% at 5 mg/mL). All extracts showed dose-dependent inhibitory activities. The results indicated that the PW extract had the highest polyphenol content, the RW and RE extracts had the best nitrite scavenging ability, and the RE extract showed the most pronounced effect on EDA, SOD-like activity and tyrosinase inhibition.
An in vitro bioassay-guide revealed that the methanol (MeOH) extract of the stem bark of Populus davidiana showed considerable inhibitory activity against cyclooxygenase (COX-1, COX-2). Continuous phytochemical study of the MeOH extract of this plant led to the isolation of ten flavonoids; sakuranetin (1), rhamnocitrin (2), 7-O-methylaromadendrin (3), naringenin (4), eriodictyol (5), aromadendrin (6), kaempferol (7), neosakuranin (8), sakuranin (9) and sakurenetin-5,4'-di-${\beta}$-D-glucopyranoside (10). Their structures were identified on the basis of their physicochemical and spectroscopic analyses. The isolated compounds, 1-10, were tested for their inhibitory activities against COX-1 and COX-2. Compound 7 was found to have potent inhibitory effect on COX-1 and a moderate effect on COX-2, meanwhile, compounds 1-6 showed moderate inhibition against COX-1 only. Moreover, compounds 5-8 exhibited suppressive effects on xanthine oxidase (XO). These results may explain, in part, the traditional uses of P. davidiana in ethnomedicine.
