Rheumatoid arthritis (RA) is a representative autoimmune disease that is primarily characterized by persistent inflammation and progressive destruction of synovial joints. RA has a complex and heterogeneous pathophysiology, involving interactions among various immune and joint stromal cells and a diverse network of cytokines and intracellular signaling pathways. With improved understanding of RA, over the past decades, therapeutic strategies have become considerably advanced and now included targeted molecular therapies, such as tumor necrosis factor inhibitors, IL-6 blockers, B-cell depletion agents, as well as inhibitors of T-cell co-stimulation and Janus kinases. However, a considerable proportion of RA patients experience refractory disease and interrupted treatment owing to the associated risk of developing serious infections and cancers. In contrast, although IL-1β, IL-17A, and p38α play significant roles in RA pathogenesis, several drugs targeting these factors have not been approved because of their low efficacy and severe adverse effects. In this review, we provide an overview of the working mechanism, advantages, and limitations of the currently available targeted drugs for RA. Additionally, we suggest potential mechanistic causes for clinically approved and failed drugs. Thus, this review provides perspectives on approaches for basic and translational studies that hold promise for identifying future next-generation therapeutics for RA.
Purpose: This paper investigated the effects of issue leadership on employees' work performance. It also explored the mediating role of job challenge in the above relationship. We also tested the moderating effect of psychological ownership in the relationship between issue leadership and job challenge. Finally, this paper examined the moderated mediation effect of psychological ownership in the course of affecting issue leadership on work performance via job challenge. Methods: This paper used a cross-sectional design with questionnaires administered to 390 employees working in Korean firms. It applied an hierarchical and multi-regression analysis by using SPSS 22.0 and Hayes' Macro Process to test the proposed hypotheses including a moderated mediation model. Results: As a result of the analysis, first, it was found that the issue leadership had a positive effect on the work performance. Second, it was also found that issue leadership had a positive effect on the job challenge. In addition, job challenge was found to positive mediating effect in the relationship between issue leadership and work performance. Third, we found that the psychological ownership positively moderated the relationship between issue leadership and job challenge. Finally, the moderated mediating effect was found by confirming that the higher the degree of psychological ownership, the stronger the effect of issue leadership on the work performance via job challenge. Conclusion: This paper contributes to the leadership and work performance studies by identifying the role of issue leadership enhancing employee work performance while confirming connection mechanism and boundary conditions strengthening the positive effect of issue leadership on employee work performance.
Background : Since tumor necrosis factor was discovered in 1975, TNF has been well known about its cytotoxic effect on tumor cells in vivo and in vitro. According to the recent improvement of molecular biological techinques, it is possible that exogenous TNF gene is transferred to tumor cells and is expressed in theirs. By virtue of TNF gene transfer, we have expected that TNF expressed in TNF-gene-transferred tumor cells would kill tumor cells in vivo without systemic side effect. The expected mechanisms in which antitumor effects of TNF expressed in TNF-gene-transferred tumor cells are working would be as followings. In the first mechanism, TNF expressed in TNF-gene-transferred tumor cells would kill tumor cells around(like homicide). In the second mechanism, TNF expressed in TNF-gene-transferred tumor cells would kill themselves(like suicide). In the third mechanism, TNF expressed in TNF-gene-transferred tumor cells would recruit immune effector cells and kill tumor cells indirectly. In the last mechanism, TNF expressed in TNF-gene-transferred tumor cells would augment cytokine such as interferon-$\gamma$ to kill tumor cells. Among these four mechanisms of antitumor effect, only the second mechanism has not been established yet. Therefore, to elucidate the second mechanism, We performed this study. Method : We transferred TNF-$\alpha$ gene to NCI-H2058, a human mesothelioma cell line and WEHI164, a murine fibrosarcoma cell line by using retroviral vector(pLT12SNTNF). And, We determined by using MTT assay whether TNF expressed in TNF-gene-transferred tumor cell lines would kill themselves like suicide or not. Then, if TNF-gene-transferred tumor cell lines would not suicide themselves, I would know more about the TNF sensitivity of TNF-gene-transferred tumor cell lines to exogenous TNF also by MTT assay. Result : NCI-H2058 and WEHI164 which were sensitive to TNF, became far less sensitive to endogenous and exogenous TNF after being transferred TNF-$\alpha$ gene to. Conclusion : TNF-gene-transfer to NCI-H2058 and WEHI164 gave them resistance to TNF.

The use of cable tunnels for electric power transmission as well as their construction in difficult conditions such as in subsea terrains and large overburden areas has increased. So, in order to efficiently operate the small diameter shield TBM (Tunnel Boring Machine), the estimation of advance rate and development of a design model is necessary. However, due to limited scope of survey and face mapping, it is very difficult to match the rock mass characteristics and TBM operational data in order to achieve their mutual relationships and to develop an advance rate model. Also, the working mechanism of previously utilized linear cutting machine is slightly different than the real excavation mechanism owing to the penetration of a number of disc cutters taking place at the same time in the rock mass in conjunction with rotation of the cutterhead. So, in order to suggest the advance rate and machine design models for small diameter TBMs, an EPB (Earth Pressure Balance) shield TBM having 3.54 m diameter cutterhead was manufactured and 19 cases of full-scale tunneling tests were performed each in 87.5 ㎥ volume of artificial rock mass. The relationships between advance rate and machine data were effectively analyzed by performing the tests in homogeneous rock mass with 70 MPa uniaxial compressive strength according to the TBM operational parameters such as thrust force and RPM of cutterhead. The utilization of the recorded penetration depth and torque values in the development of models is more accurate and realistic since they were derived through real excavation mechanism. The relationships between normal force on single disc cutter and penetration depth as well as between normal force and rolling force were suggested in this study. The prediction of advance rate and design of TBM can be performed in rock mass having 70 MPa strength using these relationships. An effort was made to improve the application of the developed model by applying the FPI (Field Penetration Index) concept which can overcome the limitation of 100% RQD (Rock Quality Designation) in artificial rock mass.

Today, a shortage of the skilled operator has been intensified gradually and the necessity of an earthwork in extreme environment operators are difficult to access is increasing for the purpose of resource development and new living space creation. For this reason, an effort to develop an unmanned excavation robot for fully automated earthwork system is continuing globally. In Korea, a research consortium called 'Intelligent Excavation System' has been formed since 2006 as a part of Construction Technology Innovation Program of Ministry of Land, Transport and Maritime Affairs of Korea. Among detailed technologies of the Task Planning System is one of the core technologies of IES, this paper explains research and development process of the Local Area Design Module, which provides informatization unit to create automated excavators' work command information at operation level such as location, range, target, and sequence for excavation work. Designing of Local Area should be considered various influential factors such as excavator's specification, working mechanism, heuristics, and structural stability to create work plan guaranteed safety and effectiveness. For this research, conceptual and detail design of the Local Area is performed for analyzing design element and variable, and quantization method of design specification corresponding with heuristics and structural safety is generated. Finally, module is developed through constructed algorithm and developed module is verified.
Purpose - The purpose of this study is to identify team commitment affecting employees' innovative activities and factors affecting team commitment including empowerment by leaders and job enrichment factors. In other words, so as to explain outcome variables of innovative activities, this study aims to emphasize employees' attachment roles towards their groups within nomological network, and identify the motives encouraging employees' innovative activities. The research purpose is significant due to the realistic situation of hotel industry. The reason why innovative activities are important can be found in recent changes of business environment. Also, unlike other various studies on precedence factors encouraging employees' innovative activities, this study classified those precedence factors into job and leader characteristics, and it emphasized the importance of team commitment as the process that job and leader characteristics are connected to innovative activities. Research design, data and methodology - The survey for this study was conducted during October 6th ~ November 10th in 2014 to the employees who are working in 5-star hotels in Korea. As for the selection of hotels and sampling method, convenience sampling method was used to the employees in 5-star hotels. Self-report method was used in the survey, judging that the employees' characteristics would be relatively homogeneous. 311 questionnaires were distributed in total, and 275 reponses were collected. After excluding the missing and unreliable responses, 245 questionnaires were used in the research. SPSS and AMOS programs were used for the analysis. Results - First, empowering leadership had positive effects on hotel employees' team commitment. It indicates that hotel employees are more committed to their team when their leaders set examples, provide information, and involve employees in decision-making process. Second, as a result of the relationship analysis in task diversity, task significance, task identity and team commitment, task diversity and task significance had significant effect on team commitment, while task identity had no significant effect on team commitment. It indicates that team commitment is enhanced when the employees can conduct diverse types of jobs and get more opportunities to talk with the guests. Also, the repetition for the same jobs in hotel rooms and the space for preparing food and beverage do not lead to team commitment, even though the employees fulfill their duties to the end. Third, hotel employees' team commitment has positive effect on their innovative activities. It indicates that employees voluntarily conduct innovative activities when they are attached to their team and identifies themselves with the team. Conclusions - There are theoretical and practical implications in this study. First, in terms of the theoretical perspective, this study proposes structural framework in team commitment, and it identifies the psychological mechanism in team commitment from the aspect of social exchange, which resulted in identification of precedence factors related to team commitment. In addition, this study presents new possibilities for relevant studies about team commitment by examining the effect on team commitment when the importance of innovative activities is emphasized in recent business environment.

In our previous report, we indicated that not only BSA but also BGG played an important role in the allergenicity of beef. In this study, the effect of heat or high-pressure treatments to beef extract on the SDS-PAGE patterns was examined. The antigenicity of each treated samples was also investigated by Western blots assay with the sera of BGG-positive beef allergic patients. The BGG band and its antigenicity slightly disappeared but not generally in $100^{\circ}C$ group, indicating $100^{\circ}C$ treatment is not sufficient to totally eliminate the antigenicity of beef allergens. Compared with BGG band, BSA band significantly disappeared in SDS-PAGE with $100^{\circ}C$ treatment, indicating BSA is more heat- sensitive than BGG. When the beef extract was heated at $120^{\circ}C$, not only BSA but also BGG bands was largely disappeared in both SDS-PAGE and Western blots. High pressure (HP) treatment even at 600 MPa did not affect SDS-PAGE and Western blots pattern of BSA. On the contrary, BGG treated with HP showed visible changes in SDS-PAGE. 600 MPa treatment significantly reduced the antigencity. Interestingly, these behaviors of BGG were not found in the same experiments with pure BGG treated with HP. From these results, it was speculated that some kinds of proteolytic enzymes in beef extracts were involved in the BGG molecular degradation by HP treatment. The aging experiments of beef extracts treated with HP supported this hypothesis. Further studies are needed to clarify the function and working mechanism of enzymes associated with BGG degradation in beef extracts by HP treatment.

Objectives: We investigated the impacts of indoor plants on indoor air quality and occupational health, focusing on allergic rhinconjunctivitis and stress among employees in new office buildings. Methods: A total of 34 employees working at new public office buildings were enrolled as subjects (n=17, with indoor plants) and as a control (n=17) group. Before and after introducing indoor plants for three months, indoor air quality measurements including VOCs and aldehydes and questionnaires on sick building syndrome, AR symptoms (ARIA based), stress (DASS 42, KOSS, and SACL), and indoor characteristics were performed and statistically analysed. Results: Among the 34 enrolled subjects, 19 were included in the probable AR subject group (subjects with indoor plants, n=8, control n=11) and completed all questionnaires. Statistical analyses were done for total, AR subject groups, and controls. As a result, it was confirmed that major indoor air pollutants decreased after the introduction of indoor plants (p<0.5). Among major symptoms of allergic rhinoconjunctivitis, watery rhinorrhea, nasal stuffiness, and nasal itching indexes decreased (p<0.5, respectively). A decrease was noted in some areas of work-related stress indexes (mainly KOSS) among the subject group (total and AR) and a decrease of indoor environmental attractiveness among the control group (total and AR) (p<0.5, for all). Conclusions: Indoor plants may help reduce indoor air pollutants and decrease AR symptoms and work-related stress of employees in newly built office buildings. Various further follow-up studies on the mechanism of environmental, physical, and emotional influences and utilization of indoor plants in association with allergic diseases will be needed.
Many studies of heat transfer on the swirling flow or unswirled flow in a abrupt pipe expansion are widely carried out. The mechanism is not fully found evidently due to the instabilities of flow in a sudden change of the shape and appearance of turbulent shear layers in a recirculation region and secondary vortex near the corner. The purpose of this study is to obtain data through an experimental study of the swirling flow and heat transfer downstream of an abrupt expansion in a circular pipe with uniform heat flux. Experiments were carried out for the turbulent flow nd heat transfer downstream of an abrupt circular pipe expansion. The uniform heat flux condition was imposed to the downstream of the abrupt expansion by using an electrically heated pipe. Experimental data are presented for local heat transfer rates and local axial velocities in the tube downstream of an abrupt 3:1 & 2:1 expansion. Air was used as the working fluid in the upstream tube, the Reynolds number was varied from 60, 00 to 120, 000 and the swirl number range (based on the swirl chamber geometry, i.e. L/d ratio) in which the experiments were conducted were L/d=0, 8 and 16. Axial velocity increased rapidly at r/R=0.35 in the abrupt concentric expansion turbulent flow through the test tube in unswirled flow. It showed that with increasing axial distance the highest axial velocities move toward the tube wall in the case of the swirling flow abrupt expansion. A uniform wall heat flux boundary condition was employed, which resulted in wall-to-bulk temperatures ranging from 24.deg. C to 71.deg. C. In swirling flow, the wall temperature showed a greater increase at L/d=16 than any other L/d. The bulk temperature showed a minimum value at the pipe inlet, it also exhibited a linear increase with axial distance along the pipe. As swirl intensity increased, the location of peak Nu numbers was observed to shift from 4 to 1 step heights downstream of the expansion. This upstream movement of the maximum Nusselt number was accompanied by an increase in its magnitude from 2.2 to 8.8 times larger than fully developed tube flow values.
Plant construction projects usually take much higher uncertainty and risks than the projects from other domains. This implies the importance of plant construction project management should be more emphasized than the other domain. Especially, the overall successes of the projects often depend on the performance of process planning and scheduling performed at the initial stage of the project. However, most plant construction projects suffer great difficulties in establishing proper process planning and scheduling timely because of unstructureness and dynamicity of environment of the project itself In this paper, we propose a knowledge-based process planning and scheduling approach in a plant construction domain to cope this problem. First, we modulize process planning knowledge and present the knowledge representation scheme. Second, we propose an inferencing mechanism to build a process planning for plant construction based on the represented process planning knowledge. Since our approach automate the initial process planning, which was usually done by manual way, it can improve the correctness and also completeness of the process plan and schedule by reducing the time to plan and allowing simulations on the various situation. We also design and implement this our approach as a real working system, and it is successfully applied to real plant construction cases from a leading construction company in Korea. Based on this success, we expect our approach can be easily applied to the projects of other areas, while contributing to enhancement in productivity and quality of project management.
