Effect of curcumin in the prevention of experimentally induced nephrolithiasis in rats by ethylene glycol and Vitamin D3

  • Curcumin (CMN) is known to have beneficial role in anorexia, coryza, cough, diabetic wounds, and hepatic disorders apart from its inherent antioxidant effects. Therefore, the present study was aimed to evaluate antioxidant effect of CMN in prevention of nephrolithiasis in rats-induced by ethylene glycol (EG) and Vitamin D3 (Vit. D3). Male Wistar rats (175 - 200 g) were randomized in groups like control, EG + Vit. D3 induced nephrolithiatiatic rats, CMN treated rats, CMN + EG + Vit. D3 treated rats, Vit. E + EG + Vit. D3 treated rats. Urine was collected weekly throughout the experimental protocol and estimated for calcium oxalate (CaO) count. After completion of experimental protocol serum was estimated for blood urea nitrogen and creatinine. Both the kidneys were excised and used to evaluate levels of biomarkers of oxidative stress and calcium oxalate crystal deposition by histopathological studies. Administration of EG and Vit. D3 to rats resulted in increased oxidative stress, hyperoxaluria and renal deposition of CaO crystals. Supplementation with CMN improves kidney function, reduces elevated oxidative stress, urinary oxalate level and renal deposition of CaO which shows its protective action in nephrolithiasis. The increased deposition of stone in the kidney and stone forming constituents of nephrolithiatic rats were effectively lowered by treatment of CMN.

Antihyperlipidemic Effects of Bupleuri Radix, padix and Uncariae Ramulus et Uncus on Experimental Hyperlipidemia in rats (고지혈증 랫트를 이용한 시호, 작약, 조구등의 항고지혈 효과)

  • Effects of Bupleuri Radix, Paeonie Radix or Uncariae Ramulus et rncus on experimental yperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high holesterol diet for one week, as previously described by the authors. Blood lipid profile was verified on these ats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low ensity lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, ethanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitored. Methanol extract of 5Bupleuri radix reduced the TC value significantly at 2 week point and Paeoneae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.

Relationship between Hypertension and Central Dopaminergic Neural Activity in Spontaneously Hypertensive Rats (선천성 고혈압 쥐의 고혈압 현상과 중추 도파민 신경계 활성의 상관성)

  • The role of central dopaminergic neural activity for the maintenance of hypertension was tested. Two groups of animals were prepared; 1) Spontaneously hypertensive rats (SHR) at 14 weeks of age which reveal hypertension and 2) age-matched normotensive Wistar Kyoto rats (WKY). Blood pressures and dopamine turnover rate were measured from animals in each group. Dopamine turnover rate was determined in four brain regions such as telence-phalon, hypothalamus/thalamus, midbrain and pons/medulla, from concentration of dopamine at time zero, 1 and 2 hours after alpha-methyl-para-tyrosine (AMPT) was injected into animals. In SHR dopamine turnover rate was greater than normotensive rats in only midbrain. The result from the present study implicates that dopaminergic neural activity in midbrain may be positively coupled to the manifestation of hypertension in SHR.

An in vivo Study of Lipid Peroxidation in Rats under Conditions of Oxidative Stress and the Antioxidant Effects of Probucol

  • The purpose of this study was to investigate in vivo lipid peroxidation in rats under conditions of streptozotocin-induced oxidative stress and the antioxidant effects of probucol. In vivo lipid peroxidation was observed by measuring low molecular weight aldehydes and related carbonyl compounds in rat urine. Three groups of male Wistar rats weighing 165-190 g were used: normal (N), streptozotocin-induced oxidative stress (OS) and oxidative stress plus probucol treatment (P). following streptozotocin treatment of the rats, a variety of secondary lipid peroxidation products were increased. The levels of butanal, hexanal, hex-2-enal, kept-2-enal, octanal, non-2-enal, deca-2,4-dienal, 4-hydroxyhex-2-enal, 4-hydroxyno n-2-enal, malondi aldehyde(MDA), and unknown carbonyl compounds were significantly increased in the oxidative stress group compared to the control group. Treatment with probucol resulted in significant decreases in buoal, hexanal, hex-2-enal, octanal, deca-2,4-dienal, 4-hydroxyhex-2-enal, MDA and unknown carbonyl compounds. Hept-2-enal, hepta-2,4-dienal and non-2-enal appeared to have a tendency to decrease due to pobucol treatment.

  • The relationship between the maintenance of hypertension and the central noradrenergic nervous system activity in spontaneously hypertensive rats (SHR) was studied. The norepinephrine turnover rates in 5 brain areas; telencephalon, hypothalamus/thalamus, midbrain, pons/medulla, cerebellum as a measure of noradrenergic neuronal activity were measured at the ages of 14 weeks in SHR and normotensive Wistar rats. In 14-week old SHR, blood pressure was significantly higher than in normotensive rat, and central norepinephrine turnover rates were significantly greater in telencephalon, hypothalamus/thalamus, midbrain. There were no differences between norepinephrine turnover rates in pons/medulla, cerebellum of SHR and those of normotensive rats.

  • Relationship between the maintenance of hypertension and central serotonergic nervous system activity in opontaneously hypertensive rats (SHR) was studied. Serotonin turnover-rates were measured in 5 brain areas as an index of serotonergic neuronal activity and compared at the ages of 14 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) (2) normotensive wistar kyoto rats (WKY). In 14-week old SHR, central serotonin turnover rate was significantly lower in telencephalon, hypothalamus/thalamus and midbrain than normotensive rat, but significantly higher in cerebellum. There were no significant differences between serotonin turnover rate in pons/medulla of SHR and that of normotensive rat. THese data suggest that the abnormally lower turnover rates of serotonin in telencephalon, hypothalamus/thalamus and midbrain may be one of the underlying neuronal factors for manifestation of hypertension in SHR.

Heme Oxygenase-l Induced by Aprotinin Inhibits Vascular Smooth Muscle Cell Proliferation Through Cell Cycle Arrest in Hypertensive Rats

  • Spontaneous hypertensive rats (SHR) are an established model of genetic hypertension. Vascular smooth muscle cells (VSMC) from SHR proliferate faster than those of control rats (Wistar-Kyoto rats; WKY). We tested the hypothesis that induction of heme oxygenase (HO)-1 induced by aprotinin inhibits VSMC proliferation through cell cycle arrest in hypertensive rats. Aprotinin treatment inhibited VSMC proliferation in SHR more than in normotensive rats. These inhibitory effects were associated with cell cycle arrest in the G1 phase. Tin protoporphyrin IX (SnPPIX) reversed the anti-proliferative effect of aprotinin in VSMC from SHR. The level of cyclin D was higher in VSMC of SHR than those of WKY. Aprotinin treatment downregulated the cell cycle regulator, cyclin D, but upregulated the cyclin-dependent kinase inhibitor, p21, in VSMC of SHR. Aprotinin induced HO-1 in VSMC of SHR, but not in those of control rats. Furthermore, aprotinin-induced HO-1 inhibited VSMC proliferation of SHR. Consistently, VSMC proliferation in SHR was significantly inhibited by transfection with the HO-1 gene. These results indicate that induction of HO-1 by aprotinin inhibits VSMC proliferation through cell cycle arrest in hypertensive rats.

Study of moxibustion stimulation in the ovariectomized rat model

  • In order to examine the effectiveness of moxibustion stimulation for climacteric disturbance, we administered moxibustion stimulation to ovariectomized (OVX) rats, and compared the tail surface temperature, laboratory parameters, and the level of locomotor activity with those of untreated rats. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g) were divided into three groups. The rats in the OVX-M group underwent moxibustion stimulation after ovariectomy. The rats in the OVX-C group underwent ovariectomy but did not receive moxibustion stimulation. The rats in the Normal group received neither ovariectomy nor moxibustion stimulation. The level of locomotor activity was determined by a metabolism measuring system. The tail surface temperature was significantly lower in the OVX rats before moxibustion stimulation than in the Normal group. In the OVX rats before moxibustion stimulation, there was no clear difference in the level of locomotor activity between the active and resting phases, and the pattern of locomotor activity was irregular. After moxibustion stimulation, the tail surface temperature of the OVX-M group did not significantly differ from that of the Normal group, and the pattern of locomotor activity of the OVX-M group became diphasic with clear active and resting phases, similar to that observed in the Normal group. These results demonstrate that moxibustion stimulation is effective for the treatment of climacteric disturbance.

Effects of Atrial Natriuretic Peptide on Renal and Endocrine Functions in Spontaneously Hypertensive Rats (Spontaneously Hypertensive Rat에 있어서 Atrial Natriuretic Peptide의 신장기능과 몇가지 호르몬 분비에 미치는 영향)

  • The present study was undertaken to clarify the involvement of atrial natriuretic peptide in the development of hypertension in spontaneously hypertensive rats. Plasma concentration of immunoreactive atrial natriuretic peptide was higher in spontaneously hypertensive rats than in normotensive Sprague-Dawley and Wistar rats. Plasma renin concentration was lower in SHR than in normotensive rats, as observed in earlier experiments. Hydration-induced increase in urine flow and urinary excretions of sodium and potassium were smaller in SHR than in normotensive control rats. Intraarterial infusion of atrial natriuretic peptide resulted in increases in urine flow, urinary excretions of sodium and potassium in both hypertensive and normotensive rats. Renal response to atrial natriuretic peptide was markedly suppressed in SHR. Plasma renin and aldosterone concentration were suppressed by atrial natriuretic peptide in both SHR and normotensive rats. The responses were not significantly different in both groups. These results suggest that the renal responsiveness to atrial natriuretic peptide may be suppressed in SHR by some mechanisms still remaining obscure.

Protective Effect of DA-9601, an Artemisiae Herba Extract, on Radiation-induced Colitis in Wistar Rats

  • This study was performed to examine the effects of DA-9601, a novel antiulcer agent extracted from Artemisiae Herba, on radiation colitis in the rat. Female Wistar rats received a 30 Gy dose of irradiation to the 2 cm of distal colon in length using an intrarectal applicator system. 30 mg/tg or 100 mg/kg of DA- 9601 was administered orally 30 min before and 4 h after radiation on day 1. And the same dose of DA-9601 was given to the animals twice a day from day 2 to 14. As a reference control, sucralfate suspension (100 or 300 mg/head) was given as an enema based on the same treatment schedule of DA-9601. Body weight change and the frequency of diarrhea were recorded during the observation period as markers of radiationinduced injury, All animals were sacrificed on day 15 for evaluation of macro- and microscopic findings and mucosal myeloperoxidase (MPO) activity. Radiated animals showed diarrhea, mucosal redness and histologic changes characterized by edema and eosinophilic infiltration of the periglandular lamina propria with loss of colonic epithelium. Radiation also significantly increased mucosal MfO activity of affected colon f\\\\\\\\`<0.05). However, most of these changes were completely protected by oral administration with DA-9601. DA-9601 reduced radiation-induced histologic alteration significantly in a dose-related manner (P<0.05). In addition, mucosal MPO activity in rats receiving high dose of DA-9601 decreased significantly when compared with that in radiated control. High dose of sucralfate (300 mg/head) alleviated radiation-induced histologic lesion, but failed to reach statistical significance. The results of this study suggest that DA-9601 can be useful for the prevention of acute clinical symptoms of radiation proctocolitis and that decrease of mucosal MPO by DA-9601 plays a role in its protective mechanism(s), at least in part.
