야생초 추출물에 의한 간장내 활성산소 생성과 항산화 효소계 조절에 관한 연구 (Modulation of Hepatic Lipid Peroxidation and Antioxidant Defenses by Wild Plants Extracts)

  • This study was performed to elucidate the possible antioxidative effects of several wild plant extracts. Wild plants were extracted with methanol or water using general method. In first experiments, antioxidative effects were measured by lipid peroxidation using rat brain homogenate. Coptis japonica extract showed the highest antioxidative activity among the 15 wild plant extracts. In second experiments, rats were fed on the semipurified diets with or without Coptis japonica extracts at the level of 0.5% for 4 weeks. MDA production of liver homogenate were significantly lower in the rats fed Coptis japonica extracts (P<0.05). Cytosolic catalase. GPX, and SOD activities were not changed, whereas the activities of GST and glutathione level were significantly higher in rats fed Coptis japonica extracts (P<0.05). These results suggest that Coptis japonica extract has an antioxidative effect through increasing GST activity and glutathione level and decreasing MDA production.

Spontaneously Hypertensive Rats에 있어서 산삼배양근 추출물의 항고혈압 효과 (Antihypertensive Effect of Wild Mountain Ginseng Cultured Root Extract in Spontaneously Hypertensive Rats)

  • The antihypertensive effects of the Wild Mountain Ginseng Cultured Root Extract (WMGCE) were investigated in spontaneously hypertensive rats (SHR). Daily oral administration of the WMGCE (100 and 200 mg/kg) exhibited a significant decrease in blood pressure in SHR rats during for 8 weeks. The systolic blood pressure was dose- and time-dependently decreased significantly from the second weeks (p < 0.05) to the end of WMGCE treatment in SHR. The WMGCE decreased the plasma levels of sodium, potassium, chlorides, urea and osmolarity in SHR rats but no statistically significant change was observed. Furthermore, no significant changes were noted on heart weight, heart rate and diameter of aorta after WMGCE treatment in SHR. Our results suggest that daily oral administration of WMGCE at the dose of 100 and 200 mg/kg for 8 weeks exhibited antihypertensive activities.

산삼약침이 자궁내막증 유발 흰쥐에 미치는 영향 (Effects of Cultivated Wild Ginseng Pharmacopuncture on Surgically Induced Endometriosis in Rats)

  • Purpose : This study was carried out to identified the effects of Cultivated Wild Ginseng Pharmacopuncture(CWG Pharmacopuncture) to the rats with surgically induced endometriosis. Methods : Endometrial tissue was implanted in the serosal wall of the small intestine in rats. The rats were divided randomly into an experimental and control group. The experimental group was treated with CWG Pharmacopuncture intravenous injection to the vein of tail and the control group was given same dose of normal saline for 4 weeks. And the size of the ectopic uterine tissue was estimated, and the serum progesterone, estradiol and cytokine($TNF-{\alpha}$, IL-2, IL-4, IL-6, IL-10) concentrations were analyzed. Results : 1. The size of the ectopic uterine implants in the experimental group was much smaller than that in the control group. 2. In analysis of serum hormone, the concentration of progesterone showed higher value than that of control group and estradiol showed lower value. 3. In analysis of cytokine, $TNF-{\alpha}$, IL-2 and IL-6 concentrations showed lower value than that of control group. 4. In analysis of cytokine, IL-4 concentration showed no significant difference and IL-10 concentrations showed higher value than that of control group. Conclusion : These results indicated that CWG Pharmacopuncture is useful treatment for endometriosis.

산양산삼약침이 고지방식이에 의한 당뇨병 유발 흰쥐에 미치는 영향 (Effects of distilled Cultivated Wild Ginseng Herbal Acupuncture in Rats with Diabetes Induced by High Fat Diet)

  • Objectives : This study was to verify the effects of distilled cultivated wild ginseng herbal acupuncture(CWGHA) on diabetes by hematological analysis. Methods : Rats were fed with high fat diet for 8 weeks and the rats with hyperglycemia were selected for the experiment. Various treatments of distilled cultivated wild ginseng herbal acupuncture were administered intravenously and glucose, ${\beta}-lipoprotein,$ triglyceride, total-cholesterol, HDL-cholesterol, LDL-cholesterol, Free Fatty acid(FFA), TBARS, superoxide dismutase(SOD), catalase and glutathione peroxidase activities in the liver were analyzed. Results : 1. Experiment group 3(0.1 ml of CWGHA was injected intravenously 10 times) showed significant decrease in serum glucose, ${\beta}-lipoprotein,$ triglyceride, LDL-cholesterol levels and liver TBARS compared to the control group, whileas showed significant increase in liver glutathione peroxidase activity. 2. Experiment group 2 and 3 (treated with 0.5 ml, 1 ml, respectively), showed significant decrease in serum FFA, total cholesterol and TBARS levels compared to the control group, and showed significant increase in liver superoxide dismutase and catalase activities. 3. Serum HDL-cholesterol didn't show significant changes in both experiment and control groups. Conclusions : Above results indicate that distilled cultivated wild ginseng herbal acupuncture plays significant role as a hypoglycemic agent and in lipid metabolism. Increase in the number of administrations yielded more significant results.

국내 야생 집쥐(Rattus norvegicus)의 murine typhus 감염조사 (Murine typhus infection of wild rats(Rattus norvegicus) in Korea)

  • Sixty eight wild rats, Rattus norvegicus, were caught from Seoul city, Kyonggi, Kangwon, Cholla, and Kyongsang-provinces. All sera collected from rats were examined by immunofluorescent test to find antibody against murine typhus. The antibody prevalance to murine typhus was 4.4%(3/68). Sero-positive rate was 12.5%(2/16) in Kyonggi province and 12.5%(1/8) in Cholla province. The sero-positive rate difference between sexes of rats was not recognized. Three Rattus norvegicus having antibody to murine typhus were morphologically classified as subadult with 8.3%(1/12), middle-aged adult with 5.9%(1/17), and old adult with 4.8%(1/21), respectively.

Protective effect of wild ginseng cambial meristematic cells on ᴅ-galactosamine-induced hepatotoxicity in rats

  • Background: Panax ginseng has a wide range of biological activities including anti-inflammatory, antioxidant, and immunomodulatory functions. Wild ginseng cambial meristematic cells (CMCs) were obtained from P. ginseng cambium. This study examined the protective mechanism of wild ginseng CMCs against $\small{D}$-galactosamine (GalN)-induced liver injury. GalN, a well-known hepatotoxicant, causes severe hepatocellular inflammatory damage and clinical features similar to those of human viral hepatitis in experimental animals. Methods: Hepatotoxicity was induced in rats using GalN (700 mg/kg, i.p.). Wild ginseng CMCs was administered orally once a day for 2 wks, and then 2 h prior to and 6 h after GalN injection. Results: Wild ginseng CMCs attenuated the increase in serum aminotransferase activity that occurs 24 h after GalN injection. Wild ginseng CMCs also attenuated the GalN-induced increase in serum tumor necrosis factor-${\alpha}$, interleukin-6 level, and hepatic cyclooxygenase-2 protein and mRNA expression. Wild ginseng CMCs augmented the increase in serum interleukin -10 and hepatic heme oxygenase-1 protein and mRNA expression that was induced by GalN, inhibited the increase in the nuclear level of nuclear factor-kappa B, and enhanced the increase in NF-E2-related factor 2. Conclusion: Our findings suggest that wild ginseng CMCs protects liver against GalN-induced inflammation by suppressing proinflammatory mediators and enhancing production of anti-inflammatory mediators.

The Therapeutic Effect of Tissue Cultured Root of Wild Panax ginseng C.A. Mayer on Spermatogenetic Disorder

  • This study examined the possibility of using a tissue cultured root of wild Panax ginseng (tcwPG) as a fertility agent. The effect of tcwPG on spermatogenesis was studied using male rats. The tcwPG crude powder was administered orally to 7-week-old rats over a 6-week period. The number of sperm in the testes and epididymides was significantly higher than the control. A histological examination did not reveal any morphological changes in the testes from the tcwPG powder treated rats. Moreover, there were no significant differences in the weights of the heart, spleen, liver, kidney, brain, testes and epididymides. Oligospermia was also induced by administering 2,3,7,8-tetrachlorodaibenzo-p-dioxin (TCDD) to the rats in order to estimate the feasibility of using tcwPG as treatment for infertility caused by spermatogenic disorders. After exposing the rats to TCDD, the tcwPG saponin fraction treated rats showed some improvement in the body weight, sperm number and testis morphology. It was estimated that tcwPG had feasibility as a therapeutic agent on spermatogenic disorder.

한국산 야생식용식물이 당뇨유발 흰쥐의 혈당 및 간과 근육내 에너지원 조성에 미치는 영향 (The Effects of Korean Wild Vegetables on Blood Glucose Levels and Liver-muscle Metabolism of Streptozotocin-induced Diabetic Rate)

  • Attempts were made to determine the effects of 5-Korean wild vegetabel consumptions on blood glucose leveles and orgen-energy metabolisms in streptozotocin-induced diabetic rats. The 5-Korean wild vegetables were : Cassia tora L.(C.t), Lycium chinese Mill(L.c), Trichosanthes kirilowii Max(T.k), Polygonatum odoratum var. Pluriflorum Ohwi(P.o) and Arctium lappa L(A.l). Sixty male Spargue-Dawley rats(160-220g) were induced diabetes mellitus by streptozotocin injection into the tail vein and were devided into 6 groups : a diabetic control and 5-experimental groups. All groups of the rats were fed on a AIN-76 diet, and the 5-experimental groups were fed with each wild vegetable (10%) for four weeks. An increased tendency in body weight of all the groups was observed and the tendency was more significant in L.c, T.k. and P.o. groups. The organ weight of liver and kidney were higher in L.c. and A.l. groups and lower in T.k. group which has shown the improvement from diabetes. Plasma glucose levels were markedly decreased from the 1st week in C.t, T.k. and P.o. groups and the tendency has lasted throughout the four weeks experimental period. The consumption of P.o. has decreased plasma cholesterol level while any significant difference was not seen in plasma protein levels from all the experimental groups. The level of plasma triglyceride was decreased in P.o. group and the levels of plasma free fatty acids were also significantly lower in P.o. and T.k. groups. The liver protein levels were significantly higher in P.o. and T.k. groups and these two groups also showed the negative or relatively small amount of urinary glucose excretion. The experimental group of T.k. has revealed the decreased level of muscles protein and the increased level of muscle glycogen. The 5-Korean wild vegetables contained dietary fiber and 9-analyzed minerals comparable to the ordinary use vegetables.

국내 야생 집쥐류(Rattus norvegicus와 R rattus)의 spotted fever group rickettsia 감염에 관한 역학조사 (Epidemiological study for spotted fever group rickettsia of wild rats(Rattus norvegicus and R rattus) in Korea)

  • Seventy wild rats were caught from Seoul city, Kyonggi, Kangwon, Honam, and yongnam provinces. All of them were in same species, Rattus norvegicus, except two R rattus from Kyonggi province. Seventy sera from wild rats were studied by immunofluorescent antibody assay for evidence of infection by spotted fever group rickettsia. The antidody prevalance was 37.14%(26/70) for spotted fever group rickettsia. The sero-prevalence rates for spotted fever group rickettsia antibody was the hightest in Kyonggi province with 55.56%(10/18), yongnam province with 50.00%(10/20), Kangwon province with 25.00%(2/8), Seoul city with 18.75%(3/16), and Honam province with 12.50%(1/8). The sero-positive rates difference between sexes were higher in female with 46.15%(12/26) than in male with 31.81%(14/44) for spotted fever group rickettsia. Twenty six of 68 Rattus norvegicus with antibody for spotted fever group rickettsia were in subadult with 50.00%(6/12), young adult with 38.89%(7/18), middle-aged adult with 35.29%(6/17), and old adult with 33.33%(7/21). No antibody was detected from R rattus.

산양산삼 추출물이 지방세포의 분화에 미치는 영향 (The Effect of Cultivated Wild Ginseng Extract on Preadipocyte Proliferation)

  • Objectives The purpose of this study is to investigate the effects of cultivated wild ginseng extract on primary cultured preadipocyte and adipocytes. Methods Diminish preadipocyte proliferation does primary role to reduce obesity. So, preadipocytes and adipocytes were performed on cell cultures with using Sprague-Dawley rats and treated with 0.01-1mg/ml cultivated wild ginseng extract. Result At all concentrations, cultivated wild ginseng extract wasn't show the suppress proliferation of preadipocytes significantly and failed to show effects on decomposition of adipocytes except high dosage. Conclusion Based on these findings, cultivated wild ginseng is not a suitable choice for the treatment of localized obesity.
