취학전(就學前) 아동(兒童)의 식품기호(食品嗜好)와 식습관(食習慣) 조사연구(調査硏究) (A Study on Food Attitude and Dietary Habits of Pre-school Children)

  • A group of 132 pre-school children with a mean age of six years and one month was the subjects of this investigation. These children were members of three nursery schools locating in Kwang-ju city. Each child's mother replied to a questionaire which included 42 food items and 4 questions. The results showed that the food preference of each child was variable and the group as a whole expressed more positive than negative feelings. The average number of foods liked per child was 22.9(range: $7{\sim}42$) among 42 food items. Peach, Apple, Strawberry, Laver, Egg, Tomato, Sweet Potato, Tangerine Orange and Beef were ranked in descending order of 'like' ratings. The average number of foods refused was 4.9 (range: $0{\sim}25$). Onion, Liver, Red Pepper Leaf, Root of Bellflower, Taro, Wild Seasame Leaf, Egg Plant, Cabbage, Water Cress and Bracken were ranked in descending order of 'refuse' ratings. The odor of hurting feelings, the hot taste and the flabby texture were the reasons why the children refused these foods. The average number of foods unexperienced was 4.4 (range: $0{\sim}14$). Kidney, Dried Small Whitebail, Tangle (DA SHI MA), Liver, SA RI Mushroom, Acorn Mook, Dried Fragrant Mushroom, Pine Agaric, Root of Bellflower and Ped Pepper Leaf were ranked in descending order of 'unexperience' ratings. Children's 'like' rating toward total foods was 57.3% and the 'refuse' rating was 11.7%. Among eight food groups, fruits showed the highest favority (91.4%) and mushroom showed the least (25.9%). The difference between male and female in the preference of total foods did not showed significant level. But the difference were significant in those of individual food groups; other vegetables, green and yellow leafy vegetables, mushrooms(above, p.<01), fishes and shellfish and sea weeds (above, p<.05). Children who had experienced meals missed rated 59.1% and 34.1% of these children missed meals once a week. The main reason for maels missed was due to the heavey snacks before meal time. Children who had snacks twice a day rated 45.6%. Main foods used as their snacks were starch foods (Cake, Biscuit, Sweet Potato) and citrous foods (Fruits, Apple, Tangerine).

Gene Expression Analyses of Mutant Flammulina velutipes (Enokitake Mushroom) with Clogging Phenomenon

  • Regulation of proper gene expression is important for cellular and organismal survival, maintenance, and growth. Abnormal gene expression, even for a single critical gene, can thwart cellular integrity and normal physiology to cause diseases, aging, and death. Therefore, gene expression profiling serves as a powerful tool to understand the pathology of diseases and to cure them. In this study, the difference in gene expression in Flammulina velutipes was compared between the wild type (WT) mushroom and the mutant one with clogging phenomenon. Differentially expressed transcripts were screened to identify the candidate genes responsible for the mutant phenotype using the DNA microarray analysis. A total of 88 genes including 60 upregulated and 28 downregulated genes were validated using the real-time quantitative PCR analysis. In addition, proteomic differences between the WT and mutant mushroom were analyzed using two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF). Interestingly, the genes identified by these genomic and proteomic analyses were involved in stress response, translation, and energy/sugar metabolism, including HSP70, elongation factor 2, and pyruvate kinase. Together, our data suggest that the aberrant expression of these genes attributes to the mutant clogging phenotype. We propose that these genes can be targeted to foster normal growth in F. velutipes.

소나무림에서 간벌률이 산림 내 환경과 외생균근성 버섯 발생에 미치는 영향 (Effect of thinning ratio on the forest environment and fruiting of ectomycorrhizal mushrooms in a Pinus densiflora stand)

  • 간벌률이 산림 내에서 환경 요인과 외생균근성 버섯 발생에 미치는 영향을 알아보기 위해 34%, 45%, 60%의 간벌 처리 후 10년이 지난 소나무림에서 수관 울폐율, 수관 통과우량, 토양 온도, 토양 수분량, 하층 식생의 광반응과 외생균근성 버섯 발생 특징을 조사하였다. 수관 울폐율은 4월에 34% 처리구와 대조구가 80~85%로 유의적으로 높았고 11월에는 모든 간벌 처리구들이 대조구에 비하여 5~10%가량 낮아졌다. 60% 처리구의 수관통과우량은 대조구에 비하여 총 200 mm이상 많았으며 월별 수관통과 우량은 10월에 100 mm 이상 큰 차이가 나타났다. 각 처리구의 토양 온도는 60% 처리가 45% 처리에 비하여 0.3~0.5℃ 정도 높아 큰 차이는 없었으나 대조구와는 1℃이상 유의적으로 높아졌다. 간벌 처리구에서 강우 시 토양 수분량이 5% 이상 더 증가하며 34% 처리구에서는 수분 감소 속도도 느려졌다. 소나무를 제외한 주요 수종의 광합성량은 신갈나무가 7 µmolCO2·m-2·s-1로 가장 많았다. 800 µmol·m-2·s-1의 동일한 광도에서 신갈나무는 45% 처리구에서 6±0.3 µmolCO2·m-2·s-1로 가장 높은 광합성 수준을 보여주었으며 때죽나무와 쇠물푸레는 간벌률이 높을수록 광합성량이 증가하였다. 균근성 버섯의 SWI에서 종수는 각 처리구간에는 큰 차이가 나타나지 않았으나 버섯의 발생량은 34%와 45% 처리구가 대조구에 비하여 약 360~840 g 더 많이 발생하였고 60% 처리구에서는 대조구에 비하여 860 g 더 적은 양이 발생하였다. 특정 종에 따라 애광대버섯은 대조구가 간벌 처리구에 비하여 더 많은 개체수가 발생하였으나 황소비단그물버섯은 34% 간벌 처리구가 대조구에 비하여 10배 이상 증가하였다. 이상의 결과를 통하여 소나무림에서 간벌률에 따른 수관 울폐율의 감소는 산림 내에 광량과 수관통과우량을 증가시키며 토양 온도 상승 및 토양 수분량의 증가와 하층 식생의 광합성량을 증가시켜 특정 버섯의 발생량 변화에 영향을 주는 것으로 추정된다. 그러나 산림생태계의 복잡성을 고려할 때 간벌과 버섯 발생에 대한 정확한 상관관계를 알기 위해서는 장기간의 연구가 지속되어야 할 것으로 생각된다.

한국산 야생버섯 추출물의 항돌연변이원성 및 암세포 성장억제 효과 (Antimutagenic and Cytotoxic Effects of Korean Wild Mushrooms Extracts)

  • 13종의 한국산 야생버섯 추출물의 항돌연변이 효과의 검토는 S. typhimurium TA98과 TA100을 이용한 Ames 실험계로 확인하였다. 그 결과, 직접변이원인 MNNG에 대해 산속그물버섯 아재비와 구름버섯의 메탄올 추출물은 TA100에서 90-94%의 강한 항변이원성을 보였고, 갓버섯과 구름버섯의 열수 추출물도 58-61%의 저해효과를 보였다. 간접변이원인 B(a)P에 대해서는 TA98과 TA100에서 구름버섯과 턱받이금버섯 메탄올 추출물이 가장 강한 저해효과를 나타내었고, 열수 추출물중 잔피막흑주름버섯과 수원무당버섯은 TA98에서 60-70% 정도의 저해활성을, 갓버섯은 TA100에서 45% 정도의 돌연변이 억제효과를 볼 수 있었고, 나머지 버섯시료는 저해활성을 거의 보이지 않았다. 한편 야생버섯 메탄올 추출물 0.5mg/mL 투여시 결장암세포인 HT29에 대해 그물버섯, 산속그물버섯아재비, 방패외대버섯, 황소비단그물버섯, 구름버섯, 턱받이금버섯, 덕다리버섯에서 55-78% 이상의 억제 효과를 보였고, 1 mg/mL의 시료농도에서는 11종의 버섯에서 50% 이상의 억제 효과를 나타내었다. 간암세포인 HepG2에서 대해서는 방패외대버섯 메탄올 추출물 1 mg/mL에서만 58.4%의 저해능을 보였고, 나머지 시료에서는 45% 정도의 낮은 저해활성을 보였다. 혈액암세포주인 U937의 경우도 그물버섯, 산속그물버섯아재비, 수원무당버섯, 황소비단 그물버섯, 밀버섯, 잔피막흑주름버섯 메탄올 추출물에서 60% 이상의 높은 저해 활성을 나타내었다. 구름버섯 열수추출물의 경우 HepG2와 U937에 대해서 53-65% 정도의 비교적 높은 저해활성을 보였고, 대부분의 버섯 열수 추출물은 40%이하의 낮은 저해활성을 보였다. 정상 임파구에 대해서 야생버섯 추출물 0.5-1 mg/mL 농도에서 75-80% 이상의 생존율을 보여, 야생버섯 추출물을 1 mg/mL 농도로 처리할 경우 암세포에 대한 높은 억제효과에 비해 정상세포에 대해서는 비교적 낮은 독성을 가짐을 확인하였다.

여러 가지 비빔밥의 섭취가 생쥐의 각종 면역 활성에 미치는 효과 (The Effects of Several Types of Bibimbabs on Immune Activities in Mice)

  • The purpose of this study was to evaluate the effects of several types of bibimbab (a Korean traditional meal of mixed rice with assorted vegetables), on various immune activities. Compared to control animals in a mouse model (given hamburgers), the oral administration of a portion of bibimbab containing wild plants significantly increased splenic B/T, thymic Th lymphocyte subpopulations, serum IFN-${\gamma}$ production, and enhanced hemagglutination titers up to 300%. Also, a consumption of mushroom-bulgogi bibimbab and Jeonju-style bibimbab markedly decreased compound 48/80-induced systemic anaphylaxis (immediate hypersensitivity), while bibimbab with wild plants inhibited SRBC-induced delayed type hypersensitivity. These results suggest that bibimbab with wild plants both up-regulate on immune activities and have anti-allergenic properties.

전라북도 지역의 주요 특산물 중 식품에 대한 조사 (A Survey on Foods of Principal Products in Jeollabukdo Area)

  • This survey was performed to investigate the distribution of specialities which cultivated importantly in Jeollabukdo area in order to understand food culture of this area. Foods in speciality were divided into 4 groups, agricultural, aquatic, animal and the others products. Total number of 93 species of specialities, including 40 species of agricultural products, 29 species of aquatic products, 8 species of animal products and 16 species of the miscellaneous products, were identified from Jeollabukdo area. Especially, there were various fruits and vegetables among agricultural products and fishes among aquatic products in this region. Rice, pepper, pear, Mandarin fish, beef, pine mushroom, lentinus edodes and honey were determined as specialities in Jeollabukdo area. Various aquatic products were specialities in Gochang, Gunsan, Buan and Gimje region. Fruits, mushrooms, wild edible greens and animal products were specialities in Muju, Jangsu, Jeongeup and Jinan. Sweet fish, cat fish and freshwater crab were caught off in Sumjin river region only such as Namwon, Sunchang and Imsil, Recently, pumpkin(Cucurbita maxima) and paprika were determined as specialities in Jeollabukdo. In case of aquatic products, mushrooms and wild edible greens, their wild type products decreased, while cultivated type increased Commercialization by brand naming of agricultural and animal products were rapidly increasing. The kinds of specialities in Jeollabukdo area were changing by the influence of geographical, religious and social factors.

팽이버섯(Flammulina velutipes) 교잡분리집단의 형태학적 특성 분석 (Analysis of phenotypic characterization of segregation population developed by crossing in Flammulina velutipes)

  • 팽이버섯의 야생 갈색 균주에서 유래한 단핵 계통 ASI 4019-20과 상용 품질에서 유래한 백색 단핵균주 M3를 교잡하여 분리집단을 만들고 그 특성을 조사하여 육종의 기초 자료를 작성하였다. 4019-$20{\times}M3$ 교잡으로 육성된 $F_1$인 M3-Sn 계통으로부터 단핵균주 94개를 분리하고 다시 교배 모본의 하나인 M3와 교잡하여 $BC_1F_1$인 M3-n 집단을 육성하여 생장특성을 조사하였다. 분리된 단핵균주는 형태상으로는 균사의 생장 형태가 불규칙한 것이 있었으며 톱밥 생장이 늦은 것이 많았다. 그 반면 이핵균주인 $BC_1F_1$(M3-n) 계통의 균사는 생장형태가 대부분 균일하며 톱밥에서 생장이 양호하였다. 갈색 야생 균주와 백색 상용 품종 유래의 단핵균주간 교잡으로 육성된 $F_1$의 자실체 갓색은 연갈색으로 나타나 유색이 백색에 우성이었으며, 백색 회보친을 교잡하여 육성한 $BC_1F_1$ 분리집단에서 색깔이 분포는 진갈색 7%, 갈색 25%, 연한갈색 27%, 아이보리 16%, 흰색 25%로 나타나 전체 유색에 백색의 비율은 3:1이었다.

Chemical Constituents of the Fruiting Bodies of Clitocybe nebularis and Their Antifungal Activity

  • During a continuing search for antimicrobial substances from Korean native wild mushroom extracts, we found that the methanolic extract of the fruiting body of Clitocybe nebularis exhibited mild antifungal activity against pathogenic fungi. Therefore we evaluated the antifungal substances and other chemical components of the fruiting body of Clitocybe nebularis, which led to the isolation of nebularine, phenylacetic acid, purine, uridine, adenine, uracil, benzoic acid, and mannitol. Nebularine showed mild antifungal activity against Magnaphorthe grisea and Trichophyton mentagrophytes, and phenylacetic acid potently inhibited the growth of Pythium ultium and displayed moderate antifungal activity against Magnaphorthe grisea, Botrytis cinerea, and Trichophyton mentagrophytes. The other isolated compounds showed no antimicrobial activity.

Identification of Single Nucleotide Polymorphism Markers in the Laccase Gene of Shiitake Mushrooms (Lentinula edodes)

  • We identified single nucleotide polymorphism (SNP) markers in the laccase gene to establish a line-diagnostic system for shiitake mushrooms. A total of 89 fungal isolates representing four lines, including Korean registered, Korean wild type, Chinese, and Japanese lines, were analyzed. The results suggest that SNP markers in the laccase gene can be useful for line typing in shiitake mushrooms.

Characterization of a Fibrinolytic Serine Protease from a Wild Mushroom, Lepista nuda

  • Fibrinolytic enzyme was purified from the fruiting bodies of Lepista nuda, using DEAE-Cellulose chromatography, Phenyl Sepharose chromatography, and Mono-S column chromatography. The substance has a molecular weight of 30006.62 Da as measured by MALD-TOF mass spectrometry. The N-terminal amino acid sequence of the enzyme was Tyr-Pro-Ser-Pro-Ser-His-Gln-Thr-Ala-Val-Asn-Ala-Ile-Ile-X. The activity of the enzyme was inhibited by PMSF, indicating that the enzyme is a serine protease. No inhibition was found with E-64, pepstatin, and EDTA. It has broad substrate specificity for synthetic peptides. The enzyme was stable up to $30^{\circ}C$. The enzyme hydrolyzes both Aa and y chains of human fibrinogen but did not show any reactivity for $B{\beta}$ chain of human fibrinogen.
