Effects of Reduced Glutathione and Ginseng Extract on Non-Protein Sulfhydryl, and Non-Protein Disulfide of Mouse Liver and Blood Following Whole Body X-Irradiation (Reduced glutathione 및 인삼추출액(人蔘抽出液)이 X-선전신조사(線全身照射)를 입은 마우스 간조직(肝組織) 및 혈중(血中) NP-SH 및 NP-SS에 미치는 영향(影響))

  • In an attempt to better understand the radioprotective effect of reduced glutathione(GSH), and to observe a possible radioprotective effect of Ginseng extract, whole body X-irradiation of 1,200 r was administered to the mouse either independently or immediately following the injection of GSH or Ginseng extract to the mouse intraperitoneally. The non-protein sulfhydryl (NP-SH) and non-protein disulfide (NP-SS) levels of the liver, and NP-SH level of NP-SH of the blood of the mouse were measured at 30, 60 and 120 minutes, and results were compared with the normal. The results thus obtained are summarized as follows; 1) The normal values of NP-SH and NP-SS of the mouse liver were $5.90{\pm}0.46\;{\mu}\;mol/gm\;wet\;wt.,\;and\;3.02{\pm}0.42\;{\mu}\;mol/ml$ wet wt., respectively, and the normal value of NP-SH of NP-SH of the mouse blood was $3.98{\pm}1.29\;{\mu}\;mol/ml$ 2) The injection of both GSH and Ginseng extract produced the highest values of NP-SH in the liver at 30 minutes, but a gradual decrease to the normal was observed thereafter. When X-irradiation alone was applied, the liver NP-SH value was lower than the normal at 60 minutes post-irradiation and thereafter. When Ginseng extract was injected immediately prior to X-irradiation, the liver NP-SH was lower than the normal throughout the experiment with the lowest value at 60 minutes. However, the combination of GSH and X-irradiation produced higher than the normal values throughout the entire experiment. 3) The liver NP-SS value was most significantly elevated at 30 minutes after the injection of GSH, hut the recovery to the normal was observed thereafter. The injection of Ginseng extract produced slightly higher liver NP-SS values at 30 and 60 minutes, but the value at 120 minutes was similar to the normal. The single application of X-irradiation resulted in the lower then normal liver NP-SS values throughout the entire experiment. When GSH was injected price to X-irradiation, the liver NP-SS values were higher than the normal at 30 and 60 minutes followed b the recovery to the normal at 120 minutes. The combination of Ginseng extract and X-irradiation showed generally lower liver NP-SS values throughout the experiment. 4) The blood NP-SH showed the higher than the normal values in all the experimental groups except when GSH was injected prior to X-irradiation alone produced e significantly elevated blood NP-SS value at 30 minutes post-irradiation.

  • The effect based upon changes of body weight and effect of X-ray irradiation on tryptophan pyrrolase (TPO) activity in liver homogenate of albino rats was studied. 1. The average TPO activity of control group of 33 rats weighing $95{\sim}172g$\;was\;2.372{\pm}0.165{\mu}$ moles kynurenine/g protein/hr. 2. Correlation between body weight of 33 rats and its TPO activity was not showed practically(r=0.011). 3. TPO activity of whole-body X-ray irradiated rats (700r) was increased about double at 3hours and increased for a period of 4 days after irradiation, but after 6 days it was decreased gradually. This result is that whole-body X-ray irradiation showed dual effect on TPO activity in rat liver.

  • Oxygen consumption rate $(QO_2)$ and protein content of liver and kidney of the Ehrlich ascites tumor-bearing mouse were measured from 6th till 14th day after the inoculation of $4{\times}10^6$ Ehrlich ascites tumor cells. The results thus obtained were compared with those of the groups in which; 1) Whole body x-irradiation with 400 r was done to mouse prior to the inoculation of $4{\times}10^6$ Ehrlich ascites tumor cells, 2) Same number of the irradiated tumor cells were inoculated after subjecting the tumor cells to x-irradiation with 400 r or 900 r in vitro, and 3) the normal, and the following results were obtained; 1. $QO_2$ of the liver and kidney of the tumor-bearing mouse were all lower than the normal and a gradual decrease of $QO_2$ in both liver and kidney was noted as the ascites tumor was progressively developing. 2. In the groups where whole body x-irradiation with 400 r was done, or x-irradiation of ascites tumor cells in vitro with either 400 r or 900 r, $QO_2$ of the liver and kidney were lower than the normal, and the pattern of the decrease was similar in the case of the tumor-bearing mouse. 3. Protein contents in all the groups showed lower values than the normal, and the decrease was gradual as the ascites tumor was developing. 4. $QO_2$ and protein levels in the liver were generally lower than those in the kidney. 5. A certain cancerous metabolism was, therefore, noted in the remote organs of Ehrlich ascites tumor-bearing animal.

Effect of Single or Fractionated X-Irradiation on the NP-SH and NP-SS of Rabbit Liver and Lung Tissues (단회(單回) 및 분획(分劃) X선(線) 조사(照射)가 가토(家兎)의 간(肝) 및 폐조직(肺組織)의 NP-SH 및 NP-SS에 미치는 영향(影響))

  • In view of the importance of sulfhydryl groups in producing a chemical protection against ionizing radiation, an attempt was made to evaluate the changes of intrinsic non-protein sulfhydryl (NP-SH) and non-Protein disulfide(NP-SS) of the lung and liver tissues of rabbits following the whole body X-irradiation with 900 r either in single or fractionated $(300\;r{\times}3)$ dose. NP-SH was measured by Ellman's method, and NP-SS was measured by the electrolytic reduction method described by Dohan. Experiment was performed at 1,3,5,24 and 48 hours post·irradiation, and the results were compared with the control. The results thus obtained are summarized as follows; 1) Intrinsic levels of NP-SH and NP-SS of normal rabbits were $0.77{\pm}0.10$ and $0.61{\pm}0.07\;{\mu}mol/gm$ wet weight in liver, and $0.28{\pm}0.03$ and $0.54{\pm}0.03$ in lung tissues respectively. 2) NP-SH of liver after single X-irradiation showed no significant change in general, but at 48 hours post·irradiation, it was elevated comparing with the normal value. 3) Levels of NP-SS in liver was decreased than the normal value in the irradiated groups, and the lowest level was observed at 3 hours after single X-irradiation and at 5 hours after fractionated irradiation. 4) In lung tissues, levels of NP-SS showed no significant change from the control at earlier experimental hours, but a great decrease was observed at later Part of the experiment.

Studies on the Chromatin Isolated from the Organs of Animals Received Whole-body X-ray Irradiation

  • 1. Within experimental chromatin, the total protein: DNA ratio did not vary in the same organs of control and irradiated rats. However, the amount of RNA and total protein associated with the DNA varied considerably among the different types of chromatin. In particular, the content of chromatin was the control tissue. RNA and total protein ratio of chromatins from brtain, liver, testis and spleen declined with experimental I organs. 2. There was the same quantitative relationship between the amount of RNA and the amount of histone-protein associated with DNA in chromatin. 3. RNA: DNA ratio of chromatin showed 1.5-2 times increas in the irradiated organs except brain. However, RNA: DNA ratio was decreased in chromatin by irradiation. 4. Histone-protein:residual protein ratio was greatly varied among the organs. However, the effect was not found by irradiation. 5. Priming activity of chromatin showed a higher value in testis and the activity was greater in organs with higher metabolic activity: 6. Inhibition of Actinomycin D is observable in chromatin from testis, liver, spleen and brain declined without relationship between irradiated and non-irradiated conditions. Ammonium sulfate showed increased priming activity by the electrostatic dissociation of DNA and histone in chromatin on the stimulation depending on property of chromatins. 7. It is suggested that the results support a proposal that testis and spleen of highly sensitive to irradiation should an increase in the priming activity whereas brain and liver of lower sensitivity decreased in the activity.

Whole-liver Radiotherapy Concurrent with Chemotherapy as a Palliative Treatment for Colorectal Patients with Massive and Multiple Liver Metastases: a Retrospective Study

  • The purpose of this study was to investigate whether whole-liver radiotherapy plus a tumor-boost dose with concurrent chemotherapy is beneficial for colorectal cancer patients with massive and multiple liver metastases. From January 2007 to December 2012, 19 patients who exhibited massive (with a longest diameter > 5 cm) and invasive liver metastases and multiple metastases were treated with radiotherapy and concurrent chemotherapy. The total radiation dose was 53.4 Gy (range 38.8 Gy-66.3 Gy). All of the patients received a continuous intravenous dose of 5 fluorouracil (5-FU) 225 mg/m2 concurrently with radiation. The median survival time was 19 months. The 1- and 2- year overall survival rates were 78.3% and 14.3%, respectively. Of all of the patients who presented with abdominal pain, 100% experienced a decrease in pain. Decreases in the rates of ascites and jaundice were confirmed by ultrasound and bilirubin levels. No cases of Grade 4 or 5 acute or late toxicity were recorded. There were only two cases of Grade 3 toxicity (elevated bilirubin). These data provide evidence that whole-liver radiotherapy plus a tumor-boost dose with concurrent chemotherapy is beneficial for colorectal cancer patients with massive and multiple liver metastases.

Radioprotective Effect of Ginseng Components on Antioxidant Enzymes, Glutathione and Lipid Peroxidation of Liver in ${\gamma}$-Irradiated Mice (홍삼 분획물이 감마선을 비사한 생쥐 간에서 항산화물질과 지질과산화에 미치는 방사선 보호효과)

  • In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.

Whole Liver Palliative Radiotherapy for Patients with Massive Liver Metastases

  • Background: The purpose of this retrospectively study was to examine the effectiveness and tolerability of a radiotherapy technique for the palliation of symptomatic liver metastases. Materials and Methods: Twentyseven patients with liver metastases were enrolled and received targeted whole liver irradiation consisting of mean 1, 8 Gy in five to twelve fractions to a total mean dose 17Gy. Symptoms at baseline were hepatic pain (26 patients), lost of weight (6), lack of appetite (2), and night sweats (1). Seventeen patients (63%) had failed previous treatment with chemotherapy and/or high-dose steroids. Results: Individual symptom response rates were 100% at 4 weeks. Partial or complete global symptomatic responses were noted in 11 patients (40%) after 2 months. After 3 months, 8 patients (28%) reported loss of pain. The treatment was well tolerated with one patient (3%) experiencing grade 3 toxicity (one vomiting and one diarrhoea). Overall the median survival time was 4.9 months (range 1 - 14 months). One year survival was 39%. Conclusions: This is simple and well-tolerated treatment but to achieve good palliation effects we should carefully selected patients whose conventional treatment does not bring good analgesic control.

Radioprotective Effect of Ascorbate in the Liver of ${\gamma}-Irradiated$ Mice (Ascorbate의 방사선 보호효과에 관한 연구)

  • In the present study, to determine whether the ascorbate protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of ascorbate(240 mg/kg, i.p) pretreatment of mice on the survival ratio, splenic weight, major antioxidant enzymes(SOD, catalase and glutathione peroxidase) activities, glutathione contents and lipid peroxidation in the liver were examined for 2 weeks after whole-body ${\gamma}-irradiation$(6.5 Gy). The 30-day survival ratio Increased from 10% to 47% for mice treated with ascorbate. The ascorbate decreased the extent of loss in splenic weight and stimulated recovery of splenic weight in irradiated mice(p<0.01). On the day of 14 after ${\gamma-irradiation}$, the ascorbate pretreatment produced a slight increase of antioxidant enzymes activities and significantly increased reduced glutathione(GSH) contents(P<0.05) in the liver compared with non-treated group. Pretreatment with the ascorbate significantly decreased GSSG/total GSH ratio(p<0.05) without the change of GSSG in the liver and inhibited the radiation-induced increase in the hepatic malondialdehyde levels(p<0.05). In these results, we found that its radioprotective effect by protecting antioxidant enzyme activities and glutathione contents from radiation induced a decrease, and thereby suppressing lipid peroxidation which is induced by free radicals.

Effect of Reduced Glutathione and Cold Exposure on the Levels of NP-SH and NP-SS of Some Organs of the Whole-Body X-Irradiated Mouse (Reduced glutathione 및 한랭노출(寒冷露出)이 X선(線) 전신조사(全身照射)를 입은 마우스수종장기(數種臟器)의 NP-SH 및 NP-SS에 미치는 영향(影響))

  • In an attempt to observe the Possible radioprotective actions of single or combined application of reduced glutathione (GSH) and cold exposure in mammals, the albino mouse was subjected to GSH injection, cold exposure at $-1{\pm}0.2C$ and whole-body X-irradiation with 900r either singularly or in combination, or the X-irradiation following the cold exposure and/or GSH injection. The levels of intrinsic NP-SH and NP-SS of the liver, brain and heart were measured at one hour after each application, and the results were compared with the control, i.e., non-irradiated and non-cold exposed normal animal. NP-SH was measured by the Ellman's method, and NP-SS was measured by the electrolytic reduction method described by Dohan and Woodward. The results thus obtained are summarized as follows: 1) The levels of NP-SH in the liver, brain and heart of the normal mouse was $6.35{\pm}0.61,\;2.65{\pm}0.15\;and\;3.17{\pm}0.10{\mu}\;mol/gm\;wet\;wt.$, respectively, and NP-SS was $3.09{\pm}0.11,\;2.95{\pm}0.20\;and\;0.18{\pm}0.24{\mu}\;mol/gm\;wet\;wt.$, respectively. 2) Though there were some degrees of difference among the tissues studied, a general tendency of (1) elevated NP-SH and NP-SS levels in the GSH injection group, (2) similar or slightly elevated NP-SH and NP-SS levels in the cold-exposed group, and (3) markedly decreased levels of NP-SH and NP-SS in the X-irradiated group, was observed. When GSH was injected prior to the X-irradiation, NP-SH and NP-SS in all the tissues studied showed generally higher values than in the group where the X-irradiation was given alone, and the values were close to the normal. In the group where the cold exposure was applied immediately after the X-irradiation, no significant difference was observed in the NP-SH and NP-S5 levels comparing with the X-irradiation group. On the contrary, when GSH was injected immediately prior to the X-irradiation or cold exposure, NP-SH and NP-SS levels were either similar to, or higher than, the normal value.
