In this study, we investigated antioxidant activities and whitening effects of Acer mono sap by encapsulation of nanoparticles. Acer mono sap was through ultra high pressure process and then encapsulated by lecithin. Nano-encapsulated The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 89.7% in adding sample (1.0 mg/ml), compared to sap of non-encapsulation. It was showed strong inhibition effect on melanin production test by Clone M-3 cells as 47.8%. High inhibitory of tyrosinase was also measured as 85.8% by adding lecithin nano-particle of 1.0 mg/ml. The nano-particles also showed 14.8% of low cytotoxicity against human normal fibroblast cells in adding 1.0 mg/ml of the highest concentration. These results indicate that Acer mono sap may be a source of cosmetic agents capable of improving whitening effect and antioxidant activites.

Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.
Melanin biosynthesis inhibitors are useful not only for the materials used in cosmetics as skin-whitening agents but also for the remedy of hyperpigmentation. In order to find the new skin-whitening compounds from the natural products. screening of tyrosinase and melanin biosynthesis inhibitory activities in vitro has been carried out. The MeOH extracts and/or fractions of Polygoni multiflori Radix, Dalbergiae odoriferae Lignum. Solnani nigri Herba. Polygoni cuspidati Radix. POlygoni multiflori Ramulus. Salviae Radix showed tyrosinase inhibitory effects. Four methanolic extracts also showed melanin biosynthesis inhibitory effects in B-16 melanoma cell line.

In an attempt to find natural sources of antioxidants and whitening agents, Comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex were carried out. Comparison of the four ethanol extracts revealed that, Astragali Radix had the highest electron-donating ability(72.5%) and the highest SOD-like ability(26.1%). The xanthine oxidase experiment exhibited a hindrance effect of 88.5% in Atractylodis Rhizoma Alba, 81.1% in Acanthopanacis Cortex, 75.8% in Astragali Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 42.1% in the Acanthopanacis Cortex, 37.2% in the Atractylodis Rhizoma Alba, 6.0% in the Astragali Radix. Based on these results, we suggest that the ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex can be used as food and cosmetic ingredients.

To identify the natural and novel cosmeceutical ingredients which have whitening function, we examined the effects of tyrosinase activity and melanin production on halophyte Limonium tetragonum (Thunb.) A. A. Bullock extracts. As a result, the extracts showed whitening effect with no cytotoxicity. Melanin contents of B16F10 melanoma cells were decreased in a dose-dependent manner at 50, 100, $200{\mu}g/mL$ treatment of the extracts. In tyrosinase activity inhibition test, L. tetragonum (Thunb.) A. A. Bullock extracts showed decreased tyrosinase activity as the concentration of ${\alpha}-MSH$ was increased. Furthermore, it was observed that the tyrosinase and MITF expression was significantly downregulated by adding L. tetragonum (Thunb.) A. A. Bullock extracts in ${\alpha}-MSH$, a melanogenesis inducing material, treated B16F10 melanoma cells. These results indicate that L. tetragonum (Thunb.) A. A. Bullock extract might be an effective whitening agent by inhibit melanin formation.

In this study, we evaluated the anti-oxidative, anti-wrinkle and whitening effect of Duchesnea indica (Andr.) Focke extract. Duchesnea indica (Andr.) Focke was extracted by two different solvents which were n-hexane and ethyl acetate. The anti-oxidant activity was measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical). And the inhibitory activities of tyrosinase for whitening effect and collagenase for anti-wrinkle were investigated. For anti-oxidant activity and whitening activity, ethyl acetate fraction of Duchesnea indica (Andr.) Focke extract showed more significant activity than n-hexane fraction of Duchesnea indica (Andr.) Focke extract. For anti-wrinkle activity, ethyl acetate fraction of Duchesnea indica (Andr.) Focke extract exhibited strong inhibition effects compared with reference. Therefore, Duchesnea indica (Andr.) Focke extract may be useful as a new antioxidant and anti-aging agent.

In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix were carried out. Comparison of the three ethanol extracts revealed that, Paeoniae Radix had the highest electron-donating ability(79.3%),; however, Rubi Fructus had the highest SOD-like ability(31.1%). The xanthine oxidase experiment exhibited a hindrance effect of 74.3% in Cuscutae Semen, 80.4% in Rubi Fructus and 60.8% in Paeoniae Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 20.1% in the Cuscutae Semen, 54.2% in the Rubi Fructus, 56.3% in the Paeoniae Radix. Based on these results, we suggest that the ethanol extracts of Cuscutae Semen, Rubi Fructus and Paeoniae Radix can be used as food and cosmetic ingredients.

The demand for natural substances with anti-melanogenic activity is increasing due to the recent interest in skin whitening. Intensive investigation on the culture broth of Streptomyces sp. SCO-736, a marine bacterium from the Antarctica coast, has led to the isolation of a new natural product named antaroide (1). The chemical structure was established through the interpretation of MS, UV, and NMR spectroscopic data. Antaroide is a nine-membered macrolide with lactone and lactam moieties. To investigate its applicability in skin whitening cosmetics, its anti-melanogenic activity in B16F10 murine melanoma cells was examined. As a result, antaroide displayed strong inhibitory activities against melanin synthesis and also attenuated the dendrite formation induced by the α-melanocyte stimulating hormone (α-MSH). Antaroide suppressed the mRNA expression of the melanogenic enzymes such as tyrosinase, TRP-1 and TRP-2. This suggests that it may serve as a transcriptional regulator of melanogenesis. Collectively, the discovery of this novel natural nine-membered macrolide and its anti-melanogenic activity could give new insights for the development of skin whitening agents.

This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

In this study, we investigated whitening and anti-inflammatory constituents from a watermelon (Citrullus lanatus, C. lanatus) vines (leaves and stems). As anti-melanogenesis and anti-inflammatory activities were screened for the ethanol extract and solvent fractions, n-hexane (n-Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex and EtOAc fractions of C. lanatus; ${\alpha}-linolenic$ acid (1), sigmast-7-en-O-${\beta}$-D-glucopyranoside (2), 1-feruloyl-${\beta}$-D-glucopyrinoside (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Whitening and anti-inflammatory effects were studied for the isolated compounds. Upon the anti-melanogenesis tests using ${\alpha}-MSH$ stimulated B16F10 melanoma cells, the compounds 1 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. For the anti-inflammation tests using lipopolysaccharide (LPS)-induced RAW 264.7 cells, the isolates 1 and 3 were determined to decrease the production of nitric oxide (NO) and pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6). Based on these results, C. lanatus vines extract could be potentially applicable as whitening and anti-inflammatory ingredients in cosmetic formulations.
