Pathogenicity and Host Range of a Potential Mycoherbicide, Isolate BWC98-105, Causing White Root Rot on Trifoliorum repens

  • White root rot of wild white clover (Trifoliorum repens) caused by isolate BWC98-105 has been first reported in Korea. Typical symptoms on root include water-soaked and dark-brown rot, resulting in complete blight of the whole plant. The fungus grew well at $20-28^{\circ}C$ and produced abundant sclerotia at 10-15 days after full mycelial growth on potato dextrose agar. Sclerotia were brown to dark-brown in color and 1-3 mm in length. When white clover plants were inoculated with mycelial suspension ($10^5$ cfu/ml) of isolate BWC98-105, the plant shoots were killed within 4-6 days and the roots were completely blighted. Sclerotia were also formed on the surface of the root covered with whitish mycelia within 10-15 days in the field. All nine isolates developed high incidences of white root rot disease on white clover seedlings, of which the symptoms were similar to those observed in the fields. Hence, their pathogenicity was confirmed on white clover. The infection rate of the fungal isolates varied from 78.5% to 95.2%, among which BWC98-105 was the most virulent isolate. The weeding efficacy of the fungus was maintained until the following year, leading to a significant reduction of reshooting. The fungus was specifically parasitic to white clover, but not to four lawn species including zoysiagrass (Zoysia japonica) under greenhouse test. The fungus also had no response to some Gramineae species including rice, but caused little damage to five species of Leguminosae.

  • Stromata of the white root rot fungus, Rosellinia necatrix, were produced on diseased roots although they were reported to develop rarely in nature. Forty-two (42) out of 47 samples produced synnemata while 23 developed stromata. Forty-seven (47) isolates obtained from diseased root samples were divided into 24 mycelium compatibility groups (MCGs). Sixteen (16) out of 24 MCGs produced stromata. Single ascospore isolates from 10 stroma samples produced dsRNA-containing isolates from diseased tissue beneath stromata. The frequency of synnema production on axenic culture varied among isolates with different origin. The dsRNA was not transmitted vertically to the ascospore offspring despite the infection of various dsRNA in the parental isolates. The dsRNA was absent in 35 ascospore isolates in two stroma samples that originated from the isolates, in which dsRNA was not eliminated by hyphal tip isolation. Consequently, sexual reproduction in the white root rot fungus was suggested to produce propagules as a new infection source and to have the function to eliminate infectious factors such as mycoviruses.

Dieback Reality of Apple Trees Resulting from Soil-Borne Fungal Pathogens in South Korea from 2016 to 2019

  • Recently, the severe dieback of apple trees resulting from soil-borne diseases has occurred in South Korea. The casual agents of dieback were surveyed on 74 apple orchards that had been damaged nationwide in 2016-2019. The number of apple orchards affected alone by Phytophthora rot, violet root rot, and white root rot was 31, 34, and 3, respectively. Also, the total number of mixed infection orchards was 6. Out of 9,112 apple trees affected by dieback, the trees damaged by Phytophthora rot, violet root rot, and white root rot were 3,332, 3,831, and 44, respectively. Moreover, the total number of mixed infection apple trees was 1,905. The provinces mainly affected were Gyeongnam, Gyeongbuk, Chungbuk, and Jeonbuk. The survey on these infected apple orchards will be available to form management strategy for the dieback that had been increased by soil-borne fungal pathogens.

재배양식에 따른 더덕 병해 발생양상 (Incidence of Diseases in Codonopsis lanceolata with Different Cultivation Method)

  • Disease incidence of Codonopsis lanceolata was surveyed at the major cultivating fields in Chonbuk province in 1996 to 1997. The main diseases of Codonopsis lanceolata were ovserved as leaf spot caused by Septoria codonopsis, anthracnose by Glomerella cingulata, brown leaf spot by Cercospora sp., rust by Coleosporium koreanum, powdery mildew by Erysiphe sp., Fusarium wilt caused by Fusarium oxyporum, and white root rot by Sclerotium rolfsii. Anthracnose, leaf spot and brown leaf spot occurred severely on leaves from early July to late August. They were caused early fallen leaves. Fusarium wilt and white root rot occurred severely on stem and below the soil line in late August. They resulted in withering to death or chlorosis and fallen of leaves. Disease incidence of Codonopsis lanceolata was also substantially different in occurrence with a method of cultivation in late growth stage. Fusarium wilt and white root rot were more severe with a method of no support cultivation than those with a method of support cultivation with a stick. Fusarium wilt occurred 48.8% in a method of no support cultivation but 3.1% in a method of support cultivation with a stick. And white root rot occurred 18.9% in a method of no support cultivation but 0.3% in a method of no support cultivation with a stick. Thus, it proved that soil-borne diseases could be controlled support cultivation with a stick.

Fusarium oxysporum Causes Root Rot on Gastrodia elata in Korea: Morphological, Phylogenetic, and Pathogenicity Analyses

  • Gastrodia elata infected with root rot disease was collected from cultivated G. elata fields in Gimcheon, Korea, in 2018. G. elata tuber surfaces exhibited root rot disease symptoms of dark-grey lesions and white fungal mycelial growth. The fungus was isolated from symptomatic tubers and cultured. Based on morphological characteristics and molecular analysis of the internal transcribed spacer region of ribosomal DNA and translation elongation factor 1-alpha, the isolated fungus was Fusarium oxysporum. This is the first report of root rot caused by F. oxysporum on G. elata tubers in Korea.

  • Among 131 isolates of Strptomyces obtained from ginseng cultivating soil, the two isolates ST59 and ST129 showing high antagonistic activity to Fusarium solani(Mart.) Appel & We. causing ginseng root rot were identified. The two isolates were identified Streptomyces alboniger Porter, et al. and Strptomyces reseolilacinus Pridham, et al., respectively, based on mrophology, cultural, and physiological characteristics on various culture media. Spore chains of ST59 and ST129 were flexuous(RF) and coiled(S). Spore surfaces of two isolates were all smooth. Aerial mass color of ST59 was white series and ST129 red series.

충북에서 사과 주요 토양병에 의한 고사율과 재배환경과의 상관관계 (Correlation between the Dieback Ratio and Cultivation Environment for Apple Orchards Infected by Soil-Borne Diseases in Chungbuk Province)

  • 이전 연구에서 충북지방의 사과과수원에서 토양병에 의한 사과나무 고사율이 매우 높게 나타났다. 본 연구에서는 토양병에 의한 사과나무 고사율과 재배환경과의 상관관계를 조사하였다. 자주날개무늬병에 의한 사과나무 고사율은 토양 내 칼슘 함량과 유의하게 정의 상관관계를 보였고, 흰날개무늬병에 의한 사과나무 고사율은 토양 내 유효 인산 함량과 유의하게 정의 상관관계를 보였다. 반면에 역병 피해 사과원 14개소에서는 사과나무 고사율과 재배환경과는 유의성 있는 상관관계가 없었다. 세분화한 재배환경과의 상관관계 분석결과는 토양 내 칼슘함량 외에 과원 경사도와 착과수가 자주날개무늬병에 의한 고사율에 영향을 미쳤고, 토양 내 유효인산 함량 외에 칼슘 함량이 흰날개무늬병에 의한 고사율에 영향을 미쳤다. 과원경사도, 토성, 마그네슘과 칼슘 함량이 사과역병에 의한 고사율에 영향을 주었다. 이 결과는 사과과원의 재배법 변경에 의해 토양병에 의한 사과나무 고사율을 감소시키는 데 적용 가능할 것이다.

  • This study was carried out to select effective fungicides against white and violet root rot caused by Rosellinia necatrix and Helicobasidium mompa with nine fungicides including thiophanate-methyl from 1993 to 1994. Through laboratory, greenhouse and field trials on inhibitory effect of mycelial growth and disease incidence against the two fungal pathogens, 5 fungicides have been selected finally. Thiopanate-methyl, benomyl, iminoctadine-triacetate and isoprothiolane were proven to have high control effect against R. necatrix. In addition to thiopanate-methyl and benomyl, tolclofos-methyl has been selected for effective control of H. mompa, since it showed prominent control effect in field trial than in laboratory or green house test.

Sclerotirum cepivorum에 대한 마늘 재배종의 저항성 검정 (Screening for Resistance of Garlic Cultivars to White Rot Caused by Sclerotium cepivorum)

  • The optimal quantity of inoculum was determined to screen resistance of garlic cultivars against Sclerotium cepivorum and 30 cultivars was tested. The growth of the pathogen in detached roots, Disease incidence was increased when the inoculum density was raised form 10 to 100 sclerotia. The optimal inoculum density to differentiate resistance or susceptibility of garlic cultivars was seemed to be 50 sclerotia. The cultivars collected from England, Japan, Nepal and Turkey, and cultivars such as common red, PI1356104 and PI135693 were less than the other cultivars in their disease incidence. The growth of S. cepivorum in detached roots varied from 23 to 33 mm according to garlic cultivars. There was no relationship between the disease incidence and the growth in detached roots. The sclerotial germination was increased significantly when root extract was extract was added. The addition of only distilled water resulted in 13% germination, but the addition of 0.25 g of root extract in 100 ml distilled water resulted in more than 85% germination. There was no difference in the stimulation of sclerotial germination among cultivars which showed different resistance.
