Cultural Condition for the mycelial growth of Ganoderma lucidum on Cereals (곡물을 이용한 영지버섯의 균사체 배양조건)

  • Cereals were used as solid-substrate for the cultivation of Ganoderma lucidum. The hydration time with cold water appeared to be 10, 11 and 12 hours for Malt, Danyeob and Black soybeans respectively, and the water content was enough for mycelial growth in this condition. The hydration times required for sorghum, job's tears, barley, brown rice and wheat were 2.5, 4, 5, 10 and 12 hours respectively, but the final water content was much less than optimum water content (65%). Hot water reduced the hydration time of soybeans, and the water content reached to 65% within $120{\sim}150$mins. This condition showed the optimum for the mycelial growth. For the other cereals, it took about $17{\sim}120$ mins to reach the optimum water content (65%). From this result, hot water was better than cold water for the hydration of cereals. We attempted to develop a practically applicable process by combining the soaking and sterilization. This process was successful with soybean and about 1.1 times of water based on the weight of soybean appeared to be suitable. In all varieties of cereal, the water content of 65% appeared to be the best for the growth of the fungi and production of glucosamine related to the amount of mycelium. The mycelial growth rate in accordance with kinds of solid-state materials was in the order of barley > wheat > job's tears > sorghum > brown rice > soybean. The glucosamine content for determing the mycelial growth in solid material was in the order of wheat> barley > brown rice > job's tears > sorghum > soybean.

Optimum mixing rate of used media for saving the production cost of Flammulina velutipes (팽이버섯(Flammulina velutipes) 생산비 절감을 위한 폐배지 적정 혼합비율)

  • These experiments were conducted to find the optimum mixing rate of used media for saving the production cost of Flammulina velutipes. The materials for media formation was used needle-leaf tree sawdust, media that finished 1th cultivation of F. velutipes, corncob meal, and rice bran, wheat bran as nutrition source. We inoculated the F. velutipes in 14 kinds of different media types and checked the spawn growth speed, fruit body quality and quantity. Two nutrition agents, which is rice bran and wheat bran, did not affected the incubation period, but the effective stem number, quality and quantity of fruit body was better at rice bran than wheat bran. The quality of fruit body produced at mixed 20% of used media (needle-leaf tree sawdust 60% + used media 20% + rice bran 20%) was similar to control plot (needle-leaf tree sawdust 80% + rice bran 20%), but the yield was improved 10% than control plot 130g. According as the used-media mixing amount increases, quality and quantity of fruit body became low remarkably. Therefore, the optimum mixing amount of used-media was 20% and it increased 10% of fruitbody yield.

Classification of Crop Lands over Northern Mongolia Using Multi-Temporal Landsat TM Data

  • Although the need of crop production has increased in Mongolia, crop cultivation is very limited because of the harsh climatic and topographic conditions. Crop lands are sparsely distributed with relatively small sizes and, therefore, it is difficult to survey the exact area of crop lands. The study aimed to find an easy and effective way of accurate classification to map crop lands in Mongolia using satellite images. To classify the crop lands over the study area in northern Mongolia, four classifications were carried out by using 1) Thematic Mapper (TM) image August 23, 2) TM image of July 6, 3) combined 12 bands of TM images of July and August, and 4) both TM images of July and August by layered classification. Wheat and potato are the major crop types and they show relatively high variation in crop conditions between July and August. On the other hands, other land cover types (forest, riparian vegetation, grassland, water and bare soil) do not show such difference between July and August. The results of four classifications clearly show that the use of multi-temporal images is essential to accurately classify the crop lands. The layered classification method, in which each class is separated by a subset of TM images, shows the highest classification accuracy (93.7%) of the crop lands. The classification accuracies are lower when we use only a single TM image of either July or August. Because of the different planting practice of potato and the growth condition of wheat, the spectral characteristics of potato and wheat cannot be fully separated from other cover types with TM image of either July or August. Further refinements on the spatial characteristics of existing crop lands may enhance the crop mapping method in Mongolia.

Improvement of Hairy Vetch Seed Production by Mixture Cropping of Hairy Vetch and Triticale

  • Demand for the domestic hairy vetch seed production will be increased with the increasing interest of environment-friendly agriculture in Korea. This study was conducted during from 2000 fall to 2003 spring at upland field of National Institute of Crop Science in Suwon, Korea to compare wheat and triticale (TC) as stake crop of hairy vetch (HV), and to know proper seeding rates and ratios between TC and HV for the maximum HV seed production. As supporting crop of HV, TC was superior to wheat at the points of higher HV seed yield, stronger TC stalk for supporting, consistence of ripening stage of two seeds. In seeding method, row seeding was superior to broadcast seeding at the points of less lodging and higher HV seed yield. HV seed yield decreased with the increase of TC seeding rate in mixture cropping (row seeding), particularly at TC seeding rates over 5kg/10a. HV seed yield increased with the increase of HV seeding rate at the condition of TC seeding rates under 5kg/10a in spite of higher lodging of mixed crops at higher HV seeding rate due to higher HV aboveground dry matter. Maximum HV seed yield was obtained at TC seeding rate of $1\~1.5kg/10a$ as indicating HV seed yield 176kg/l0a (CV. Madison) at seeding rate of TC 1kg/10a + HV 2 kg/10a in 2001, and HV seed yield 96kg/10a (CV. Common) at seeding rate of TC 1.5kg/10a + HV 4.5kg/10a in 2003. Use of all-purpose combine harvester for harvesting and appliance for separation of mixed seeds using centrifugal force, which are prerequisite for HV seed production, was excellent in the simultaneous seeds production system of HV and TC.

Saccharification and Ethanol Fermentation from Uncooked Starch using Aspergillus niger Koji (생전분의 당화와 주정발효)

  • The energy for cooking starch prior to saccharification could be saved by fermenting raw starch into ethanol using Aspergillus niger koji of wheat bran. Optimum cultivation time to produce glucoamylase was 4 days in wheat bran medium. The rate of saccharification from uncooked corn starch were optimum at pH 3.3 and 40-$50^{\circ}C$. Corn and sweet potato starch were saccharified more efficiently by wheat bran koji than other tested starch sources. 5 days of fermentation were required for optimum yield of ethanol using a mixture of AspergiUus niger koji and dried yeast. Final ethanol yields from raw corn, sweet potato, and rice starch with agitation at the rate of 100 rpm were about 95% at $30^{\circ}C$.

Studies on the optimal conditions for the high yield in the production of Ganoderma lucidum (영지버섯 다수확 생산을 위한 최적조건에 관한 연구)

  • The results were as follows; 3 additives(rice bran, wheat bran and soybean bran) to the Quercus dentata sawdust were applied at 10, 20 and 30%. High yield was recorded at 20% of rice bran and wheat bran and 30% of soybean bran. Among various sawdust media, the cultivation with the Quercus dentata sawdust alone showed the most favored growth index, such as 22 days of full mycelial growth, 11 days of pinheading from spawning and 23.5g per $1,200m{\ell}$ bottle. In the size of bottles, the lowest infection ratio was 5 % at $1,200m{\ell}$ bottle. In the growth characteristics with the bottle size, Reishi at $1,600m{\ell}$ bottle showed best result when it is observed for 13~15 days in pinheading from spawing, 36~37 days in fruit bodies growing and 26.7~29.3g yield per bottle. The highest yield per area was 6.4kg at horizontal bed with 218 bottles per $3.3m^2$. The Reishi produced in $1,600m{\ell}$ bottle with horizontal cultivation had less number of fruit bodies and best yield, thus it is determined to be the most economic method.

  • Artificial cultivation of Lyopyllun decastes is costly because the high expense is required for soil covering and bark. Therefore, most farmers of Lyopyllun decastes have recognized it is difficult to cultivate it. The purpose of this study is to reduce the cost of cultivation of Lyopyllun decastes by applying new cultivation method and introducing mutation of fungi. In this study, a new method of practical artificial cultivation was developed through many experiments using fermented pine sawdust and wheat bran. In conclusion, the method of practical artificial cultivation of Lyopyllun decaste is simple and cost efficient because neither soil covering nor bark is required.

Production of Lovastatin in Solid Culture (고체 배양법에 의한 Lovastatin생산)

  • Cultivation conditions for overproduction of lovastatins were investigated from the lovastatin producing strain N-03 which was obtained with NTG (N-methyl-N'-nitro-nitrosoguanidine) treatment from Aspergiliu ferrous ATCC 20542. Produced lactone and acid form of lovastatin were detected, and analyzed by HPLC method. In liquid culture, medium No. 2 containing soy protein produced higher amounts of the lovastatins than medium No. 1 (contained rapeseed oil). In solid culture, maximum production was obtained at 28$^{\circ}C$ for 15 days cultivation using cooked wheat bran. For the overproduction of lovastatin from this strain, solid culture method using plastic bag is more superior than liquid culture.

Selection of Some Species for Artificial Cultivation from the Wild Mushrooms (야생 버섯의 인공재배 가능성 검사)

  • Three edible species and one species utilized as drug were selected from wild mushrooms collected from mountains and fields throughout Korea during 1977 to 1979 for artificial cultivation. The media, MES for G. lucidum, MHS for P. rudis and MSES and MHS for P. adipose and Pluteus sp. were selected. The range of optimum pH was 4.0 to 5.0 in citric acid-sodium phosphate dibasic for the tested species. The range of optimum temperature for the mycelial growth of G. lucidum and P. adiposa was 25 to 3$0^{\circ}C$ and 30 to 35$^{\circ}C$ for P. rudis and Pluteus sp. For the moisture content 75 to 80% (w/w) in the substrates, the mycelial growth of most tested species was rapid. As the spawning substrates, poplar sawdust for G. lucidum and Pluteus sp., oak sawdust for P. rudis and wheat for P. adipose were selected, respectively. In this experiment, the fruit bodies of all tested species were appeared. Among them, G. lucidum for medicinal purpose and P. adiposa and Pluteus sp. for sources of new edible mushrooms seems to be possible. However, the sporophores of P. rudis were not adequate as an edible mushroom for the commercial due to the leathery flesh.

Assessment of the Effect of Substrates from Different Wood Species on the Cultivation of Edible Mushroom

  • Mushroom is very important to rural dwellers due to the various economic, health and nutritional benefits that can be derived from its consumption, sales and utilization. Hence, this experiment was carried out with a view to evaluating the influence of locally available substrate i.e., sawdust of different wood species on the cultivation of Pleurotus ostreatus (oyster mushroom). Pleurotus ostreatus was grown on sawdust (indigenous and exotic species) using two pretreatment methods, which included hot water treatment to remove (a water-soluble extract) and non-extracted sawdusts (substrate). The result of this study showed that there was a significant difference in the effect of different wood species selected. Parkia biglobosa produced a better yield (71.06 g), followed by Anogeissus leiocarpus (53.17 g) and Gmelina arborea (51.39 g) in a non-treated unextracted states. In the treated samples, Parkia biglobosa has the highest yield (50.61 g) followed by Anogeissus leiocarpus (49.28 g) and Gmelina arborea (45.00 g). Thus, Parkia biglobosa and Anogeissus leiocarpus produced a higher yield of mushroom either in a treated or non-treated state and they could be recommended for growing oyster mushroom (Pleurotus ostreatus).
