녹두(Vigna radiata L.) Trypsin Inhibitor의 정제 및 약물학적 특성 (Characterization and Pharmacological Effect of Mung Bean Trypsin Inhibitor)

  • 우리나라에서 식용으로 뿐만아니라 한방재료로 널리 사용되고 있는 녹두(vigna radiata L. wilczek) 로부터 trypsin inhibitor (Mung bean trypsin inhibitor, MBTI)를 분리정제하여 그 특성을 조사하였다 또한 병태동물모델 즉, septic shock induced guinea pig model을 이용하여 MBTI의 약물학적 효과를 평가하였다. MBTI의 분리 및 정제과정은 Sephadex C-50 chromatography, DEAE-celluloseion exchange chromatography 및 trypsin affinity column 을 차례로 이용하였다. 정제한 MBTI는 전기영동 및 아미노산 서열분석결과 분자량 약 8,000 Da 의 BBI-type (Bowman-birk inhibitor type)임을 알 수 있었으며 이들의 생화학적 특성을 구명하였다. 또한 pseudomonal elastase로 유도된 septic shock guinea pig model에서 MBTI 10 mg/kg를 전처치한 결과 hypotention shock 유발이 억제됨을 알 수 있었다.

축방향 충격흡수 향상을 위한 소형구형 투척 로봇구조 설계 (Design of a Miniature Sphere Type Throwing Robot with an Axial Direction Shock Absorption Mechanism)

  • In this paper, we propose a novel surveillance throwing robot which is compact, light-weight and has an efficient shock absorption mechanism. The throwing robot is designed in a spherical shape to be easily grabbed by a hand for throwing. Also, a motor-wheel linking mechanism is designed to be robustly protected from shocks upon landing. The proposed robot has a weight of 2.2kg and the diameter of its wheels is 150 mm. Through the field experiments, the designed robot is validated to withstand higher than 13Ns of impulse.

PC/PET 합금의 열충격 특성에 관한 연구 (An Experimental Study on the Thermal Shock Behavior of PC/PET Alloy)

  • Tests were performed to evaluate the effect of thermal shock behavior on the mechanical properties of PC(poly-carbonate) and PET(polyethylene-terephthalate) with MBS(methylmethacrylate-butadiene-styrene) alloy. Five different material weight fraction for PC/PET were employed : 0/100, 25/75, 50/50, 75/25, and 100/0. Three different weight fraction of MBS were added to each PC/PET : 0, 3, and 9. Therefore fifteen different types of PC/PET/MBS were prepared using single screw extrude. and injection molding machine. One thermal shock cycle consisted of each one hour stay at -$40^{\circ}C$ chamber and $+80^{\circ}C$ chamber without delay. Specimens were thermal shocked up to 20 and 40 cycles. Specific mechanical properities considered in this study include tensile, izod impact, and high rate Impact behaviors. In addition, the morphology of the fractured surface after Izod impact testing was investigated by the SEM (scanning electron microscope).

Numerical Analysis of Hypersonic Shock-Shock Interaction using AUSMPW+ Scheme and Gas Reaction Models

  • The flowfield of hypersonic shock-shock interaction has been simulated using a two-dimensional Navier-Stokes code based on AUSMPW+ scheme. AUSMPW+ scheme is a new hybrid flux splitting scheme, which is improved by introducing pressure-based weight functions to eliminate the typical drawbacks of AUSM-type schemes, such as non-monotone pressure solutions. To study the real gas effects, three different gas models are taken into account in the present paper: perfect gas, equilibrium flow and non equilibrium flow. It has been investigated how each gas model influences on the peak surface loading, such as wall pressure and wall heat transfer, and unsteady structure of flowfield in the region of shock-shock interaction. With the results, the value of peak pressure is not sensitive to the real gas effects nor to the wall catalyticity. However, the value of peak heat transfer rates is affected by the real gas effects and the wall catalyticity. Also, the structure of the flowfield changes drastically in the presence of real gas effects.

Characterization of a Low Molecular Weight Heat-Shock Protein cDNA Clone from Nicotiana tabacum

  • We characterized a cDNA clone for a low molecular weight heat-shock protein (LMW HSP) from tobacco named TLHS-l. Nucleotide sequence determination of TLHS-1 identified an open reading frame for 159 amino acids. To the upstream of the open reading frame, a sequence of 124 nucleotides was determined. To the 3' downstream of the open reading frame, 212 nucleotides were identified which carried poly(A)-tail. Comparison of the open reading frame and hydropathy plot of TLHS-1 with the previously reported class I LMW HSPs showed high identity which classified TLHS-1 as a class I LMW HSP cDNA clone. We proposed that there are six consensus regions in class I LMW HSPs. RNA blot hybridization for TLHS-1 showed a typical expression pattern of heat-shock-inducible gene from three common tobacco cultivars. The open reading frame of TLHS-1 was overexpressed in Escherichia coli. TLHS-1 protein confers thermal protection of other proteins in vitro and in vivo. Thermal induced aggregation of citrate synthase was reduced by purified TLHS-1 protein, and thermal death rate at $50^{\circ}C$ was reduced in E. coli expressing TLHS-l. From these data, we can expect that TLHS-1 acts as a molecular chaperone.perone.

가미천마구등음의 항스트레스 효과에 대한 실험적 연구 (The Study of KCG Extract Under the Anti-Stress Effect of Rats)

  • To determine the anti-stress effect of KCG(加味天麻鉤藤飮) extract on sprague-dawley rats. we conducted a research about the change of weight, activity, reactivity, c-fos protein, cytotoxicity against PC12 cell line and heal shock protein. 1) KCG extract siginificantly inhibited the decrease of body weight induced by stress, compared with the control group. 2) KCG extract had no siginificant effect in the activity and reactivity of rats between the control and the experimental groups. 3) KCG extract siginificantly restrained c-fos protein manifestation, compared with the control group. 4) KCG extract siginificantly restrained heat shock protein, compared with the control group. These results suggested that KCG might be usefully anti-stress effect.

모의 수송 환경에서의 청과물 골판지 상자의 진동 피로에 따른 내구성 (Durability of Corrugated Fiberboard Container for Fruit and Vegetables by Vibration Fatigue at Simulated Transportation Environment)

  • The compression strength of corrugated fiberboard container for packaging the agricultural products rapidly decreases because of various environmental conditions during distribution of unitized products. Among various environmental conditions, the main factors affecting the compression strength of corrugated fiberboard are absorption of moisture, long-term accumulative load, and fatigue caused by shock and vibration. An estimated rate of damage for fruit during distribution is about from 30 to 40 percent owing to the shock and vibration. This study was carried out to characterize the durability of corrugated fiberboard container for packaging the fruit and vegetables under simulated transportation environment. The vibration test system was constructed to simulate the land transportation using truck. After the package with corrugated fiberboard container was vibrated by vibration test system at various experimental conditions, the compression test for the package was performed. The compression strength of corrugated fiberboard container decreased with loading weight and vibrating time. The multiple nonlinear regression equation for predicting the decreasing rate of compression strength of corrugated fiberboard containers were developed using four independent variables such as input acceleration level, input frequency, loading weight and vibrating time. The influence of loading weight on the decreasing rate of corrugated fiberboard container was larger than other variables.

Sn-xAg-0.5Cu 무연 솔더의 파손특성에 관한 실험적 연구 (An Experimental Study on the Failure Characteristics of Sn-xAg-0.5Cu Lead-free Solder)

  • This paper presents an experimental study on the failure characteristics of SnAgCu lead-free solder balls. To estimate the effect of Ag, three types of SnAgCu balls are first prepared by varying the weight percent of Ag(1.0, 3.0, 4.0 wt%) and then analyzed by reliability tests such as thermal shock, high speed ball shear, and drop tests. Thermal shock test reveals that the higher the weight percent of Ag is, the longer the fatigue lift becomes. To the contrary, high speed ball-shear test and drop test show that the shear strength and the fracture toughness of solder balls are inversely proportional to the weight percent of Ag, respectively, Reasons for these observations will be further investigated In the future work.

  • The purpose of this study was to investigate the shock attenuation mechanisms while varying the loads in a backpack during drop landing. Ten subjects (age: $22.8{\pm}3.6$, height: $173.5{\pm}4.3$, weight: $70.4{\pm}5.2$) performed drop landing under five varying loads (0, 5kg. 10kg. 20kg. 30kg). By employing two cameras (Sony VX2100) the following kinematic variables (phase time, joint rotational angle and velocity of ankle, knee and hip) were calculated by applying 2D motion analysis. Additional data, i.e. max vertical ground force (VGRF) and acceleration, was acquired by using two AMTI Force plates and a Noraxon Inline Accelerometer Sensor. Through analysing the power spectrum density (PSD), drop landing patterns were classified into four groups and each group was discovered to have a different shock attenuation mechanism. The first pattern that appeared at landing was that the right leg absorbed most of the shock attenuation. The second pattern to appear was that subject quickly transferred the load from the right leg to the left leg as quickly as possible. Thus, this illustrated that two shock attenuation mechanisms occurred during drop landing under varying load conditions.

요통 유무에 따른 달리기 시 충격과 충격 흡수율 (Impact and Shock Attenuation of the Runners with and without Low Back Pain)

  • Objective: The purpose of the study was to compare the acceleration and shock attenuation (SA) of the runners with/without low back pain (LBG vs. NLBG) while running at 2.5 m/s, 3.0 m/s, 3.5 m/s and 4.0 m/s. Method: 15 adults without low back pain (age: 23.13±3.46 years, body weight: 70.13±8.94 kg, height: 176.79±3.68 cm, NLBG) and 7 adults with low back pain (age: 27.14±5.81 years, body weight: 73.10±10.74 kg, height: 176.41±3.13 cm, LBG) participated in this study. LBG was recruited through the VAS pain rating scale. All participants ran on an instrumented treadmill (Bertec, USA). Results: The LBG shows statistically greater vertical acceleration at the distal tibia during running at 3.5 m/s and 4.0 m/s and greater shock attenuation from the distal tibia to the head during running at 3.5 m/s compared with the NLBG during running (p<.05). As the speed increased, there was a statistically significant increase in vertical/resultant acceleration and shock attenuation for both groups. Conclusion: The findings indicated that the runners with low back pain (LBG) experience greater impact and shock attenuation compared with non-low back pain group (NLBG) during fast running. However, it is still inconclusive whether high impact on the lower extremity during running is the main cause of low back pain in the population. Thus, it is suggested that the study on low back pain should observe the characteristics of impact during running with individuals' low back pain experience and clinical symptoms.
