The present study was conducted to determine whether the two-step time-restricted feeding improves the fattening traits of one-step time-restricted feeding in geese. Thirty-six 8-wk-old geese were allotted into one of three groups. Group R1 (the 1-step restricted feeding group) was allowed access to feed for 2 h in the morning from 8 wk to 14 wk of age. Group R2 (the 2-step restricted feeding group) was treated as Group R1, but was additionally fed for 2 h in the afternoon from 12 wk to 14 wk of age. Group C (the control group) was fed ad libitum from 8 wk to 14 wk of age. Feed intake and body weight (BW) were recorded daily and weekly, respectively. At 14 wk of age, the blood samples were collected to determine the fasting plasma levels of glucose, triacylglycerols and uric acid before sacrifice. The results showed that daily feed intake (DFI) was lower, feed efficiency (FE) was higher in both Groups R1 and R2 than in Group C, and daily gain (DG) in Group R2 was higher than in Group R1 during the whole experimental period (p<0.05). Group R1 exhibited lower abdominal and visceral fat weights in carcass than did Group C (p<0.05), and Group R2 was in intermediate. The fasting plasma glucose levels in Group C were higher, and triacylglycerol levels in Group R1 were higher, compared with the other groups (p<0.05). It is concluded that time-restricted feeding in the fattening period not only increases FE but reduces DFI, and the additional meal during the late fattening period improves the DG without the expense of FE in geese.
To investigate heavy metals (Hg, Pb and Cd) and their potential health risks in commercial dried laver (Porphyra sp.), we collected 45 samples from the major production areas on the western and southern coasts of Korea (Hwaseong, Seocheon, Gunsan, Muan, Shinan, Jindo, Haenam, Wando, Jangheung, Goheung and Busan). The Hg, Pb and Cd concentrations were measured using inductively coupled plasma spectrometry (ICP-MS) or a mercury analyzer. The average Hg, Pb and Cd concentrations in the dried laver were $0.006{\pm}0.0017$, $0.196{\pm}0.0614$ and $0.894{\pm}0.4882$ mg/kg, respectively. Based on the 2007 Korean Public Nutrition Report, these levels are 0.02, 0.11 and 2.47% of the provisional tolerable weekly intake (PTWI) for Hg, Pb and Cd, respectively, established by the FAO/WHO. The hazard quotient (HQ) determined from the ratio of exposure and safe levels were less than 1.0. Therefore, the levels of overall exposure to Hg, Pb and Cd for dried laver were below the recommended JECFA (Joint FAO/WHO Expert Committee on Food Additives) levels, which indicate safe levels for public health.
This study was designed to examine the response of feeding early weaning diets without and with amino acids supplemented cottonseed meal on growth performance and digestibility of early weaned cow calves. Fifteen 14-21 days old cross bred cow calves were randomly allotted to three experimental diets. Diet A comprised of milk replacer and concentrate feed whereas two isocaloric and iso-nitrogenous early weaning diets viz., B and C were prepared without and with lysine and methionine supplemented cottonseed meal, respectively. These early weaning diets were used as substitute of milk replacer for calves. Calves were placed in individual cages and fed twice daily for a period of 60 days. Daily feed intake, feed refused and weekly weight gain was recorded. Two digestibility trials I and II were performed at 5th and 9th week of the experiment, respectively. During the digestibility trial I, calves were fed on ad libitum basis whereas in trial II, calves were fed at 90% of their voluntary DMI. Feed, orts and feces samples were collected, weighed daily, composited, subsampled and analysed for DM and CP. Results indicated that weight gain of calves was (p<0.05) higher on diets A (0.63 kg/d/calf) and C (0.64 kg/d/calf) compared to calves on diet B (0.57 kg/d/calf). Significantly (p<0.05) less daily DMI was observed on diet A (1.48 kg) compared to early weaning diets B (1.70 kg) and C (1.72 kg). The feed efficiency was (p<0.05) better on diet A (2.33) compared to diets B (2.95) and C (2.65). The economic efficiency was noticed to be better on diets B and C compared to diet A. In trial I, digestibility of DM and CP of diet A was (p<0.05) higher than diets B and C. Whereas in trial II, digestibility of DM and CP of diet A was (p<0.05) less than diets B and C. It was concluded that early weaning diet based on lysine and methionine supplemented cottonseed meal produced better weight gain and feed efficiency compared to non-supplemented cottonseed meal based diet.
The trial was carried out on twenty-one Friesian cows at the end of eight months gestation, nine multiparous and twelve primiparous; allocated into three groups (1 control, 2 and 3 experimental). The same diet was administred to all three groups before partum (12.8 kg DM/head/day) and after partum (18.8 kg DM/head/day). The cows in groups 2 and 3 received two different daily quantities of amino-acid chelated chromium (0.6 and 1.2 mg Cr/kg DM) from 4 weeks prior to presumed parturition to 6 weeks after. The milk yield control was carried out at 15, 30, 42 and 60 days. All animals were immunised two weeks prior to the presumed parturition and two weeks after with the following antigens: ovalbumin and brucellergene. Blood samples were collected weekly to monitor humoral and cell-mediated immune responses. When analysing the results of antibody immunity (ovalbumin) in the sixth blood collection both treated groups significantly increased compared to group 1 (0.5230 and 0.4536 vs. 0.1812 OD; p<0.05). The results of the cell-mediated immune response (brucellergene) had significant differences (p<0.10) in correspondence to the third (between group 2 and control) and the fifth (between groups 3 and 2) blood collection. Significant differences in fat corrected milk were observed at 42 days between group 3 and the other two groups (31.01 vs. 26.99 and 28.66 kg/d, p<0.05) and at 60 days between group 3 and control (30.88 vs. 26.69 kg/d, p<0.05). Before partum and at partum a positive immune response was obtained with a lower dose of chromium. After partum a positive immune response, anti-OVA indicator, was obtained with the higher dose of chromium while, $\gamma$-IFN indicator, with the lower dose. A significant increase of the milk yield resulted at both 42 and 60 days with the highest level of chromium.
An experiment was conducted to establish a large scale production method of anti-serum against chicken IgM and to profile the developmental changes of serum IgM levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, three days of age, and weekly thereafter till to 7 weeks of age. The pure IgM was isolated from ammonium sulfate treated chicken serum by both sephadex G-200 and sepharose CL-6B chromatography. The breaking-through peak containing IgM appeared from the fraction 26 to 28. These fractions consisted mainly of IgM when tested by anti-chicken IgM(Nordic, Netherlands). Immunized with the heavy chain of this purified IgM, the rabbit immune sera(anti-chicken IgM) were formed a reaction only with the purified chicken IgM. The quantitative assay of serum IgM were carried by RID method. The optimal time for diffusion was 14 hours and the coefficient of determination($R^{2}$) for regression equation of standard curve was 0.992. It was observed that IgM concentrations were the highest at hatching(3.23 mg /mL), after that decreased gradually. From 2 to 5 weeks of age the levels unchanged(2.0 ~ 2.3mg /mL), and gradually decreased to 7 weeks of age(1.3 mg /mL).
The aim of this study was to identify the method on extended canine semen exposed to freezing as assessed by motility, survival and acrosomal changes following different freezing ramp rates. Five ejaculates collected by digital manipulation twice weekly from three dogs (Shih-Tzu) were added Tris-Egg Yolk (TE) buffer and divided into 4 aliquots according to formulation of our laboratory. After cooling to 4$^{\circ}C$ by ramp rate of 0.6$^{\circ}C$/min, the samples frozen by ramp rates of 1.6$^{\circ}C$/min to -$25^{\circ}C$, 3$^{\circ}C$/min to -35$^{\circ}C$, 8.9$^{\circ}C$/min to -7$0^{\circ}C$ and 19$^{\circ}C$/min to -11$0^{\circ}C$, respectively, and then stored in L$N_2$for 2days. Each sample was evaluated on their motility, survivability and acrosome integrity at different thawing temperature. The ramp rate of 3$^{\circ}C$/min to -35$^{\circ}C$/h for freezing and thawing temperature of 37$^{\circ}C$ obtained the highest results to improve survivability, motile spermatozoa and intact acrosome appearance than other onditions. In conclusion, we may suggest freezing semen for canine artificial insemination is more efficient with freezing at a ramp rate of -3$^{\circ}C$/min to -35$^{\circ}C$ and thawing with a water bath adjusted to 37$^{\circ}C$.
An experiment was conducted to evaluate the effect of form and particle size of feed supplemented with L-threonine on growth performance, carcass characteristic and blood biochemical parameters of broiler chickens. The experimental design was a $2{\times}2{\times}2$ factorial arrangement of treatments evaluating two feed forms (pellet or mash), two feed particle sizes (fine or course), and two inclusion rates of dietary L-threonine (with or without) which adopted from 7 to 42 days of age. In this experiment, 360 a day old chicks in two sexes were assigned in each treatment and each experimental unit was included 15 chicks. Feed consumption and weight gain were measured weekly. At 35 days of age, blood samples were taken to analysis blood biochemical parameters. At the end of the experimental period, two birds were slaughtered in each treatment and carcass analysis was carried out. The results showed that the effect of feed form on body weight gain and feed intake in whole of experimental period was significant (P < 0.05). Broilers fed pelleted diets had more weight gain than the mash group. Growth performance parameters were not affected by feed particle size and dietary L-threonine supplementation in whole of experimental period (P > 0.05). The results of carcass analysis showed that liver and gizzard relative weights were influenced by feed form (P < 0.05). However, pancreas and liver relative weights were affected by feed particle size and dietary L-threonine supplementation, respectively (P < 0.05). Triglyceride and VLDL levels were affected by feed form and dietary L-threonine supplementation (P < 0.05). The effect of feed particle size on blood biochemical parameters was not significant (P > 0.05). In conclusion, the experimental results indicated that feed form increased feed consumption and weight gain in whole of experimental period (1 to 42 days of age) while feed particle size and dietary L-threonine had no effect on broiler performance.
In order to understand the taxonomic and ecologic characteristics on ciliates plankton, surface water samples were collected once a weekly from March 2001 to December 2001 except once a monthly from December 2001 to January 2001 and twice a monttdy at February 2001 at Pal'tang reservoir in Korea. Totally 12 species belonging to 10 genera in 7 families of 6 order, were identified and all species new record in Korea. All of them had been recorded in eutrophic water bodies. Succession of dominant species occurred obviously according to seasonality. Large -sized Codonella cratera, Rimostrombidium lacustris, Strobilidium caudatum, Limnostrombidium viride, Pelagostrombidium fallax, Stylonychia sp., Pseudostrombidium plaktonticum and Phascolodon vorticella dominated during the period of outbreak of nanophytoplankton in winter to spring. However, small-sized Rimostrombidium hyalinum, rimostrombidium brachykinetum, Halteria gradinella and Vorticella convallaria occurred when the bacteria abundance increased in summer to autumn.
The tributaries of Daechung Lake play an important role in controlling eutrophication in the lake, which is used for agricultural purposes and as potable water. However, water quality properties were not extensively studied in the tributaries of Daechung Lake. The objectives of this study are to investigate spatial and temporal properties of water quality and to characterize streams which could threaten water quality of Daechung Lake. For this study, water samples were weekly or monthly collected from February 2014 to October 2014 in 9 streams. Water quality parameters analyzed in this study include biochemical oxygen demand (BOD), chemical oxygen demand (COD), total organic carbon (TOC), total nitrogen and phosphorus (TN and TP), suspended solids (SS), and chlorophyll a. Based on temporal distribution and principal component analysis, BOD, COD, TOC, SS, and TP were controlled by not only river discharge that increased during summer due to heavy rain fall, but also due to anthropogenic input (e.g., bridge construction and/or agricultural activity). Dilution is also one of the factors explaining TN and conductivity, both of which decreased with increased discharge. Generally, concentrations of contaminants (BOD, COD, TOC, TN and TP) in the tributaries were higher than those of Daechung Lake. However, pollution load indicated that only the main channel of Geum River and Sook Stream may largely influence lake waters, attributed mostly to their large volumes. This implies that the main channel and Sook Stream are the major influences on the water quality of Daechung Lake.
This study estimated the heavy metal concentrations in octopus (Octopus minor) and conducted a food safety assessment of octopus. Octopus, a benthic cephalopod, was collected from the Seosan intertidal zone on the west coast of Korea. The samples were digested with acids, and then the cadmium (Cd), copper (Cu), and zinc (Zn) contents were analyzed using inductively coupled plasma mass spectrometry (ICP-MS). The Cd, Cu, and Zn range of concentrations in octopus were 0.06-19 (mean 5.8), 44-1,463 (mean 354.8), and 76-929 (mean 247.9) mg/kg on a dry weight basis, respectively. The concentrations of heavy metals were higher in the internal organs than in the mantle. Of the three heavy metals, copper had the highest concentrations in the internal organs because of the existence of hemocyanin bound with copper in octopus blood, whereas zinc had the highest concentrations in the mantle. No relationship between the concentration of heavy metals and biological parameters (length, weight, and sex) was found. The ratios (I/M) of the heavy metal concentrations in internal organs and mantle were highest for cadmium, although cadmium had much lower concentrations in the internal organs compared with copper and zinc. Considering the provisional tolerable weekly intake (PTWI) of the three heavy metals and the average intake of octopus per day, all three elements should have no adverse effects on humans.
