Soil Physical Properties and Organic Matter (토양(土壤)의 물리성(物理性)과 유기물(有機物))

  • The effects of organic material application on soil physical properties were reviewed in relation to soil productivity. The organic matter contents and soil physical properties of the cultivated land in Korea were summarized and the effects of organic matter were compared in terms of land uses and soil types. Soil physical properties related to crop yield potential, such as soil aggregation, permeability, water holding capacity, erodibility, and compactibility, were used in evaluating the effects of organic materials as a soil physical amendment. The benefical effects of organic matter addition on soil physical conditions include (1) better aeration and increased infiltration in silty and clayey soils, (2) increased water holding capacity and moisture availability in sandy soils, (3) decreased soil erodibility, and (4) increased resistance to compaction. It is, therefore, concluded that continuous application of organic materials could greatly improve the various soil physical properties and favor the growth and yield of crops. A high rate of organic matter addition could contribute to reducing not only the soil erosion on sloping land, but also the possible detrimental effect of farm mechanization. In general, the effects of organic matter on soil physical improvement were estimated to be much higher in upland than in paddy. Organic matter would have a more pronounced effect on low productive lands such as heavy clayey or sandy soils and newly reclaimed soil. The optimum level of soil organic matter content was estimated to be about 3.0 to 3.5% for the best soil physical condition. Since the organic matter contents of the cultivated lands in Korea are much lower than optimum level, it would be desiable to use more organic materials to soil for the increase of soil productivity, continuation of stabilized high productivity and soil erosion control.

Development of a Simultaneous Analytical Method for Determination of Trinexapac-ethyl and Trinexapac in Agricultural Products Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 식물생장조절제 Trinexapac-ethyl과 대사산물 Trinexapac의 동시분석법 개발)

  • BACKGROUND: Trinexapac-ethyl is a plant growth regulator (PGR) that inhibits the biosynthesis of plant growth hormone (gibberellin). It is used for the prevention of lodging, increasing yields of cereals, and reducing mowing of turf. The experiment was conducted to establish a determination method for trinexapac-ethyl and its metabolites trinexapac in agricultural products using LC-MS/MS.METHODS AND RESULTS: Trinexapac-ethyl and trinexapac were extracted from agricultural products with methanol/ distilled water and the extract was partitioned with dichloromethane and then detected by LC-MS/MS. Limit of detection(LOD) was 0.003 mg/kg and limit of quantification(LOQ) was 0.01 mg/kg, respectively. Matrix matched calibration curves were linear over the calibration ranges (0.01-1.0 mg/L) for all the analytes into blank extract withr2> 0.997. For validation purposes, recovery studies were carried out at three different concentration levels (LOQ, 10LOQ, 50LOQ,n=5). Recoveries of trinexapacethyl and trinexapac were within the range of 73.6-106.9%, 72.7-99.2%, respectively. The relative standard deviations (RSDs) were less than 9.0%. All values were consistent with the criteria ranges requested in the CODEX guideline(CAC/GL 40, 2003).CONCLUSION: The proposed analytical method was accurate, effective and sensitive for trinexapac-ethyl and trinexapac determination and it can be used to as an official method in Korea.

Sensory quality, antioxidant, and inhibitory activities of XO and AO of Smilax china leaf tea fermented by Aspergillus oryzae (Aspergillus oryzae 발효 청미래덩굴잎 분말차의 관능적 품질 및 항산화능과 xanthine oxidase 및 aldehyde oxidase 저해활성)

  • This study was conducted in order to investigate the optimal fermentation periods of the Smilax china L. leaves as a fermented tea via Aspergillus oryzae for 0 (non-fermented), and 10, 20, and 30 days (NF, F10, F20, F30). It was also observed for its quality characteristics. In the color and spectrum (400~700nm) of 1% tea water extract, NF was light yellow, whereas fermented tea (F10~F30) was light red color, and the F10 among F10~F30 has the clearest color and spectrum. Furthermore, acceptabilities of aroma and brightness were insignificantly different between NF and F10~30, while the mouth feel and overall acceptabilities were insignificantly distinct among all of the fermented teas. Therefore, these results suggest that the appropriate fermentation period for tea fermentation is 10 days. On the other hand, the total polyphenol and flavonoid content in the NF was the highest among all of the fermented teas. In the antioxidant parameters, EDA (electron donating ability), FRAP (ferric reducing antioxidant power), and LPOIA (lipid peroxidation inhibitory activity) in the NF were the highest among all fermented teas. Meanwhile, the XOI (xanthine oxidase inhibitory activity) was low, as well as insignificantly different from NF and F10~F30, whereas the AOI (aldehyde oxidase inhibitory activity) was markedly higher (38.09~41.70%) by the hot water tea extract (with or without fermentation), particularly the AOI that has increased via fermentation. In conclusion, the overall antioxidant activity tended to be reduced by fermentation; however, the EDA, FRAP and LPOIA in the fermented tea for 10 days was higher than the activities during 20~30 days of fermentation. There was a similar result in the color and acceptability of fermented tea for 10 days, which was remarkably better than those of 20-30 days. Therefore, fermented tea from the leaves of Smilax china L. could be expected to be used as a functional tea without the loss of inhibitory activity of both the XO and AO via fermentation.

Anti-thrombotic Activities of Hot-water Extracts Prepared from Various Parts of Nelumbo nucifera Gaertner (다양한 부위의 연(Lotus) 열수 추출물의 항혈전 활성)

  • To investigate the anti-thrombotic activities of the lotus (Nelumbo nucifera Gaertner), various hot-water extracts were prepared from the leaf, pod of seed (PS), seed, embryo of seed (ES), root, and the node of root (NR) of the lotus. The highest extraction ratio was found in the NR (20.3%), followed by the seed, root, leaf, ES, and PS. These extracts had pH and acidity levels ranging from 5.6~6.5 and 0.06~0.20%, respectively. The seed extract showed 70% brix, whereas the leaf and PS extracts showed less than 0.1% brix. The highest contents of total polyphenol (179.7 mg/g), total flavonoids (161.4 mg/g), and reducing sugar (161.4 mg/g) were observed in the leaf extract, and the highest total sugar content (873.0 mg/g) in the seed extract. Anti-coagulation assay of the extracts of NR, leaf, and PS showed strong activities. In particular, at a concentration of 5 mg/ml, the PS extract had 15-fold extended thrombin, prothrombin, and activated partial thromboplastin times. However, only the ES extract showed activities inhibitory to platelet aggregation, with treatment with 0.25 mg/ml of ES extract decreasing platelet aggregation to 25.1%, a reduction comparable to that effected by aspirin. The extracts other than the seed extract showed no hemolysis activities against human RBC at treatments of up to 1 mg/ml. These results suggest that the NR, PS, seed, and ES, all byproducts of the lotus agriculture industry, have high potential as novel sources of anti-thrombotic agent.

Measurement of Two-Dimensional Velocity Distribution of Spatio-Temporal Image Velocimeter using Cross-Correlation Analysis (상호상관법을 이용한 시공간 영상유속계의 2차원 유속분포 측정)

  • Surface image velocimetry was introduced as an efficient and sage alternative to conventional river flow measurement methods during floods. The conventional surface image velocimetry uses a pair of images to estimate velocity fields using cross-correlation analysis. This method is appropriate to analyzing images taken with a short time interval. It, however, has some drawbacks; it takes a while to analyze images for the verage velocity of long time intervals and is prone to include errors or uncertainties due to flow characteristics and/or image taking conditions. Methods using spatio-temporal images, called STIV, were developed to overcome the drawbacks of conventional surface image velocimetry. The grayscale-gradient tensor method, one of various STIVs, has shown to be effectively reducing the analysis time and is fairly insusceptible to any measurement noise. It, unfortunately, can only be applied to the main flow direction. This means that it can not measure any two-dimensional flow field, e.g. flow in the vicinity of river structures and flow around river bends. The present study aimed to develop a new method of analyzing spatio-temporal images in two-dimension using cross-correlation analysis. Unlike the conventional STIV, the developed method can be used to measure two-dimensional flow substantially. The method also has very high spatial resolution and reduces the analysis time. A verification test using artificial images with lid-driven cavity flow showed that the maximum error of the method is less than 10 % and the average error is less than 5 %. This means that the developed scheme seems to be fairly accurate, even for two-dimensional flow.

Chemical Components and Antioxidative Activity of Korean Gold Kiwifruit (한국산 골드키위의 화학성분 및 항산화 활성)

  • The chemical components and antioxidative activity of Korean gold kiwifruit were investigated. The values of pH, soluble solid and total acidity were 4.43${\pm}$0.16, 17.01${\pm}$0.04$^{\circ}Brix$, and 0.082${\pm}$0.02%, respectively. Hunter L, a, and b values were 49.80${\pm}$0.24, -6.79${\pm}$0.02, and 19.72${\pm}$0.18 value, respectively. Proximate compositions were as follows; moisture 78.62${\pm}$2.26%, crude protein 1.34${\pm}$0.25%, crude lipid 0.70${\pm}$0.06%, crude fiber 1.99${\pm}$0.13%, crude ash 0.99${\pm}$0.26%, and carbohydrate 16.36${\pm}$1.23%, respectively. Mineral elements were K 265.86${\pm}$5.00, P 71.82${\pm}$29.18, and Ca 23.84${\pm}$2.10 mg%, respectively. Free sugar compositions were sucrose (1.04${\pm}$0.18%), glucose (2.17${\pm}$0.21%) and fructose (1.86${\pm}$0.11%). Amino acid contents of Korean gold kiwifruit was very rich in glutamic acid 86.51 ${\pm}$5.58 mg/100 g and deficient in tyrosine 15.00${\pm}$4.91 mg/100 g. Organic acid compositions were quinic acid (6.65${\pm}$0.21 mg/g), malic acid (1.62${\pm}$0.13 mg/g) and citric acid (4.82${\pm}$0.21 mg/g). Contents of ascorbic acid and total phenols were 0.27${\pm}$0.06 mg/g and 0.047${\pm}$0.002 mg/g, respectively. The DPPH radical scavenging activity and reducing power of the water extract from Korean gold kiwifruit was 86.87% and 1.96 at a concentration of 25 mg/mL. The water extract showed considerable antioxidative activity against linoleic acid autoxidation in a dose-dependent manner.

  • Objective : The purpose of this study is to establish a category for acupuncture therapy by appropriate etiological analysis and differenciation of edema. Methods : We arrange Huang Di Nei Jing and thirty four kinds of literature about edema. Results : 1. The cause of Edema is functional disorder of lung, spleen, kidney, bladder and tri-energizer by six kinds of natural factors, internal injury and loss of nutritions. 2. Edema compartmentalize into the water of five zang organs, several kinds of edema(e,g. 5, 10, 12, 24) and yang & yin edema. 3. An ultimate cause of edema pathogenesis is the disturbance of Qi function in kidney. 4. In view of the results so far achived,'GV26(水淸)' is a vitally important acupoint in acupuncture treatment of edema.'GV26(水淸)' and 'CV9(水分)' are very useful acupoints in moxibustion. 5. In the acupuncture and moxibustion treatment of yang edema, we can use acupoints as like 'GV26(水溝)', 'S36(足三里)', 'B20(脾兪)' and 'SP9(陰陵泉)' by purgation and reduction for expelling wind, reducing fever and eliminating dampness. In an instance of yin edema, we can use acupoints as like 'CV9(水分)' 'S36(足三里)', 'CV6(氣海)', 'B20(脾兪)', 'B23(腎兪)' and 'K3(太谿)' by reinforcement for tonifying spleen yang-middle energizer-, qi-transmission and water promoting.

Optimizing Surfactant-Enhanced Solubilzation of LNAPL from Soil in Saturated Zone (포화지층내 저비중 비수용성 유기용매의 용해제거를 위한 계면활성제법의 최적 조작인자 도출)

  • The solubilization of BTEX was evaluated in aqueous surfactant solutions with and without several additives. Anionic surfactant(Sodium Dodecyl Sulfate, SDS) and nonionic surfactants (NEODOL(equation omitted)25-3 and $SOFTANOL\circledR-90$ were used as test surfactants. The effects of surfactant HLB(Hydrophile-Lipophile Balance) Number and hydrocarbon molar volume and polarity of BTEX on the MSR(Molar Solubilization Ratio), micelle-water partition coefficient of BTEX, and CMC(C,itical Micelle Concentration) were investigated. Optimizing treatment conditions applicable to enhanced solubilization was also studied by manupulating salinity or electrolyte control with additives of ethyl alcohol, hydrotrope, and electrolyte solution. The most effective surfactant for solubilization was found $SOFTANOL\circledR-90$, since HLB number of 13.6 is similar to those values of BTEX ranging between 11.4 and 12.2, which was also proved experimentally. Ethyl alchohol of 3% was the most effective additives in reducing CMC and improving solubilization among the conditions using SDS, NEODOL(equation omitted)25-3, and $SOFTANOL\circledR-90$ with three additives. The partitioning of BTEX between surfactant micelles and aqueous solutions was characterized by a mole fraction micelle-phase/aqueous phase partion coefficient, $K_m$. Values of log $K_m$. for BTEX compounds in surfactant solutions of this study range from 2.95 to 3.76(100mM SDS) and 2.95 to 3.49(117mM $SOFTANOL\circledR-90$. Log $K_m$ appears to be a linear function of log $K_{ow}$ for SDS and $SOFTANOL\circledR-90$. A knowledge of partitioning of BTEX in aqueous surfactant system can be a prerequisite for the understanding of the behavior of hydrophobic organic compounds in soil-water systems in which surfactants play a role in remediation of contaminated soil and facilitated transport.

Antibacterial, Antioxidant, and Antiaging Effects of the Ethanol Extract of Dolnamul (Sedum sarmentosum) and the Production of the Oil in Water Cream (돌나물(Sedum sarmentosum) 에탄올 추출물의 항균, 항산화, 항노화 효과와 수중유적형 크림의 제조)

  • This study was performed to investigate the functional properties and characteristics of Dolnamul (Sedum sarmentosum) as a cosmetic ingredient. Lyophilized sedum powder was extracted with ethanol and stored at $-20^{\circ}C$ for the following experiments. Total polyphenol compounds of the ethanol extract of sedum (SE) was $27.98{\pm}0.34g/kg$(dry weight): epicatechin ($162.14{\pm}5.07mg/kg$), epigallocatechin ($55.99{\pm}2.49mg/kg$), and kaempferol ($47.96{\pm}3.02mg/kg$) were contained in the SE. The SE had organic radical scavenging capacity ($78.43{\pm}1.08%$) and metal reducing power (FRAP value $2.54{\pm}0.12$). FTC and TBARS assays confirmed that the SE inhibited the early stage of lipid peroxidation ($62.03{\pm}0.38%$) as well as the final stage of lipid peroxidation ($55.36{\pm}2.05%$), respectively. The SE (5 mg/mL, dry weight) was proved to have antibacterial effect on the growth of Propionibacterium acnes. The inhibitory percentages of the SE on elastase and collagenase activities were $38.94{\pm}7.09%$ and $78.94{\pm}2.49%$, respectively. Compare to the control group, the SE treated group induced an increase of Col3A1 expression and collagen production ($58.11{\pm}1.07%$). The oil in water emulsion (0.5% SE adding group) showed pH 6.88 and 1.47 g/mL of density. The hardness changes of the SE adding emulsions were not detected during the stored periods at various temperatures ($-20-45^{\circ}C$) for four weeks. It is considered that the SE has antibacterial, antioxidant, and antiaging activities.

Effects of Heat and pH Treatments on Antioxidant Properties of Ishige okamurai Extract (패 추출물의 항산화능에 미치는 열과 pH의 영향)

  • This study was carried out to determine the optimum extraction conditions for Ishige okamurai by comparing the yields, total phenolic compound content (TPC), and antioxidant properties of its 95%, 70%, 50% fermented ethyl alcohol and water extracts. Additionally, the effects of heat and pH treatments on the antioxidant properties of the extracts were evaluated by their TPC and 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging capabilities. The yields of the extracts were greatest in the order of water > 50% > 70% > 95% fermented ethyl alcohol, and the TPC of the 70% (26.18%) and 50% fermented ethyl alcohol (27.56%) extracts were higher than those of the others. However, in terms of DPPH radical scavenging and ferrous-reducing power, the 70% fermented ethyl alcohol extract of Ishige okamurai showed the highest antioxidant effects. Additionally, in the results for the heat and pH treatments, the antioxidant properties of the 70% fermented ethyl alcohol extract were not influenced by the treatment conditions except at pH 10.
